Abstract The skin of anuran species is a protective barrier against predators and pathogens, showing also chemical defense by substances that represent a potential source for bioactive substances. This review describes the current chemical and biological knowledge from the skin secretions of Leptodactylidae species, one of the most diverse neotropical frog families. These skin secretions reveal a variety of substances such as amines (12), neuropeptides (16), and antimicrobial peptides (72). The amines include histamine and its methylated derivatives, tryptamine derivatives and quaternary amines. The peptides of Leptodactylidae species show molecular weight up to 3364 Da and ocellatins are the most reported. The peptides exhibit commonly glycine (G) or glycine-valine (GV) as C-terminal amino acids, and the most common N-terminal amino acids are glutamic acid (E), lysine (K), and valine (V). The substances from Leptodactylidae species have been evaluated against pathogenic microorganisms, particularly Escherichia coli and Staphylococcus aureus, and the most active peptides showed MIC of 1-15 µM. Furthermore, some compounds showed also pharmacological properties such as immunomodulation, treatment of degenerative diseases, anticancer, and antioxidant. Currently, only 9% of the species in this family have been properly studied, highlighting a large number of unstudied species such as an entire subfamily (Paratelmatobiinae). Abstract Bauhinia forficata Link aqueous extract is usually recommended as a phytomedicine to reduce blood glucose levels and its biological activity has been linked to the presence of phenolic compounds from B. forficata preparations. Several drying processes are used in the production of dry herbal extracts, which may influence the chemical composition and efficacy of final herbal medicines. Due to significant chemical changes, defining appropriate drying processes is essential for phytopharmaceutical drug development. In view of this, we analyzed dried B. forficata leaf infusion (BFLI) extracts by HPLC-UV-MSn, followed by molecular networking analysis to evaluate the chemical profiles from dried extracts yielded by freeze-and spray-drying processes. The main metabolites detected included 11 ferulic/isoferulic acid derivatives and 13 glycosylated flavonoids. The qualitative chemical profiles were alike for both drying processes, whereas the relative abundance of some flavonoids was higher using spray-drying. Taken together, our results showed that freeze-and spray-drying preserved the phenolic profile of BFLI and suggested that spray-drying may be the most suitable to obtain its dried products. Along with studying the chemical profiles of dried herbal extracts, evaluating the influence of drying processes on the quality and chemical profiles of final products is pivotal and may benefit future research. ABSTRACTThe genus Erythrina, Fabaceae, is widely distributed in tropical and subtropical regions. Their flowers, fruits, seeds and bark are frequently used in folk medicine for its effects on the central nervous system such as anticonvulsant, antidepressant, analgesic, sedative, and hypnotic effects. Erythraline has been reported as one of the active compounds from Erythrina, but until now there are no pharmacokinetics data about this compound and only few results showing a putative metabolism were reported. To improve the information about erythraline metabolism, this article reports and discusses, for the first time, the in vitrometabolism biotransformation of erythraline by cytochrome P450 enzymes.

<p>The phytochemical investigation and evaluation of the phytotoxic effect of the extract and fractions obtained from the leaves of <italic>Annona nutans</italic>(R. E. Fr.) R. E. Fr. were performed. Phytotoxic activity was assessed on radicle and hypocotyl of <italic>Allium cepa</italic> and <italic>Lactuca sativa</italic>, where chloroform and ethyl acetate fractions proved active. Phytochemical investigation of the chloroform fraction was allowed identification of polyketides derivatives: triacontanal, 16-hentriacontane, octacosanol and triacontanol, using the ¹H NMR technique associated with data from GC/MS. Using ethyl acetate fraction, with low activity, flavonoids 3-<italic>O</italic>-β-D-galactopyranosyl-isorhamnetin, 3-<italic>O</italic>-β-D-galactopyranosyl-quercetin and 3-<italic>O</italic>-β-D-apiofuranosyl-(1→2)-galactopyranosyl-quercetin were identified, determined by spectrometric techniques one and two-dimensional NMR, combined with mass spectral data. All substances are being reported for the first time in <italic>Annona nutans</italic>. The phytotoxic activity of chloroform fraction may be related to the presence of triacontanol and similar substances. Triacontanol stimulates growth at very low concentrations, but can have an inhibitory effect at higher concentrations, such as those reported for auxin analogs. The toxicity assay using <italic>Artemia salina</italic> (BST) was also performed, with the chloroform fraction showing a negligible lethal dose, LD₅₀ = 500 mg mL^-1, while the other fractions and extracts showed no activity. Thus, the presence of acetogenins was ruled out.</p>

The best time of production of the cyclohexadepsipeptide beauvericin by the endophytic fungus Fusarium oxysporum SS46 in Czapek medium was evaluated. The highest level of beauvericin production was found on day 21 of fermentative culture, as assessed by quantitative analysis by high performance liquid chromatography coupled with a photodiode array detector. Precursor-directed biosynthesis experiments were carried out to produce new analogues of beauvericin by feeding F. oxysporum with ten analogues of L-phenylalanine. In order to evaluate which precursor analogues were incorporated by the microorganism, the obtained extracts were analyzed using matrix-assisted laser desorption ionization - time-of-flight mass spectrometry (MALDI-TOF/TOF). The precursor-directed biosynthesis studies led to the biosynthesis of novel beauvericin derivatives by replacement of one, two, or all three L-phenylalanine residues in beauvericin with DL-3-fluorophenylalanine, L-3-fluorophenylalanine, L-4-fluorophenylalanine, or L-tyrosine. Beyond these precursor analogues, one unit of L-4-aminophenylalanine, L-4-chlorophenylalanine, DL-4-bromophenylalanine, or L-4-bromophenylalanine was also incorporated by the endophyte F. oxysporum SS46. Units of L-4-nitrophenylalanine and L-histidine were not incorporated by the microorganism to produce unnatural beauvericins.

Erythrina verna is a medicinal plant used to calm agitation popularly known as mulungu. We purchased the barks of E. verna from a commercial producer and analyzed the alkaloid fraction of the bark by CG-MS and HRESI-MS. Five erythrinian alkaloids were identified: erysotrine, erythratidine, erythratidinone, epimer, and 11-hydroxieritratidinone. Here we report the compound 11-hydroxieritratidinone for the first time as a natural product.

A different methodology was used to isolate and purify oxoaporphine alkaloids, as they are difficult to separate by the usual workup when in mixture. Alkaloid extracts from Annonaceae species were obtained by base/acid extraction. The extracts were concentrated and submitted to partition in solutions of acids of different pKa values, followed by separation by preparative TLC using 1 mm thick silica gel impregnated with oxalic acid (11.2% w/w). Liriodenine, lisycamine, lanuginosine, and O-methylmoschatoline were obtained and tested against tumoral cells (line Hep2, ATCC-CCL 23, larynx carcinoma). Only O-methylmoschatoline (IC50 12.4 µM) was more active than cisplatin (18.0 µM).
