Vinte e quatro potros Puro Sangue de Corrida (PSC), com dois anos de idade foram avaliados ultra-sonograficamente, durante o período final da doma e início de treinamento, através da imagem transversal dos tendões flexor digital superficial (TFDS) e profundo (TFDP). As avaliações foram realizadas com intervalos de 15 dias. A área transversal (AT), a textura dos ecos e a ecogenicidade dos tendões foram avaliadas nas sete zonas da região metacarpiana através de um programa de mensuração de imagens do próprio aparelho de ultra-sonografia. Durante a doma e treinamento, houve diminuição da AT na zona IA e IIIA (P<0,05, f=0,001 e 0,0266 resp.) e aumento da zona IIIC (P< 0,05 f=0,0391) do TFDS. Ao considerar apenas o período do treinamento, o resultado foi semelhante (P<0,05) na zona IA (f=0,010), IIIA (f=0,023) e IIIC (f =0,0391). O TFDP apresentou uma diminuição na AT durante a doma e treinamento nas zonas IA, IIIA e IIIB (P<0,05, f=0,027; f=0,0001 e f=0,0031 resp.) e nas zonas IA e IIIA durante o treinamento (P<0,05, f=0,006 e f=0,006, resp.). No período da doma, a AT do TFDP não variou significativamente. A textura dos ecos se manteve homogênea e não houve variação significativa na ecogenicidade dos tendões entre o período da doma e após quatro meses de treinamento. A avaliação quinzenal da AT indicou uma adaptação diferente entre as zonas ultrasonográficas dos TFDS e TFDP. A textura e ecogenicidade das zonas IA a IIIC dos TFDS e TFDP não apresentaram variações ultra-sonográficas que sugerissem adaptação ao regime de exercício ao qual os potros PSC estavam sendo submetidos. A ultrasonografia não é o método mais indicado para acompanhamento da adaptação dos TFDS e TFDP em potros PSC em treinamento.

The cross-sectional area (CSA) of the superficial digital flexor tendon (SDFT) and deep digital flexor tendon (DDFT) of twenty four thoroughbreds was ultrasonographically evaluated before and during their initial training to determine the effect of exercise on the tendon CSA, texture and echogenicity to characterize the response to training. Ultrasonographic transverse images of the left forelimb were obtained every fifteen days. The SDFT CSA of zones IA and IIIA showed a decrease (P<0.05, f=0.010 e f=0.023 res.) during the breaking and training period and an increase (P< 0.05) of zone IIIC (f=0.039). Evaluating just the training period, SDFT CSA results of zones IA, IIIA and IIIC were similar. The DDFT showed no variation during the breaking phase but when the training phase was evaluated there was a decrease (P<0.05) in zones IA (f=0.006) and IIIA (f=0.006). Evaluating both breaking and training periods the DDFT showed a decrease (P<0.05) in zones IA (f=0.027), IIIA (f=0.0001) and IIIB (f=0.0031). Fiber texture and tendon echogenicity showed no significant difference between breaking and last reading during training. This study provides evidence of variation of adaptation among the ultrasonographic zones of both SDFT and DDFT to exercise during training of thoroughbred horses. Ultra-sonography is not the most indicated method to evaluate the adaptation of the SDFT and DDFT to training in thoroughbred horses.

Oócitos bovinos têm sido mantidos em fluido folicular como meio para transporte e para aumento de sua competência, antes da maturação. Oitocentos e oitenta e um (881) oócitos foram aspirados de folículos de 2 a 8mm, de ovários de abatedouro, para avaliar o efeito da manutenção de oócitos bovinos em fluido folicular bovino de folículos de diferentes tamanhos sobre o desenvolvimento embrionário. Os oócitos foram distribuídos aleatoriamente em quatro tratamentos, com sete repetições cada. No grupo controle (n=217), os oócitos foram maturados por 24h em TCM-199 com Soro de Égua em Estro (SEE), piruvato e rFSH-h, em estufa a 39°C, com 5,00% de CO2 e umidade saturada. No tratamento FFpequeno (n=216), os oócitos foram mantidos por 6h em fluido folicular de folículos de 3 a 5mm a 30°C e posteriormente maturados por 18h nas mesmas condições do grupo controle. Os oócitos dos tratamentos FFmédio (n=226) e FFgrande (n=222) foram mantidos em fluido folicular de folículos com 5,1 a 8mm e folículos maiores de 8,1mm, respectivamente e, após, maturados por 18h. Após a fecundação por 18h, os zigotos foram cultivados por 8 dias em SOFaaci com 5,00% de SEE, em estufa a 39°C, em bolsas gaseificadas com 5,00%CO2, 5,00%O2 e 90,00%N2. Oócitos do grupo FFpequeno resultaram em menor (P<0,05) taxa de blastocistos em D7 que os grupos Controle e FFgrande. No D9, as taxas de blastocistos e de eclosão não diferiram (P>0,05) entre os grupos. O fluido folicular de folículos médios e grandes pode ser utilizado para o manutenção de oócitos bovinos por 6h a 30°C, antes da maturação por 18h.

Bovine oocytes have been maintained in the follicular fluid to be transported and to increase their competence, before maturation. Eight hundred eighty-one (881) oocytes, aspirated from bovine slaughterhouse ovaries, were used to evaluate the effect of holding bovine oocytes in follicular fluid (FF) of bovine follicles of different diameters on the rate of embryo development. The oocytes were randomly distributed in four treatments with seven replicates each: The control group (n=217) was constituted by oocytes matured for 24h in modified TCM-199 with Estrus Mare Serum (EMS), pyruvate and rFSH-h in incubator with 5,00% CO2, 39°C and saturated humidity. In the FFsmall group (3 to 5mm follicles; n=216), the oocytes were held for 6h in follicular fluid at 30°C and matured for 18h in the same conditions of the Control-group. The oocytes of the FFmedium group (5,1-8mm follicles; n=226) and of the FFlarge group (>8,1mm follicles; n=222) were held in follicular fluid and matured like FFsmall. Fertilization was accomplished during 18h and, after this, the zygotes were cultured for 8 days in SOFaaci medium + 5,00% EMS in incubator at 39°C using plastic bags gasified with 5,00%CO2, 5,00%O2 and 90,00%N2. FFsmall oocytes produced a lower (P<0,05) blastocyst rate than Control and FFlarge groups, at D7. At D9, blastocyst and hatching rates were similar (P>0,05) between the groups. Follicular fluid of medium and large follicles could be used to hold for 6h at 30°C bovine oocytes before their maturation for 18h.

Complexos Cumulus-Oócito (CCO) bovinos foram divididos em 4 grupos para avaliar o seu comportamento durante a manutenção em LFb e maturação in vitro (MIV) em TCM-199 com ou sem Hepes. CCO MIV por 24h em TCM-199 em estufa a 39°C com 5,00% de CO2 (Controle) tiveram o seu desenvolvimento comparado ao de CCO MIV em tubos repletos de TCM-HEPES (5,95 mg/mL), em banho-maria (BM) a 39ºC por 24h (Grupo 1 - BM24h), ao de CCO mantidos em líquido folicular bovino (LFb), por 6h a 30ºC seguido de maturação por 18h nas mesmas condições que o grupo Controle (Grupo 2 - LFb6C18h) e ao de CCO mantidos em LFb seguido da maturação por 18h sob as mesmas condições que o grupo 1 (Grupo 3 - LFb6BM18h). A fecundação foi realizada em FERT-TALP por 18h. Os zigotos foram cultivados em SOFaaci, sob óleo mineral, em bolsas plásticas gaseificadas. A taxa de clivagem no Grupo Controle foi superior a do Grupo 3 (P<0,05), mas não houve diferença no percentual de blastocistos no D7 e D9 e no de blastocistos eclodidos entre os 4 grupos. Portanto, oócitos podem ser mantidos por 6h em LFb, a 30ºC, antes da maturação em TCM-HEPES por 18h ou ser maturados por 24h, em TCM-HEPES, em banho-maria a 39ºC, sem atmosfera gasosa controlada. A simplificação da MIV aqui introduzida através do preenchimento de tubos de 1,0mL com TCM-HEPES e manutenção em banho-maria a 39ºC, poderá ser uma opção viável e prática para os programas de OPU/PIV em bovinos.

In order to evaluate the effect of a transport medium on the rate of in vitro embryonic development, 1381 Cumulus-oocyte Complexes (COC) were obtained by aspiration of 2-8mm diameter follicles witch were randomly divided in 4 treatment groups. The Control group was formed by oocytes matured in modified TCM-199 for 24h, incubated at 39°C and 5,00% CO2 with saturated humidity. The group 1 (WB24h), included oocytes matured in 1.0mL tubes containing TCM-HEPES (5.95mg/mL), in water bath (WB) at 39°C for 24h. The group 2 (FFb6C18h), included oocytes kept in bovine follicular fluid (FFb) for 6h at 30°C followed by a period of 18h maturation under the same conditions as the Control group and with the oocytes maintained in FFb followed by 18h IVM under the same conditions as the group 1, group 3 (FFb6WB18h). Fertilization was performed in FERT-TALP for 18h. Zygotes were cultured in SOFaaci under mineral oil within gasified bags. The cleavage rate differed (P<0.05) between the Control and FFb6BM18h groups. However, there was no difference on the D7 and D9 blastocyst rates and on the percentage of blastocyst ecloded. It was concluded that it is possible to maintain the oocytes in FFb for 6h at 30°C before 18h IVM, or to proote the transport and maturation of the oocytes for 24h, in TCM-HEPES and water-bath at 39°C, without compromising embryonic development. The simplification of MIV showed in this experiment through of tubes (1.0mL) replete with TCM-HEPES and holding in water bath at 39°C, could be a viable and practice for the bovine programs of OPU/PIV.

Foi avaliada a influência da citocalasina B na vitrificação de oócitos e embriões bovinos produzidos in vitro. No Experimento I, 956 oócitos foram maturados por 22h, sendo imediatamente vitrificados (tratamento Vitri) ou expostos por 15 a 20 minutos, à solução com 7,5µg/mL (tratamento CB7,5Vitri) ou 45µg/mL (tratamento CB45Vitri) de citocalasina B, antes da vitrificação. Após 30 segundos de exposição à SV1 [400µl TCM-Hepes com 10% soro fetal (SF), 50µl etilenoglicol (EG) e 50µl DMSO], e 20 segundos à SV2 (300µl trealose 1,0M + 20% SF, 100µl EG e 100µl DMSO), os oócitos foram vitrificados em palhetas estiradas (OPS). O reaquecimento foi realizado a 37-38ºC em duas etapas de 5 minutos cada, em trealose 0,3M e 0,15M. Não houve diferenças (P>0,05) nos percentuais de clivagem e desenvolvimento embrionário entre os tratamentos Vitri, Cito7,5Vitri e Cito45Vitri, os quais foram inferiores (P<0,05) ao controle. No Experimento II, 269 blastocistos expandidos foram distribuídos em 3 tratamentos. No tratamento Vitri, os embriões foram expostos por 1 minuto à SV1 e 20 segundos à SV2, a qual foi modificada pela substituição da trealose por TCM-Hepes. No tratamento CB45Vitri, a vitrificação ocorreu após exposição de 10 minutos à solução com 45µg/mL de citocalasina B. Não foram observadas diferenças (P>0,05) no percentual de re-expansão e eclosão entre os grupos Vitri e CB45Vitri, os quais foram inferiores (P<0,05) ao grupo controle. Os resultados indicam que, independentemente da dose utilizada, a exposição a citocalasina B não produz efeito benéfico na vitrificação de oócitos ou blastocistos bovinos produzidos in vitro.

In this study, the influence of cytochalasin B on the survival and development after vitrification of oocytes and in vitro produced (IVP) bovine blastocysts was evaluated. In the Experiment I, 956 oocytes were matured for 22h and were immediately vitrified (Vitri treatment) or exposed for 15-20 minutes, to 7.5µg/ml (CB7.5Vitri treatment) or 45µg/ml (CB45Vitri treatment) cytochalasin B solutions, before vitrification. After 30 seconds of exposure to SV1 [400µl TCM-Hepes with 10% fetal serum (SF), 50µl ethylene glycol (EG) and 50µl DMSO], and 20 seconds to SV2 (300µl trehalose 1,0M + 20% SF, 100µl EG and 100µl DMSO) solutions, the oocytes were vitrified in Open Pulled Straws (OPS). The rewarming was performed at 37-38ºC in two steps of 5 minutes each, into 0.3 and 0.15M trehalose solutions, respectively. There was no difference (P>0.05) in the cleavage and embryo rates between Vitri, Cito7.5Vitri and Cito45Vitri treatments, which were inferior to control group (P<0.05). In the Experiment II, 269 expanded blastocysts were distributed into three treatments. In the Vitri treatment blastocysts were exposed for 1 minute to SV1 and for 20 seconds to SV2 solution, the later one modified using TCM-Hepes instead of trehalose. In the CB45Vitri treatment, embryos were vitrified after being exposed to a 45µg/ml cytochalasin B solution for 10 minutes. The re-expansion and hatching rates were similar to Vitri and CB45Vitri treatments (P>0.05), which were lower than those observed in the Control group (P<0.05). The results show that, independently of dose studied (7.5 or 45mg/ml), cytochalasin B has not a beneficial effect to the vitrification of oocytes and IVP bovine blastocysts.

RESUMO Os endométrios de 923 éguas foram revisados histologicamente para se verificar a presença de dilatações glandulares. O material se originou de 873 biópsias uterinas e 50 amostras uterinas de matadouro. Esta alteração foi detectada em 164 (18,8%) biópsias e em 23 (46%) casos de matadouro. Destas, 30 amostras de biópsia e 17 de matadouro foram analisadas morfológica e morfometricamente. A dilatação das glândulas endometriais estava associada à fibrose peri-glandular em 68,8% dos casos de biópsia e em 86,1% dos de matadouro. Não foi possível estabelecer a(s) causa(s) da dilatação glandular, mas os dados morfológicos sugerem um distúrbio local na relação entre os hormônios, as células do estrema, as células glandulares e a matriz extracelular.

SUMMARY Endometria of 923 mares were histological reviewed for changes of glandular dilatation. The material originated from 873 uterine biopsies and 50 uterine samples of abattoir. This change was detected in 164 (18,8%) ofthe biopsy cases and in 23 (46%) of the abbatoir cases. From these 30 biopsies and 17 abattoir samples were morphological and morphometrical analysed. Dilatation of endometrial glands were associated with periglandular fibrosis in 68,8% of the biopsy cases and in 86,1% of the abattoir cases. It was not possible to stabilish the cause(s) of glandular dilatation but morphological data suggest a local disturb in the hormonal - stroma cells -extracellular matrix relationship.

RESUMO Descreve-se o acompanhamento clínico de um caso de tumor de células da granulosa na égua através da ultra-sonografia. Foi constatada uma evolução rápida durante 18 dias para o tumor confirmado, pelo exame macroscópico e histopatológico, após remoção cirúrgica.

SUMMARY The follow up of a case of granulosa cell tumour in a mare through ultrasonography is reported. It has been observed a fast growth rate reaching its final size around 18 days. The diagnostic of granulosa cell tumour was confirmed by gross and histopathologic examination, after surgical removal.
