Em estudo prévio mostrou-se que o extrato aquoso das folhas de Averrhoa carambola (ExAq) reduziu o inotropismo atrial da cobaia. Por isso, este trabalho avaliou se o ExAq interfere com o influxo de cálcio através da membrana celular. A investigação foi conduzida em átrio esquerdo de cobaia, montado em cuba (5 mL, Tyrode, 27 ± 0,1 ºC, 95 % O2, 5 % CO2), estirado para uma tensão de repouso de 10 mN e submetido a uma estimulação de 2 Hz (0,5 ms, 400 V). O efeito do ExAq sobre a entrada de cálcio nas células foi avaliado em átrio de cobaia e em células GH3, estas submetidas a 'patch clamp' na configuração 'whole cell'. No átrio, o ExAq (1500 µg/mL) deslocou para direita a curva concentração-efeito do (±) BAY K 8644 (agonista dos canais de cálcio tipo-L), aumentando a CE50 (concentração capaz de produzir 50 % do efeito máximo) de 7,8 ± 0,38 para 115,1 ± 0,44 nM (N = 3, p < 0,05). Em células GH3, este extrato (500 µg/mL) reduziu de 282 para 190 pA (30 %) a corrente de cálcio, sem contudo alterar a voltagem de pico da curva desta corrente. Estes resultados mostram que, pelo menos em parte, o efeito inotrópico negativo do ExAq em átrio de cobaia se deve a uma diminuição do influxo de cálcio pelos canais tipo-L.

It was previously showed that aqueous leaf extract (AqEx) of Averrhoa carambola depresses the guinea pig atrial inotropism. Therefore, experiments were carried out on guinea pig left atrium and on pituitary GH3 cells in order to evaluate the effect of AqEx on the cellular calcium influx. The atrium was mounted in an organ chamber (5 mL, Tyrode, 27 ± 0.1 ºC, 95 % O2, 5 % CO2), stretched to 10 mN, and paced at 2 Hz (0.5 ms, 400 V) and GH3 cells were submitted to a whole cell voltage clamp configuration. In the atrium, the AqEx (1500 µg/mL) shifted to the right the concentration-effect curve of the positive inotropic effect produced by (±) BAY K 8644, an L-type calcium channel agonist. The AqEx increased EC50 (concentration required to promote 50% of the maximum effect) of the inotropic effect of BAY K 8644 from 7.8 ± 0.38 to 115.1 ± 0.44 nM (N = 3; p < 0.05). In GH3 cells assayed with 500 µg/mL of AqEx, the L-type calcium inward current declined 30 % (from 282 to 190 pA). Nevertheless, the extract did not change the voltage correspondent to the peak current. These data suggest that, at least in part, the negative inotropic effect of AqEx on the guinea pig atrium is due to a reduction of the L-type calcium current.

O lapachol, α e β-lapachona são naftoquinonas obtidas de espécies de Tabebuia, apresentam propriedades antiinflamatória, antibacteriana, anticâncer e tripanossomicida. O objetivo deste trabalho foi investigar um possível efeito espasmolítico destas naftoquinonas em íleo de cobaia, uma vez que, outras naftoquinonas inibem a atividade contrátil de músculos lisos. O lapachol, α e β-lapachona inibiram as contrações fásicas induzidas tanto por carbacol (CI50 = 1,5 ± 0,2 x 10-4; 7,3 ± 0,9 x 10-5 e 3,2 ± 0,5 x 10-5 M, respectivamente) quanto por histamina (CI50 = 3,6± 0,5; 3,6 ± 0,7 e 3,3 ± 0,6 x 10-5 M, respectivamente). Estes compostos também relaxaram o íleo pré-contraído com KCl (CE50 = 1,2 ± 0,4; 4,3 ± 0,8 e 2,7 ± 0,2 x 10-5 M, respectivamente); carbacol (CE50 = 2,6 ± 0,7; 3,5 ± 0,5 e 2,2 ± 0,7 x 10-5 M, respectivamente) ou histamina (CE50 = 3,0 ± 0,8; 1,1 ± 0,3 e 3,3 ± 0,6 x 10-5 M, respectivamente) de maneira dependente de concentração. Este efeito é provavelmente devido à inibição do influxo de Ca2+ através dos canais de Ca2+ dependentes de voltagem (CaV). β-lapachona antagonizou (pD'2 = 5,73 ± 0,12; "slope" = 1,51 ± 0,05) as contrações induzidas por CaCl2 em meio despolarizante nominalmente sem Ca2+. O achado de que a β-lapachona inibiu as contrações tônicas induzidas por S-(-)-Bay K8644 (CE50 = 1,4 ± 0,1 x 10-5 M) é sugestivo que o CaV envolvido é o do tipo L. Em conclusão, lapachol, α e β-lapachona apresentam atividade espasmolítica não seletiva em íleo de cobaia, e β-lapachona exerce este efeito pelo bloqueio dos canais CaV tipo L.

Lapachol, α and β-lapachone are naphthoquinones extracted from species of Tabebuia that have shown antiinflammatory, antibacterial, anticancer and trypanosomicidal properties. The aim of this work was to investigate the spasmolytic effect of these naphthoquinones on the guinea-pig ileum, since other naphthoquinones are known to depress the contractile activity of smooth muscles. Lapachol, α and β-lapachone inhibited the phasic contractions induced by both carbachol (IC50 = 1.5 ± 0.2 x 10-4; 7.3 ± 0.9 x 10-5 and 3.2 ± 0.5 x 10-5 M, respectively) and histamine (IC50 = 3.6± 0.5; 3.6 ± 0.7 and 3.3 ± 0.6 x 10-5 M, respectively). These compounds also relaxed the ileum pre-contracted with KCl (EC50 = 1.2 ± 0.4; 4.3 ± 0.8 and 2.7 ± 0.2 x 10-5 M, respectively); carbachol (EC50 = 2.6 ± 0.7; 3.5 ± 0.5 and 2.2 ± 0.7 x 10-5 M, respectively) or histamine (EC50 = 3.0 ± 0.8; 1.1 ± 0.3 and 3.3 ± 0.6 x 10-5 M, respectively) in a concentration-dependent manner. This effect is probably due to inhibition of calcium influx through voltage-gated calcium channels (Ca v). β-lapachone antagonized (pD'2 = 5.73 ± 0.12; slope = 1.51 ± 0.05) CaCl2-induced contractions in depolarizing medium nominally without Ca2+. The finding that β-lapachone inhibited the tonic contractions induced by S-(-)-Bay K8644 (EC50 = 1.4 ± 0.1 x 10-5 M) is suggestive that the L-type CaV is involved. In conclusion, lapachol, α and β-lapachone showed non-selective spasmolytic activity in guinea-pig ileum, and β-lapachone exerts this effect by to blockade of L-type CaV channels.

A espécie Xylopia langsdorffiana St. Hil.. & Tul. é popularmente conhecida como "pimenteira-da-terra" no Sudeste do Brasil. A partir do fracionamento do extrato etanólico, obtido das cascas do caule desta espécie, foi isolado um diterpeno tipo labdano, identificado como sendo o ácido 8(17),12E,14-labdatrieno-18-óico, e que neste trabalho é codificado como labdano302. O labdano302 relaxou o tônus basal dos anéis de traquéia isolada de cobaia com um valor de CE50 de 6,7 ± 0,5 x 10-8 M. O diterpeno labdano302 relaxou de maneira dependente de concentração os anéis pré-contraídos com carbacol (10-6 M), tanto na presença (CE50 = 1,4 ± 0,7 x 10-5 M) como na ausência de epitélio funcional (CE50 = 1,5 ± 0,3 x 10-5 M), bem como anéis pré-contraídos com 18 ou 60 mM de KCl, apresentando valores de CE50 de 2,3 ± 0,4 x 10(7) M e 1,8 ± 0,8 x 10-5 M, respectivamente. Este efeito relaxante, sobre as contrações induzidas por 18 mM de KCl, tanto foi significantemente mais potente quanto mais eficaz quando comparado ao efeito sobre as contrações induzidas por 60 mM de KCl. Assim, labdano302 mostra um efeito relaxante em traquéia isolada de cobaia, tanto em seu tônus basal como sob estímulo contrátil, aparentemente sem a participação dos fatores relaxantes derivados do epitélio, contudo com possível participação dos canais de K+.

Xylopia langsdorfiana St. Hil. & Tul. is popularly known as "pimenteira-da-terra" in Southeast of Brazil. The fractionation of the ethanol extract obtained from the stem-bark of this species yielded a labdane-type diterpene identified as 8(17),12E,14-labdatrien-18-oic acid, referred here as labdane302. In this study, we investigated the effect of labdane302 in guinea-pig trachea. labdane302 relaxed the basal tonus of trachea rings with EC50 value of 6.7 ± 0.5 x 10-8 M. The diterpene labdane302 relaxed the pre-contracted rings by carbachol 10-6 M both in the presence (EC50 = 1.4 ± 0.7 x 10-5 M) and absence of functional epithelium (EC50 = 1.5 ± 0.3 x 10-5 M), as well as pre-contracted by KCl 18 mM or 60 mM, presented EC50 values of 2.3 ± 0.4 x 10-7 M and 1.8 ± 0.8 x 10-5 M, respectively. This relaxant effect, upon contractions induced by KCl 18 mM, was more potent as well as more efficient than the one presented with KCl 60 mM pre-contracted rings. The labdane-type diterpene labdane302 shows the relaxant effect in isolated guinea-pig trachea, pre-contracted or upon basal tonus, apparently without participation of epithelium-derived relaxant factors, but apparently involving activation of K+ channels.

Pomacea sp. e sua desova têm uso popular para tratar diarréia e doenças respiratórias. Este trabalho objetivou avaliar a toxicidade aguda e os efeitos citotóxico e espasmolítico dos liófilos de Pomacea lineata e de sua desova. Os liófilos não apresentaram toxicidade aguda (até 2 g/kg v.o.) ou efeito citotóxico (até 1 mg/mL). P. lineata relaxou a traquéia pré-contraída com CCh na presença (Emax = 48,8 ± 6,4%) e na ausência (Emax = 47,3 ± 9,1%) de epitélio, já a desova relaxou apenas na presença (Emax = 36,3 ± 2,5%) de epitélio. Os liófilos foram ineficazes sobre o tônus espontâneo da traquéia. P. lineata foi mais potente em inibir as contrações do íleo induzidas por ACh (logCI50 = 2,5 ± 0,04 µg/mL) que por hist. (logCI50 = 2,7 ± 0,04 µg/mL). A desova inibiu igualmente as contrações induzidas por ACh (logCI50 = 2,5 ± 0,02 µg/mL) e hist. (logCI50 = 2,5 ± 0,06 µg/mL). P. lineata foi mais potente em relaxar o íleo pré-contraído com ACh (logCE50 = 1,7 ± 0,12 µg/mL) do que com KCl (logCE50 = 2,4 ± 0,06 µg/mL) ou hist. (logCE50 = 2,2 ± 0,18 µg/mL). A desova relaxou equipotentemente o íleo pré-contraído com KCl (logCE50 = 2,3 ± 0,15 µg/mL), ACh (logCE50 = 1,9 ± 0,14 µg/mL) ou hist. (logCE50 = 2,2 ± 0,16 µg/mL), sugerindo um bloqueio dos CaV. P. lineata e sua desova apresentam efeito espasmolítico justificando a sua utilização no tratamento de diarréia e de doenças respiratórias.

Pomacea sp. and its eggs are used against diarrhea and respiratory diseases in folk medicine. The aim of this study was to investigate acute toxicity, cytotoxic and spasmolytic activity of lyophilized Pomacea lineata and its eggs. P. lineata and its eggs present no acute toxicity (until 2 g/kg p.o.) or cytotoxic effect (until 1 mg/mL). P. lineata and its eggs have no effect on guinea-pig trachea spontaneous tonus. P. lineata relaxed trachea pre-contracted with CCh in the presence (Emax = 48.8 ± 6,4 %) and absence (Emax = 47.3 ± 9,1 %) of epithelium, the eggs relaxed only in the presence (Emax = 36.3 ± 2.5 %) P. lineata was more potent to inhibit contractions induced by ACh (logIC50 = 2.5 ± 0.04 µg/mL) than histamine (logIC50 = 2.7 ± 0.04 µg/mL). The eggs inhibited contractions induced by ACh (logIC50 = 2.5 ± 0.02 µg/mL) and histamine (logIC50 = 2.5 ± 0.06 µg/mL) in a non-selective manner. P. lineata was more potent in relax ileum pre-contracted with ACh (logEC50 = 1.7 ± 0.12 µg/mL) than KCl (logEC50 = 2.4 ± 0.06 µg/mL) or histamine (logEC50 = 2.2 ± 0.18 µg/mL). The eggs were equipotent in relax ileum pre-contracted with of KCl (logEC50 = 2.3 ± 0.15 µg/mL), ACh (logEC50 = 1.9 ± 0.14 µg/mL) or histamine (logEC50 = 2.2 ± 0.16 µg/mL), that is suggestive of the blockade of the voltage-operated calcium channels. Collectively, the results validate folk use of P. lineata and its eggs to treat diarrhea and respiratory diseases.

O medicamento fitoterápico brasileiro - BPF é composto de corantes das plantas Eucalyptus globulus Labill, Peltodon radicans Pohl e Schinus terebinthifolius Raddi em alcool a 13,3° GL. Este medicamento é popularmente usado no estado da Paraíba, Brasil desde 1889 como anti-séptico e antiinflamatório. O objetivo deste estudo foi avaliar a propriedade antiinflamatória deste medicamento fitoterápico. Para tal, foram utilizadas as técnicas de edema de orelha em camundongos induzido por 12-O-tetradecanoilforbol 13-acetato (TPA) ou capsaicina e o edema de pata de rato induzido por carragenina. O BPF na dose de 26 mL/kg inibiu tanto edema de orelha induzido por TPA como por capsaicina a 49% (p < 0.05) e 24% (p < 0.01) respectivamente. Estudos preliminares utilizando a técnica de edema de pata induzido por carragenina demonstraram que a administração oral também inibiu o edema de pata em aproximadamente 29%. Os resultados demonstraram que o medicamento fitoterápico brasileiro (BPF) apresentou propriedades antiinflamatórias e a melhor dose foi aquela que é usada pela população.

The Brazilian polyherbal formulation (BPF) is composed by dyes of Eucalyptus globulus Labill, Peltodon radicans Pohl and Schinus terebinthifolius Raddi in alcohol at 13.3° GL. The formulation is popularly used in Paraíba state, Brazil since 1889 and it is used as an antiseptic and anti-inflammatory medicine. The aim of this study was to evaluate the anti-inflammatory property of the polyherbal formulation. For this purpose it was used the12-O-tetradecanoylphorbol 13-acetate (TPA) and capsaicin-induced mouse ear edema and the carrageenan-induced rat paw edema. The BPF at dose of 26 mL/Kg inhibited both 12-O-tetradecanoylphorbol 13-acetate (TPA) and capsaicin-induced ear edema by 49% (p < 0.05) and 24% (p < 0.01) respectively. Preliminary results on carrageenan-induced rat paw edema demonstrated that oral administration also inhibited the paw edema by approximately 29%. The results demonstrate that the Brazilian polyherbal formulation has anti-inflammatory activity and the better dose was the one used by the population.

O presente trabalho visou estudar o efeito eletrocardiográfico produzido pela fração aquosa (AqF) obtida do extrato acético das folhas de Psidium guajava L. em coração isolado de cobaia. Os traçados eletrocardiográficos foram obtidos em corações batendo espontaneamente ou então sob estimulação elétrica. Os corações foram montados em uma sistema de perfusão do tipo Langendoff de fluxo constante. A AqF, usada em concentrações menores que 20 mg/mL, não alterou a freqüência espontânea do coração (controle: 180 ± 9 bpm, teste: 182 ± 10 bpm; N = 3; p > 0,05). Todavia, concentrações iguais ou maiores que 20 mg/mL produziram bloqueio atrioventricular completo (BAV). Este efeito, contudo, desapareceu prontamente quando se removeu a AqF do fluido de perfusão coronariana (N = 3 corações). O BAV promovido pela AqF se faz mediado pelos receptores muscarínicos porque o sulfato de atropina (1,5 mM) impediu o aparecimento deste efeito.

This paper aimed to study the electrocardiographic effect produced by the aqueous fraction (AqF) obtained from the acetic extract of Psidium guajava L. leaf on the isolated guinea pig heart. Electrocardiographic records (ECG) were obtained on isolated hearts beating spontaneously or under regular electrical stimulation. The hearts were mounted in a constant flow Langendorff perfusion system. Until 20 mg/mL, AqF did not change the spontaneous cardiac rate (control: 180 ± 9 bpm, test: 182 ± 10 bpm; N = 3; p > 0.05). Concentrations equal or greater then 20 mg/mL induced complete atrioventricular block (AVB). However, this effect promptly disappeared when AqF was removed from the perfusion fluid (N = 3 hearts). The AVB induced by AqF involves heart muscarinic receptors because atropine sulfate (1.5 mM) could prevent the appearance of such disturbance.
