A utilização de proteínas miofibrilares de carne na elaboração de filmes comestíveis implica, inicialmente, na obtenção desta macromolécula, uma vez que ela é indisponível comercialmente. Assim, os objetivos deste trabalho foram a obtenção e caracterização de proteínas miofibrilares de carne bovina, em escala de laboratório, e a elaboração e caracterização da microestrutura de filmes à base dessas proteínas. As proteínas miofibrilares foram extraídas do músculo Semitendinosus bovino, pós rigor mortis, empregando-se uma técnica de extração por centrifugação diferencial em solução tampão, com diferentes concentrações salinas, segundo metodologia de EISELE & BREKKE [13] modificada neste trabalho. As proteínas, assim obtidas, foram liofilizadas (PML) e analisadas para determinação do teor de proteínas, de lipídios, de sais minerais, de cinzas e da umidade. A composição de aminoácidos foi determinada por cromatografia de troca iônica e o perfil eletroforético foi determinado em gel de poliacrilamida. Alguns filmes foram elaborados com 1g de PML/100g de solução, 60g de glicerol/100g de proteínas e ácido acético glacial para ajuste do pH=2,8. A microestrutura desses filmes foram analisadas por microscopia eletrônica de varredura. A PML, que apresentou teor de proteínas de 84,3%, foi composta de diversas frações de proteínas miofibrilares, com destaque para frações da miosina e da actina, segundo os resultados da eletroforese. Trabalhando-se a composição de aminoácidos, verificou-se que a maior contribuição energética das interações potenciais dos aminoácidos, foram as provenientes de ligações covalentes (58,3%), seguidas das interações iônicas (35,72%). A microestrutura do filme revelou uma estrutura pouco homogênea, porém coesa e densa e uma matriz protéica compacta e fina.

The use of meat myofibrillar proteins for edible film production involves initially the extraction of the macromolecule, since it is not commercially available. Thus, the objectives of this work were the extraction and characterization of bovine meat myofibrillar proteins, in laboratory scale, and the elaboration and characterization of the microstructure of these films. The myofibrillar proteins were extracted form the Semitendinosus bovine muscle, post rigor mortis, using a differential centrifugal technique in buffer solution, with different saline concentrations, modifying the Eisele and Brekke [13] method. The proteins were lyophilized (PML) and analyzed for protein, fat, mineral, ash and moisture content. The amino acid composition was determined by ionic exchange chromatography and the eletrophoretic profile was done using SDS-PAGE. Films having 1g of PML/100g of solution, 60g of glycerol/100g of proteins and pH maintained at 2.8 with acetic acid were used for the microstructure study using a scanning electronic microscope. The PML which presented 84.3% of protein content, was composed of several fractions of myofibrillar proteins, with emphasis for the myosin and actin fractions, according to the SDS-PAGE results. From the amino acid composition values, was verified that the main energetic contribution came from covalent links (58.3%), followed by the ionic interactions (35.7%). The films microstructure showed a poorly homogeneous structure, but with a protein matrix compact and smooth.

As propriedades viscoelásticas de biofilmes podem fornecer informações à nível de estrutura dos biopolímeros usados. O objetivo deste trabalho foi testar três modelos simples, da teoria da viscoelasticidade linear (Maxwell, Maxwell generalizado com duas unidades em paralelo, e Burgers) em resultados de testes de relaxamento de tensão em biofilmes de proteínas miofibrilares de Tilápia do nilo. Os filmes foram elaborados segundo uma técnica tipo casting e condicionados em 58% de umidade relativa e 22°C por 4 dias, antes dos testes. Os corpos de prova (15mm x 118mm) foram submetidos aos testes de relaxamento em um equipamento de medidas físicas TA.XT2i. A deformação imposta na amostra foi de 1%, garantindo-se a permanência no domínio da viscoelasticidade linear. Os modelos foram ajustados aos dados experimentais (tensão x tempo) por regressão não linear. Os modelos de Maxwell generalizado com duas unidades em paralelo e o de Burgers representaram satisfatoriamente, as curvas de relaxamento de tensão. As propriedades viscoelásticas foram pouco dependentes da espessura dos filmes.

The viscoelastic properties of edible films can provide information at the structural level of the biopolymers used. The objective of this work was to test three simple models of linear viscoelastic theory (Maxwell, Generalized Maxwell with two units in parallel, and Burgers) using the results of stress relaxation tests in edible films of myofibrillar proteins of Nile Tilapia. The films were elaborated according to a casting technique and pre-conditioned at 58% relative humidity and 22ºC for 4 days. The testing sample (15mm x 118mm) was submitted to tests of stress relaxation in an equipment of physical measurements, TA.XT2i. The deformation, imposed to the sample, was 1%, guaranteeing the permanency in the domain of the linear viscoelasticity. The models were fitted to experimental data (stress x time) by nonlinear regression. The Generalized Maxwell model with two units in parallel and the Burgers model represented the relaxation curves of stress satisfactorily. The viscoelastic properties varied in a way that they were less dependent on the thickness of the films.

Biopolímeros como polissacarídeos e proteínas, têm despertado interesse recentemente, como matérias primas para embalagens comestíveis e/ou biodegradáveis. Os materiais a serem desenvolvidos devem possuir boas propriedades mecânicas e aparência, para proteger e apresentar o produto embalado. Este trabalho teve como objetivo a caracterização das propriedades mecânicas e óticas de biofilmes à base de proteínas miofibrilares de tilápia-do-nilo, em função do pH (2,0-3,0), da concentração de proteína (Cp= 0,5-2,0g/100g solução) e do teor de glicerol (Cg= 30-70g/100g proteína) da solução filmogênica (SF). Foram determinadas a força e a deformação na ruptura por teste de perfuração e a opacidade aparente através de espectrofotometria, à 22ºC e umidade relativa ambiente. Os biofilmes obtidos nessas condições de ensaio apresentaram-se manuseáveis e transparentes. A força e a deformação na ruptura apresentaram comportamentos opostos em função da Cg (pH= 2,5 e Cp= 1,25 g/100g solução): a variação da Cg de 30 a 70% provocou uma variação na força e na deformação na ruptura entre 6,67N e 2,94N e 2,71% e 7,5%, respectivamente. A Cg e o pH exercem influência significativa sobre a força na ruptura (p< 0,05). Porém nenhum dos fatores estudados demonstrou ter influência significativa sobre a deformação na ruptura ou sobre a opacidade aparente dos biofilmes.

During the last couple of years, biopolymers such as polysaccharides and proteins have been arousing scientists’ interest as raw materials for biodegradable and/or edible packaging. Packaging materials to be developed should have good mechanical and visual properties, to protect and to offer the packed product a presentable look. This work was aimed at characterization of the mechanical and optical properties of nile tilapia myofibrillar proteins-based biofilms, as a function of filmogenic solution (fs) [composition: ph (2 to 3), protein (cp = 0.5 to 2.0g/100g solution) and glycerol concentration (cg = 30 to 70g/100g protein)]. Force and percentage deformation at break were determined by a puncture test and the apparent opacity by spectrophotometry, at 22ºc and ambient relative humidity. Easy handling and transparent biofilms were obtained in these conditions. Force and percentage deformation at break presented opposite behaviors as a function of cg (ph = 2.5 and cp = 1.25g/100g solution): increasing it from 30 to 70% and causing a variation in force and percentage deformation at break between 6.67n and 2.94n and 2.71% and 7.5%, respectively. Glycerol concentration and ph had significant effect over force at break (p <0.05). None of the studied factors showed any significant influence on the percentage deformation of biofilms at break, neither on their apparent opacity.
