A mosca branca Bemisia tabaci (Hemiptera: Aleyrodidae) é considerada uma das pragas agrícolas mais ameaçadoras em todo o mundo, pelo seu efeito direto na sucção de seiva das plantas e principalmente pelo seu efeito indireto de transmitir vírus, em especial os begomovírus. No Brasil, a espécie de begomovírus predominante em solanáceas é o Tomato severe rugose vírus - ToSRV. Uma possibilidade de manejo da doença é a utilização de cultivares resistentes ou tolerantes tanto ao vírus, como ao inseto vetor. Foram avaliados neste trabalho a dispersão primária e secundária do ToSRV por Bemisia tabaci, espécie MEAM1, também conhecida como biótipo B, em cultivares tolerantes ao ToSV Capsicum baccatum (IAC-1357) C. annuum (IAC-1566) e C. frutescens (IAC-1544). O genótipo (IAC-1544) apresentou menor número de plantas infectadas pelo ToSRV no ensaio de dispersão primária e nenhuma eficiência na transmissão do isolado na dispersão secundária. Estes resultados colaboram para o melhoramento de pimentão e pimentas visando a resistência ao ToSRV e seu inseto vetor.

The whitefly Bemisia tabaci (Hemiptera : Aleyrodidae) is considered one of the most threatening agricultural pests in the world due to its direct effect on the suction of sap from plants and especially due to its indirect effect in transmitting viruses, particularly the begomovirus. In Brazil, the predominant species of begomovirus within Solanaceae is Tomato severe rugose virus - ToSRV. A possibility of management of this disease is the use of cultivars that are resistant or tolerant to both the virus and the insect vector. This study evaluated the primary and secondary dispersal of ToSRV by B. tabaci, MEAM1 species, also known as biotype B, usin ToSRV-tolerant species Capsicum baccatum (IAC-1357), C. annuum (IAC -1566) and C. frutescens (IAC-1544). The genotype IAC -1544 showed a smaller number of plants infected by ToSRV during the primary dispersal assay and no efficiency in the isolate transmission in the secondary dispersal. These results contribute to capsicum and pepper breeding forresistance to ToSRV and its insect vector.

Bemisia tabaci (Genn.) é considerada uma das mais importantes pragas em cultivos de hortaliças e ornamentais em todo o mundo. Baseado na análise da seqüência mitocondrial (citocromo oxidase I - mtCOI) foi proposto recentemente que B. tabaci deva ser considerado um complexo críptico de espécies, contendo 11 grupos e 24 espécies. Dois destes grupos: Middle East-Asia Minor e Mediterranean englobam os biótipos B e Q, respectivamente. Avaliou-se a sequência mtCOI de espécimes de B. tabaci coletados em regiões do estado de São Paulo, Brasil. Por PCR-RFLP utilizando-se a enzima Taq I, pôde-se observar somente o padrão típico de clivagem para o biótipo B. Comparando-se com sequências consenso, todas as moscas brancas foram classificadas no grupo Middle East-Asia Minor e puderam ser separadas em quatro haplótipos, indicando prevalência do biótipo B em áreas de pimentão (Capsicum annuum L.), tomate (Solanum lycopersicum L.), cucurbitáceas e berinjela (Solanum melongena L.) do Estado de São Paulo.

Bemisia tabaci (Genn.) is one of the most important pests in cultivated areas of vegetables and ornamental crops around the world. Based on the mitochondrial cytochrome oxidize I (mtCOI) sequence, there is evidence that B. tabaci should be considered a cryptic species complex of 11 groups containing 24 species. Two of the groups, Middle East-Asia Minor 1 and Mediterranean include biotypes B and Q, respectively. In this study we evaluated the mtCOI sequence of B. tabaci populations collected in sites of the state of São Paulo, Brazil. Using PCR-RFLP with Taq I, a typical biotype B profile was obtained for all specimens. Based on the comparison with mtCOI reference sequences we found four haplotypes all belonging to the Middle East-Asia Minor 1. They occurred in the hosts pepper (Capsicum annuum L.), tomato (Solanum lycopersicum L.), eggplant (Solanum melongena L.) and cucurbitaceae plants.

Um levantamento para avaliar a ocorrência de begomovírus nas culturas de pimentão e tomateiro no estado de São Paulo foi realizado entre janeiro/2007 e julho/2008. O DNA total de amostras de pimentão (710) e de tomateiro (103) foi extraído e a presença de begomovírus foi testada por PCR. Paralelamente, as mesmas amostras foram avaliadas por amplificação por círculo rolante (RCA) seguidas de PCR, e algumas amostras positivas analisadas por RCA-RFLP com a enzima de restrição HpaII, a fim de se conhecer a variabilidade genética dos isolados. Os resultados demonstraram que, para a técnica de PCR, 99 amostras de pimentão (13,94%) e 39 de tomateiro (37,86%) foram positivas para a presença de begomovírus, enquanto que por RCA-PCR, 333 (46,90%) de pimentão e 82 (79,61%) de tomateiro mostrando a maior sensibilidade desta técnica. Seqüências correspondentes à região 5' da capa protéica (CP) e um segmento de gene da região intergênica foram analisadas e indicaram apenas a presença da espécie Tomato severe rugose virus (ToSRV). Porém, seqüenciamento parcial de clones obtidos a partir de produto RCA de tomateiro permitiu a detecção de infecção mista de ToSRV e Tomato yellow vein streak virus (ToYVSV). Por RCA-RFLP quatro padrões de restrição foram observados para o ToSRV em pimentão, enquanto que em tomateiro observaram-se 18 padrões.Os resultados indicam maior diversidade genética dos begomovírus em tomateiro quando comparada com os de pimentão.

From January/2007 to July/2008 a survey was carried out to evaluate the occurrence of begomoviruses in pepper and tomato crops from São Paulo State. Total DNA was extracted from 710 pepper and 103 tomato samples, and the presence of begomoviruses was tested by Polymerase chain reaction (PCR). The same samples were tested by Rolling Circle Amplification (RCA) followed by PCR, and some positive samples analyzed by RCA-RFLP and cleaved by the restriction enzyme HpaII to evaluate the genetic variability of these isolates. By PCR, 99 (13.94%) samples collected from pepper and 39 (37.86%) from tomato were positives for the presence of begomovirus, while by RCA-PCR 333 (46.90%) and 82 (79.61%) from pepper and tomato, respectively, indicating higher sensitivity of this technique. The 5' region of the coat protein (CP) gene and a segment of the intergenic region was analyzed indicating the presence of Tomato severe rugose virus (ToSRV) in pepper and tomato plants. However, the partial sequencing of clones from RCA products from a tomato sample indicated mixed infection of ToSRV with Tomato yellow vein streak virus (ToYVSV). By RCA-RFLP four restriction profiles were observed for ToSRV in pepper, while 18 profiles for begomovirus from tomato plants, indicating higher degree of genetic variability for begomovirus found in tomato plants compared to that in pepper plants.

O presente trabalho teve por objetivo estudar os aspectos biológicos, morfológicos e comportamentais de uma linhagem unipariental de Aspidiotus nerii Bouché, 1833 (Hemiptera: Diaspididae), sob condições controladas (25±1oC, 70±10% de UR e 12 horas de fotofase), tendo abóboras híbridas "cabotiá" como hospedeiro. Ninfas recém-eclodidas foram transferidas da criação massal para abóboras "cabotiá", devidamente higienizadas, dispostas em câmaras climatizadas. O período médio para fixação das ninfas móveis foi de 2,4±0,33 horas. A duração média do 1° e 2° estádios, após a fixação das ninfas, foi de 8,3±0,06 e 19,4±0,11 dias, respectivamente, totalizando fase ninfal média de 27,9±0,11 dias. Das ninfas que atingiram a fase adulta, 36 fêmeas foram acompanhadas diariamente até o final do ciclo. A duração média dos períodos pré-reprodutivo e reprodutivo foi de 17,3±0,18 e 68,2±4,33 dias, respectivamente. O número total médio de ninfas/fêmea foi 175,5±10,29 e o número médio de ninfas/fêmea/dia foi 2,7±0,11. A longevidade média das fêmeas foi 100,5±4,51 dias.

The objective of this work was to study the biology, morphology and behavior of Aspidiotus nerii Bouché, 1833 (Hemiptera: Diaspididae) under controlled conditions (25±1°C, 70±10% R.H. and 12 hours of photophase), using "cabotiá" cucumber pumpkins as hosts. Nymphs of the 1st stage obtained of mass rearing (mother culture) were transfered to "cabotiá" pumpkins, properly cleaned and disposed in acclimatized cameras. The medium period for fixation of the nymphs was of 2.4±0.33 hours. The 1st and 2nd stages, after the fixation of the nymphs, lasted 8.3±0.06 and 19.4±0.11 days, respectively, totalizing a nymphal phase of 27.9±0.11 days. 36 females were observed daily until the end of the cycle. The pre-reproductive and reproductive periods lasted 17.3±0.18 and 68.2±4.33 days, respectively. The medium total number of nymphs/female was 175.5±10.29 and the medium number of nymphs/female/day was 2.7±0.11. The female's longevity was 100.5±4.51 days.
