RESUMO: Adenovírus aviário (FAdV) é um importante patógeno na indústria avícola global e a etiologia da síndrome da hepatite por corpúsculo de inclusão-hidropericárdio (SHH) em galinhas. Desde a década de 1990, vários surtos de SHH foram descritos em todas as áreas de produção de aves, incluindo na América do Sul, e a coinfecção entre FAdV e vírus da anemia das galinhas (CAV) pode ser agravante para todos os aspectos da SHH. Objetiva-se descrever um surto de SHH em matrizes de frangos corte, caracterizar a estirpe de FAdV envolvida e destacar a coinfecção com CAV. Foram avaliados machos reprodutores de corte com três semanas de idade, com medula óssea pálida, fígado aumentado e amarelado e esplenomegalia, timo atrofiado e um com hidropericárdio. Fígado e timo foram coletados para isolamento do vírus em embriões de galinhas SPF, PCR e histopatologia. Todas as aves acometidas e embriões inoculados foram positivos para FAdV e CAV. Os embriões inoculados tiveram óbito de 30% em até sete dias após a inoculação e alterações hepáticas por fígados esverdeados e aumentados. A análise filogenética de FAdV com base em parte da sequência do gene que codifica a proteína hexon revelou identidade com a espécie E, que se tornou disseminada em vários países. A coinfecção de FAdV e CAV resulta em maior intensidade de lesões, maior morbidade e mortalidade e em reprodutores tem alta relevância epidemiológica, em razão da transmissão vertical de ambos e da ampla distribuição geográfica das progênies infectadas. Na última década, ocorreu um aumento mundial na ocorrência de SHH em frangos de corte relacionado a falhas em biosseguridade, especialmente em reprodutores, condição que pode ter ocorrido neste episódio. A presença de FAdV e CAV em reprodutores é motivo para preocupação por reflexos negativos à imunidade e viabilidade das progênies. ABSTRACT: Fowl aviadenovirus (FAdV) is an important pathogen in the global poultry industry and the etiology of inclusion body hepatitis-hydropericardium syndrome (HHS) in chickens. Since the 1990s, several outbreaks of HHS have occurred in poultry producing areas, including South America. The coinfection of FAdV and chicken anemia virus (CAV) may markedly impact the incidence of HHS. This study describes an outbreak of HHS in coinfection with CAV in industrial broiler breeders and characterizes the FAdV isolate. The three-week-old male broiler breeders had pale bone marrow, enlarged and yellowish liver, splenomegaly, and atrophied thymus; one chicken was also found with hydropericardium. Virus isolation was performed in SPF chicken embryos of liver and thymus. Tissues of the naturally infected chickens and the inoculated embryos were evaluated by PCR and histopathology. All affected chickens and inoculated embryos were positive for FAdV and CAV. The inoculated embryos had enlarged, greenish and hemorrhagic livers, and 30% died within 7 days of inoculation. Phylogenetic analysis of the FAdV isolate hexon gene partial sequence enabled grouping with E species. The E species has recently become a relevant species in several countries. The association of FAdV with CAV in breeders is of further concern due to both being capable of vertical transmission. Within the last decade, a worldwide upsurge of HHS in broiler breeders owing to failed biosecurity has occurred. In this episode, the failure on biosecurity may have enabled challenge with both FAdV and CAV, with pathological synergism. The CAV-impaired adaptive immunity may have benefited the FAdV infection. RESUMO: A efetividade das vacinas recombinantes vetorizadas para o controle da laringotraqueíte infecciosa (LTI) nas aves de uma região (Minas Gerais, Brasil) com aproximadamente 10 milhões de poedeiras foi avaliada em condições de campo, no período de 2014 a 2018. Durante este período, somente as vacinas recombinantes “turkey herpesvirus” (rHVT) ou “fowl poxvirus” (rFPV), que expressam antígenos do vírus da laringotraqueíte (Gallid herpesvirus-1; GaHV-1) foram utilizadas. Galinhas poedeiras (n=1.283), de oito diferentes granjas produtoras de ovos, foram individualmente amostradas e examinadas por monitoramento ativo e, na ocorrência de notificação de doença respiratória aos veterinários do serviço oficial, por monitoramento passivo. Exames clínicos, macroscópicos e histopatológicos foram realizados para o diagnóstico de LTI, bem como técnicas moleculares para a detecção e caracterização do DNA de GaHV-1 da traqueia e gânglio trigêmeo. As galinhas poedeiras pertenciam a lotes e granjas que usavam diferentes protocolos de vacinação (não vacinadas, uma dose ou tipo de vacina e duas doses ou tipos de vacina). Este é o primeiro longo estudo a campo sobre a efetividade das vacinas vetorizadas em uma região com população elevada de poedeiras de múltiplas idades. Utilizando vários métodos de diagnóstico, a ocorrência da infecção por GaHV-1 e a LTI clínica em poedeiras de uma região interditada do Brasil foi investigada. O número de galinhas positivas para o vírus GaHV-1 e para casos clínicos de LTI nas granjas foi menor quando todas as aves estavam vacinadas com, pelo menos, um tipo ou dose de vacina. Entretanto, a diferença na taxa de detecção da infecção por GaHV-1 foi significativa somente quando a comparação foi realizada entre granjas com aves vacinadas com duas doses e aves de granjas vacinadas com uma única dose de HVT-LT.

ABSTRACT: The effectiveness of vectored recombinant vaccines to control infectious laryngotracheitis (ILT) in chickens from a region (State of Minas Gerais, Brazil) with ~10 million layers was evaluated under field conditions from 2014-2018. During this period, only recombinant turkey herpesvirus (rHVT) or fowl poxvirus (rFPV) vaccines that express antigens of infectious laryngotracheitis virus (Gallid herpesvirus-1; GaHV-1) were used. Layer chickens (n=1,283), from eight different egg-producing companies, were individually sampled and examined (active surveillance), and in instances when government poultry health veterinarians were notified due to respiratory disease (passive surveillance). Clinical, macroscopic, and histopathology examinations were performed to diagnose ILT as well as molecular techniques for the detection and characterization of the GaHV-1 DNA from the trachea and trigeminal ganglia (TG). The layer hens sampled and examined belonged to flocks and farms that used different vaccination protocols (non-vaccinated, single dose vaccination, and prime/boost vaccination). This is the first long-term field study of the effectiveness of ILT vectored vaccines in a high-density multiple age layer hen region. Using various diagnostic methods, the occurrence of GaHV-1 infection and ILT clinical disease in layer hens vaccinated with vectored recombinant vaccines in one quarantined region of Brazil were investigated. The number of ILTV positive chickens by PCR and ILT clinical disease cases was lower in farms when all chickens were vaccinated with at least one vaccine. However, the difference in the detection rates of GaHV-1 infection was significant only when compared farms with prime/boost and farms using single dose of HTV-LT.

ABSTRACT Computers and Intractability: A Guide to the Theory of NP-Completeness, by Michael R. Garey and David S. Johnson, was published 40 years ago (1979). Despite its age, it is unanimously considered by many in the computational complexity community as its most important book. NP-completeness is perhaps the single most important concept to come out of theoretical computer science. The book was written in the late 1970s, when problems solvable in polynomial time were linked to the concepts of efficiently solvable and tractability, and the complexity class NP was defined to capture the concept of good characterization. Besides his contributions to the theory of NP-completeness, David S. Johnson also made important contributions to approximation algorithms and the experimental analysis of algorithms. This paper summarizes many of Johnson’s contributions to these areas and is an homage to his memory.

Abstract Thirty-six isolates of psittacid herpesvirus (PsHV), obtained from 12 different species of psittacids in Brazil, were genotypically characterized by restriction fragment length polymorphism (RFLP) analysis and PCR amplification. RFLP analysis with the PstI enzyme revealed four distinct restriction patterns (A1, X, W and Y), of which only A1 (corresponding to PsHV-1) had previously been described. To study PCR amplification patterns, six pairs of primers were used. Using this method, six variants were identified, of which, variants 10, 8, and 9 (in this order) were most prevalent, followed by variants 1, 4, and 5. It was not possible to correlate the PCR and RFLP patterns. Twenty-nine of the 36 isolates were shown to contain a 419 bp fragment of the UL16 gene, displaying high similarity to the PsHV-1 sequences available in GenBank. Comparison of the results with the literature data suggests that the 36 Brazilian isolates from this study belong to genotype 1 and serotype 1.

Polymer-modified mortars are highly popular for setting exterior tile systems and low-porosity ceramic tiles because they possess greater flexibility and higher bond strengths than conventional mortars which increases cladding serviceability. However, the long-term performance of this application is not completely understood and many pathologies has been reported. The negative influence of water saturation on polymer modified mortar is well documented and this paper investigates the influence that progressive water intake on bond strength by applying pull out tests on several different mortars. Four of those mortars were especially prepared with well known raw materials and concentrations, and three others were ready-made commercial products designed for setting ceramic tile on building façades. The tests were performed during twenty-five day drying period in laboratory conditions after specimens saturation for ten days of immersion in water. The results revealed that: (i) the ready-made commercial and the prepared mortars have similar behavior; (ii) their bond strengths are highly influenced by small increases in moisture content; (iii) moisture content higher than 10% is enough to reduce mortar bond strength by one-half; (iv)during the drying process bond strength increases continuously until reaching values similar to the initial ones.

Random key genetic algorithms are heuristic methods for solving combinatorial optimization problems. They represent solutions as vectors of randomly generated real numbers, the so-called random keys. A deterministic algorithm, called a decoder, takes as input a vector of random keys and associates with it a feasible solution of the combinatorial optimization problem for which an objective value or fitness can be computed. We compare three types of random-key genetic algorithms: the unbiased algorithm of Bean (1994); the biased algorithm of Gonçalves and Resende (2010); and a greedy version of Bean's algorithm on 12 instances from four types of covering problems: general-cost set covering, Steiner triple covering, general-cost set k -covering, and unit-cost covering by pairs. Experiments are run to construct runtime distributions for 36 heuristic/instance pairs. For all pairs of heuristics, we compute probabilities that one heuristic is faster than the other on all 12 instances. The experiments show that, in 11 of the 12 instances, the greedy version of Bean's algorithm is faster than Bean's original method and that the biased variant is faster than both variants of Bean's algorithm.

É relatado um surto de febre catarral maligna (FCM) em Minas Gerais, que resultou na morte de 5 búfalas e uma vaca de uma mesma propriedade. Quatro búfalas morreram com 10-15 dias após o início dos sinais clínicos e uma búfala foi sacrificada in extremis, após manifestar sinais clínicos semelhantes. O exame histopatológico revelou lesões comumente observadas em búfalos com FCM como epicardite e miocardite histiolinfocítica multifocal e pneumonia linfocítica intersticial. Além disso, vasculite linfocítica, principalmente na camada adventícia, com necrose fibrinóide da camada muscular, foi observada no rim, fígado, baço, linfonodos e cérebro. A seqüência de nucleotídeos amplificada pela técnica de PCR revelou 98% de homologia entre o fragmento de DNA amplificado da amostra do sistema nervoso central (SNC) da búfala com seqüências de OHV-2 previamente depositadas no Genbank. Adicionalmente, a técnica de PCR revelou a presença do DNA viral no sangue total de 2 búfalas, clinicamente sadias. O diagnóstico de FCM foi baseado em dados epidemiológicos, clínicos, patológicos, histopatológicos e semi-nested PCR.

An outbreak of Malignant Catarrhal Fever (MCF) resulted in death of five female buffaloes and one domestic cow from the same farm. Four buffaloes died 10-15 days after the appearance of clinical signs, while the fifth was euthanized in extremis, after similar clinical signs. Histopathological lesions included multifocal histiolymphocytic epicarditis, myocarditis and lymphocytic interstitial pneumonia, which are commonly seen in cases of MCF in buffaloes. Furthermore, lymphocytic vasculitis centered in the adventitia, with occasional fibrinoid necrosis in the muscular layer, was found in the kidneys, liver, spleen, lymph nodes and brain. Nucleotide sequencing of DNA fragments from the central nervous system amplified by PCR revealed 98% similarity with known OHV-2 sequences from Genbank. Additionally, PCR analysis also revealed the presence of OHV-2 DNA in the peripheral mononuclear blood cells of two clinically healthy buffaloes. The diagnosis of MCFwas based on epidemiological, clinical, gross and histopathological findings and on the results of a semi-nested PCR followed by nucleotide sequencing.

Anticorpos monoclonais (AcM) foram produzidos contra o extrato EDTA obtido de Leptospira interrogans, sorovar icterohaemorrhagiae. Pelo teste de precipitação foram caracterizados como IgM e IgG (IgGl e IgG2). A eletroforese em gel de poliacrilamida do extrato EDTA revelou diversas bandas quando corada pela prata. No "Western blot", as bandas em torno de 20 kDa reagiram com o AcM 47B4D6, foram oxidadas pelo periodato e não digeridas pela pronase, sugerindo que o determinante é de natureza carboidrato. O determinante reconhecido pelo AcM 47B4D6 estã localizado sob o envelope externo como revelado pela imunocitoquímica usando marcação com ouro coloidal. O AcM contra extrato EDTA do sorovar icterohaemorrahagiae não protegeu hamsters quando inoculados com lepstopira homóloga virulenta.

Monoclonal antibodies (MABs) ivere produced against an etbylenediaminetetraacetate (EDTA) extract of Leptospira interrogans serovar icterohaemorrhagiae being characterized by gel precipitation as IgM and IgG (IgGl and IgG2b). The EDTA extract was detected as several bands by silver staining in SDS-PAGE. In the Western blot the bands around 20 KDa reacted with a monoclonal antibody, 47B4D6, and was oxidized by periodate and was not digested by pronase, suggesting that the determinant is of carbohydrate nature, lmmunocytochemistry, using colloidal gold labeling, showed that an EDTA extract determinant recognized by monoclonal antibody 47B4D6, is localized under the outer envelope of serovar icterohaemorrhagiae. Hoe AIAB raised against the EDTA extract was not able to protect hamsters from lethal challenge with virulent homologous leptospires.
