ABSTRACT: The present study evaluated the cryoprotectant efficacy of dimethylacetamide (DMA) and ethylene glycol in a one-step protocol to freeze boar sperm. The sperm-rich portion of the ejaculates from two boars were collected once a week, for 10 weeks. After collection, the ejaculates were diluted (1:1; v/v) in the cooling extender. After determining their spermatozoa concentration, the ejaculates were pooled with the same number of spermatozoa from each boar and stabilized at 20°C for 120 min. Distinct cryoprotectants were added to the cooling extender at 20 °C, at different concentrations, composing six treatments: 1.25% and 2.5% glycerol (control); 1.25% and 2.5% ethylene glycol; 2.5% and 5.0% DMA. The samples were stored in 0.25 mL straws, containing 35 × 106 spermatozoa. After 90 min at 20 °C, the straws were submitted to a cooling curve until 5 °C (0.3 to 0.5 °C/min) and kept at 5°C for 60 min. Freezing was conducted by placing the straws horizontally 5 cm above the liquid nitrogen for 10 min, followed by immersion on liquid nitrogen. After thawing at 37 °C for 30 seconds, sperm quality was evaluated through a computer-assisted semen analysis system and flow cytometry. Sperm motility was greater (P< 0.05) in treatments with 5.0% and 2.5% DMA (22.2 ± 2.6% and 20.0 ± 2.8%, respectively) than in treatment with 2.5% ethylene glycol (8.2 ± 1.0%). The integrity of the plasma membrane (P = 0.08) and mitochondrial membrane potential (P = 0.27) was similar among the treatments. The treatment with 2.5% ethylene glycol was the least efficient to maintain intact acrosome membrane (P< 0.01). Some kinetics parameters (DAP, DCL, DSL, VAP, VCL, VSL e ALH) were positively affected by 5.0% DMA. The one-step freezing protocol resulted in unsatisfactory boar sperm motility after thawing, regardless of the cryoprotectant. RESUMO: O presente estudo objetivou avaliar a eficácia de um protocolo one-step de congelamento do sêmen suíno utilizando dimetilacetamida (DMA) e etilenoglicol como crioprotetores. Durante 10 semanas, a fração rica dos ejaculados de dois machos suínos foram coletados, uma vez por semana. Após a coleta, os ejaculados foram diluídos (1:1; v/v) no diluidor de resfriamento. Após a avaliação da concentração espermática, os ejaculados foram agrupados em um pool com o mesmo número de espermatozoides de cada macho e estabilizados a 20 °C por 120 min. Os criopropetores foram adicionados ao diluidor de congelamento a 20 °C, em diferentes concentrações, compondo seis tratamentos: glicerol (controle), 1,25% e 2,5%; etilenoglicol, 1,5% e 2,5%; e DMA, 2,5% e 5,0%. As amostras foram armazendadas em palhetas de 0,25 mL contendo 35 x 106 espermatozoides. Após 90 min a 20 °C as palhetas foram submetidas a uma curva de resfriamento até 5 °C (0,3 a 0,5 °C/mim) e mantidas a 5 °C por 60 min. O congelamento foi realizado a partir da colocação das palhetas horizontalmente a 5 cm acima do nitrogênio líquido por 10 min, com sua posterior imersão no nitrogênio líquido. Após o descongelamento a 37 °C por 30 segundos a qualidade espermática foi avaliada através de um sistema computadorizado e por citometria de fluxo. A motilidade espermática foi maior (P < 0,05) nos tratamentos com 5,0% e 2,5% DMA (22,2 ± 2,6% e 20,0 ± 2,8%, respectivamente) do que no tratamento com 2,5% etilenoglicol (8,2 ± 1,0%). A integridade da membrana plasmática (P = 0,08) e potencial de membrana mitocondrial (P = 0,27) foi similar entre os tratamentos. O tratamento com 2,5% de etilenoglicol foi menos eficiente em manter membrana acrossomal intacta (P< 0,01). ABSTRACT: The association of amoxicillin and clavulanic acid has shown high efficacy to treat mastitis worldwide, also promoting a shorter milk disposal period compared to other antimicrobials. However, no previous investigations regarding its application to treat mastitis in Brazilian dairy cows were developed. This study evaluated the effectiveness of amoxicillin-clavulanic acid to treat Brazilian dairy cows with mastitis, comparing it with a reference protocol treatment combination of tetracycline, neomycin and bacitracin. Holstein cows (n = 468) from three different dairy herds in Brazil were assigned to two groups: combination of tetracycline, neomycin and bacitracin (n = 178; positive control group) and amoxicillin-clavulanic acid protocol (n = 290). Before the treatment, milk samples were collected and cultivated in chromogenic media. After finishing the antimicrobial treatment (intramammary), milk samples were also collected and cultivated in chromogenic media. Results from microbiological analysis obtained before and after treatment were compared to determine the healing rate. Nine different microbiological agents were identified: eight of environmental origin and one of contagious origin; being eight gram-positive and one gram-negative bacteria. TThe positive control group and the treatment group showed similar healing rate (86.5% and 90.3%, respectively; P > 0.05). No differences were found between the groups for the healing rate, when the causing agent was considered. Therefore, it is possible to indicate the amoxicillin-clavulanic acid-based protocol to treat intramammary mastitis in Brazilian dairy cows, achieving great healing rates and providing a substantial reduction in milk disposal. RESUMO: A associação de amoxicilina e ácido clavulânico tem mostrado alta eficácia para o tratamento mastite em diversos países, além de promover um menor período de carência em comparação a outros antimicrobianos utilizados no tratamento da mastite. Apesar disso, estudos considerando a sua utilização para o tratamento de mastite bovina nos rebanhos leiteiros brasileiros ainda não foram realizados. Portanto, esse estudo teve o objetivo de avaliar a efetividade do uso de amoxicilina associado ao ácido clavulânico no tratamento da mastite bovina em rebanhos leiteiros do Brasil e compará-los com os resultados obtidos a partir do uso de um protocolo referência (contendo tetraciclina, neomicina e bacitracina). Um total de 468 vacas da raça Holandesa de três diferentes rebanhos brasileiros foi utilizado no estudo, sendo 178 animais designados para o grupo tetraciclina, neomicina e bacitracina (controle positivo) e 290 animais designados para o grupo amoxicilina-ácido clavulânico. Abstract Treating lactating sows with chorionic gonadotropins may allow controlling their post-weaning reproductive function, despite the occurrence of anestrous during lactation. This article reviews the potential effectiveness of treatment with both equine and human chorionic gonadotropins (eCG and hCG, respectively) during lactation on the control of estrus expression and ovulation in weaned sows. The use of 1,000 IU hCG at 24 and 48 h postpartum may induce ovulation in the treated sows, but the ovulation rate may be variable. Pregnancy rates may be improved with combined treatment after the second week of lactation with both chorionic gonadotropins: 1,500 IU eCG plus 500 – 1,000 hCG; or 1,000 IU eCG plus 1,000 IU hCG. Treatment with eCG (1,000 – 2,000 IU) at the end of lactation may result in acceptable estrus expression and ovulation rates, although with marginal benefit for pregnancy rates. ABSTRACT Artificial insemination success in swine is mainly associated with semen dose quality. Thus, this study compared quality control parameters in 11 Brazilian boar studs after applying audit services for 24 months (1,650 boars). An extensive checklist was applied in each audit, registering ‘compliance’ or ‘noncompliance’ for 75 items. Semen doses produced were analyzed as regards volume and sperm concentration, and microbiological analyses were conducted for semen and water samples collected at distinct production stages. On average, boar studs produced 112.9 semen doses per boar per month, and the odds of raw semen contamination increased when boars were inadequately housed and doses were collected under increased temperatures, with no anti-slip rubber mat or after a poor prepuce cleaning (p < 0.05). Collection from boars with locomotor problems and no regular change of reverse osmosis filters increased the contamination odds in semen doses produced and stored at the stud (p < 0.05). As regards the water submitted to the osmosis reverse process, contamination odds increased as a result of deficient cleaning and disinfection of the purification equipment (p < 0.05). Risk factors for reduced sperm motility (< 70 %) were: no anti-slip rubber mat for semen collection, no cleaning program for automatic feeding system (drops) and bins, and inadequate intervals between semen collections (≤ 2 days or > 7 days; p < 0.05). RESUMO: A inseminação artificial em tempo fixo (IATF) surge como uma biotecnologia para definir o melhor momento para realizar uma única IA, reduzindo o número de células espermáticas por fêmea inseminada. Os protocolos de IATF podem se basear na detecção do estro ou na data de desmame e têm como objetivo sincronizar a ovulação a partir do uso de indutores da ovulação. Protocolos baseados na detecção de estro comumente utilizam o hormônio luteinizante suíno (pLH) como indutor e, de maneira geral, resultados satisfatórios têm sido observados quanto à performance reprodutiva. No caso dos protocolos baseados na data de desmame, os principais hormônios utilizados são os análogos do hormônio liberador de gonadotrofina: triptorelina e buserelina. Independentemente do protocolo, o número de nascidos totais normalmente não é afetado pelo uso da IATF. Porém, um possível comprometimento na taxa de parto deve ser considerado. Já a aplicação da IATF em leitoas requer o fornecimento de um progestágeno, para sincronização do estro, aumentando o manejo e o custo do protocolo. A IATF pode proporcionar diversos benefícios para a indústria suinícola, uma vez que é possível reduzir o número de doses de sêmen produzidas, melhorar o planejamento de coberturas e, consequentemente, otimizar a mão de obra. No entanto, essa biotecnologia apresenta limitações devendo ser considerado a redução nos dados de fertilidade, uma vez que a taxa de parto pode ser comprometida em alguns casos e, o custo do protocolo e a dificuldade de estimar todos os benefícios conceituais da IATF quando aplicada sob condições comerciais. O objetivo dessa revisão é abordar o desempenho reprodutivo dos mais recentes protocolos de IATF, considerando os benefícios e as limitações dessa tecnologia na produção de suínos.

ABSTRACT: Fixed-time artificial insemination (FTAI) is a reproductive technology that aids in obtaining an appropriate time to perform single artificial insemination (AI), thus reducing the number of inseminations per sow bred. FTAI protocols can either be based on estrus detection or day of weaning, aiming to synchronize ovulation using ovulation inducers. The protocols involving estrus detection usually employ porcine luteinizing hormone (pLH) as an inducer and, in general, satisfactory reproductive performance is observed. For protocols based on weaning day, the main hormone used is analog of gonadotropin-releasing hormone such as triptorelin and buserelin. Regardless of the protocol, the number of piglets born is usually not affected by FTAI. However, a possible compromise in the farrowing rate should be considered. The FTAI in gilts requires progestogen treatment for estrus synchronization, increasing the labor requirement and cost of protocol. Some of the benefits of FTAI are a reduced number of semen doses required, advantage of planning the breeding time and; consequently, optimizing labor involved. However, the limitations include a slight reduction in the fertility index due to the compromised farrowing rate in some cases, costs incurred by following the protocol, and difficulty in measuring all the conceptual benefits under commercial conditions. The aim of this review is to approach the reproductive performance of the current protocols of FTAI, considering the benefits and limitations of this technology in swine production.
