ABSTRACT Lippia alba (Mill.), popularly known bushy matgrass, is considered an aromatic and medicinal plant with physiotherapeutic characteristics, leading to its use in the chemical and pharmaceutical industries. Therefore, the study aimed to evaluate the chemical composition and yield of the essential oil of Lippia alba (Mill.) after the occurrence of frost in southern Brazil. The study was carried out in an experimental area located at the Federal University of Santa Maria, Frederico Westphalen Campus, in July 2014. The essential oil was extracted using the hydrodistillation technique for three hours after the start of boiling. The chemical composition of Lippia alba (Mill.) essential oil was identified through chromatographic analysis. The chemical composition of Lippia alba (Mill.) essential oil was not altered by frost. ABSTRACT: This study evaluated the effectiveness of the essential oil of Hesperozygisringens (EOHR) for anesthesia of Colossomamacropomum by documenting hematological and blood biochemical responses after biometric handling. In Experiment 1, juveniles (14.12 ± 3.53 g) were exposed to different concentrations of EOHR: 0 (control), 75, 150, 300 and 450 µL L-1 (n=10 fish for each concentration), to determine times for induction and recovery from anesthesia, as well as its effects on ventilatory frequency (VF). Based on these results, Experiment 2 evaluated the effects of 0 (control), 75 (with induction and recovery times outside that recommended for fish anesthesia) and 150 µL L-1 EOHR (within recommended times) on hematological and biochemical variables of juveniles (20.52 ± 3.47 g) after anesthesia and after 24 h of recovery (n = 6 fish for each concentration and collection time). Survival was 100%. Induction time showed a quadratic effect of EOHR concentration. Recovery time did not differ among EOHR concentrations. Concentrations between 150 and 450 µL L-1 EOHR caused rapid induction (< 3 min) and recovery (< 5 min). EOHR concentration affected VF. RESUMO: Este estudo avaliou a eficiência do óleo essencial de Hesperozygis ringens (EOHR) para anestesia de Colossoma macropomum, documentando as respostas hematológicas e bioquímicas do sangue após o manuseio biométrico. No experimento 1, juvenis (14,12 ± 3,53 g) foram expostos a diferentes concentrações de EOHR: 0 (controle), 75, 150, 300 e 450 µL L-1 (n = 10 peixes para cada concentração), para determinar os tempos de indução e recuperação da anestesia, bem como seus efeitos na frequência ventilatória (VF). RESUMO O ergosterol é responsável por importantes funções na membrana plasmática dos fungos. A influência de agentes fungitóxicos no conteúdo de ergosterol na membrana é um dos mecanismos mais importantes de ação antifúngica e seu conhecimento permite a geração de produtos que associam compostos ativos de diferentes mecanismos, consequentemente, melhorando a eficácia dos conservantes de madeira. Portanto, este estudo otimizou um método para quantificar ergosterol em fungos apodrecedores da madeira. As espécies de podridão-branca selecionadas foram Ganoderma applanatum e Trametes versicolor, enquanto que as de podridão-parda foram Gloeophyllum trabeum e Lentinus lepideus. Discos miceliais de cada espécie foram transferidos para placas de Petri contendo meio batata-dextrose-ágar coberto por celofane. O micélio de cada fungo foi coletado, pesado e transferido para tubos de ensaio com 5 mL de hidróxido de potássio alcoólico 25%. Os tubos foram agitados em vortex por 5 min, submetidos a ultrassom por 5 min, incubados a 85 °C por 4 h, seguido da adição de 2 mL de água destilada estéril e 5 mL de n-heptano e subsequente agitação em ultrassom por 2 min. A camada de n-heptano foi analisada por espectrofotometria no UV entre 230 e 300 ηm. O branco continha apenas n-heptano. A massa micelial úmida dos fungos variou de 0,061 a 0,296 g. O teor de ergosterol foi de 0,007% para Lentinus lepideus e 0,004% para as demais espécies. A absorbância foi maior que a observada no branco para todas as amostras. O método adaptado foi eficiente para extração de ergosterol.

ABSTRACT Ergosterol is responsible for important functions in the fungal plasma membrane. The influence of fungitoxic agents on membrane ergosterol content is one of the most important mechanisms of antifungal action and its knowledge allows the generation of products that associate active compounds of different mechanisms, consequently improving the effectiveness of wood preservatives. Therefore, this study optimized a method for quantifying ergosterol in wood-decay fungi. The white-rot species selected were Ganoderma applanatum and Trametes versicolor, while the brown-rot were Gloeophyllum trabeum and Lentinus lepideus. Mycelial discs of each species were transferred to Petri dishes containing a cellophane-covered potato-dextrose-agar medium. Mycelia of each fungus were collected, weighed, and transferred to test tubes with 5 mL of 25% alcoholic potassium hydroxide. The tubes were vortexed for 5 min, subjected to ultrasound for 5 min, incubated at 85 °C for 4 h, followed by the addition of 2 mL of sterile distilled water and 5 mL of n-heptane and subsequent ultrasound shaking for 2 min. The n-heptane layer was analyzed by UV spectrophotometry between 230 and 300 ηm. The blank sample only contained n-heptane. The mycelia wet weight of the fungi ranged from 0.061 to 0.296 g. Ergosterol content was 0.007% for Lentinus lepideus and 0.004% for the other species. The absorbance was higher than the ones observed in the blank for all samples. The adapted method was efficient for ergosterol extraction.

RESUMO. O objetivo deste estudo foi investigar o efeito da suplementação na dieta com óleo essencial de Cymbopogon flexuosus (EO) sobre o desempenho produtivo, os parâmetros metabólicos e a composição corporal e de filés de jundiás (Rhamdia quelen) adultos. O comprimento total, o fator de condição, o ganho de peso, a taxa de crescimento específico, o índice hepatossomático, o teor de gordura visceral e a composição dos filés não foram afetados pela suplementação com EO. Os níveis plasmáticos de proteína total, globulina e triglicérides foram reduzidos pela suplementação com EO. A suplementação com 1 mL de EO kg-1 de dieta aumentou o rendimento de carcaça e a deposição de proteína na carcaça, mas reduziu o índice gonadossomático e a deposição de gordura em comparação ao grupo controle. Além disso, 1 mL de EO kg-1 de dieta também melhorou o consumo de ração em comparação com 3 mL de EO kg-1 de dieta. Os resultados sugerem que se pode utilizar até 1 mL de EO kg-1 de dieta na alimentação de peixes para melhorar a deposição de proteína e a produtividade de carcaça de jundiás em fase de engorda.

ABSTRACT. The aim of this study was to investigate the effect of dietary supplementation with Cymbopogon flexuosus essential oil (EO) on productive performance, metabolic parameters and body and fillet composition of adult silver catfish, Rhamdia quelen. Total length, condition factor, weight gain, specific growth rate, hepatosomatic index, visceral fat content and fillet composition were not affected by EO supplementation. Plasma total protein, globulin and triglycerides were reduced by EO supplementation. Supplementation with 1 mL EO kg-1 diet increased carcass yield and protein deposition in the carcass, while reducing gonadosomatic index and fat deposition in the carcass when compared to control group. In addition, 1 mL EO kg-1 diet also improved feed intake when compared to 3 mL EO kg-1 diet. Our results suggest that EO up to 1 mL kg-1 diet can be used in fish feed to improve protein deposition and carcass yield of silver catfish.

RESUMO O efeito anestésico dos óleos essenciais (OEs) da casca de Citrus x aurantium (OECA) e Citrus x latifolia (OECL) em jundiá Rhamdia quelen foi investigado. Os peixes foram expostos a diferentes concentrações de OECA e OECL para determinar o tempo de indução e recuperação da anestesia. Todos peixes expostos a 400, 600 ou 800 μL L−1 OECA e 300, 400 ou 500 μL L−1 OECL foram anestesiados. Outro grupo de peixes foi exposto aos OEs durante 8 h a 50, 100 ou 200 μL L−1. Peixes expostos ao etanol e aos OEs apresentaram VF maior que o grupo controle durante as 8 h de exposição. Os efluxos líquidos de Na+, K+, Cl− e a excreção de amônia foram significativamente menores nos peixes expostos a 50, 100 ou 200 μL L−1 dos OEs em comparação com o grupo controle. A mortalidade foi de 37% nos peixes expostos a 200 μL L−1 de ambos os OEs após 8 h. Os resultados sugerem que OECA e OECL são anestésicos e sedativos úteis para o jundiá, mas sua utilidade como alternativa para reduzir o estresse no transporte de peixes nas concentrações mais baixas testadas (50-100 μL L−1) necessita de estudos adicionais.

ABSTRACT This study investigated the anesthetic effect of the essential oils (EOs) from the peel of Citrus x aurantium (EOCA) and Citrus x latifolia (EOCL) on silver catfish Rhamdia quelen. Fish were exposed to different concentrations of EOCA and EOCL to determine time of anesthesia induction and recovery. Induction of anesthesia was observed in all fish exposed to 400, 600 or 800 μL L−1 EOCA and 300, 400 or 500 μL L−1 EOCL. Another group of fish were exposed for 8 h to 50, 100, or 200 μL L−1 of either EOs. Overall, fish exposed to ethanol and both EOs presented higher ventilatory frequencies (VF) than the control group throughout the 8 h of exposure. Net ion (Na+, K+ and Cl−) effluxes and ammonia excretion were significantly lower in fish exposed to 50, 100 or 200 μL L−1 of either EOs compared to control fish. Mortality was 37% in fish exposed to 200 μL L−1 of either EOs after 8 h. These findings suggest that EOCA and EOCL are useful anesthetics and sedatives for Rhamdia quelen, but their usefulness as alternatives to reduce stress in fish transportation at the lower concentrations tested (50-100 µL L−1) deserves further study.

RESUMO: Este estudo teve como objetivo avaliar os possíveis efeitos relaxantes e/ou moluscicidas do eugenol e óleos essenciais de Origanum majorana, Ocimum americanum, Hesperozygis ringens e Piper gaudichaudianum no gastrópode Pomacea canaliculata. Os compostos foram testados nas concentrações de 100, 250, 500 e 750µL L-1 para avaliar os efeitos relaxantes. Em um segundo experimento, os animais foram expostos a 10, 25 e 50µL L-1 por 24 horas aos óleos essenciais de H. ringens e P. gaudichaudianum para avaliação dos efeitos moluscicida. O eugenol e os óleos essenciais de O. majorana e O. americanum apresentaram efeito relaxante nas concentrações ≥250µL L-1, mas os óleos essenciais de H. ringens e P. gaudichaudianum não promoveram efeito relaxante ou moluscicida no tempo e concentrações estudadas. Portanto, apenas o eugenol e os óleos essenciais de O. majorana e O. americanum podem serem usados para fins de relaxamento em P. canaliculata.

ABSTRACT: This study evaluated the potential relaxing and/or molluscicidal effects of eugenol and essential oils of Origanum majorana, Ocimum americanum, Hesperozygis ringens, and Piper gaudichaudianum in the gastropod Pomacea canaliculata. Compounds were tested at concentrations of 100, 250, 500, and 750µL L-1 to evaluate the relaxing effects. In the second experiment, animals were exposed to 10, 25, and 50µL L-1 of essential oils of H. ringens and P. gaudichaudianum for a period of 24h for the evaluation of molluscicidal effects. Eugenol and essential oils of O. majorana and O. americanum showed relaxing effects at ≥250µL L-1, but the essential oils of H. ringens and P. gaudichaudianum did not promote relaxing or molluscicidal effects within the times and concentrations studied. Therefore, only eugenol and the essential oils of O. majorana and O. americanum can be used for relaxation purposes in P. canaliculata.

This study evaluated the seasonal influence on the yield and chemical composition of the essential oil (EO) of Nectandra megapotamica. Fresh young (YL) and old leaves (OL) obtained from three trees in each season (Nov/2010 to Sep/2011) collected in Santa Maria-RS were hydrodistilled in triplicate. The chemical composition was determined by the gas chromatography coupled to mass spectrometry (GC-MS) and the yield on dry basis was evaluated by two-way ANOVA (seasons, development stage). Spring (Sp) and summer (Su) showed higher average incomes (0.45 and 0.33%), which occurred when flowering, fruiting, and growth of YL and senescence of OL took place, while autumn (Au) presented the lowest yield (0.25%) during the rustification of OL. The highest yield was obtained for the YL in Sp (0.59%) and the lowest for the OL in Au (0.21%). The major constituents of the EO were independent from the season and were identified as α-pinene, bicyclogermacrene, β-pinene, germacrene D, and limonene. Seasonality and phenology influenced the production of EO probably due to morphological and metabolic alterations in the leaves as well as due to the needs of the tree, such as attraction and/or protection.
