Resumo As florestas urbanas fornecem cobertura vegetal de médio e grande porte em áreas urbanas. O plantio de árvores nativas nas calçadas é uma abordagem viável para reduzir os danos causados pela extensa urbanização. O uso de hidrogéis parece aumentar o sucesso do transplante de mudas em ambientes urbanos. Assim, este estudo tem como objetivo avaliar a eficácia de Schinus terebinthifolia na arborização urbana, focando na sua adaptação e sobrevivência pós-transplante utilizando hidrogel e sendo monitorado por sistema arduino. As concentrações do hidrogel utilizadas foram 0,75; 1,5; 3,0 e 6,0 g L-1, sendo também estabelecidos dois controles: um tratamento controle sem irrigação e sem hidrogel, e um controle com irrigação diária com água. A avaliação foi contínua durante 14 dias, e os parâmetros analisados foram temperatura e umidade do substrato, teor relativo de água (CR), extravasamento de eletrólitos (EEE), teor de clorofila e compostos bioquímicos. Adotou-se delineamento inteiramente casualizado, composto por 6 tratamentos com 9 repetições cada. Os resultados indicam que S. terebinthifolia é uma espécie altamente resiliente e adequada para arborização urbana, apresentando notável tolerância ao transplante e à restrição hídrica. Observou-se que o uso de hidrogéis contribui significativamente para a manutenção da umidade do substrato, resultando em maior estabilidade das mudas transplantadas. Abstract Urban forest provide medium and large vegetation cover in urban areas. Planting native trees on sidewalks is a viable approach to reduce damages caused by extensive urbanization. The use of hydrogels seems to increase the success of seedling transplantation in urban environments. Thus, this study aims to evaluate the efficacy of Schinus terebinthifolia in urban afforestation, focusing on its adaptation and post-transplant survival using hydrogel and being monitored by an arduino system. The concentrations of the commercial hydrogel used were 0.75, 1.5, 3.0, and 6.0 g L-1, and two controls were also established: a control treatment without irrigation and without hydrogel, and a control with daily water irrigation. The evaluation was continuous for 14 days, and the parameters analyzed were substrate temperature and moisture, relative water content (RWC), electrolyte extravasation (EEE), chlorophyll content, and biochemical compound. Resumo Clitoria ternatea L. é valorizada por suas características ornamentais, propriedades medicinais e usos culinários. No entanto, a produção eficiente desta espécie é limitada pela dormência do tegumento, que impede a germinação e a produção de mudas. Este estudo teve como objetivo explorar os aspectos físico-químicos e morfológicos de sementes de C. ternatea, com foco na superação da dormência através da aplicação de nanotubos de carbono de paredes múltiplas (MWCNTs) funcionalizados com ácido carboxílico. As sementes foram caracterizadas quanto à sua composição física, química e mineral. A caracterização incluiu análises de dominância de cor, geometria, peso de mil sementes e teor de umidade. Abstract Clitoria ternatea L. is valued for its ornamental characteristics, medicinal properties, and culinary uses. However, efficient production of this species is constrained by seed coat dormancy, which impedes germination and seedling production. This study aimed to explore the physicochemical and morphological aspects of C. ternatea seeds, focusing on overcoming dormancy by applying multi-walled carbon nanotubes (MWCNTs) functionalized with carboxylic acid. The seeds were characterized by their physical, chemical, and mineral composition. Characterization included color dominance, geometry, thousand seed weight, and moisture content analyses. The seeds were treated with different concentrations (0, 100, 200, 400, 800 mg L-1) of multi-walled carbon nanotubes (MWCNT) during 24 hours of soaking. Then the germination rates were evaluated and morphological analysis was performed using scanning electron microscopy. RESUMO O armazenamento em baixa temperatura é uma das técnicas mais eficazes para manter a qualidade e a durabilidade das flores cortadas. Tanto a manutenção da qualidade quanto a durabilidade são dependentes de fatores do metabolismo da planta como o sistema antioxidante. O objetivo foi avaliar o efeito da baixa temperatura no pré-resfriamento e armazenamento pós-colheita, no metabolismo do copo-de-leite [Zantedeschia aethiopica (L.) K. ABSTRACT Low-temperature storage is one of the most effective techniques to maintain the quality and durability of cut flowers. Both quality maintenance and durability are dependent on plant metabolism factors such as antioxidant system. RESUMO Campomanesia rufa (O. Berg) Nied. é uma espécie mirtácea nativa do Cerrado brasileiro. Considerada uma espécie ameaçada de extinção, a propagação in vitro de C. rufa apresenta-se como uma alternativa para sua conservação. No entanto, a falta de protocolos eficazes para a desinfecção de microrganismos endofíticos naturalmente presentes nos tecidos vegetais dificulta o sucesso da micropropagação. Neste contexto, o objetivo deste estudo foi o isolamento, a identificação molecular e o controle de bactérias endógenas de C. rufa propagadas in vitro. Produtos de PCR purificados dos isolados bacterianos foram sequenciados pelo método de Sanger. ABSTRACT Campomanesia rufa (O. Berg) Nied. is a Myrtaceae species native to the Brazilian Cerrado. As C. rufa is considered endangered, in vitro propagation is an alternative for its conservation. However, the lack of effective disinfection protocols for endophytic microorganisms naturally present in plant tissues hinders the success of micropropagation. In this context, the objective of this study was to isolate, molecular identify, and control endogenous occurring bacteria of C. rufa propagated in vitro. Purified PCR products of bacterial isolates were sequenced by the Sanger method and aligned with homologous sequences using the Basic Local Alignment Search Tool (BLAST) available in National Centre for Biotechnology Information (NCBI) database. MEGA7 software was used to align all sequences and to draw phylogenetic trees. Survival, shoot height, and efficiency of antibiotics (streptomycin, ampicillin, and chloramphenicol) at different concentrations in the culture medium were evaluated. ABSTRACT Genipa americana L. is a forest species of high socioeconomic potential. However, predatory extractivism actions threaten its existence, making it necessary to adopt conservationist practices. G. americana seeds show sensitivity to desiccation and cooling, making it unfeasible for conservation by conventional methods. Thus, cryopreservation is an promising alternative for the long-term conservation of species that produce unorthodox seeds, such as the genipap. In this sense, the objective was to cryopreservation of seeds of G. americana and to evaluate the effects of desiccation and freezing on germination and establishment of seedlings. Initially, seeds were dehydrated in silica gel 0, 16, 18, 20, 22 and 24 h, and then were cryopreserved in liquid nitrogen (LN) (-196 °C) during 24 h. After thawing, the viability and germination were analyzed. Dehydrated and non-cryopreserved seeds were also analyzed. The silica gel desiccation caused a reduction in viability and germination of the seeds of G. americana. The initial seed water content was so high (47%) that storage in LN (+LN) without prior dehydration treatment resulted in seed mortality. It was verified that, the dehydration in silica gel for the minimum time 20 h (corresponding to 14% water content) provides greater freezing tolerance, allowing the successful cryopreservation. Resumo Strelitzia ou ave-do-paraíso, é uma importante planta ornamental tropical de difícil propagação devido aos problemas de dormência das sementes e ao reduzido número de mudas obtidas nas divisões das plantas. Com o maior conhecimento de sua estrutura, é possível desenvolver estratégias para melhorar o processo de propagação dessa espécie, uma vez que as informações sobre estes aspectos são limitadas. Assim, o objetivo foi caracterizar sementes e embriões de Strelitzia reginae. A caracterização das sementes foi realizada por meio de análise de imagens em equipamento GroundEye, Raio-X e análises anatômicas e bioquímicas. Adicionalmente, foram obtidas as seguintes características biométricas das sementes: área de 0,33 cm2, diâmetro máximo de 0,736 cm, diâmetro lateral máximo de 0,59 cm, diâmetro mínimo de 0,58 cm e perímetro de 2,47 cm. As sementes de S. reginae podem ser consideradas amiláceas por conterem 15% de amido. Pela análise anatômica do tegumento, observou-se a presença de uma exotesta (ex) e uma camada parenquimática posterior que foi dividida em mesotesta (m) e endotesta (en). A caracterização de sementes e embriões de estrelícia apresentou observações relevantes para a taxonomia e fisiologia desta espécie. As sementes são aleuro-amiláceas e apresentam área média de 0,33 cm2, com diâmetro lateral máximo de 0,59 cm. Como resultado de estudos anatômicos, foi possível identificar o tegumento responsável pela dormência.

Abstract Strelitzia or bird-of-paradise, is an important tropical ornamental plant that is difficult to propagate because of seed dormancy issues and the low number of seedlings obtained from plant divisions. With greater knowledge of its structure, it is possible to develop strategies to improve the propagation process for this species, since information on this subject is currently limited. Thus, the objective was to carry out an anatomical, morphological and biochemical characterization of Strelitzia reginae seeds and embryos. The characterization of the seeds was performed through image analysis using GroundEye equipment, X-rays, and anatomical and biochemical analyses. Additionally, the following biometric characteristics of the seeds were obtained: an area of 0.33 cm2, a maximum diameter of 0.736 cm, a maximum lateral diameter of 0.59 cm, a minimum diameter of 0.58 cm, and a perimeter of 2.47 cm. The seeds of S. reginae can be considered starchy since they contain 15% starch. By integument anatomical analysis, we observed the presence of an exotesta (ex) and a posterior parenchymatic layer that was divided into a mesotest (m) and endotest (en). The characterization of Strelitzia reginae seeds and embryos showed relevant observations for the taxonomy and physiology of this species. The seeds are aleurostarches and present an area of 0.33 cm2 on average, with maximum lateral diameter of 0.59 cm. As result of anatomical studies, it was possible to determine coat responsible for integument dormancy.

RESUMO As espécies tropicais podem exigir temperaturas mais altas e maiores concentração de reguladores de crescimento para o desenvolvimento in vitro. Em função dessas condições afetarem o metabolismo vegetal, o objetivo foi identificar como diferentes temperaturas na sala de crescimento e concentrações de giberelinas podem afetar os embriões no desenvolvimento in vitro de estrelícia, analisando o efeito sobre EROS e a estabilidade citogenética. Os embriões zigóticos foram cultivados em meio MS suplementado com 0, 5, 10 e 20 µM de GA3 e mantidos em sala de crescimento sob temperaturas de 25 °C, 30/25 °C e 30 °C. Após 60 dias, observou-se maior germinação dos embriões (72%) e comprimento das brotações (3,14 cm) em meio adicionado com 20 µM de GA3. Nessa concentração, houve um aumento na atividade da redutase do nitrato sem alterar a estabilidade citogenética. A temperatura influenciou apenas o comprimento da parte aérea e da raiz, que foram maiores a 25 °C. A 30 °C, a atividade de SOD e APX aumentou em comparação com o tratamento a 25 °C. Assim, a adição de 20 µM de GA3 ao meio de cultura e temperatura de 25 °C na sala de crescimento deve ser utilizada para cultura de embriões zigóticos de estrelitzia.

ABSTRACT Tropical species may require higher temperatures as well as higher growth regulator concentrations for in vitro development. Since these conditions may affect plant metabolism, the objective of this study was to identify how different temperatures and gibberellin concentrations may affect the in vitro development of strelitzia embryos, analyzing the effect on ROS and cytogenetic stability. Zygotic embryos were cultivated on MS medium supplemented with 5, 10 and 20 µM GA3 under temperatures of 25 °C, 30/25 °C and 30 °C. After 60 days, higher embryonic germination rate (72%) and shoot length of plantlets (3.14 cm) were observed on medium containing 20 µM gibberellic acid (GA3). At this concentration, there was an increase in nitrate reductase activity with no change in the cytogenetic stability. The temperature influenced only shoot and root lengths, which were highest at 25 °C. At 30 °C, superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities increased compared with those at 25 °C. Thus, the addition of 20 µM GA3 to the culture medium and a temperature of 25 °C in the growth room should be used for zygotic embryo culture of strelitzia.

RESUMO Rugosa rugosa tem alta tolerância a várias estresses; no entanto, os mecanismos moleculares dessa aptidão sob condições adversas não são claros. O objetivo deste estudo é investigar padrões de expressão de genes relacionados ao estresse em resposta a salinidade. Alterações nos níveis de transcrição de mudas de R. rugosa, cultivadas sob diferentes condições de estresse salino (0, 25, 50 e 100 mM de NaCl) durante um longo período de exposição (30 dias), foram investigadas. Além disso, os efeitos do estresse por choque salino após a exposição das mudas a um nível alto (200 mM) de NaCl por um período relativamente curto (3 h) também foram investigados. Os níveis de expressão de genes expressos diferencialmente selecionados foram determinados usando a reação em cadeia da polimerase com transcrição reversa relativa (RT-PCR). Observou-se que as mudas expostas ao estresse salino por um longo período não apresentaram sinais de estresse nas folhas e nas raízes. Além disso, a expressão de NHX1 em R. rugosa aumentou na presença de NaCl. Além disso, os transcritos dos genes EXP4, GPP, NHX1, NAC e DREB também aumentaram em mudas cultivadas sob altos níveis de NaCl. ABSTRACT Rugosa rugosa has high tolerance to various stresses; however, the molecular mechanisms of this behavior under adverse conditions are unclear. The objective of this study is to investigate expression patterns of stress-related genes in response to salinity stress. Changes in transcript levels of R. rugose, grown under different salt stress conditions (0, 25, 50, and 100 mM NaCl) over a long exposure period (30 days), have been investigated. In addition, the effects of salt shock stress on seedlings exposed to a high level (200 mM) of NaCl for a relatively short duration (3 h) have also been investigated. Expression levels of selected differentially expressed genes have been determined using relative reverse transcription polymerase chain reaction (RT-PCR). It has been observed that seedlings exposed to salt stress for a long duration exhibited no signs of stress in both leaves and roots. In addition, expression of NHX1 in R. rugosa increased in the presence of NaCl. Furthermore, transcripts of EXP4, GPP, NHX1, NAC, and DREB genes also increased under high levels of NaCl. In contrast, expression levels of MYB and TIR decreased during this salt shock treatment. Of particular interest is the increase in levels of transcripts of NHX1 in leaves of seedlings grown under both salt stress and salt shock conditions, thus suggesting that this gene plays an important role in salt stress tolerance in R. rugosa. These findings will support efforts in enhancing salt tolerance in roses, and perhaps in other members of the Rosaceae family.

RESUMO Campomanesia rufa (O. Berg) Nied. é uma espécie nativa do Cerrado que apresenta grande potencial comestível. No entanto, é uma espécie “em perigo de extinção”, segundo a União Internacional para a Conservação da Natureza (IUCN). Não existe informação técnica e científica sobre a espécie, demonstrando a importância das pesquisas. O presente trabalho teve como objetivo a caracterização físico-química de frutos imaturos e maduros de C. rufa. Os frutos apresentaram alteração na coloração de verde (b * = 25.11, h = 122.43) para verde amarelado (b * = 34.26, h = 115.73), aumento na massa (6.54 g para 10.88 g), diâmetro (23.76 mm a 28.03 mm) e sólidos solúveis (8.00 a 10.80%). Os frutos apresentaram altos teores de pectina total (1246.35 mg 100 g-1) e solúvel (195.93 mg 100 g-1), alto teor de água (78.86 g 100 g-1), baixo valor de pH (3.40) e alto ácido cítrico conteúdo (1.2%). No entanto, os frutos apresentaram baixo teor de proteína (0.81 g 100 g-1), teor de lipídios e baixo valor calórico (64.76 kcal 100 g-1). Os frutos apresentaram valores significativos de carotenóides, compostos fenólicos (312.47 mg 100 g-1), vitamina C (263.60 mg 100 g-1) e boa atividade antioxidante in vitro (1862.81 µM g-1). Os resultados obtidos indicam que os frutos de C. rufa apresentam composição semelhante aos frutos de outras espécies de Campomanesia, e suas propriedades biológicas devem ser investigadas adicionalmente em condições in vivo.

ABSTRACT Campomanesia rufa (O. Berg) Nied. is a native Cerrado species that presents great edible potential. However, it is a species “in danger of extinction” as recommended by the International Union for the Conservation of Nature (IUCN). No technical and scientific information about the species exists, thus demonstrating the importance of its research. The present work aimed at the physical and chemical characterization of immature and mature C. rufa fruits. The fruits showed a change in coloration from green (b * = 25.11, h = 122.43) to yellowish-green (b * = 34.26 , h = 115.73), an increase in mass (6.54 g to 10.88 g), diameter (23.76 mm to 28.03 mm) and soluble solids (8.00 to 10.80%). The fruits presented high levels of total (1246.35 mg 100 g-1) and soluble pectin (195.93 mg 100 g-1), high water content (78.86 g 100 g-1), low pH value (3.40), and high citric acid content (1.2%). However, the fruits had low protein (0.81 g 100 g-1), lipid contents, and low caloric values (64.76 kcal 100 g-1). The fruits presented significant values of carotenoids, phenolic compounds (312.47 mg 100 g-1), vitamin C (263.60 mg 100 g-1) as well as good in vitro antioxidant activity (1862.81 µM g-1). The results obtained indicate that C. rufa fruits showed a similar composition to the fruits of other Campomanesia species, and their biological properties should be investigated additionally under in vivo conditions.

RESUMO A Campomanesia rufa é uma espécie frutífera nativa do Brasil considerada como vunerável pela IUCN (International Union for Conservation of Nature) e poucas informações sobre a propagação são encontradas. Neste contexto, o objetivo do presente estudo foi desenvolver protocolos de germinação in vitro, micropropagação e calogênese para a espécie. Para germinação in vitro, as sementes foram inoculadas em meio MS suplementado com GA3 (Ácido Giberelico), para a indução da parte aérea, o meio de cultura foi suplementado com BA (benzilamina), BAP (6-Benzilamina Purina) ou TDZ (Thidiazuron). Para o crescimento da parte aérea, o meio de cultura contendo BA, BAP e TDZ foi mantido sob lâmpadas fluorescentes brancas e sob diodos emissores de luz (LED). O GA3 também foi testado para elongação de brotações in vitro. Para o controle da oxidação, o meio foi suplementado com PVP (polivinilpirrolidona) e para a indução de calos, 2,4-D (Ácido 2,4-diclorofenoxiacético) . Os resultados mostraram 68% de germinação, independentemente da concentração de GA3. No estágio de propagação, BAP a 4,5 µM induziu maior número de brotações (4,53) e lâmpadas de LED como fonte de luz combinada com o meio de cultura com 1 μM de BAP induziu maior número de brotações (4,08). A maior média de comprimento de C. rufa (31,9 mm) foi obtida usando 8,8 µM de GA3. O uso de PVP (584,3 µM) controla até 27,3% de oxidação em explantes de folhas jovens. O uso de 10 µM de 2,4-D leva a uma maior formação de calos (58,7%). Portanto, pode-se concluir que o uso de BAP é eficiente na indução de brotações, o PVP controla a oxidação de segmentos foliares e o 2,4-D induz calos em C. rufa.

ABSTRACT The Campomanesia rufa is a fruitful species native from Brazil considered as endangered by the IUC (International Union for Conservation of Nature) and low information regarding its propagation is available. In this context, the aim of the present study was to develop in vitro germination, micropropagation and callogenesis protocols for the species. For in vitro germination, seeds were inoculated in MS medium supplemented with GA3 (gibberellic acid) and for shoot induction, the medium was supplemented with three different cytokinins BA (benzyladenine), BAP (6-Benzylaminopurine) or TDZ (Thidiazuron). For shoot growth, culture medium containing BA, BAP, TDZ were maintained under white fluorescent lamps and blue:red light-emitting diodes (LED). GA3 was also tested on in vitro shoot elongation. For oxidation control, the medium was supplemented with PVP (polyvinylpyrrolidone) and for callus induction, 2.4-D (dichlorophenoxyacetic acid). The results showed 68% germination regardless of GA3 concentration. In the propagation stage, BAP at 4.5 µM induced a higher number of shoots (4.53) and LED lamps as the light source combined with the culture medium with 1 μM BAP induced a higher number of shoots (4.08). The highest average of C. rufa length (31.9 mm) was obtained using 8.8 µM GA3. The use of PVP (584.3 uM) controls up to 27.3% oxidation in young leaf explants. The use of 10 µM 2.4-D leads to a higher callus formation (58.7 %). Therefore, it can be concluded that the use of BAP is efficient in the induction of shoots, PVP controls oxidation leaf segments, and 2.4-D induces callus in C. rufa.

RESUMO A germinação in vitro de sementes é uma alternativa eficaz para obtenção de explantes com rapidez e qualidade sanitária. No entanto, sementes de jenipapeiro apresentam germinação lenta e desuniforme. Para tanto, fatores internos e externos às sementes que interferem diretamente no processo, devem ser identificados, visando à adaptação de melhores técnicas para obtenção de mudas. Neste sentido, objetivou-se otimizar a germinação in vitro de sementes de Genipa americana L. por meio da avaliação de diferentes fatores (qualidade de luz, tratamento com GA3, pré-embebição em água destilada, diferentes composições no meio de cultivo e estratificação no escuro). Verificou-se que a germinação das sementes foi indiferente a luz, no entanto, os melhores resultadados foram obtidos sob condições de escuro contínuo; Não houve efeito da aplicação de GA3 exógeno; A pré-embebição em água destilada durante 48h contribui para obtenção de melhores taxas de germinação; E a redução de sais do meio MS, bem como o pré-tratamento de estratificação no escuro maximizam o potencial germinativo das sementes. Portanto, as condições ótimas para germinação in vitro de sementes de G. americana L. requer a pré-embebição em água destilada por 48 horas e a inoculação em meio de cultivo constituído de 1/2 MS + 15 g L-1 de sacarose, com estratificação no escuro por 16 dias, seguida pela transferência a salas de crescimento com iluminação fornecida por lâmpadas fluorescentes branca.

ABSTRACT The in vitro seed germination is an effective alternative for quickly obtaining explants with sanitary quality. However, jenipapo seeds present slow and uneven germination. Therefore, internal and external factors to seed which directly interfere in the process, they must be identified, in order to adapt better techniques to obtain seedlings. In this sense, this work aimed to optimize the in vitro germination of Genipa americana L. seeds by evaluating different factors (light quality, GA3 treatment, pre-soaking in distilled water, growing media and stratification in the dark). It was found that the seed germination of G. americana was indifferent to light, however, the best results were obtained under conditions of continuous darkness; There was no effect of the application of exogenous GA3; The pre-soaking in distilled water for 48 h contributes to obtaining better germination rates; And the reduction in MS medium salts, and laminating the pretreatment in the dark maximizes the germination potential of seeds.Therefore, the optimal conditions for in vitro germination of G. americana L. seeds requires pre-soaking in distilled water for 48 hours and inoculation into culture media consisting of 1/2 MS + 15 g L-1 sucrose, with stratification in the dark for 16 days, followed by the transfer to growth chambers with lighting provided by white fluorescent lamps.

RESUMO A conservação dos recursos genéticos vegetais é uma medida importante para prevenção do processo de erosão genética. Bancos de sementes são as formas mais comuns de conservação ex situ, entretanto, sementes de cafeeiro não podem ser armazenadas por métodos convencionais. A criopreservação é uma alternativa viável para conservação em longo prazo de espécies, como o cafeeiro, que produzem sementes intermediárias ou recalcitrantes. O objetivo deste trabalho foi criopreservar embriões zigóticos de Coffea arabica L. cv Catuaí Vermelho avaliando o efeito da desidratação antes da criopreservação, e da reidratação osmótica após o descongelamento. Antes da criopreservação foram testados e determinados os teores de umidade em diferentes tempos de desidratação (0, 15, 30, 60 e 120 min). Após a desidratação os embriões foram criopreservados por uma hora em nitrogênio líquido, e após o descongelamento foram reidratados utilizando solução osmótica. Para avaliação da viabilidade de embriões desidratados e/ou criopreservados, utilizou-se o teste com o cloreto de 2,3,5-trifenil tetrazólio. Embriões não desidratados não sobreviveram ao congelamento. Embriões desidratados até cerca de 20% de umidade que não passaram por uma reidratação osmocondicionada, não germinaram. Embriões desidratados até cerca de 20% de umidade, criopreservados e reidratados lentamente após o descongelamento, apresentaram 98% de germinação. A máxima viabilidade obtida através do teste de tetrazólio foi 75% após 60 minutos de desidratação. Portanto, embriões zigóticos de cafeeiro podem ser criopreservados utilizando desidratação física em sílica gel por 60 minutos (23% de teor de umidade), seguida de reidratação com solução osmótica após o descongelamento. Esse método permitiu a germinação de 98% dos embriões criopreservados.

ABSTRACT Conservation of plant genetic resources is important to prevent genetic erosion. Seed banks are the most common method of ex situ conservation; however, coffee seeds can not be stored by conventional methods. Cryopreservation is a viable alternative for long-term conservation of species that produce intermediate or recalcitrant seeds, as coffee. The aim of this work was to cryopreserve Coffea arabica L. cv Catuaí Vermelho IAC 144 zygotic embryos, and analyse the effects of dehydration prior cryopreservation and osmotic rehydration after thawing, in embryos germination and seedlings formation after cryopreservation. Prior to cryopreservation, different dehydration times (0, 15, 30, 60 and 120 min) were tested. Dehydrated embryos were cryopreserved in liquid nitrogen for 1 hour, and after thawing were rehydrated by osmotic solutions. Dehydrated and non-cryopreserved embryos were also analysed. The test with 2,3,5 triphenyl tetrazolium chloride (TTC) was used to evaluate the embryos viability. Non-dehydrated embryos did not survive after freezing. Embryos that were dehydrated until 20% of the moisture content did not germinate when osmotic rehydration was not performed. In contrast, cryopreserved embryos with the same moisture content presented 98% germination when they were rehydrated slowly in osmotic solution. According to tetrazolium tests, embryos presented maximum viability (75%) after dehydration for 60 minutes (23% moisture content). Therefore, coffee zygotic embryos (Coffea arabica L. cv. Catuaí Vermelho) can be successfully cryopreserved using physical dehydration in silica gel for 60 minutes (23% moisture content), followed by osmotic rehydration after thawing. This method allowed a germination of 98% of cryopreserved zygotic embryos.

RESUMO O bastão-do-imperador (Etlingera elatior Jack Smith RM) pode ser propagado in vitro, mas não há informações sobre as fases de aclimatização e adaptação no campo. O objetivo foi estudar as respostas de crescimento e sobrevivência durante a aclimatização ex vitro da espécie, quando mantida em diferentes substratos: Plantmax florestal(r); areia; Plantmax florestal(r) e areia 1:1; e malhas de sombreamento a 50% na cor vermelha; azul e controle sem a presença de malha, assim como estudar respostas fotossintéticas durante a aclimatização em casa de vegetação. A maior taxa de sobrevivência das plantas, número de folhas e número de brotos por planta, foram observados no tratamento com substrato tipo Plantmax florestal(r) puro, na ausência de sombreamento. Quanto à taxa de fotossíntese líquida, eficiência de carboxilação, condutância estomática e taxa de transpiração, plantas de cultivo in vitro apresentaram comportamento semelhante ou superior quando comparadas com plantas controle obtidas a partir de rizomas. Em geral, a eficiência do funcionamento bioquímico e do fotossistema II foram semelhantes em plantas de cultivo in vitro e controle. Conclui-se que tais resultados mostram que as plantas derivadas do cultivo in vitro apresentam rendimento fisiológico satisfatório, conferindo capacidade de aclimatação no campo.

ABSTRACT Torch ginger (Etlingera elatior (Jack) RM Smith) can be propagated in vitro, but there is currently little information about the stages of acclimatization and adaptation for torch ginger in a greenhouse. The objective was to study the growth responses, survival and photosynthetic response during the acclimatization of plants maintained on three different substrates (Plantmax forestry(r), sand and 1:1 Plantmax forestry(r) and sand) under three shading conditions (50% red and blue shading nets and control without shading nets). The highest leaf and shoot numbers per plant and survival rate were observed in treatments with the Plantmax forestry(r) type substrate in the absence of shading. In vitro culture plants behaved similarly or better than rhizome propagated control plants with regard to the net photosynthesis rate, carboxylation efficiency, stomatal conductance and transpiration rate. In general, for both conditions, in vitro and control plants had similar efficiency of biochemical functions and of photosystem II. These results show that plants derived from in vitro culture exhibit satisfactory physiological yield and greenhouse acclimation capacity.

O objetivo deste trabalho foi avaliar a eficiência da droplet vitrification e vitrificação na criopreservação de ápices caulinares de mangabeira. Foi avaliada a retomada de crescimento de ápices de mangabeira criopreservados conforme a técnica de droplet vitrificationutilizando diferentes tempos de tratamento com a solução de vitrificação (15, 30, 45 e 60 min.) e submetidos a diferentes períodos de pré-cultivo (ausência, 24 ou 48h) em meio com 0,3 M de sacarose antes da criopreservação. Além desta técnica, foi avaliado o efeito da técnica clássica de vitrificação utilizando quatro tempos de exposição à solução de vitrificação (15, 30, 45 e 60 min.). Foram observadas diferenças significativas no tempo de exposição para a técnica de vitrificação e droplet vitrification.Para ambas as técnicas o tempo de imersão de 60 minutos promoveu uma retomada de crescimento superior a 70% nos ápices criopreservados. O pré-cultivo promoveu o aumento na sobrevivência dos explantes criopreservados. As técnicas de criopreservação de ápices caulinares por droplet vitrification e vitrificação mostraram-se viáveis para a conservação em tempo prolongado de mangabeira.

The aim of this study was to evaluate the efficiency of vitrification and droplet vitrification for the cryopreservation of Hancornia speciosa shoot tips. The shoot tips were subjected to four different periods of exposure (15, 30, 45 and 60 min.) to plant vitrification solution 2 (PVS2) before plunging into liquid nitrogen. We evaluated the regrowth of H. speciosa shoot tips that were cryopreserved by the classical vitrification technique and by the droplet vitrification technique. Shoot tips were submitted to different periods of pre-culture (absence, 24 or 48h) in a medium containing 0.3 M sucrose prior to cryopreservation. With a PVS2 exposure period of 60 min., significant differences were observed between the two techniques used in this study with respect to regrowth rates; however, both cryopreservation methods allowed for a shoot tip regrowth rate of over 70%. Use of a preculture step increased the regrowth rate of cryopreserved shoot tips. Shoot tip cryopreservation techniques employing vitrification and droplet vitrification proved to be viable for the long-term storage of mangaba tree samples.

O Brasil é um dos centros de origem do maracujazeiro e possui uma importante variabilidade natural do gênero Passiflora. Muitas espécies selvagens desse gênero são resistentes a algumas pragas e doenças e várias são consideradas medicinais. O objetivo deste trabalho foi induzir calos in vitro a partir de folhas de Passiflora gibertiipara a quantificação de vitamina C e fenóis totais. Uma vez que o balanço adequado entre auxina/citocinina promove a formação de calos e pode otimizar a produção de compostos secundários e vitaminas, calos foram induzidos em meio MS meia-força com uma combinação da auxina Picloram e da citocinina Cinetina, todos na ausência de luz. O teor de vitamina C e de fenóis totais foi quantificado por métodos colorimétricos após diferentes períodos de cultivo dos calos. A indução de calos foi fortemente influenciada pela combinação de Picloram/Cinetina. Foi observado um alto teor de vitamina C (94,8mg 100g-1) durante o período de indução de calos em MS+4,14µM de Picloram+0,207µM de Cinetina. Altas concentrações de Picloram/Cinetina conduzem a um aumento no teor de vitamina C após três subcultivos. Quanto maior a concentração de Picloram (8,28µM)/Cinetina (0,828µM), maior é a produção de fenóis (66mg de ácido tânico 100g-1 de calos) totais durante o período de indução de calos.

Brazil is one of the center of origin of passion fruit and has an important natural variability of the genus Passiflora. Several wild species of this genus are resistant to some pests and diseases and many are considered as medicinal. The aim of this research was to induce callus from in vitro Passiflora gibertiileaf explants for quantification of vitamin C and total phenols. Once the appropriate auxin/cytokine balance promotes callus formation and may optimize the production of secondary compounds and vitamins, calli were induced using a half-strength MS medium with a combination of the auxin Picloran (PIC) and the cytokine Kinetin (KIN). The vitamin C and total phenols were quantified by colorimetric methods from calli after different culture periods. The calli induction was strongly dependent of the combination PIC/KIN. It was observed high vitamin C content (94.8mg 100g-1) during the calli induction period in MS+4.14µM PIC+ 0.207µM KIN. Higher PIC/KIN concentrations promoted an increase in the vitamin C content after three subcultures. The higher PIC (8.28µM)/KIN (0.828µM) concentration was the higher was the total phenols production (66mg tannic acid 100g-1of fresh callus) during the calli induction period.
