Abstract Objectives To determine the concentration of calcium, iron, manganese and zinc ions after the application of chelator to Enterococcus faecalis biofilms. Material and Methods Fifty bovine maxillary central incisors were prepared and inoculated with E. faecalis for 60 days. The following were used as irrigation solutions: 17% EDTA (pH 3, 7 and 10), 2.5% sodium hypochlorite (NaOCl) combined with 17% EDTA (pH 3, 7 and 10), distilled water (pH 3, 7 and 10), and 2.5% NaOCl. Each solution was kept in the root canal for five minutes. Fifteen uncontaminated root canals were irrigated with 17% EDTA (pH 3, 7 and 10). Six teeth were used as bacterial control. The number of calcium, iron, manganese and zinc ions was determined using flame atomic absorption spectrometry. Mean ± standard deviation (SD) values were used for descriptive statistics. Results Calcium chelation using 17% EDTA at pH 7 was higher than at pH 3 and 10, regardless of whether bacterial biofilm was present. The highest concentration of iron occurred at pH 3 in the presence of bacterial biofilm. The highest concentration of manganese found was 2.5% NaOCl and 17% EDTA at pH 7 in the presence of bacterial biofilm. Zinc levels were not detectable. Conclusions The pH of chelating agents affected the removal of calcium, iron, and manganese ions. The concentration of iron ions in root canals with bacterial biofilm was higher after the use of 17% EDTA at pH 3 than after the use of the other solutions at all pH levels.

O objetivo deste estudo foi detectar a presença de istmos em molares superiores e inferiores, e avaliar a frequência usando dinâmica de navegação em imagens de tomografia computadorizada de feixe cônico (TCFC). Duzentos molares superiores e inferiores humanos foram usados em ensaio ex vivo. Uma amostra consecutiva de duzentos molares (superiores e inferiores, primeiros e segundos) foi selecionada a partir de exames de TCFC. Os istmos foram detectados a partir do orifício de entrada da cavidade pulpar em direção ao ápice, de acordo com o início e o fim, nas categorias: 1. início e término no terço cervical; 2. início no terço cervical e término no terço médio; 3. início no terço cervical e término no terço apical; 4. início e fim no terço médio; 5. início no terço médio e término no terço apical; 6. início e término no terço apical ; 7. ausência de istmo radicular. Os exames de TCFC foram obtidos em diferentes planos com estratégias de navegação em cortes axiais de 0,5 mm/0.5 mm de coronal para a direção apical. As frequências de istmo radicular foram analisadas de acordo com o terço da raiz e avaliadas pelo teste do qui-quadrado. O nível de significância estabelecido foi de 5%. A presença de istmo radicular em molares superiores foi de 86% em ensaio ex vivo e 62% em ensaio in vivo, enquanto que, em molares inferiores foram observados 70% em ensaio ex vivo e 72% em ensaio vivo. A frequência radicular de istmo foi elevada em ambos os modelos de estudo. A dinâmica da estratégia de navegação em imagens de TCFC é precisa para detectar a localização do istmo radicular.

The aim of this study was to detect root isthmuses in maxillary and mandibular molars and evaluate their frequencies using map-reading dynamics in CBCT images. Two hundred extracted human maxillary and mandibular molars were used in ex vivo assay. A consecutive sample of two hundred maxillary and mandibular molars (first and second) was selected from CBCT exams. The isthmuses were detected from the pulp orifice to the apex and were recorded according to their beginning and their end, into categories: 1. begin and end in cervical third; 2. beginning in cervical third and end in middle third; 3. beginning in cervical third and end in apical third; 4. beginning and end in middle third; 5. begin in middle third and end in apical third; 6. beginning and end in apical third; 7. no isthmus. The scans were obtained in different planes with map-reading in axial slices of 0.5 mm/0.5 mm involved the coronal to apical direction. The frequencies of isthmus were analyzed according to the level of root and evaluated by Chi-square test. The level of significance was set at α=0.05. The presence of isthmus detected in maxillary molars was 86% in ex vivo assay and 62% in vivo assay, whereas in mandibular molars was observed 70% in ex vivo assay and 72% in vivo assay. The frequency of isthmus was high in both study models. The map-reading dynamics in CBCT images was found to be precise to detect the localization of isthmus.

Este estudo avaliou in vitro a influência da termociclagem (TC) e do armazenamento em água (AA) na resistência ao cisalhamento de resina composta ao esmalte de dentes decíduos preparados com broca convencional e laser Er:YAG. As superfícies de trabalho foram obtidas de 48 molares decíduos e divididas aleatoriamente em dois grupos (n=24), de acordo com o tipo de preparo cavitário: A - preparo com broca; B - irradiação com laser Er:YAG. Os espécimes foram restaurados com um sistema adesivo etch-and-rinse e resina composta. Cada grupo foi dividido em 4 subgrupos (n=6) de acordo com o tempo de armazenamento em água (AA) e o número de termociclagens (TCs): I - 24 h AA/0 TCs, II - 7 dias AA/500 TCs; III - 1 mês AA/2000 TCs; IV - 6 meses AA/12000 TCs. O teste de cisalhamento foi realizado em máquina de ensaio universal a uma velocidade de 0,5 mm/min. Os dados foram analisados estatisticamente pelo teste ANOVA a dois critérios e teste de Tukey. As médias de resistência ao cisalhamento (D.P.), em MPa, foram: AI: 17,45 (2,03), AII: 16,38 (1,49), AIII: 6,88 (0,66), AIV: 7,77 (1,53), BI: 12,32 (0,99), BII: 15,37 (2,24), BIII: 15,05 (2,01) e BIV-5,51 (1,01). O tempo de armazenamento em água quanto a termociclagem influenciou significativamente os valores de resistência ao cisalhamento só para o grupo BIV (p<0,05). AI apresentou o maior valor de SBS, que foi estatisticamente semelhantes aos de AII, BII e BIII. Em conclusão, a adesão de um sistema adesivo etch-and-rinse ao esmalte de dente decíduo irradiado com Er: YAG foi afetada pelos métodos empregados para simulação da degradação da interface adesiva somente quando armazenamento em água por 6 meses e 12.000 ciclos de termociclagem foram empregados.

This study evaluated in vitro the influence of thermocycling and water storage (WS) on the shear bond strength (SBS) of composite resin in cavities prepared in primary tooth enamel with conventional bur or Er:YAG laser. The test surfaces were obtained from 48 primary molars and randomly assigned to 2 groups (n=24), according to cavity preparation: A: bur-preparation and B: Er:YAG laser irradiation. The specimens were restored with an etch-and-rinse adhesive system and composite resin. Each group was divided into 4 subgroups (n=6) according to WS duration and number of thermal cycles (TCs): I: 24 h WS/no thermocycling; II: 7 days WS/500 TCs; III: 1 month WS/2,000 TCs; IV: 6 months WS/12,000 TCs. The specimens were tested to failure in shear strength at a crosshead speed of 0.5 mm/min. Data were analyzed statistically by two-way ANOVA and Tukey's test. SBS means (S.D.) in MPa were: AI: 17.45 (2.03), AII:16.38 (1.49), AIII: 6.88 (0.66), AIV: 7.77 (1.53), BI: 12.32 (0.99), BII: 15.37 (2.24), BIII: 15.05 (2.01) and BIV-5.51 (1.01). WS duration and number of TCs influenced significantly the SBS values only for BIV (p<0.05). AI presented the highest SBS value, which was statistically similar to those of AII, BII and BIII. In conclusion, the adhesion of an etch-and-rinse adhesive to Er:YAG laser-irradiated primary tooth enamel was affected by the methods used to simulate degradation of the adhesive interface only when 6 months WS/12,000 TCs were employed.

OBJECTIVE: This ex vivo study evaluated the effect of pre-flaring and file size on the accuracy of the Root ZX and Novapex electronic apex locators (EALs). MATERIAL AND METHODS: The actual working length (WL) was set 1 mm short of the apical foramen in the palatal root canals of 24 extracted maxillary molars. The teeth were embedded in an alginate mold, and two examiners performed the electronic measurements using #10, #15, and #20 K-files. The files were inserted into the root canals until the "0.0" or "APEX" signals were observed on the LED or display screens for the Novapex and Root ZX, respectively, retracting to the 1.0 mark. The measurements were repeated after the preflaring using the S1 and SX Pro-Taper instruments. Two measurements were performed for each condition and the means were used. Intra-class correlation coefficients (ICCs) were calculated to verify the intra- and inter-examiner agreement. The mean differences between the WL and electronic length values were analyzed by the three-way ANOVA test (p<0.05). RESULTS: ICCs were high (>0.8) and the results demonstrated a similar accuracy for both EALs (p>0.05). Statistically significant accurate measurements were verified in the pre-flared canals, except for the Novapex using a #20 K-file. CONCLUSIONS: The tested EALs showed acceptable accuracy, whereas the pre-flaring procedure revealed a more significant effect than the used file size.

Os autores estudaram a atividade antimicrobiana do cimento de Grossman e de seus componentes sobre 13 diferentes cepas pelo método de difusão de poço em camada dupla. Os resultados revelaram que o cimento de Grossman apresentou atividade antimicrobiana contra todas as cepas utilizadas. Dos componentes do cimento, o tetraborato de sódio foi o que apresentou maior atividade antimicrobiana, tanto por tipo como por tamanho do halo e aro de inibição. O pó do cimento, o breu e o eugenol apresentaram atividades semelhantes, sendo que eles não tiveram ação sobre P. aeruginosa e C. albicans e, dos três componentes, somente o eugenol teve ação sobre E coli. O óxido de zinco somente teve ação sobre S. sobrinus e E. coli. O sulfato de bário e o subcarbonato de bismuto não tiveram nenhuma ação antimicrobiana.

The antimicrobial activity of Grossman’s sealer and its components was evaluated on 13 different strains using the double layer well-diffusion method. Results revealed that Grossman’s sealer presented antimicrobial activity against all the tested strains. Among the components of the cement, sodium tetraborate presented the greatest antimicrobial activity, both in type and diameter of the halo and ring of inhibition. Sealer powder, rosin, and eugenol presented similar activity, with no effect on P. aeruginosa and C. albicans. Among these, only eugenol had an effect on E. coli. Zinc oxide was only active against S. sobrinus and E. coli. Barium sulfate and bismuth subcarbonate did not show any antimicrobial effect.

As variações da anatomia interna de cada canal radicular podem interferir no sucesso da terapêutica endodôntica devido ao fato de que em canais radiculares achatados, pode persistir remanescentes teciduais em istmos, reentrâncias e ramificações dificultando a execução das técnicas de instrumentação. O sucesso do tratamento dos canais radiculares depende fundamentalmente do preparo da forma, limpeza, desinfecção e obturação. Poucos trabalhos avaliaram a limpeza dos canais radiculares após a instrumentação rotatória. O presente trabalho verificou a qualidade de limpeza dos canais radiculares, por meio da microscopia óptica, promovida pela técnica de instrumentação rotatória associada ao hipoclorito de sódio 0,5%, HCT20 e clorexidina, em canais achatados no sentido mésio-distal. Doze incisivos centrais inferiores humanos foram divididos aleatoriamente em três grupos para que fossem instrumentados com instrumetação rotatória (ProFile .04). A análise estatística evidenciou que os valores da porcentagem de limpeza para as diferentes soluções irrigantes foram estatisticamente diferentes entre si. Comparações duas a duas permitiram dispor as soluções irrigantes em ordem crescente de efetividade na limpeza, sendo: hipoclorito de sódio a 0,5% > clorexidina > HCT20.

Variations in the internal anatomy of root canals may interfere with the success of root canal because tissue remnants may remain in the isthmus, re-entrances and ramifications of flattened root canals making instrumentation more difficult. Successful root canal treatment depends fundamentally on shaping, cleaning, disinfection and obturation. This study verified the quality of cleaning of ProFile .04 rotary technique associated with different irrigating solutions: 0.5% sodium hypochlorite, calcium hydroxide + Tergentol and 2% chlorhexidine in canals with mesio-distal flattening. Twelve human mandibular central incisors were randomly divided into 3 groups and instrumented up to ProFile 0.4 #35 file after cervical preparation, and processed histologically. After optical microscopic evaluation, statistical analysis showed that the percentage of cleaning of the three irrigating solutions was statistically different. Two-by-two comparisons classified the irrigating solutions in increasing order of cleaning: calcium hydroxide + Tergentol < chlorhexidine < 0.5% sodium hypochlorite.
