Abstract Obtaining transgenic sugarcane lines in vitro is a laborious, expensive, and time-consuming process. To minimize these disadvantages, the presence of transgenes must be checked as soon as possible. Here, we describe a method for direct PCR amplification of DNA from sugarcane plantlets, using smaller quantities of vegetal material, low-cost lab reagents, and rapid processing. By using this methodology, we were able to identify 100% of the transgenic lines generated. ABSTRACT The propagation of sugarcane (Saccharum spp.) Requires seed with physiological and phytosanitary quality. The work was carried out with the objective of establishing a bank of sugarcane mother plants under semi-controlled conditions to guarantee the efficiency of in vitro propagation with plants free of Xanthomonas albilineans. A greenhouse was modified and the structure was remodeled, a Sarán shade mesh (50%) and a sprinkler irrigation system were placed. The plants were grown in plastic containers (20 m3 volume) with a mixture of cachaça compost and zeolite 3:1. Stems of cultivars C98-357, C97-445 and C86-156, without symptoms of leaf scald and with six months of culture, were cut into segments with a bud. Hydrothermal treatment was applied and subsequently the biostimulant VIUSID Agro® (0.8 ml l-1) before the buds planting. At three months of culture, a serological diagnosis was made to detect the presence of X. albilineans. The negative plants were treated with biostimulants and the response of the in vitro establishment was evaluated. Adequate plant growth was achieved under the conditions described. The establishment of a bank of sugarcane mother plants under semi-controlled conditions and its management guaranteed that microbial contamination and phenolic oxidation decreased in the in vitro establishment phase. The effectiveness of the hydrothermal treatment combined with the semi-controlled conditions and the management of the plants make it possible to reduce the incidence of X. albilineans.

RESUMEN La propagación de caña de caña de azúcar (Saccharum spp.) requiere de semilla con calidad fisiológica y fitosanitaria. El trabajo se realizó con el objetivo de establecer un banco de plantas madre de caña de azúcar en condiciones semicontroladas para garantizar la eficacia de la propagación in vitro con plantas libres de Xanthomonas albilineans. Se modificó una casa de cultivo y se remodeló la instalación, se colocó malla de sombra Sarán (50%) y un sistema de riego por aspersores. El cultivo de las plantas se realizó en recipientes de plástico (volumen de 20 m3) con una mezcla de compost de cachaza y zeolita 3:1. Tallos sin síntomas de escaldadura foliar y con seis meses de cultivo de los cultivares C98-357, C97-445, y C86-156 se cortaron en segmentos con una yema. Se aplicó tratamiento hidrotérmico y posteriormente el bioestimulante VIUSID Agro® (0.8 ml l-1) antes de la plantación de las yemas. A los tres meses de cultivo se realizó un diagnóstico serológico para detectar la presencia de X. albilineans. Las plantas negativas se trataron con productos bioestimulantes y se evaluó la respuesta del establecimiento in vitro. En las condiciones descritas se logró el crecimiento adecuado de las plantas. El establecimiento de un banco de plantas madre de caña de azúcar en condiciones semicontroladas y su manejo garantizaron que disminuyera la contaminación microbiana y la oxidación fenólica en la fase de establecimiento in vitro. La efectividad del tratamiento hidrotérmico combinado con las condiciones semicontroladas y el manejo de las plantas permiten disminuir la incidencia de X. albilineans.

Abstract: A prerequisite for commercial release of a transgenic crop variety in Argentina and other countries in the world is the identification and sequence analysis of the transgene(s) insertion site in the plant genome. Southern blot analysis showed a high number of transgene inserts in the glyphosate-resistant transgenic sugarcane line TUC 87-3RG. By using a targeted sequence enrichment approach, coupled with second-generation sequencing (SGS) and bioinformatic analyses, we were able to identify and subsequently analyze all flanking sequences for these transgene insertions in the sugarcane genome. PCR reactions confirmed 18 out of 29 candidate contigs as junction sequences that did not match essential genes in published sugarcane genomes. The methodology outlined here represents an efficient strategy for characterization of flanking sequences corresponding to multiple inserts of a transgene in a large, complex, and not fully sequenced plant genome such as that of sugarcane.

The Sugarcane Breeding Program of Agroindustrial Experimental Station Obispo Colombres continuously generates new varieties of sugarcane, especially for the province of Tucumán, Argentina. Accurate varietal identification is essential to protect the intellectual property rights of new varieties. In this work, Target Region Amplified Polymorphism (TRAP) and Simple Sequence Repeat (SSR) molecular markers were used, in combination with phenotypic markers proposed by the International Union for Plant Variety Protection (UPOV), to characterize three sugarcane varieties recently released by the local breeding program. Five combinations of TRAP primers and five pairs of SSR primers were used. The amplification products were separated by electrophoresis in polyacrylamide gels under denaturing conditions. For the phenotypic characterization, the nine mandatory UPOV descriptors were used. The bands and morphological data were transformed into binary matrixes of 0 and 1, the similarity was calculated and the dendrograms were generated by the Unweighted pair-group method using arithmetic averages (UPGMA) analysis. Both molecular markers clearly revealed the phylogenetic relationship among genotypes. The morphological characterization correctly differentiated the varieties analyzed; only revealing the external similarity between genotypes. The entire characterization allowed us to unambiguously differentiate the recently released cultivars to register and protect plant breeders rights.

El Programa de Mejoramiento Genético de la Caña de Azúcar (PMGCA) de la Estación Experimental Agroindustrial Obispo Colombres (EEAOC) genera continuamente nuevas variedades de caña de azúcar, especialmente para la provincia de Tucumán, Argentina. Una precisa identificación varietal es esencial para proteger los derechos de propiedad intelectual de los nuevos cultivares. Para caracterizar tres cultivares de caña de azúcar recientemente liberados por el PMGCA, en este trabajo se utilizaron marcadores moleculares TRAP y SSR (de sus siglas en inglés deTarget Region Amplified Polymorphism ySimple Sequence Repeat , respectivamente), en combinación con marcadores fenotípicos propuestos por la Unión Internacional para la Protección de las Obtenciones Vegetales (UPOV). Se utilizaron cinco combinaciones de cebadores TRAP y cinco pares de cebadores SSR. Los productos de amplificación se separaron mediante electroforesis en geles de poliacrilamida en condiciones desnaturalizantes. Para la caracterización fenotípica se utilizaron los nueve descriptores obligatorios de la UPOV. Las bandas y los datos morfológicos se transformaron en matrices binarias de 0 y 1, se calculó la similitud y se generaron los dendrogramas utilizando el análisis UPGMA (de sus siglas en inglés deUnweighted Pair-Group Method using Arithmetic averages ). Ambos marcadores moleculares revelaron claramente la relación filogenética entre genotipos. La caracterización morfológica permitió diferenciar correctamente las variedades analizadas, revelando únicamente la semejanza externa entre los genotipos. La caracterización completa permitió diferenciar inequívocamente los cultivares recientemente liberados para registrarlos y proteger los derechos de los mejoradores.

The Vitroplantas Project of sugarcane of Estación Experimental Agroindustrial Obispo Colombres (EEAOC) was started in 2001. Its main goal is to provide sugarcane producers with seed cane of high phytosanitary quality and guaran-teed genetic purity. Since its beginning, improvements in the process such as molecular techniques for disease diagnosis and assessment of genetic purity, have been incorporated. Until 2004 the vitroplant sanitary evaluation for two systemic diseases was carried out by serological techniques. However, since 2005, molecular diagnoses based on PCR for the most regionally important systemic diseases of sugarcane were incorporated. In 2014, the genetic identity analysis of vitroplants undertaken since 2007 with AFLP was replaced by TRAP since it was cheaper and revealed similar polymorphisms. Recently, a Quality Management System, which allows effective administration and increases the efficiency at each step of the micropropagation process, was implemented. Over the 2001-2019 period, 13 commercial and promising sugarcane clones were micropropagated, reaching a total of 1,210,519 Vitroplants. Currently, around 70% of the area under cultivation in Tucumán is occupied by high quality sugarcane seeds produced by the Vitroplantas Project, resulting in lower disease incidence and a gradual increase in yields in commercial fields. This Project has constantly looked at improving the process to obtain a product able to satisfy sugarcane producersdemands.

El Proyecto Vitroplantas de caña de azúcar de la Estación Experimental Agroindustrial Obispo Colombres (EEAOC) se inició en 2001, con el objetivo principal de proporcionar a los productores caña semilla de alta calidad fitosanitaria y pureza genética garantizada. Desde el inicio se incorporaron mejoras en el proceso, como técnicas moleculares para el diagnóstico de enfermedades y evaluación de la pureza genética. Hasta el 2004, la evaluación sanitaria de las vitroplantas para dos enfermedades sistémicas se llevaba a cabo mediante técnicas serológicas. A partir del 2005 se incorporó el diagnóstico molecular basado en PCR para las enfermedades sistémicas más importantes de la región. En 2014, el análisis de identidad genética de vitroplantas realizado desde 2007 por AFLP fue reemplazado por TRAP, que es menos costoso y revela un polimorfismo similar. Recientemente, se implementó un Sistema de Gestión de Calidad que permitió administrar y aumentar la eficiencia en cada paso del proceso de micropropagación. En el período 2001-2019 se micropropagaron 13 variedades comerciales y clones promisorios de caña de azúcar, alcanzando un total de 1.210.519 Vitroplantas. Actualmente, alrededor del 70% de la superficie plantada en Tucumán se cultiva con caña semilla de alta calidad producida por el Proyecto Vitroplantas, lo que se tradujo en una menor incidencia de enfermedades y un aumento gradual de los rendimientos en los campos comerciales de caña de azúcar. Desde el inicio, el Proyecto Vitroplantas ha buscado constantemente mejorar el proceso a fin de obtener un producto capaz de satisfacer la demanda de los productores de caña de azúcar.

Molecular markers could be useful to determine the occurrence of genetic changes during the genetic transformation process. Among them, functional markers randomly distributed throughout the genome may reflect genetic variations of direct interest. For this reason, the objective of this work was to determine similarity to parental genotype of sugarcane of different transgenic events by using Target Region Amplified Polymorphism (TRAP) markers. DNAs from transgenic events, wild type genotypes and other sugarcane varieties were characterized by seven to nine combinations of TRAP primers. Molecular markers were separated by electrophoresis on polyacrylamide denaturing gels in a DNA Analyzer (Li-cor). Amplified fragments were transformed into either a 0/1 matrix. Similarity was calculated by using Jaccard coefficient and dendrograms were generated. At first instance, markers confirmed the preliminary phenotypic evaluations of herbicide resistant events of RA 87-3 variety since transformed events with close growth resemblance to its parental variety exhibited none or only minor genetic changes whereas events with growth aberrations showed a significant degree of polymorphism. The incorporation of other genotypes allowed validating the technique assuring that a significant number of polymorphic bands were analyzed. Considering the accuracy of the methodology, it was routinely applied to characterize transgenic events of LCP 85-384, TUCCP 77-42, TUC 95-10 and TUC 03-12 at early stages of the process. In conclusion, the use of TRAP markers is a quick and recommendable strategy to characterize and identify transgenic events genetically close to their parental genotypes. This makes possible the selection at the first stages of evaluation of those of the most valuable events to carry out field tests.

Los marcadores moleculares son útiles para determinar la presencia de cambios genéticos durante el proceso de transformación. Entre ellos, los marcadores funcionales distribuidos al azar en todo el genoma pueden reflejar variaciones genéticas de interés directo. Por este motivo, el objetivo de este trabajo fue determinar la similitud con la variedad de caña de azúcar sin transformar de diferentes eventos transgénicos mediante el uso de marcadores TRAP (" Target Region Amplified Polymorphism"). Para ello, ADNs provenientes de los eventos transgénicos, los genotipos sin transformar y de otras variedades de caña de azúcar se caracterizaron mediante amplificación con siete a nueve combinaciones de cebadores TRAP. Los marcadores se separaron mediante electroforesis en geles de poliacrilamida en condiciones desnaturalizantes en el equipo Li-cor DNA Analyzer. Los fragmentos amplificados fueron transformados en matrices binarias de 0/1, utilizadas para calcular el coeficiente de Jaccard y construir árboles de similitud. En primer lugar, los marcadores permitieron confirmar las evaluaciones fenotípicas preliminares de eventos resistentes al herbicida glifosato de la variedad RA 87-3, dado que aquellos eventos fenotípicamente similares a la variedad sin transformar no mostraron cambios genéticos o solo algunos menores, mientras que eventos con aberraciones de crecimiento presentaron un alto nivel de polimorfismo. La incorporación en el análisis de otros genotipos permitió validar internamente la técnica asegurando el análisis de un número significativo de bandas polimórficas. Considerando la precisión de esta metodología, se la aplicó de rutina para caracterizar eventos transgénicos de las variedades LCP 85-384, TUCCP 77-42, TUC 95-10 y TUC 03-12 en las primeras etapas del proceso de evaluación. En conclusión, el uso de marcadores TRAP constituye una estrategia rápida y recomendable para caracterizar e identificar eventos transgénicos genéticamente próximos a su genotipo sin transformar. Esto posibilita la selección en las primeras etapas de evaluación de aquellos eventos más adecuados para realizar los ensayos a campo.

Sugarcane borer Diatraea saccharalis (Fabricius) (Lepidoptera: Crambidae) is the most important sugarcane pest in Tucumán, Argentina. Older larvae bore into the stalks, facilitating microorganism colonization which indirectly reduces yield and quality of sugar. The aim of the present work was to optimize a sugarcane plant phenotyping method with D. saccharalis under controlled conditions to evaluate different strategies to manage the pest. Different numbers of neonate larvae were placed in the leaf whorl of sugarcane seedling (2-months-old) of cultivars TUC 95-10, TUC 03-12 and LCP 85-384. On the other hand, on single plants 6-month old of TUC 95-10, 10 larvae of several instars were added and the number of perforations and damaged sheath, the tunnel total length and the dead heart symptom were evaluated. In in vitro tests, immature leaf roll disks cut from seedlings were inoculated with two neonate larvae and the larval survival percentage was determined. Each assay was repeated twice or three times with 5 -10 replicates per treatment and conducted under controlled conditions (28-30ºC; 50-70% RH). In seedlings the symptom of the dead heart was observed in all the treatments with different larval number. In the case of the 6-month-old plants, damage in the sheath and stem were observed when infested with L2, L3 and L4 instars, whereas neonate larvae only produced sheath damage. In the laboratory tests, the survival percentage of larvae obtained was high. The results suggest that several methodologies were optimized to evaluate different types of plant material, which are available to study potential strategies for sugarcane borer management.

El barrenador de caña de azúcar Diatraea saccharalis (Fabricius) (Lepidoptera: Crambidae) es la plaga más importante del cultivo en Tucumán, Argentina. Los estadios larvales más avanzados perforan los tallos facilitando la colonización de microorganismos que reducen indirectamente el rendimiento y la calidad del azúcar. El objetivo del presente trabajo fue optimizar sistemas de fenotipado de D. saccharalis en caña de azúcar, en condiciones controladas, con el fin de evaluar diferentes estrategias de manejo de la plaga. En primer lugar, diferentes números de larvas neonatas fueron colocadas en el cogollo de plantines de caña de azúcar de dos meses de edad de las variedades TUC 95-10, TUC 03-12 y LCP 85-384. Por otro lado, en plantas individuales de seis meses de edad de TUC 95-10 se inocularon 10 larvas de diferentes estadios y se determinó el número de perforaciones y de vainas dañadas, la longitud total del túnel y el síntoma del corazón muerto. En ensayos in vitro se inocularon dos larvas neonatas en ápices caulinares extraídos de plantines y se determinó el porcentaje de supervivencia de larvas. Todos los ensayos se realizaron en condiciones controladas (28-30ºC; 50-70% de HR), con dos o tres repeticiones y con 5 - 10 unidades experimentales por tratamiento. En plantines se observó el síntoma del corazón muerto en todos los tratamientos con diferente número de larvas neonatas. Las plantas de seis meses de edad presentaron daños en vaina y tallo cuando se infestaron con los estadios larvales L2, L3 y L4, mientras que las larvas neonatas solo produjeron daño en la vaina. En los ensayos de laboratorio el porcentaje de supervivencia de larvas obtenido fue elevado. Los resultados sugieren que diferentes sistemas de fenotipado empleando material vegetal de distinta edad fueron optimizados y están disponibles para evaluar potenciales estrategias de manejo del barrenador.

A previous development of transgenic glyphosate-resistant sugarcane suitable for commercial release was carried out through genetic transformation of cultivar RA 87-3. During the time elapsed to develop this technology, new commercial cultivars produced by the local breeding program have been adopted by farmers. The complex genetics of modern sugarcane cultivars, which are interspecific hybrids, highly polyploid and frequently aneuploid, make the introgression by backcrossing of the transgene into other varieties extremely difficult. Direct transformation of new commercial cultivars or promising clones at final stages of a breeding scheme could greatly improve the development and adoption of transgenic sugarcane by both farmers and millers. The aim of this study was to obtain glyphosate-resistant transgenic events of the two recently released cultivars, TUC 95- 10 and TUC 03-12, through the introduction via microprojectile bombardment of plasmids harbouring the epsps and nptII genes. A total of 23 and 8 independent bombardments experiments were carried out on TUC 95-10 and TUC 03-12, respectively. The stable transformation and integration of both epsps and nptII genes were determined by using PCR with specific primers. Transgenic events were evaluated for glyphosate tolerance under ex vitro conditions and for genetic similarity with donor plant by using target region amplification polymorphism (TRAP) molecular markers. Although plantlets of both varieties regenerated from calli, TUC 95-10 showed a low tissue culture-response since only two events were obtained, whilst a total of 22 plantlets regenerated for TUC 03-12. One line of TUC 95-10 and three lines of TUC 03-12 were PCR-positive for both genes and different levels of herbicide-tolerance were observed. Genetic analyses of 213 TRAP loci from transgenic lines derived from TUC 03-12 indicate that two events show 99.30% of similarity with the non-transformed TUC 03-12 control and the third had 98.7% similarity. These are encouraging results as, based on our previous experience, the molecular-marker data suggest that the events are practically identical to their parental cultivar, and are suitable for future comparative field testing.

El previo desarrollo de un evento transgénico de caña de azúcar resistente al glifosato aceptada para su liberación comercial fue llevado a cabo a partir de la transformación genética de la variedad RA 87-3. Durante el tiempo transcurrido para desarrollar esta tecnología, nuevas variedades comerciales liberadas por el programa de mejoramiento local están siendo adoptadas por los agricultores. La complejidad genómica de las variedades modernas de caña de azúcar, caracterizadas por ser híbridos interespecíficos, con un alto grado de poliploidía y frecuente aneuploidía, hace prácticamente imposible la introgresión del transgén por retrocruzamientos en otras variedades. La transformación directa de nuevas variedades comerciales o clones promisorios que se encuentren en etapas finales de un esquema de mejoramiento, podrían mejorar enormemente el desarrollo y la adopción de la caña de azúcar transgénica tanto por los agricultores como por los industriales. El objetivo de este estudio fue obtener eventos transgénicos resistentes al glifosato de las variedades TUC 95-10 y TUC 03-12 a partir del bombardeo de microproyectiles recubiertos por plásmidos que contengan los genes epsps y nptII. Se realizaron experimentos independientes con un total de 23 y 8 bombardeos de las variedades TUC 95-10 y TUC 03-12, respectivamente. La transformación e integración estable de los genes epsps y nptII se determinaron mediante el uso de la técnica de PCR con cebadores específicos. Los eventos transgénicos fueron evaluados en condiciones ex vitro para determinar la tolerancia al glifosato, mientras que también se analizó su similitud genética con la variedad que les dio origen mediante marcadores moleculares TRAP. Aunque se obtuvieron plántulas de ambas variedades, la variedad TUC 95-10 mostró una menor respuesta al cultivo de tejido, ya que solo se obtuvieron dos eventos, mientras que para la variedad TUC 03-12 se regeneraron un total de 22 plántulas. Un evento regenerado a partir de TUC 95-10 y tres eventos derivados de TUC 03-12 fueron PCR positivos para ambos genes, observándose diferentes niveles de tolerancia al herbicida. El análisis genético de 213 loci TRAP en las 3 líneas transgénicas derivadas de TUC 03-12 indican que dos eventos comparten un 99,30% de similitud con el control no transformado, mientras que el evento restante presenta un 98,7%. Estos resultados son alentadores, ya que basados en conocimientos previos, los marcadores moleculares indicarían que los eventos obtenidos son prácticamente idénticos a su variedad donante, con lo cual serían adecuados para conducir ensayos comparativos adicionales a campo.

The commercial variety of sugarcane RA 87-3 was transformed by biobalistic with a genetic construct that carries the epsps gene of the CP4 strain of Agrobacterium tumefaciens, and with the nptII gene that confer resistance to glyphosate and resistance to kanamycin / geneticin, respectively. Transformed lines were multiplied in greenhouse and resistance to the herbicide was evaluated using different concentrations of glyphosate (3, 4 and 8 l / ha). Resistant lines to the herbicide (RH) were evaluated in the field to confirm resistance to glyphosate and to carry out a preliminary analysis of their behavior with respect to the parental cultivar. All transformed lines maintained resistance to the herbicide but many showed phenotypic changes. Ten RH lines that were very similar to the RA 87-3 variety were analyzed phenotypically and genetically using the 9 compulsory morphological markers proposed by The International Union for the Protection of New Varieties of Plants (UPOV) and with 339 molecular markers, respectively. Six lines of the previous ones presented minor morphological and genetic changes and were selected for field trials during two vegetative cycles (cane plant and ratoon 1), in two production areas of Argentina. The six RH lines presented agronomic, industrial and chemical composition characteristics indistinguishable from the parental cultivar. The stable inheritance of the CP4 epsps gene was confirmed by RT-qPCR and Southern blot assays in different clonal generations (growth cycles). Our studies confirmed the usefulness of genetic transformation as a complementary tool to classical improvement and highlight the advantage of the use of UPOV descriptors together with molecular markers for an early selection of transgenic events that have a high similarity with the parental genotype. Taking into account the results, one of the six lines evaluated was proposed for a possible commercial release, under the requirements of regulatory affairs offices (CONABIA, SENASA and DNMA).

La variedad comercial de caña de azúcar RA 87-3 se transformó mediante biobalística con una construcción genética que porta el gen epsps de la cepa CP4 de Agrobacterium tumefaciens, y el gen nptII que confieren resistencia a glifosato y resistencia a kanamicina/geneticina, respectivamente. Las líneas transformadas fueron multiplicadas en invernadero y se evaluó la resistencia al herbicida utilizando diferentes concentraciones de glifosato (3, 4 y 8 l/ha). Las líneas resistentes al herbicida (RH) se evaluaron en campo para confirmar la resistencia a glifosato (3 l/ha) y realizar un análisis preliminar del comportamiento de estas con respecto al cultivar parental. Todas las líneas transformadas mantuvieron la resistencia al herbicida pero muchas mostraron cambios fenotípicos. Las diez líneas RH que resultaron muy parecidas a la variedad RA 87-3 se analizaron fenotípica y genéticamente utilizando los nueve descriptores morfológicos obligatorios propuestos por la “International Union for the Protection of New Varieties of Plants” (UPOV) y con 339 marcadores moleculares, respectivamente. Seis líneas de las anteriores presentaron cambios morfológicos y genéticos menores y fueron seleccionadas para ensayos en campo durante dos ciclos vegetativos (caña planta y soca 1), en dos áreas de producción de Argentina. Las seis líneas RH presentaron características agronómicas, industriales y de composición química indistinguibles respecto del cultivar parental. La herencia estable del gen CP4 epsps fue confirmada mediante ensayos de RT-qPCR y Southern blot en diferentes generaciones clonales (caña planta y soca 1). Los estudios confirmaron la utilidad de la transformación genética como una herramienta complementaria a la mejora clásica, y destaca la ventaja del uso de los descriptores de UPOV junto con los marcadores moleculares para una selección temprana de eventos transgénicos que tengan un alto parecido con el genotipo parental. Tomando en cuenta los resultados, una de las seis líneas estudiadas se seleccionó para una posible liberación comercial, la cual debe ser sometida a evaluación por parte de los entes regulatorios (CONABIA, SENASA y DNMA).

Since 2001, the Estación Experimental Agroindustrial Obispo Colombres (EEAOC) has been working on the“Vitroplantas” Project. Ninety thousand sugarcane plantlets of commercial varieties are produced annually through in vitro meristem culture, in order to eliminate systemic diseases caused by bacteria and viruses. Sanitation of plant material is achieved through in vitro culture of apical meristems from donor plants, previously hot water treated and grown for three years under greenhouse conditions, with anti-aphid screens. Systemic diseases are evaluated in both meristem donor plants and micropropagated plantlets using different molecular diagnostic techniques. It is well known that in vitro plant tissue culture can produce somaclonal variations, which consist of genetic changes in cultured cells and tissues. In order to guarantee that seedlings propagated in vitro are identical to the parental variety, molecular markers that quantify and detect somaclonal variation are routinely applied. Thus, the aim of this project is to guarantee healthy and genetically pure plantlets for their plantation in the field. After in vitro micropropagation and testing, plantlets undergo an acclimatization process in an especially adapted greenhouse at the EEAOC. In order to avoid dehydration of plantlets, this process takes place in a greenhouse with very high relative humidity (RH=80%-100%) and low light intensity. After acclimatization, two other stages of conventional field propagation (Basic and Registered fields) are carried out before the seed cane is finally distributed among sugarcane growers. The implementation of the “Vitroplantas” Project has greatly improved the health and yields of sugarcane plantations in Tucumán, Argentina..

En el año 2001, la Estación Experimental Agroindustrial Obispo Colombres (EEAOC) puso en marcha el Proyecto Vitroplantas. Se producen anualmente 90.000 plantines saneados de variedades comerciales de caña de azúcar mediante cultivo in vitro de meristemas, con la finalidad de eliminar enfermedades sistémicas causadas por bacterias y virus. El saneamiento del material vegetal se consigue mediante el cultivo in vitro de meristemas apicales de plantas donadoras, previamente hidro-termotratadas y mantenidas durante tres años en invernadero en condiciones controladas. Las enfermedades sistémicas son evaluadas mediante diferentes técnicas de diagnóstico molecular, tanto en las plantas donadoras de meristemas como en los plantines micropropagados. Por otro lado, el cultivo in vitro de tejidos vegetales puede producir variaciones somaclonales, que consisten en cambios genéticos en las células y tejidos cultivados. Con el fin de garantizar que los plantines propagados in vitro sean idénticos a la variedad parental, se utiliza una metodología basada en marcadores moleculares para detectar y cuantificar la variación somaclonal. Es así que el objetivo de este proyecto es garantizar la sanidad y la pureza genética de los plantines micropropagados antes de su plantación a campo. Al finalizar el proceso de micropropagación in vitro, los plantines son sometidos a un proceso de aclimatación en un invernadero, especialmente adaptado para este fin. Para evitar la deshidratación de los plantines, este proceso se lleva a cabo en un ambiente con elevada humedad relativa (HR = 80%-100%) y baja intensidad de luz. Posteriormente, se llevan a cabo dos etapas de propagación convencional en campo (semilleros Básico y Registrados) antes de que la caña semilla se distribuya a los productores. La implementación del Proyecto Vitroplantas ha mejorado en gran medida el estado sanitario y los rendimientos de caña de azúcar en Tucumán, R. Argentina.

In 1994, a group whose aims were both scientific research and technological development was set up. This team was inspired by the increasing demand of the agro-industrial sector in the North West of Argentina. Besides financial considerations inherent to the productive process, social and environmental issues were borne in mind when research had to be carried out. The conceptual premises of scientific work were the study, preservation and a judicious use of biodiversity; equally important was the relentless search for phytosanitary management alternatives which were friendly to both humans and the environment. Thus, in 1997, the frst National Program of Strawberry Genetic Breeding in Argentina started at INSIBIO (CONICET-UNT), Tucumán. This Institute coordinated this program on a national level. Thanks to an agreement between INSIBIO and the Estación Experimental Agroindustrial Obispo Colombres (EEAOC), in 2002, the EEAOC's Biotechnology Department, a Unit Associated to INSIBIO, was founded. One of the main goals of this department was both to develop and adapt technologies to support institutional ongoing programs which intended not only to increase productivity but also to confer more sustainability to those agro-industries related to sugar and alcohol, soybean and citrus. and most importantly, a new scientific and technological team constituted by professionals working with other groups from different research centers nationwide carne into existence. Their common goal has been the improvement of agro-industrial productive processes in order to help build a society which is fair and equitable for both the present and the future.

En 1994 se conformó un grupo de investigación científica y desarrollo tecnológico guiado por la demanda del sector agroindustrial de la región del noroeste argentino, que además de las consideraciones económicas intrínsecas del proceso productivo, tuvo en cuenta para sus investigaciones, aspectos sociales y ambientales. Las premisas conceptuales del trabajo científico fueron el estudio, la preservación y el aprovechamiento de la biodiversidad, en conjunción con la búsqueda continua de alternativas de manejo fitosanitario lo menos reñidas posible con la salud humana y ambiental. Fue así que en 1997 se inició el primer Programa Nacional de Mejoramiento Genético de la Frutilla en Argentina, que desde el INSIBIO (CONICET-UNT) en Tucumán coordinó el trabajo de grupos en otras instituciones. A partir del año 2002 mediante un convenio entre el INSIBIO y la Estación Experimental Agroindustrial Obispo Colombres (EEAOC), se fundó la Sección Biotecnología de la EEAOC- Unidad Asociada al INSIBIO, con el principal objetivo de desarrollar y adaptar tecnologías para apoyar programas institucionales preexistentes, tendientes a incrementar la productividad con mayor sostenibilidad en las agroindustrias suero-alcoholera, sojera, y citrícola. Sin embargo, lo más importante es que se constituyó un equipo científico y tecnológico que trabaja en forma articulada con otros grupos de filiación diversa, pero que tienen el objetivo común de mejorar los procesos productivos en la actividad agroindustrial, para contribuir a generar sociedades más justas y solidarias, no sólo en el presente sino para las futuras generaciones.

In vitro culture of plant tissue can produce somaclonal variation, which consists of genetic modifications in cultured cells and tissues. This may constrain the use of this technique in massive micropopagation, especially if such change causes an agronomically relevant phenotypical modification. In this work, a methodology based on the comparison of AFLP (Amplified Fragment Length Polymorphism) molecular marker profiles was developed for detecting somaclonal variation in in vitro propagated sugarcane plants. To optimize AFLP technique application to sugarcane plants, six conventionally propagated genotypes and two types of samples (tender leaves and meristems) were used. Somaclonal variation was determined in micropropagated lines of these genotypes after six months of micropropagation. Molecular profile differentiation of the selected genotypes was achieved with 19 primer combinations. Differential profiles were detected in LCP85-384 and TUCCP77-42 micropropagated lines with 3 of the 19 primer combinations. This result demonstrated that the technique can be used to detect somaclonal variants and that there are different susceptibility levels to in vitro culture among genotypes. Therefore, micropropagation methodology was adjusted to each multiplied genotype so as to ensure genetic purity of in vitro propagated plants.

El cultivo in vitro de tejidos vegetales puede producir variación somaclonal, fenómeno que consiste en modificaciones genéticas en las células y tejidos cultivados. Esto puede limitar la aplicación de dicha técnica para la micropopagación masiva, especialmente si la variación provoca un cambio fenotípico de importancia agronómica. En este trabajo se optimizó una metodología basada en la comparación de perfiles de marcadores moleculares AFLP (del inglés "Amplified Fragment Length Polymorphism"), para la detección de la variación somaclonal en vitroplantas de caña de azúcar. Para la optimización de la técnica de AFLP en caña de azúcar, se utilizaron plantas de seis genotipos propagados convencionalmente y dos tipos de muestras: hojas tiernas y meristemas. La variación somaclonal fue evaluada en líneas de vitroplantas de los mismos genotipos al final del cultivo in vitro, luego de seis meses de micropropagación. Con las 19 combinaciones de cebadores utilizadas, se diferenciaron los perfiles moleculares de los seis genotipos. En los plantines micropropagados se detectaron perfiles diferenciales en las variedades LCP85-384 y TUCCP77-42 con 3 de las 19 combinaciones de cebadores utilizadas. Este resultado muestra la validez de la técnica para detectar variantes somaclonales, y deja en evidencia la diferencia de susceptibilidad de los genotipos al cultivo in vitro. Esto permitió ajustar la metodología de micropropagación para cada genotipo multiplicado y asegurar la pureza genética de cada vitroplanta.
