Resumo Este trabalho teve o objetivo de avaliar a resposta morfogênica da micropropagação de Aechmea miniata e Aechmea blanchetiana após diferentes tratamentos com citocinina e auxinas. O estabelecimento in vitro se deu a partir de sementes de plantas adultas, com aproximadamente 2 anos de idade. Após descontaminação as sementes foram inoculadas em meio MS. Após 30 dias da germinação, foram isolados segmentos caulinares com aproximadamente 5 mm, e em seguida, foram transferidos para meio MS suplementado com BAP nas concentrações 4,44; 8,88 ou 13,32 µM. Ao final de 225 dias de cultivo, foi avaliado o número de brotos / explante, altura de brotações, porcentagem de enraizamento, número de raízes e comprimento de raízes. Verificou-se que, para as duas espécies, a concentração 4,44 µM de BAP mostrou maior eficiência para a multiplicação. Para a etapa de enraizamento, os brotos obtidos in vitro foram inoculados em meio MS com metade da concentração salina de macronutrientes, acrescido com 1, 2 e 3 µM de AIB ou ANA. Aos 60 dias, verificou-se que as duas espécies apresentaram 100% de enraizamento em meio de cultura sem a adição de reguladores de crescimento. Após enraizamento as plantas foram transferidas para substrato, e aos 180 dias de cultivo foi verificado sobrevivência superior a 80%, independente da espécie.

Abstract The objective of this study was to evaluate the morphogenic response of Aechmea miniata and Aechmea blanchetiana after different treatments with cytokinins and auxins during micropropagation. In vitro cultures were established from seeds of adult plants, approximately 2 years old. After sterilization, the seeds were inoculated in MS médium. 30 days after germination, caulin segments of approximately 5 mm were isolated, then, transferred to MS medium supplemented with BAP at concentrations of 4.44, 8.88, and 13.32 µM. At the end of 225 days of culture, the number of buds/explants, height of shoots, percentage of rooting, number of roots, and length of roots were evaluated. It was found that 4.44 µM BAP induced relatively higher frequency of multiplication in both species. For rooting, the shoots obtained in vitro were inoculated in MS medium with half of the concentration of macronutrient salts, and 1, 2, and 3 µM AIB or ANA. At 60 days, the two species exhibited 100% rooting in the médium without the addition of growth regulators. After rooting the plants were transferred to substrate, and at 180 days of culture, the survival rate was more than 80%, independent of the species.

RESUMO Poincianella pyramidalis (Tul.) L.P. Queiroz, é uma espécie endêmica da Caatinga, se destaca pela importância nas atividades madeireira e medicinal. Este estudo teve como objetivo avaliar o efeito dos agentes osmóticos sacarose, sorbitol e manitol e o retardante de crescimento paclobutrazol (PBZ) na indução do crescimento mínimo in vitro de P. pyramidalis, visando o estabelecimento de técnicas alternativas para conservação ex situ da espécie. No primeiro experimento, plântulas com sete dias de idade foram inoculadas em meio WPM suplementado com quatro concentrações de sacarose (87,64, 131,46, 175,28 ou 219,10 mM) combinadas com 0,0 ou 87,64 mM de sorbitol ou manitol. No segundo experimento, essas plântulas foram inoculadas em meio WPM contendo diferentes concentrações (0,0, 1,5, 3,0, 4,5 ou 6,0 µM) de PBZ e suplementadas com 87,64 mM de sacarose. Para ambos os experimentos, foi avaliado a porcentagem de sobrevivência das plantas a cada 60 dias. Ao final de 240 dias, foram analisados o número de folhas verdes e folhas senescentes, comprimento da parte aérea e da maior raiz, e massa seca da parte aérea e da raiz. Os melhores resultados para manter a qualidade das plantas de P. pyramidalis conservadas in vitro foram obtidos em meio suplementado com 219,10 mM de sacarose isolada por até 240 dias sem subcultivo. Com relação ao PBZ, a concentração de 6,0 μM pode ser recomendada para conservação in vitro de P. pyramidalis por um período de 240 dias sem a necessidade de subcultivos.

ABSTRACT Poincianella pyramidalis (Tul.) L.P. Queiroz, is an arboreal species endemic to Brazil’s Caatinga biome and an important source of lumber and also of medicinal substances. This study aimed to evaluate the effect of the osmotic agents sucrose, sorbitol and mannitol and the growth retardant paclobutrazol (PBZ) on the induction of slow growth in vitro of P. pyramidalis, seeking to establish alternative techniques for ex situ conservation of the species. In the first experiment, plantlets with seven days of age were inoculated on Woody Plant Medium (WPM) supplemented with four concentrations of sucrose (87.64, 131.46, 175.28 or 219.10 mM) combined with 0.0 or 87.64 mM sorbitol or mannitol. In the second experiment, these plantlets were inoculated on WPM medium with 0.0, 1.5, 3.0, 4.5 or 6.0 µM PBZ and supplemented with 87.64 mM sucrose. For both experiments, the survival percentage of the plants was evaluated every 60 days, and at the end of 240 days, the following parameters were recorded: number of green leaves and senescent leaves; length of the aerial part and longest root; and dry mass of the aerial part and roots. The best results to maintain the quality of the P. pyramidalis plants conserved in vitro were obtained on medium supplemented with 219.10 mM sucrose for up to 240 days without subculture. With respect to PBZ, the concentration of 6.0 μM can be recommended for in vitro conservation of P. pyramidalis for 240 days without the need of subculturing.

O cultivo in vitro de embriões zigóticos é uma técnica promissora para se avançar no estudo do desenvolvimento embrionário e da quebra da dormência de sementes. Diante do exposto, objetivou-se, com este trabalho, avaliar o efeito dos reguladores vegetais 6-benzilaminopurina (BAP) e ácido naftalenoacético (ANA) no potencial morfogenético, in vitro, de embriões zigóticos de mulungu. Embriões zigóticos maduros, oriundos de sementes foram utilizados inteiros, ou seccionados em plúmula, região intermediária e radícula, sendo posteriormente inoculados em meio de cultura WPM, suplementado com combinações de BAP (0,0; 2,0; 4,0; 8,0; 12,0 e 16,0 µM) e ANA (0,0; 1,0 e 2,0 µM), acrescido de 87,64 mM de sacarose e solidificado com 0,7% de ágar. Após 30 dias, avaliaram-se a percentagem de regeneração dos embriões e ápice plumular, o número de brotos, o número de folhas, o comprimento da parte aérea dos brotos, o número de raízes e a percentagem de formação de calos oriundos da região intermediária e da radícula. É possível a regeneração in vitro de mulungu, a partir dos explantes plúmula e embriões zigóticos inteiros, cultivados em meio de cultura WPM, suplementado com 4,0 µM de BAP. Regiões intermediárias e da radícula promoveram a formação de calos compactos (96,06%), na combinação de 10,63 µM BAP e 2,0 µM de ANA.

The in vitro culture of zygotic embryos represents a promising technique to study the embryonic development and dormancy in seeds. The objective of this study was to determine the effect of plant growth regulators, 6-benzylaminopurine (BAP) and naphthalene acetic (NAA), in the morphogenetic potential of in vitro zygotic embryos of mulungu. Mature zygotic embryos from seeds were either used as a whole or sectioned in plumule, radicle and intermediate region, and subsequently inoculated in WPM medium supplemented with combinations of BAP (0.0, 2.0, 4, 0, 8.0, 12.0 and 16.0 µM) and NAA (0.0, 1.0 and 2.0 µM), added with 87.65 mM sucrose and 0.7% agar. After 30 days, the following parameters were evaluated: regeneration percentage of embryos and plumularapice, number of buds, number of leaves, aerial part length of buds, number of roots and percentage of callifrom the intermediate region and radicle. It was feasible the in vitro regeneration of mulungu from plumule explant and whole zygotic embryos in WPM medium supplemented with 4.0 µM BAP. Moreover, intermediate and radicle regions promoted the formation of compact calli (96.06%)by combining10.63 µM BAP and 2.0 µM NAA.

Tapirira guianensis possui grande relevância medicinal, ecológica e socioeconômica, ocorrendo em todo o território brasileiro. O objetivo deste estudo foi estabelecer e determinar as melhores condições para a sua multiplicação in vitro. Os explantes, segmentos nodais, cotiledonares e epicótilos, oriundos de plântulas germinadas in vitro, foram testados em concentrações de 6-benzilaminopurina (BAP) e, ou, ácido naftalenoacético (ANA), em meio de cultura WPM. As características avaliadas foram a percentagem de explantes responsivos, o número de brotos e de gemas, o comprimento dos brotos e a matéria seca da parte aérea, aos 30 e 60 dias após inoculação. Foi observado que o segmento cotiledonar, nas condições deste estudo, foi o explante mais indicado para a multiplicação, não havendo indução de brotos adventícios nos epicótilos. O tratamento com 1,0 mg L-1 de BAP na ausência de ANA é o mais responsivo para a regeneração de T. guianensis.

Tapirira guianensis is of great medicinal, ecological and socio-economical importance occurring throughout the Brazilian territory. The objective of this study was to establish and determine the best conditions for the in vitro multiplication of T. guianensis. The explants nodal, cotyledonary and epicotyl segments from seedlings germinated in vitro were tested in diverse concentrations of 6-benzilaminopurine (BAP) and/or napthaleneacetic acid (NAA) in medium WPM. The following parameters were evaluated: the percentage of responsive explants, the number of shoots and buds, and the length of shoots and shoot dry mass at 30 and 60 days after inoculation. The cotyledonary segment, under these conditions, is the most suitable explant for the multiplication, with no induction of adventitious buds in epicotyl. The treatment with 1.0 mg L-1 BAP in the absence of NAA is the most responsive to the T. guianensis regeneration.

Bauhinia cheilantha, conhecida como "pata-de-vaca", possui grande relevância econômica e etnofarmacológica no semiárido brasileiro. Nas suas sementes constatou-se dormência, o que dificulta a obtenção de plantas uniformes e em curto período de tempo, diante disso, o presente trabalho objetivou estabelecer um protocolo de micropropagação para a espécie. Os segmentos cotiledonar e nodal de plântulas emergidas in vitro foram inoculados em meio de cultura WPM, suplementado com diferentes concentrações de 6-benzilaminopurina (BAP), thidiazuron (TDZ) ou cinetina (KIN) para induzir a regeneração de brotos adventícios. Na fase de enraizamento foram testadas diferentes concentrações de ácido indol-3-butírico (AIB) e de carvão ativado. Nos segmentos nodais houve maior capacidade organogênica do que no segmento cotiledonar. O maior número de brotos (4,3 e 2,1) foi obtido com 1,0mg L-1 de TDZ e 1,5mg L-1 de BAP, respectivamente. No entanto, na presença de TDZ foram observadas as menores brotações. A presença de 2,0mg L-1 de AIB com carvão ativado (CA) promoveu a maior porcentagem de enraizamento (cerca de 60,0%) e maior número de raízes adventícias (2,5). As brotações enraizadas foram transferidas para casa de vegetação e aclimatizadas com sucesso.

Bauhinia cheilantha, known as "pata-de-vaca", is of great economic and ethnopharmacological importance in the semiarid Brazilian. Seeds are dormant which makes it difficult to obtain uniform plants in a short time before this, our study aimed to establish a micropropagation protocol for the species. Cotyledonary and nodal segments of seedlings grown in vitro were inoculated on Woody Plant Medium (WPM) supplemented with various concentrations of 6-benzylaminopurine (BAP), thidiazuron (TDZ) or kinetin (KN) to induce adventitious shoot regeneration. In the rooting phase, different concentrations of indole-3-butyric acid (IBA) and the activated charcoal were tested. The nodal segments showed organogenic capacity greater than the cotyledonary segment. The highest number of shoots (4.3 and 2.1) was obtained at 1.0mg L-1 TDZ and 1.5mg L-1 BAP, respectively. However, in the presence of TDZ was observed the smaller shoots.The use of 2.0mg L-1 IBA within activated charcoal (AC) promoted the highest percentage of rooting (about 60.0%) and number of adventitious roots (2.5). The rooted shoots were transferred to greenhouse and successfully acclimatized.

Erythrina velutina (mulungu) é uma planta nativa do semiárido brasileiro que pode ser propagada via cultura de tecidos, o que poderá contribuir com a rápida multiplicação dessa espécie, além da redução de possíveis impactos ambientais. O objetivo deste trabalho foi estabelecer um protocolo de micropropagação para o mulungu utilizando explantes de plântulas assépticas obtidas in vitro. Para a indução de brotos, foram utilizados como explantes hipocótilos segmentos nodais e nós cotiledonares inoculados em meio de cultura WPM acrescido de diferentes concentrações de BAP e ANA. O enraizamento dos brotos foi realizado em meio WPM suplementado com 0,0; 2,46; 4,92; e 9,88µM de AIB. A melhor resposta para a multiplicação foi obtida em meio suplementado com 17,76µM de BAP, sendo utilizado segmento nodal ou nó cotiledonar como explante. Os brotos enraizaram em todas as condições testadas, inclusive no meio de cultura sem auxina.

Erythrina velutina (mulungu) is a native plant of the semi-arid region that can be propagated via tissue culture, which may contribute to the rapid multiplication of this species, reducing the possibility environmental impacts. The objective of this study was to establish a protocol for micropropagation of mulungu using aseptic explants seedlings grown in vitro. For the induction of shoots, , nodal segments and cotyledons were used as explants hypocontyl and were inoculated in a WPM culture medium supplemented with different concentrations of BAP and NAA. The rooting of shoots was achieved in a WPM medium supplemented with 0.0; 2.46; 4.92; 9.88µM IBA. The best answer to the multiplication was obtained in medium supplemented with 17.76µM BAP using nodal segment or cotyledonary node explants. The shoots rooted in all conditions tested, including the culture medium without auxin.

Este trabalho teve como objetivo avaliar o estabelecimento in vitro de Anadenanthera colubrina var. cebil, através das respostas morfofisiológicas das plântulas submetidas a várias condições de cultivo. As sementes foram desinfestadas e inoculadas em placas de Petri contendo papel germtest previamente esterilizado e umedecido com água estéril. As placas ficaram no escuro por dois dias até que ocorresse a germinação das sementes e, em seguida, transferidas para tubo de ensaio contendo meio de cultura WPM. Foram avaliados três tipos de fechamentos dos tubos de ensaio: filme de PVC, tampa plástica sem filme de PVC e tampão de algodão. No segundo experimento, o meio de cultura foi suplementado com carvão ativado (0,0; e 82,2 mM) e diferentes concentrações de sacarose (0,0; 29,21; 58,43; e 87,64 mM). Ao final de 30 dias, observaram-se efeitos altamente significativos dos tipos de fechamento sobre todas as variáveis analisadas: comprimento da parte aérea (CPA), número de folhas (NF), número de pinas (NP), matéria seca da parte aérea (MSPA), abscisão foliar (AF), comprimento da raiz (CR), matéria seca da raiz (MSR) e número de raízes secundárias (NRS). A AF foi reduzida em 7,3 vezes, quando os tubos de ensaio foram fechados com tampão de algodão em relação ao fechamento com filme de PVC. Os maiores valores de CPA (102,5 mm), NF (3,0), CR (194,7 mm) e NRS (47,88) foram obtidos com 29,21 mM de sacarose; a MSPA (59,1 mg), quando utilizou 58,43 mM e MSR (33,37 mg), com 87,64 mM de sacarose. O carvão ativado apresentou efeito significativo apenas em NRS, com maior valor (45,79) na presença desse composto.

This work aimed to evaluate the in vitro establishment of the species Anadenanthera colubrina var. cebil, throug the morphophysiological answers of the seedlings subjected to several culture conditions. The seeds were desinfested and inoculated in Petri plates containing germtest paper, previously sterilized and imbibed with sterile water. The plates were kept in the dark for two days, until the seeds germinated. Then, they were transferred to the test tube containing WPM medium. Three types of test tube closings were evaluated - PVC film, plastic lid without PVC film and cotton lid. In the second experiment, the culture medium was supplied with activated charcoal (0,0 and 82,2mM) and different sucrose concentrations (0,0; 29,21; 58,43; 87,64mM). After 30 days, highly significant effects of the types of closure were observed on all the variables analysed - aereal part length (APL), number of leaves (NL), number of pinnas (NP), dry matter of aereal part (DMAP), leaf abscission (LA), root length (RL), root dry matter (RDM) and number of secondary roots (NSR). The LA was reduced in 7,3 times when the cotton lid was used instead of the PVC film. The greatest values for APL (102,5 mm), NP (3,0), RL (194,7 mm) and NRS (47,88) were obtained with 29,21mM of sucrose. For DMAP (59,1 mg), when it used 58,43mM, and for RDM (33,37 mg), with 87,64mM of sucrose. The activated charcoal showed significant effects only for the variable SRN, presenting the greatest value (45,79) in the presence of this compound.

O trabalho teve por objetivos controlar a abscisão foliar de plântulas de angico utilizando AgNO3 e CoCl2 e avaliar o efeito do paclobutrazol sobre o comportamento in vitro das plântulas. As sementes foram desinfestadas e inoculadas em placas de Petri contendo papel germtest, previamente esterilizado e umedecido com água estéril. As placas ficaram no escuro por dois dias até que ocorresse a germinação das sementes e, em seguida, foram transferidas para tubo de ensaio contendo meio WPM. Todo esse material foi mantido em sala de crescimento com temperatura de 25 ± 2 ºC. No primeiro experimento, o meio de cultura foi suplementado com dois tipos de inibidores de etileno (AgNO3 e CoCl2) em diferentes concentrações (0,0; 5,0; 10,0; 20,0; e 40,0 µM). No segundo, o meio foi suplementado com diferentes concentrações (1,7; 3,4; 6,8; e 13,6 µM) de paclobutrazol. Verificou-se que houve aumento no número de folhas e redução na abscisão foliar com a utilização de 10 e 20 µM (respectivamente), independentemente do inibidor de etileno utilizado. Tanto o AgNO3 quanto o CoCl2 contribuíram com acréscimos no número de gemas e no número de brotações. O aumento na concentração de paclobutrazol reduziu o crescimento das plântulas, obtendo-se menores médias das seguintes variáveis: comprimento da parte aérea, número de folhas, número de folhas senescentes e matéria seca da parte aérea na maior concentração (13,6 µM) de paclobutrazol. Esse inibidor de crescimento teve efeito também no sistema radicular, reduzindo o comprimento da raiz e o número de raízes secundárias, no entanto promoveu o engrossamento das raízes.

This work aimed to control leaf abscission of seedlings of angico using AgNO3 and CoCl2 and evaluate the effect of the paclobutrazol on the in vitro behavior of seedlings. The seeds were disinfected and inoculated in Petri plates containing germtest paper, previously sterilized and imbibed in sterile water. The plates were kept in the dark for two days, until seed germination. Seeds were transferred to test tubes containing WPM medium. In the first experiment, the culture medium was supplemented with different concentrations of (0.0; 5.0; 10.0; 20.0; 40.0 µM) and types (AgNO3 and CoCl2) of ethylene inhibitors. In the second experiment, the medium was supplemented with different concentrations (1.7; 3.4; 6.8; 13.6 µM) of paclobutrazol. There was increase in the number of leaves and reduction in foliar abscission with 10 and 20 µM respectively, independent of the used ethylene inhibitor. Both the AgNO3 and the CoCl2 helped to increase the number of buds and shoots. The increase in the concentration of paclobutrazol reduced seedling growth, with lower means for the variables - length of aerial part, number of leaves, number of senescent leaves and dry matter of the aerial part in the highest concentration (13.6 µM) of paclobutrazol. This growth inhibitor had also effects on the root system, reducing root length and number of secondary roots, however promoting root thickening.
