Resumo Foram coletados carrapatos parasitando 102 animais silvestres nos estados de Mato Grosso e Goiás, Brasil, entre 2015 e 2018. Um total de 2338 carrapatos (865 machos, 541 fêmeas, 823 ninfas e 109 larvas) pertencentes a quatro gêneros (Amblyomma, Dermacentor, Haemaphysalis e Rhipicephalus) e pelo menos 21 espécies foram identificadas. A extração de DNA e pesquisa molecular para agentes riquétsiais foram realizadas em 650 carrapatos. Os resultados revelaram parasitismo pelas seguintes espécies: Rickettsia amblyommatis em Amblyomma cajennense s.s., A. cajennense s.l., Amblyomma coelebs, Amblyomma humerale, Amblyomma longirostre, Amblyomma nodosum, Amblyomma scalpturatum, Amblyomma sculptum e Amblyomma romitii; Rickettsia parkeri em Amblyomma nodosum, Amblyomma ovale, Amblyomma scalpturatum e Amblyomma triste; Rickettsia rhipicephali em Haemaphysalis juxtakochi; Rickettsia sp. em A. cajennense s.s., A. nodosum e A. sculptum e, por último, ‘Candidatus Rickettsia andeanae’ em Amblyomma parvum e Rhipicephalus microplus. Abstract Ticks parasitizing 102 wild animals in the states of Mato Grosso and Goiás, Brazil were collected between 2015 and 2018. A total of 2338 ticks (865 males, 541 females, 823 nymphs, and 109 larvae) belonging to four genera (Amblyomma, Dermacentor, Haemaphysalis, and Rhipicephalus) and at least 21 species were identified. Resumo Trypanosoma (Megatrypanum) theileri é um protozoário flagelado que infecta ruminantes e apresenta alta diversidade genética. Neste estudo, investigamos as taxas de prevalência deste protozoário com base na hemocultura e no diagnóstico molecular. Os isolados de T . theileri obtidos foram caracterizados pelos marcadores moleculares SSU rDNA e gGAPDH e o diagnóstico molecular foi baseado no gene do tipo Catepsina L (PCR-TthCATL). O PCR-TthCATL e a hemocultura indicaram uma taxa de prevalência total de 8,13% e a sequência derivada do gene Catepsina L denominada IB de T. theileri foi identificada pela primeira vez em bovinos da Amazônia Ocidental, bem como a IF no Brasil. Também descrevemos uma possível nova sequência derivada da PCR-TthCATL em bovinos, designada IL.

Abstract Trypanosoma (Megatrypanum) theileri is a flagellated protozoan that infects ruminants and it displays high genetic diversity. In this study, we investigated the prevalence rates of this protozoan based on hemoculture and molecular diagnosis. The isolates of T. theileri thus obtained were characterized by molecular markers SSU rDNA and gGAPDH and molecular diagnosis based on Cathepsin L-like gene (PCR-TthCATL). The PCR-TthCATL and hemoculture indicated an overall prevalence rate of 8.13%, and the CATL derived sequence named IB was identified for the first time in cattle in the western Amazon region, as well as IF in Brazil. We also describe a possible new PCR-TthCATL derived sequence in cattle, designated IL.

Abstract INTRODUCTION The epidemiology of Rickettsia and Ehrlichia species infection is underestimated in Mato Grosso State. METHODS: Serum samples obtained during a Dengue outbreak in 2011-2012 were tested via indirect immunofluorescence and/or ELISA. RESULTS: Samples from 19/506 (3.8%) patients presented antibodies for at least one of three Rickettsia species; 2/506 (0.4%) samples reacted against Ehrlichia canis. Most afflicted patients are residents of cities from the south-central region of the state, where these diseases have been reported in animals. CONCLUSIONS: These results show serological evidence of human exposure to Rickettsia and Ehrlichia species in Mato Grosso State.

RESUMO: O município de Ji-Paraná, Rondônia, é uma das principais áreas de produção leiteira na região norte do Brasil. Assim, é importante avaliar agentes infecciosos que apresentam potencial para afetar negativamente a produtividade na indústria. O objetivo deste estudo foi investigar a prevalência de ' Candidatus Mycoplasma haemobos' usando um método de detecção baseado na PCR e relacionar com variáveis do rebanho leiteiro (frequência de aborto, nascimento de bezerros fracos, número total de vacas, número de vacas >24 meses de idade, tamanho da propriedade e, sistema de produção) na agricultura familiar do município de Ji-Paraná, região norte, Brasil. Amostras de sangue foram coletadas de 320 vacas leiteiras localizadas em 64 fazendas (ou seja, cinco animais por exploração) de setembro de 2012 a novembro de 2013. A prevalência total de ' Ca. M. haemobos' foi de 64,2% e a prevalência por rebanho foi de 95,3%; o número de vacas >24 meses de idade nas propriedades estudadas correlacionou-se com as taxas de infecção por ' Ca. M. haemobos'. Considerando a importância da indústria de laticínios para a área de estudo, investigações adicionais são necessárias para avaliar o efeito da infecção crônica nestes animais na produção de leite e saúde do rebanho.

ABSTRACT: The municipality of Ji-Paraná, Rondônia, is one of the major dairy production areas in the north region of Brazil. Thus, it is important to evaluate infectious agents that have the potential to negatively affect productivity in the industry. The objective of this study was to investigate the prevalence of ' Candidatus Mycoplasma haemobos' by using a PCR-based detection method and correlate this with dairy herd variables (abortion frequency, weak calf birth rate, total cattle number, >24-month-old cow number, farm size, and production system) in family farms of the Ji-Paraná municipality, north region, Brazil. Blood samples were collected from 320 dairy cows located across 64 farms (i.e., five animals per farm) from September 2012 to November 2013. Overall prevalence of 'Ca. M. haemobos' was 64.2% and prevalence per herd was 95.3%; the number of >24-month-old cows in the farms studied correlated with ' Ca. M. haemobos' infection rates. Considering the importance of the dairy industry to the study area, additional investigations are necessary to evaluate the effect of chronic infection in these animals on milk production and herd health.

Resumo O presente estudo investigou evidência sorológica de agentes transmitidos por carrapatos em gambás em oito municípios do Estado de São Paulo, Brasil. Amostras de soro de 109 gambás (91 Didelphis aurita e 18 Didelphis albiventris) foram testadas para detecção de anticorpos contra Rickettsia rickettsii (cepa Taiaçu, ponto de corte 1:64) Ehrlichia canis (cepa São Paulo, ponto de corte 1:40), pela reação de imunofluorescência indireta (RIFI); e contra Borrelia burgdorferi (cepa G39/40) pelo teste imunoenzimático (ELISA). A presença de anticorpos anti-R. rickettsii, anti-E. canis e anti-B. burgdorferi foi detectada em 32 (29,35%), 16 (14,67%) e 30 (27,52%) gambás, respectivamente. Os títulos finais variaram de 64 a 1.024 para R. rickettsii, de 40 a 160 para E. canis, e de 400 a >51.200 para B. burgdorferi. Esses resultados sugerem que os gambás foram expostos a agentes relacionados à Rickettsia spp., Ehrlichia spp., e B. burgdorferi no Estado de São Paulo. Neste estudo salienta a necessidade de novas pesquisas sobre esses agentes nessas áreas de trabalho, devido à ocorrência da Febre Maculosa e da Síndrome Baggio-Yoshinari em humanos no Estado de São Paulo, Brasil.

Abstract This work involved a serological investigation of tick-borne pathogens in opossums in eight municipalities of the state of São Paulo, Brazil. Serum samples from 109 opossums (91 Didelphis aurita and 18 Didelphis albiventris) were tested to detect antibodies to Rickettsia rickettsii (Taiaçu strain, 1:64 cut-off) and Ehrlichia canis (São Paulo strain, 1:40 cut-off), by indirect immunofluorescence assay (IFA); and against Borrelia burgdorferi (strain G39/40) by enzyme-linked immunosorbent assay (ELISA). The presence of antibodies to anti-R. rickettsii, anti-E. canis and anti-B. burgdorferi was detected in 32 (29.35%), 16 (14.67%) and 30 (27.52%) opossums, respectively. Opossum endpoint titers ranged from 64 to 1,024 for R. rickettsii, from 40 to 160 for E. canis, and from 400 to >51,200 for B. burgdorferi. These serological results suggest that opossums have been exposed to Rickettsia spp., Ehrlichia spp., and B. burgdorferi-related agents in the state of São Paulo. Our study underscores the need for further research about these agents in this study area, in view of the occurrence of Spotted Fever and Baggio-Yoshinari Syndrome disease in humans in the state of São Paulo, Brazil.

Neosporose é considerada uma das principais causas de aborto em rebanhos bovinos em todo o mundo. O objetivo deste estudo foi investigar a prevalência de anticorpos anti-Neospora caninum em bovinos de leite e correlacioná-los com possíveis fatores de risco em 63 pequenas propriedades rurais (agricultura familiar), no município de Ji-Paraná, uma das principais regiões produtoras de leite do Estado de Rondônia, norte do Brasil. Para este propósito, 621 amostras de soro foram coletadas de vacas leiteiras e avaliadas por meio da reação de imunofluorescência indireta (RIFI). A prevalência total de propriedades com animais positivos a anticorpos anti-N. caninum (38/63; 60,31%) indica que o parasito está difundido entre os rebanhos leiteiros da região, apesar de infectar uma pequena proporção de animais (66/621; 10,62%). Ocorrência de abortos e nascimento de bezerros fracos foram as únicas variáveis consideradas como fatores de risco para a ocorrência de N. caninum. O resultado do modelo de análise espacial indica que o nascimento de bezerros fracos e a presença de N. caninum estão aparecendo em fazendas localizadas próximas umas das outras, indicando a ocorrência de agregação da doença.

Neosporosis is considered a major cause of abortion among cattle worldwide. The aim of this study was to investigate the prevalence of anti-Neospora caninum antibodies in dairy cattle and correlate them with possible risk factors on 63 small farms (family farms) in the municipality of Ji-Paraná, the main milk-producing region of the state of Rondônia, northern Brazil. For this purpose, 621 serum samples were collected from cows and were evaluated by means of the indirect fluorescence antibody test (IFAT). The overall herd prevalence of N. caninum antibodies among the farms (38/63; 60.31%) showed that N. caninum are widespread among the dairy herds in this region, despite only infecting a small proportion of animals (66/621, 10.62%). Occurrences of abortion and birth of weak calves were the only variables that showed as risk factors for the presence of N. caninum. The result from the spatial lag model strongly indicated that birth of weak calves and presence of N. caninum are occurring on farms that are located close to each other, indicating aggregation of disease occurrence.

Toxoplasma gondii, Neospora caninum e Sarcocystis neurona são coccídios relacionados responsáveis por causar desordens reprodutivas e neurológicas em uma ampla variedade de animais domésticos e selvagens. No presente estudo, a Reação de Imunofluorescência Indireta (RIFI) foi utilizada para investigar a presença de anticorpos contra T. gondii, N. caninum e S.neurona em soros de 11 onças-pintadas de vida livre (Panthera onca), provenientes de duas áreas protegidas na região do Pantanal do Estado de Mato Grosso, Brasil. Dez (90,9%), sete (63,6%) e oito (72,7%) onças amostradas foram soropositivas para T. gondii, N. caninum e S. neurona, respectivamente. Os resultados indicam a exposição a esses coccídios relacionados entre as onças-pintadas e a circulação ambiental desses patógenos nesse ecossistema selvagem. Este é o primeiro relato da detecção sorológica de N. caninum e S. neurona em onças-pintadas de vida livre.

Toxoplasma gondii, Neospora caninum and Sarcocystis neurona are related apicomplexan parasites that cause reproductive and neurological disorders in a wide range of domestic and wild animals. In the present study, the immunofluorescence antibody test (IFAT) was used to investigate the presence of antibodies against T. gondii, N. caninum and S. neurona in the sera of 11 free-living jaguars (Panthera onca) in two protected areas in the Pantanal region of Mato Grosso state, Brazil. Ten jaguars (90.9%) showed seropositivity for T. gondii, eight (72.7%) for S. neurona, and seven (63.6%) for N. caninum antigens. Our findings reveal exposure of jaguars to these related coccidian parasites and circulation of these pathogens in this wild ecosystem. To the best of our knowledge, this is the first serological detection of N. caninum and S. neurona in free-living jaguars.

Ehrlichiosis is a worldwide distributed disease caused by different bacteria of the Ehrlichia genus that are transmitted by arthropod vectors. Its occurrence in dogs is considered endemic in several regions of Brazil. Regarding cats, however, few studies have been done and, consequently, there is not enough data available. In order to detect Ehrlichia spp. in cats from the central-western region of Brazil, blood and serum samples were collected from a regional population of 212 individuals originated from the cities of Cuiabá and Várzea Grande. These animals were tested by the Immunofluorescence Assay (IFA) and the Polymerase Chain Reaction (PCR) designed to amplify a 409 bp fragment of the dsb gene. The results obtained show that 88 (41.5%) cats were seropositive by IFA and 20 (9.4%) cats were positive by PCR. The partial DNA sequence obtained from PCR products yielded twenty samples that were found to match perfectly the Ehrlichia canis sequences deposited on GenBank. The natural transmission of Ehrlichia in cats has not been fully established. Furthermore, tick infestation was not observed in the evaluated cats and was not observed any association between age, gender and positivity of cats in both tests. The present study reports the first serological and molecular detection of E. canis in domestic cats located in the endemic area previously mentioned.

Diversos estudos sobre erliquiose felina vêm sendo relatados na literatura. No entanto, a caracterização da infecção por Ehrlichia em gatos ainda é escassa. O presente estudo objetivou avaliar a associação entre infecção por Ehrlichia e dados hematológicos em 93 gatos atendidos no Hospital Veterinário da Universidade Federal de Mato Grosso, em Cuiabá, Brasil. A presença de infecção por Ehrlichia spp. foi avaliada pela Reação em Cadeia pela Polimerase (PCR) visando à amplificação dos genes dsb e 16S rRNA de Ehrlichia e por Reação de Imunofluorescência Indireta (RIFI). Oito (8,6%) gatos demonstraram ser positivos pela PCR, sendo suas sequências parciais de DNA 100% idênticas à E. canis. Quarenta e dois gatos (45,1%) apresentaram anticorpos reativos contra Ehrlichia spp. Alterações hematológicas como baixas contagens de eritrócitos, trombocitopenia, linfopenia e monocitose foram observadas em gatos positivos pela PCR. Dentre essas, eritropenia foi associada em gatos com títulos de anticorpos IgG entre 40 e 640, sendo cinco destes positivos pela PCR. Adicionalmente, gatos positivos na PCR apresentaram uma tendência a serem linfopênicos. Não foram observadas associações entre a presença de infecção nos gatos e suas respectivas idades e sexo. Nenhum carrapato foi observado nos gatos examinados.

The literature contains several studies on feline ehrlichiosis. However, information about the characteristics of Ehrlichia infection in cats is still scanty. This study evaluated the association between Ehrlichia spp. infection and the hematologic data of 93 cats treated at the Federal University of Mato Grosso Veterinary Hospital in Cuiabá, state of Mato Grosso, Brazil. The presence of or exposure to Ehrlichia spp. infection was evaluated by Polymerase Chain Reaction (PCR) targeting the dsb and 16S rRNA gene of Ehrlichia, and by detection of anti-Ehrlichia canis IgG antibodies in Indirect Fluorescence Assay (IFA), respectively. Eight (8.6%) cats tested positive by PCR and the partial DNA sequence obtained from PCR products was a 100% match to E. canis. Forty-two (45.1%) cats showed antibody reactivity against Ehrlichia spp. Hematological alterations such as low erythrocyte count, thrombocytopenia, lymphopenia and monocytosis were observed in PCR positive cats. Among them, low erythrocyte counts were associated with IgG antibody titers of 40 to 640 and five cats also tested positive by PCR. Furthermore, PCR-positive cats showed a tendency to be lymphopenic. No correlation was found between age and sex, and no ticks were observed in any of the examined cats.

O presente estudo avaliou a presença de DNA deEhrlichia spp. em 320 cães das áreas urbana e rural do município de Poconé, região do Pantanal de Mato Grosso, pela PCR visando o gene dsb. Os fatores de risco para a infecção em cães também foram avaliados. Quarenta e oito (15%, IC 95%: 11,4-19,5%) cães foram positivos, 25 (15,6%, IC 95%: 10,4-22,2%) da área urbana e 23 (14,37%, 95% CI: 9,3-20,8%) da área rural (P > 0,05). Sequências parciais de DNAs obtidos a partir de produtos da PCR de 18 amostras da área urbana e 16 da área rural foram 100% idênticas a E. canis do Brasil e EUA. Este estudo relata a primeira detecção molecular deE. canis em cães da região norte do Pantanal.

The present study evaluated the presence of EhrlichiaDNA in the blood samples of 320 dogs from the urban and rural areas of the municipality of Poconé, Pantanal region, Mato Grosso state, by Polymerase Chain Reaction (PCR), targeting the ehrlichial dsbgene. Risk factors for infection in dogs were also evaluated. Forty-eight (15%, 95% CI: 11.4-19.5%) dogs were positive: 25 (15.6%, 95% CI: 10.4-22.2%) from the urban area and 23 (14.4%, 95% CI: 9.3-20.8%) from the rural area (P > 0.05). Partial DNA sequence obtained from PCR products of 18 samples from the urban area and 16 samples from the rural area were 100% identical to E. canis from Brazil and the USA. This study reports the first E. canis molecular detection in dogs from the northern Pantanal region.

Descreve-se um surto de aborto em caprinos por T. gondii em uma propriedade situada na região Centro-Oeste do Brasil. Não foram observadas lesões macroscópicas em sete fetos abortados encaminhados ao Laboratório de Patologia Veterinária da Universidade Federal de Mato Grosso para necropsia. As principais lesões histopatológicas foram encefalite necrosante e pneumonia mononuclear em variados graus de intensidade. PCR para Brucella abortus e Neospora caninum e cultivos em aerobiose obtiveram resultado negativo em todos os casos. Anticorpos anti-T. gondii em titulações de 1:1024 a 1:32.768 foram detectados no soro de quatro cabras que abortaram. Amostras de todos os cérebros enviados foram positivos para T. gondii pela técnica de Nested-PCR. Estes achados indicam que a infecção por T. gondii deve ser considerada no diagnóstico de aborto em caprinos na região Centro-Oeste do Brasil.

An outbreak of abortion by Toxoplasma gondii in goats on a farm in the Brazilian Midwest is reported. Gross lesions were not observed in seven aborted fetuses submitted to the Veterinary Pathology Laboratory, Federal University of Mato Grosso, for necropsy investigation. The main histologic lesions were mononuclear cell pneumonia and necrotizing encephalitis in varying degrees of intensity. PCR for Brucella abortus and Neospora caninum and aerobic cultures were negative in all cases. Antibody titles against T. gondii varying from 1:1024 to 1:32.768 were detected in serum samples from four aborted goats. Nested-PCR assay for T. gondii were positive in brain samples of all cases submitted. These findings indicate that T. gondii infection should be considered in the diagnosis of abortion in goats in Midwest Brazil.

O objetivo deste estudo foi relatar pela primeira vez a infecção por Hepatozoon spp. e Babesia spp. em cães domésticos provenientes da cidade de Cuiabá, estado de Mato Grosso. Foram utilizados pares de primers que amplificam um fragmento de 574 pb do gene 18S rRNA de Hepatozoon spp., e 551 pb do gene 18S rRNA para Babesia spp. Dos 10 cães amostrados, 6 apresentaram-se positivos para Babesia spp., e 9 foram positivos para Hepatozoon spp. pela PCR. Co-infecção entre Babesia spp. e Hepatozoon spp. ocorreu em 5 cães. As amostras revelaram 100% de identidade com B. canis vogeli, e as amostras que foram positivas para Hepatozoon spp. foram 100% idênticas a H. canis. Esta é a primeira identificação molecular de H. canis em cães domésticos em Cuiabá. Adicionalmente, descrevemos pela primeira vez a presença de B. canis vogeli circulando entre cães em Cuiabá.

The objective of this study was to report for the first time infection by Hepatozoon spp. and Babesia spp. in 10 dogs from the city of Cuiabá, State of Mato Grosso, central-western Brazil. A pair of primers that amplifies a 574 bp fragment of the 18S rRNA of Hepatozoon spp., and a pair of primers that amplifies a 551 bp fragment of the gene 18S rRNA for Babesia spp. were used. Six dogs were positive for Babesia spp., and 9 were positive for Hepatozoon spp. Co-infection of Babesia spp. and Hepatozoon spp. was seen in 5 dogs. Sequenced samples revealed 100% identity with B. canis vogeli, and H. canis. This is the first molecular detection of H. canis in domestic dogs from Cuiabá. Additionally, it is described for the first time the presence of B. canis vogeli circulating among dogs in Cuiabá.
