A mancha-de-ramularia, doença causada pelo fungo Ramularia areola, é uma das doenças de destaque na cotonicultura brasileira. Há alguns anos atrás, esta doença era considerada de final de ciclo da cultura, mas em anos recentes tornou-se a doença de maior importância econômica nas principais regiões produtoras de algodão do Brasil. Dentre as alternativas de controle, estão o uso de fungicidas e o uso de cultivares resistentes, sendo que o último é o preferido. Visando estudar a herança da resistência do algodoeiro a R. areola, foram avaliadas populações derivadas do cruzamento entre a linhagem FMT02102996, como resistente e a cultivar FMT 701, como suscetível. As plantas foram classificadas como resistentes ou suscetíveis por meio de inoculação artificial em casa de vegetação. Realizou-se teste de segregação por meio de teste qui-quadrado. Os resultados obtidos indicam que a resistência do algodoeiro à mancha-de-ramularia é condicionada por um gene dominante. Este resultado pode auxiliar o planejamento dos programas de melhoramento do algodoeiro na incorporação da resistência a R. areola em novas cultivares, também constituindo informação básica para o início de trabalhos de mapeamento genético deste gene de resistência agronomicamente desejável.

Areolate mildew, caused by Ramularia areola is an important disease affecting cotton in Brazil. A few years ago this disease was of secondary importance since it used to occur almost at the end of the crop cycle. However, in recent years the disease has gained considerable importance in the main cotton growing areas of Brazil. Amongst control alternatives is the use of fungicides and resistant cultivars the latter being preferred. In order to study the inheritance of cotton resistance to R. areola, plant populations derived from the crossing between the resistant line FMT02102996 and the susceptible cultivar FMT 701, were evaluated. These plants were classified as resistant or susceptible by means of artificial inoculation in greenhouse. The segregation test was done through ÷2 test. Results indicated that cotton resistance to areolate mildew is controlled by a dominant gene. This finding can assist in planning cotton breeding programs aimed at incorporating resistance to R. areola in nem cultivars, constituting basic information for the beginning of the genetic mapping of this resistance gene in agronomically desirable cotton cultivars.

A mancha angular do algodoeiro, causada por Xanthomonas axonopodis pv. malvacearum (Xam), é uma doença de importância econômica e pode causar perdas apreciáveis no rendimento. A doença pode ser controlada por resistência varietal desde que haja conhecimento sobre a variabilidade das populações do patógeno. A variabilidade genética e a estabilidade patogênica entre os isolados deste patógeno não foram suficientemente estudadas, principalmente considerando a introdução de novas cultivares e a expansão da cultura. O objetivo do presente estudo foi avaliar a variabilidade genética entre os 61 isolados de Xam, provenientes de diversas cultivares e regiões do Brasil, através de ensaios moleculares. Análises de ERIC e REP-PCR demonstraram dois grupos distintos de Xam associados a região geográfica de origem. Não foram observadas diferenças nos perfis dos isolados através de PCR-RFLP da região 16S-23S rDNA. A região espaçadora 16S-23S rDNA de três linhagens de Xam foi analisada através de clonagem e sequenciamento e seis diferenças nas seqüências foram encontradas. A técnica de RAPD revelou um maior nível de polimorfismo, distinguindo 6 grupos de Xam a 85% de similaridade. Os resultados indicam a existência de variabilidade muito restrita entre os isolados analisados.

Angular leaf spot or black arm of cotton, caused by Xanthomonas axonopodis pv. malvacearum (Xam), is a disease of economic importance. The disease can be controlled by using resistant cultivars if the knowledge regarding the genetic variability of the pathogen population is available. The genetic variability and the pathogen stability among the isolates of this pathogen have not been sufficiently studied, especially considering the introduction of new cultivars and the expansion of areas under cotton cultivation. The objective of the present study was to identify genetic variability among 61 isolates of Xam collected from different cultivars and geographic regions. Analysis of ERIC and REP-PCR revealed two distinct groups of Xam based on the geographic region of isolation. PCR-RFLP of 16S-23S rDNA did not reveal differences in the restriction profiles of the isolates. The 16S-23S rDNA region of three isolates of Xam was also analysed by cloning and sequencing and 6 differences in the sequences were observed. The RAPD technique revealed a higher level of polymorphism and distinguished 6 groups of Xam. The results indicate a very narrow genetic variability among Xam isolates used in this study.

Estudou-se a eficiência do meio semi-seletivo desenvolvido para Xanthomonas axonopodis pv. malvacearum (Xam) do algodoeiro, com algumas modificações, na detecção de Xanthomonas axonopodis pv. phaseoli (Xap) em sementes de feijoeiro, bem como a eficiência do fungicida tolylfluanid na erradicação de Xap através do tratamento de sementes. Foram utilizadas sementes naturalmente infectadas por Xap, procedentes de diferentes municípios do Estado do Paraná. Houve o desenvolvimento de colônias bacterianas ao redor das sementes infectadas sob meio semi-seletivo entre seis e 12 dias de incubação. A freqüência de recuperação de colônias da bactéria em relação ao meio Agar nutriente (AN) variou entre 30 a 112%. O crescimento de Xap foi inibido a concentrações entre 54 a 1500 ppm do fungicida tolylfluanid. A bactéria foi recuperada das sementes não tratadas em meio semi-seletivo, mas não das sementes tratadas em solução do fungicida tolylfluanid (1,20g/L água). Em casa de vegetação, das 400 sementes tratadas com tolylfluanid não foi produzida nenhuma planta com sintomas da doença até 30 dias após a semeadura, enquanto que 9,75% das sementes não tratadas apresentaram sintomas de Xap. Em um outro experimento, das 1200 sementes não tratadas um total de 7,08% e 11,67% das plantas mostraram-se com sintomas da doença, enquanto que das sementes tratadas 0,5% e 2,4% apresentaram sintomas, 26 e 46 dias após a semeadura, respectivamente.

The efficiency of a semi-selective medium developed for Xanthomonas axonopidis pv. malvacearum (Xam), was verified with some modifications, in detecting the presence of X. axonopodis pv. phaseoli (Xap) in bean seed. Seed samples naturally infected with Xap were collected from the State of Paraná. The bacterial growth developed around the infected seed in the semi-selective medium after 12 days after incubation. The recovery frequency of bacterial colonies in relation to nutrient agar varied between 30 and 112%, depending upon the isolate. Efficiency of tolylfluanid to eradicate Xap from the bean seed was also verified in laboratory and glasshouse experiments. The bacteria was recovered from the untreated seeds, but not from the seeds treated in a solution of tolylfluanid (1.20g/L water), and placed in plates containing semi-selective medium. In the glasshouse, seeds treated with tolylfluanid, did not produce any symptoms of the disease till 30 days after seeding, whereas untreated seeds yielded 9,75% plants with typical disease symptoms. In an another experiment, untreated seeds yielded 7,08% and 11,67% diseased plants, whereas treated seeds yielded 0,5% and 2,4% diseased plants, 26 and 46 days after planting, respectively.

Estudou-se a resposta diferencial de 32 cultivares comerciais do algodoeiro a A. macrospora em casa de vegetação. A inoculação foi realizada em plântulas de 20-25 dias de idade com 25 repetições, utilizando-se o delineamento experimental de blocos ao acaso. As cultivares foram agrupadas através de análise de Scott & Knott. Resistência completa não foi encontrada, sendo que todas as cultivares foram suscetíveis. Houve diferença entre reação de plantas da mesma cultivar. Não obstante, quando algumas plantas das cvs. BRS Antares e Fibermax 986, que mostraram resistência, foram inoculadas com mistura de nove isolados escolhidos ao acaso, as mesmas mantiveram sua resistência a esta mistura. Trabalhos futuros deverão ser realizados para encontrar fontes de resistência a este patógeno em outras espécies de Gossypium.

The differential response of 32 commercial cotton cultivars to A. macrospora was studied under greenhouse conditions. Twenty to twentyfive days old plants were inoculated in 25 repetitions in a randomized block design. The cultivars were grouped using the Scott & Knott analysis. Complete resistance was not found and all cultivars were susceptible to the pathogen. Differential reaction within the plants of the same cultivar was observed. However, when some resistant plants of cultivars BRS Antares and Fibermax 986 were inoculated using a mixture of nine randomly selected isolates of A. macrospora, the plants maintained their resistance. Further research is necessary to find sources of resistance to this pathogen in other species of Gossypium.
