Resumo O gênero Dermocystidium infecta uma ampla variedade de espécies de animais, e o hospedeiro pode se infectar pela ingestão de endósporos. O diagnóstico depende de montagem em lâmina a fresco e análise histopatológica dos tecidos afetados. O objetivo deste estudo foi relatar infecção por Dermocystidium na pele do bagre listrado (P. hypophthalmus), cultivado em piscicultura do estado do Ceará, nordeste do Brasil. Foi detectado que 100% dos animais analisados estavam infectados por Dermocystidium sp. A montagem em lâminas a fresco e histopatologia das lesões mostrou cistos repletos de esporos entre a derme e a epiderme, encapsulados por tecidos conectivos. Devido às escassas pesquisas sobre esse parasito, em diferentes espécies de peixes no Brasil e no mundo, estudos adicionais são necessários para entender a endemicidade em diferentes pisciculturas do Brasil, consequentemente, melhorando os métodos de prevenção da doença e aumentando a produção e a produtividade.

Abstract The genus Dermocystidium infects a wide range of animals. The host infection often occurs through the ingestion of endospores. The diagnosis depends on wet mounts and histopathological analysis of the affected tissue. The aim of this study was to investigate the incidence of Dermocystidium sp. infection on the skin of farmed striped catfish (Pangasianodon hypophthalmus) from a fish farm located in Fortaleza, Ceará state, northeastern Brazil. From these observations, we determined that 100% of the analyzed animals were infected with Dermocystidium sp. The wet mount and histopathology of the fish lesions revealed spore-filled cysts between the dermis and epidermis, encapsulated by connective tissue. Owing to a lack of research on the parasite and its prevalence among different fish species in Brazil and the rest of the world, additional studies are required to understand their endemicity in fish farms of Brazil, and consequently develop better disease prevention methods and increase the overall productivity.

ABSTRACT This study evaluated the dietary supplementation of ginger Zingiber officinale essential oil for the Neotropical catfish Pseudoplatystoma reticulatum, its effects on fish hematology, immunology, the response of these variables to induction of stress (exposition to air for 3 min), and a bacterial challenge with Aeromonas hydrophila, and the effects on liver histomorphometry. Three levels of the ginger essential oil were evaluated (0.5, 1.0 and 1.5%) in addition to a control group (0%) in a feeding trial for 65 days in triplicate. Fish hematology and immunology were assessed at five different moments: after the feeding trial (0), after the stress induction (stress), and three points after the bacterial challenge (3, 6 and 24 h post-challenge). Fish fed the ginger essential oil supplemented diets did not exhibit significant differences between the hematocrit at the 0-sample point and the 24 h post-challenge point (P ≥ 0.05), while fish fed the control diet presented significantly lower values for the hematocrit when comparing those same two moments (P < 0.05). The thrombocytes count for fish fed the 0.5% essential oil supplemented diet was significantly higher than all other groups after the feeding trial (P < 0.05), and their total leukocytes count after the stress induction was also significantly higher than the other treatments (P < 0.05). Although the results mentioned above indicate that the ginger essential oil had positive health effects on the fish, no significant differences in survival after the challenge with A. hydrophila were observed (P ≥ 0.05). Lastly, histomorphometry suggests that the essential oil did not negatively impact fish hepatocytes.

Resumo Ascocotyle (Phagicola) longa é o agente etiológico da fagicolose humana. Os mugilídeos são os segundos hospedeiros intermediários. O primeiro é Heleobia australis e pássaros e mamíferos predadores de mugilídeos, os hospedeiros definitivos. A ocorrência de cistos contendo metacercárias de A. longa é descrita em mugilídeos, com até 100% de prevalência. A ampla distribuição geográfica de A. longa e seus hospedeiros intermediários, acompanhado do aumento do consumo de peixe cru ou mal cozido, pode aumentar o risco de infecção humana, portanto o objetivo do presente estudo foi verificar o padrão de distribuição de cistos, contendo metacercárias de A. longa, em mugilídeos. Amostras de tecidos e órgãos desses peixes foram processadas em liquidificador doméstico e observadas em microscópio estereoscópico à procura de cistos contendo metacercárias do digenético. Todos os 24 (100%) mugilídeos, 12 Mugil curema e 12 Mugil liza examinados apresentaram cistos contendo metacercária de A. longa. Foram observados cistos do digenético nas brânquias, coração, estômago, fígado, intestino, mesentério e tecido muscular de M. curema e de M. liza. Em M. curema, os cistos contendo metacercárias de A. longa estão distribuídos de forma aleatória por praticamente todos os tecidos e órgãos dos mugilídeos examinados.

Abstract Ascocotyle (Phagicola) longa is an etiological agent of human phagicolosis. Mugilids are the second intermediate host, the first being Heleobia australis, and mugilids predatory birds and mammals are its definitive hosts. The occurrence of cysts holding A. longa metacercariae is described in mugilids with a prevalence of up to 100%. The wide geographical distribution of A. longa and its intermediate hosts coupled with the rise in the consumption of raw or poorly cooked fish may elevate the risk of human infection. Therefore, in this study, we aimed to verify the distribution pattern of cysts holding A. longa in mugilids. The tissue and organ samples of these fish were processed in a domestic blender and examined under a stereoscopic microscope to identify the cysts holding the digenetic metacercariae. Of the 24 (100%) fish samples that were analyzed, 12 of Mugil curema and 12 of Mugil liza possessed cysts holding A. longa metacercariae. Digenetic cysts were identified to be present in the gills, heart, stomach, liver, intestines, mesentery, and muscular tissues collected from M. curema and M. liza. Conclusively, in M. curema, the cysts holding A. longa metacercariae were found to be distributed randomly throughout the fish body in almost every tissue and organ that was examined.

Resumo Este trabalho identificou espécies parasitas em peixes de produção juvenis de água doce, durante a fase de engorda, oriundos de uma piscicultura da Zona da Mata (MG), na região sudeste do Brasil, além das lesões de brânquias, causadas tanto macro quanto microscopicamente. Um total de 172 peixes juvenis de diferentes espécies (Oreochromis niloticus, Ictalurus punctatus, Ctenopharyngodon idella, Cyprinus carpio, Astyanax bimaculatus e Brycon amazonicus) foram transportados para um laboratório na cidade de São Paulo. Os peixes foram anestesiados e eutanasiados para análise parasitológica. Todos os peixes estavam acometidos por pelo menos uma espécie de parasito. Dez diferentes espécies de parasitos foram identificadas: Apiosoma sp., Epistylis sp., Ichthyobodo sp., tricodinídeos, Piscinoodinium pillulare, Ichthyophthirius multifiliis, Tetrahymena sp., monogeneas, metacercárias de Centrocestus formosanus e Dermocystidium sp. As boas práticas de manejo e inadequado controle sanitário também foram discutidos.

Abstract This study identified the parasitic species in juvenile freshwater finfishes during the fattening stage, from a fish farm located in the Zona da Mata (MG), southeastern Brazil, and revealed both macro and microscopical lesions in fish gills. A total of 172 juvenile fishes of different species (Oreochromis niloticus, Ictalurus punctatus, Ctenopharyngodon idella, Cyprinus carpio, Astyanax bimaculatus and Brycon amazonicus) were transported to a laboratory in São Paulo city. The fish were sedated and then euthanized for parasitological analysis. All fish were infected by at least one parasite species. Ten different species of parasites were identified: Apiosoma sp., Epistylis sp., Ichthyobodo sp., trichodinids, Piscinoodinium pillulare, Ichthyophthirius multifiliis, Tetrahymena sp., monogeneans, Centrocestus formosanus metacercariae, and Dermocystidium sp. The best management practices and lack of sanitary control were also discussed.

Resumo: Objetivo Foi avaliado o incremento da decomposição da matéria orgânica em amostras de sedimento de viveiros de criação de tilápias-do-Nilo com a introdução das bactérias Bacillus subtilis e B. licheniformis. Métodos Amostras de sedimento colocadas em caixas plásticas de 18L, receberam dose única de inóculos com as seguintes concentrações: 1,21 x 106 UFC g-1 (equivalente a 75 g ha-1), 2,41x 106 UFC g-1 (equivalente a 150 g ha-1), 4,82 x 106 UFC g-1 (equivalente a 300 g ha-1) e 1,61x 107 UFC g-1 (equivalente a 1000 g ha-1), além de um tratamento controle, com adição apenas de solução salina. Foram analisados teor de matéria orgânica, teores de carbono orgânico total (COT) e oxidável (COO), nitrogênio total (NT), relações de carbono: nitrogênio (COT:N e COO:N), teor de argila, pH em água, índce Shoemaker, McLean, Pratt (índice SMP), teores de fósforo (P), de potássio (K), de cálcio (Ca) e de magnésio (Mg), acidez potencial (H+Al), capacidade de troca catiônica (CTC) a pH 7,0 e saturação por bases (V). Resultados Os valores de MO apresentaram diferença significativa entre os menores valores (tratamentos 75, 150 e 300 g ha-1) e o maior valor (tratamento controle). COT, COO, NT e suas relações (COT:N e COO:N) apresentaram diferenças significativas entre os valores médios do tratamento controle e os demais tratamentos. Conclusões A adição das bactérias Bacillus subtilis e B. licheniformis incrementou a taxa de decomposição de matéria orgânica em amostras de sedimento de viveiro de criação de tilápias-do-Nilo.

Abstract: Aim The increment of decomposition of organic matter in sediment samples from Nile tilapia farms was evaluated with the introduction of Bacillus subtilis and B. licheniformis bacteria. Methods Sediment samples placed in 18L plastic boxes received single dose inoculum with the following concentrations: 1.21 x 106 CFU g-1 (equivalent to 75 g ha-1), 2.41 x 106 CFU g-1 (equivalent to 150 g ha-1), 4.82 x 106 CFU g-1 (equivalent to 300 g ha-1) and 1.61 x 107 CFU g-1 (equivalent to 1000 g ha-1), in addition to a control treatment with saline solution only. Organic matter content, total organic carbon (TOC) and oxidizable (OOC), total nitrogen (TN), ratios (TOC: N and OOC: N), clay content, pH in water, Shoemaker, McLean, Pratt index (SMP Index), phosphorus (P), potassium (K), calcium (Ca) and magnesium (Mg) contents, potential acidity (H + Al), cation exchange capacity (CEC) at pH 7.0, base saturation (V) and sum of bases (S). Results The values of OM showed significant difference, between the lowest values (treatments 75, 150 and 300 g ha-1) and the highest value, (control treatment). TOC, OOC, NT and their relationships (TOC: N and OOC: N) showed significant differences between the mean values of the control treatment and the other treatments. Conclusions The addition of Bacillus subtilis and B. licheniformis bacteria increased the decomposition rate of organic matter in sediments samples from Nile tilapia farms.

Resumo Este estudo avaliou a fauna parasitária de híbridos tambacu (Colossoma macropomum × Piaractus mesopotamicus) e patinga (P. mesopotamicus × Piaractus brachypomus) e a interação hospedeiro-parasito-ambiente durante as estações quente e fria em duas pisciculturas localizadas no estado de Mato Grosso do Sul, Brasil, América do Sul. Foram examinados um total de 120 peixes, sendo 60 peixes por espécie. A qualidade da água foi mensurada semanalmente para avaliar a interação entre condições ambientais e parasitismo. Foram encontradas quinze espécies de parasitos: Ichthyophthirius multifiliis, Chilodonella hexasticha e Trichodina sp. (Protozoa); Henneguya piaractus, Myxobolus colossomatis e Myxobolus cuneus (Myxozoa), Anacanthorus penilabiatus, Mymarothecium boegeri, Mymarothecium viatorum e Notozothecium janauachensis (Monogenoidea), Goezia spinulosa e Goezia sp. (Nematoda), Echinorhynchus jucundus (Acanthocephala), Dolops carvalhoi eLernaea cyprinacea (Crustacea). O protozoário ciliado I. multifiliis e helmintos monogenoides foram os parasitos mais prevalentes nas estações fria e quente para ambos os híbridos nos diferentes sistemas de cultivo, produção intensiva comercial e pesca esportiva. Observou-se que os híbridos de serrasalmídeos são mais suscetíveis aos parasitos e abrigam alta diversidade parasitária em relação às espécies parentais, C. macropomum, P. mesopotamicus e P. brachypomus em ambiente de cultivo com predominância de ectoparasitos.

Abstract This study evaluated the parasitic fauna of hybrids tambacu (Colossoma macropomum × Piaractus mesopotamicus) and patinga (P. mesopotamicus × Piaractus brachypomus) and their host-parasite-environment interaction during the warm and cold seasons in two fish farms located in the State of Mato Grosso do Sul, Brazil, South America. A total of 120 fish, being 60 fish for species (30 in the warm season and 30 in the cold season) were examined. Water quality was measured weekly to evaluate the interaction between environmental conditions and parasitism. Fifteen species of parasites were found: Ichthyophthirius multifiliis, Chilodonella hexasticha e Trichodina sp. (Protozoa), Henneguya piaractus, Myxobolus colossomatis and Myxobolus cuneus (Myxozoa); Anacanthorus penilabiatus, Mymarothecium boegeri, Mymarothecium viatorum, and Notozothecium janauachensis (Monogenoidea), Goezia spinulosa and Goezia sp. (Nematoda), Echinorhynchus jucundus (Acanthocephala), and Dolops carvalhoi, Lernaea cyprinacea (Crustacea). The ciliate protozoan I. multifiliis and monogenoids were the most prevalent parasites in the cold and warm seasons for both hybrids in the different culture systems intensive production and sportive fishing. It was observed that the serrasalmid hybrids are more susceptible to parasites and harbor high diversity of parasites in relation to the parental species C. macropomum, P. mesopotamicus and P. brachypomus, with a predominance of ectoparasites.

Resumo Mais de 1.450 espécies de peixes marinhos são comercializados como peixes ornamentais e, aproximadamente, 90% são extraídos na natureza. O peixe borboleta Rainford, Chaetodon rainfordi (McCulloch, 1923), é endêmico no Pacífico Ocidental. Esta espécie de peixe pertence a uma das principais famílias importadas para o Brasil. Haliotrema é um gênero da família Dactylogyridae. Espécies desse gênero têm associação com hospedeiros e distribuição geográfica. O objetivo deste estudo é relatar a ocorrência de Haliotrema aurigae exótico, em peixes-borboleta doentes, que foram importados para o Brasil. Um peixe-borboleta Rainford, suspeito de estar infectado com doença parasitária, foi eutanasiado e submetido à raspagem de pele e brânquias. As amostras foram montadas em lâminas semipermanentes com Hoyer, para análise morfológica de estruturas esclerotizadas. A morfologia e as estruturas esclerotizadas duras das amostras foram comparadas e identificadas como Haliotrema aurigae (Yamaguti, 1968). Esses resultados enfatizam a importância da implementação de medidas de controle pós-fronteira para evitar a introdução de parasitas exóticos no país. Além disso, pode-se enfatizar que a fácil adaptação do parasita a um novo ambiente é uma preocupação para os pscicultores ornamentais.

Abstract More than 1.450 marine fish species are commercialized as ornamental fish and approximately 90% of these are wild-caught. Rainford's butterflyfish, Chaetodon rainfordi (McCulloch, 1923), is endemic to the western Pacific. This fish species belongs to one of the main ornamental fish families imported into Brazil. Haliotrema is a genus of the family Dactylogyridae. Species of this genus have very interesting host associations and geographic distributions. The aim of this study was to report on occurrences of exotic Haliotrema aurigae in diseased Rainford butterflyfish that were imported to Brazil. One Rainford’s butterflyfish with suspected parasitic disease was euthanized and subjected to skin and gill scraping. The parasite specimens were mounted on semi-permanent slides with Hoyer’s solution for morphological analysis of sclerotized structures. The hard-sclerotized structures of these specimens were compared and were identified as those of Haliotrema aurigae (Yamaguti, 1968). These results emphasize the importance of implementation of correct control measures at the country’s borders, to avoid the introduction of exotic parasites. In addition, it can be emphasized that the easy adaptation of this parasite to new environments is a concern for ornamental fish farmers.

Resumo Scuticociliatose, causada pelo protozoário ciliado oportunista, é responsável por significativas perdas econômicas em peixes marinhos ornamentais. O estudo relata a ocorrência de Uronema spp., parasitando dez espécies de peixes de recife em um distribuidor de peixes ornamentais: “Blue green damselfish” (Chromis viridis), “Vanderbilt’s Chromis” (Chromis vanderbilti), “Pennant coralfish” (Heniochus acuminatus), “Threespot angelfish” (Apolemichthys trimaculatus), “Goldspotted angelfish” (Apolemichthys xanthopunctatus), “Sea goldie” (Pseudanthias squamipinnis), “Orchid dottyback” (Pseudochromis fridmani), “Threadfin butterflyfish” (Chaetodon auriga), “Vagabond butterflyfish” (Chaetodon vagabundus), e “Bluecheek butterflyfish” (Chaetodon semilarvatus). Peixes doentes apresentaram distúrbios como hemorragias e lesões ulcerativas na superfície do corpo. A análise histopatológica do músculo, fígado, intestino, rim, baço, brânquias e estômago revelou hemorragias e degeneração das fibras musculares, degeneração vacuolar de hepatócitos, processo inflamatório e granuloma no fígado, atrofia das vilosidades intestinais, processo inflamatório e granuloma no rim, centros de melanomacrófagos e processo inflamatório no baço, hiperplasia das células epiteliais, bem como formação de granuloma nas brânquias e degeneração vacuolar e eosinófilos no estômago. Devido à gravidade da doença, é necessário implementar medidas de biossegurança com diagnóstico rápido e preciso para minimizar o risco de perdas econômicas causadas por Uronema spp.

Abstract Scuticociliatosis, caused by an opportunistic ciliate protozoan, is responsible for significant economic losses in marine ornamental fish. This study reports the occurrence of Uronema spp., parasitizing ten species of marine reef fish at an ornamental fish wholesaler: Blue green damselfish (Chromis viridis), Vanderbilt’s Chromis (Chromis vanderbilti), Pennant coralfish (Heniochus acuminatus), Threespot angelfish (Apolemichthys trimaculatus), Goldspotted angelfish (Apolemichthys xanthopunctatus), Sea goldie (Pseudanthias squamipinnis), Orchid dottyback (Pseudochromis fridmani), Threadfin butterflyfish (Chaetodon auriga), Vagabond butterflyfish (Chaetodon vagabundus), and Bluecheek butterflyfish (Chaetodon semilarvatus). Diseased fish showed disorders such as hemorrhages and ulcerative lesions on the body surface. Histopathological analysis of the muscle, liver, gut, kidney, spleen, gills, and stomach revealed hemorrhages and degeneration of muscle fiber, vacuolar degeneration of hepatocyte, inflammatory process and granuloma in the liver, atrophy of intestinal villi, inflammatory process and granuloma in the kidney, melanomacrophage centers, as well as inflammatory process in the spleen, epithelial cells hyperplasia and granuloma formation in the gills, and vacuolar degeneration and eosinophils in the stomach. Due to the severity of the disease, it is necessary to implement biosecurity measures with rapid and accurate diagnosis to minimize the risk of economic losses caused by Uronema spp.

Resumo Cryptocaryon irritans é um protozoário ciliado parasita obrigatório de peixes marinhos selvagens e cultivados. Causa a doença dos “pontos brancos”. Infecções por esse patógeno pode causar perdas significativas para aquaristas e para marinocultura comercial em todo o mundo. Este estudo relata a ocorrência de C. irritans parasitando a espécie de peixe ornamental de recife Zebrasoma xanthurum. Seis dias após serem introduzidos no novo ambiente, onze peixes apresentaram pontos brancos espalhados pelo corpo e nadadeiras. Suspeitando de infecção por C. irritans foi realizado raspado de pele para confirmar o diagnóstico clínico. Após a confirmação, os peixes foram transferidos para um aquário hospital e tratados com o agente terapêutico Seachem Cupramine® por 15 dias. Durante o período de tratamento o monitoramento da concentração de cobre foi diário através da utilização de um fotômetro. Após o final do tratamento, todos os peixes já não apresentavam mais sinais clínicos de pontos brancos pelo corpo, e foram negativos no raspado de pele, sendo foram encaminhados para comercialização.

Abstract Cryptocaryon irritans is an obligate parasitic ciliate protozoan of wild and cultured marine fish. It causes white spot disease, and infections with this pathogen can cause significant losses for aquarists and commercial marine cultures worldwide. This study reports the occurrence of C. irritans parasitizing the ornamental reef fish, yellowtail tang, Zebrasoma xanthurum. Six days after being introduced to a new environment, 11 yellowtail tangs had white spots scattered across their bodies and fins. Suspicion of infection with C. irritans was evaluated by scraping the skin to confirm clinical diagnosis. After confirmation, the yellowtail tangs were transferred to a hospital aquarium and treated with the therapeutic agent Seachem Cupramine® for 15 days. During the treatment period, the copper concentration was monitored daily. At the end of the treatment, none of the yellowtail tangs showed clinical signs of white spots on their bodies, and skin scraping confirmed the yellowtail tangs were no longer infected. Subsequently, the yellowtail tangs were released for sale.

ABSTRACT This study aimed to evaluate the hematological profile and relative condition factor of naturally parasitized Nile tilapia from 12 fish farms in southern Brazil. A total of 240 fish were collected from four regions of the state of Santa Catarina. They were anesthetized, and the blood was collected by puncturing the caudal vein. Subsequently, the fish were necropsied for parasitological analysis. The results were compared with the Kruskal-Wallis test, and the correlations were verified with the Spearman test (P ˂ 0.05). The parasitological analysis revealed high infestations by Ichthyophthirius multifiliis on the fish gills of the south and west regions. The presence of Trichodina magna and Trichodina compacta was observed in low intensity on the body surface of fish from all facilities. Trichodina spp. was found in high intensity parasitizing the fish gills of northern, Itajaí valley and western regions, nonetheless, in those regions, the presence of Monogenea, Cichlidogyrus sclerosus, and Cichlidogyrus halli, on fish gills was observed. The hematological profile presented altered parameters, featuring monocytosis and thrombocytopenia, possibly associated with high parasitic intensities. The observed correlations suggest that neutrophils and monocytes are directly involved in an organic defense against parasites. Monocytes are mainly targeted in the defense against I. multifiliis.

Resumo Neobenedenia melleni é conhecido como um parasito letal para peixes teleósteos ornamentais marinhos capturados se não tratado. Este estudo relata a ocorrência de N. melleni parasitando quatro espécies de peixes ornamentais importados para o Brazil: Arabian angelfish (Pomacanthus asfur), yellowbar angelfish (Pomacanthus maculosus), regal angelfish (Pygoplites diacanthus) e bluecheek butterflyfish ( Chaetodon semilarvatus). Dez dias depois do início da quarentena, foram observados irritabilidade e opacidade na córnea dos peixes que, foram diagnosticadas com N. melleni na superfície corporal. Todos os peixes foram tratados com duas aplicações de praziquantel 2 mg L-1 em intervalo de quatro dias. Sete dias após o início do tratamento, um novo exame parasitológico foi realizado o qual não revelou a presença dos parasitos, sendo liberados para comercialização.

Abstract The capsalid monogenean Neobenedenia melleni is known as a lethal pathogen for captured marine teleost ornamental fish, if left untreated. This study reports the occurrence of N. melleni parasitizing four species of ornamental reef fish imported into Brazil and maintained in quarantine: Arabian angelfish (Pomacanthus asfur ), yellowbar angelfish (Pomacanthus maculosus), regal angelfish (Pygoplites diacanthus), and bluecheek butterflyfish (Chaetodon semilarvatus). Ten days after the beginning of quarantine, some fish showed behavioral alterations, such as irritability, and corneal opacity, which were rapidly diagnosed to be caused by monogenean parasites by body surface scraping. The fish from the same batch were treated with two applications of 2 mg L-1 praziquantel each at an interval of four days. Seven days after the first treatment, the mucus surface of the fish was re-examined, which did not reveal the parasites presence being delivered for commercialization.

Resumo A infecção pelo acantocéfalo Neoechinorhynchus buttnerae é considerada uma das preocupações mais importantes para produção de tambaqui (Colossoma macropomum). Estratégias de tratamento têm sido o foco de vários estudos in vivo ; entretanto, poucos estudos foram realizados em protocolos in vitro para manutenção do parasito. O objetivo deste estudo foi desenvolver a melhor condição de cultura in vitro para N. buttnerae para garantir sua sobrevivência e adaptação fora do hospedeiro, a ser utilizado para teste com substâncias no controle do parasito. Para isso, os parasitos foram coletados de peixes naturalmente infectados e distribuídos em placas de cultura de 6 poços sob os seguintes tratamentos em triplicata: 0.9% NaCl, água estéril do tanque, meio de cultura L-15 Leibovitz, meio de cultura L-15 Leibovitz + ágar 2%, meio de cultura RPMI 1640, e meio de cultura RPMI 1640 + ágar 2%. As placas contendo os parasitos foram mantidos a 24 °C, 28 °C, e 32 °C. O meio de cultura RPMI 1640 + ágar 2% apresentou a melhor sobrevivência de 24 dias a 24 °C. Nenhuma alteração corporal tais como inchaço dos parasitos, deformação corporal, desidratação e endurecimento foram observados no meio de cultura RPMI 1640 + ágar 2%.

Abstract Infection by the acantocephalan Neoechinorhynchus buttnerae is considered one of most important concerns for tambaqui fish (Colossoma macropomum ) production. Treatment strategies have been the focus of several in vivo studies; however, few studies have been undertaken on in vitro protocols for parasite maintenance. The aim of the present study was to develop the best in vitro culture condition for N. buttnerae to ensure its survival and adaptation out of the host to allow for the testing of substances to be used to control the parasite. To achieve this, parasites were collected from naturally infected fish and distributed in 6-well culture plates under the following treatments in triplicate: 0.9% NaCl, sterile tank water, L-15 Leibovitz culture medium, L-15 Leibovitz + agar 2% culture medium, RPMI 1640 culture medium, and RPMI 1640 + agar 2% culture medium. The plates containing the parasites were maintained at 24 °C, 28 °C, and 32 °C. The RPMI 1640 + agar 2% culture medium showed the best survival of 24 days at 24 °C. No body alterations such as swollen parasites, body deformation, dehydration and hardening were observed in the RPMI 1640 + 2% culture medium.

Resumo O objetivo deste estudo foi avaliar o impacto do parasitismo por metazoários sobre os parâmetros hematológicos, bioquímicos e fator de condição relativo de tambaquis (Colossoma macropomum) cultivados na região Norte do Brasil. Foram coletados 32 tambaquis em fase de engorda numa piscicultura comercial no município de Rio Preto da Eva, estado do Amazonas, Brasil. A prevalência de parasitos foi de 100% para Anacanthorus spathulatus, Mymarothecium boegeri e Notozothecium janauachensis, 100% para Neoechinorhynchus buttnerae e 53,13% para Dolops geayi. A maior intensidade média foi de acantocéfalos, seguida por monogenéticos e branquiúros. Correlação negativa significativa foi observada entre abundância de N. buttnerae e os parâmetros de hematócrito, concentração de hemoglobina, trombócitos totais e glicose, bem como entre a abundância de monogenéticos e glicose. A infecção por parasitos metazoários foi capaz de debilitar os tambaquis como observado por meio das alterações hematológicas; sendo este quadro de anemia hipocrômica e trombocitopenia importante para ser utilizado no diagnóstico destas parasitoses. Este estudo foi o primeiro registro da ocorrência de Dolops geayi em tambaquis cultivados na Amazônia.

Abstract The aim of this study was to evaluate the impacts of metazoan parasites on hematological and biochemical parameters and relative condition factor of tambaqui (Colossoma macropomum ) farmed in northern Brazil. A total of 32 juvenile fish were captured from a commercial fish farm located in the municipality of Rio Preto da Eva, Amazonas state, Brazil. Parasite prevalence was 100% for Anacanthorus spathulatus, Mymarothecium boegeri and Notozothecium janauachensis, 100% for Neoechinorhynchus buttnerae and 53.13% for Dolops geayi. The greatest mean parasite intensity was found in acantocephalans followed by monogeneans and branchiuran crustaceans. A negative correlation was observed between abundance of N. buttnerae and hematocrit percentage, hemoglobin concentration, total thrombocyte count and glucose and between abundance of the monogenean and glucose concentration. Parasitic infections caused damage in tambaqui in terms of the observed hematological parameters that were characterized by hypochromic anemia and thrombocytopenia, which are important parameters to be used in parasitic diagnosis. This study is the first record of the occurrence of Dolops geayi in farmed tambaqui in the Amazon.

RESUMO: A ictiofitiríase em peixes tem distribuição mundial e forte impacto econômico na piscicultura de água doce devido às altas taxas de mortalidade associadas. Este estudo avaliou o uso do peróxido de hidrogênio (H2O2) e do dióxido e cloro (ClO2) no controle do parasito protozoário Ichthyophthirius multifiliis em jundiá. A concentração letal mediana (CL50, 96h) de cada agente químico foi estabelecida, assim como a concentração mínima inibitória do peróxido de hidrogênio para a fase larval infecciosa do parasito (teronte). Os agentes foram testados assincronicamente em alevinos parasitados na forma de banhos de curta e longa duração, nas seguintes concentrações e tempos de exposição: 1. Peróxido de hidrogênio: (T1) banho contínuo - 30ppm e (T2) 50ppm; (T3) banho curto - 150ppm com duração de 1h e (T4) 250ppm com duração de 1h, além de um grupo controle (sem a adição do agente químico). 2. Dióxido de cloro: (T1) banho contínuo - 4ppm; (T2) banho contínuo - 20ppm; (T3) banho curto - 200ppm, com duração de 1min; (T4) banho curto - 400ppm, com duração de 1min, além de um grupo controle. A análise dos dados indicou a concentração de 82,54ppm do produto comercial utilizado (ou 24,76ppm do princípio ativo) como a Cl50, 96h de H2O2 e de 38,4ppm produto (ou 2,68ppm do princípio ativo) para o ClO2. A concentração de H2O2 que causou 100% de mortalidade dos terontes em 1h foi de 25ppm (do produto, ou 7,5ppm do princípio ativo). Ao final do quarto dia de experimento curativo, 98% dos animais morreram devido à ictiofitiríase. Nenhum dos tratamentos foi efetivo frente à parasitose.

ABSTRACT: Ichthyophthiriasis is a worldwide fish disease with great financial impact on freshwater fish farming due to its associated high mortality rates. Current study assesses the parasiticidal capacity of hydrogen peroxide (H2O2) and chlorine dioxide (ClO2) against the causative agent, Ichthyophthirius multifiliis, in jundiá. Median lethal concentration (LC50, 96h) of each chemical agent was established, as well as the minimum inhibitory concentration of hydrogen peroxide for the parasite´s infectious larval phase (theront). Products were tested asynchronously in parasitized fingerlings for short and long baths at the following concentrations and exposure times: 1. Hydrogen peroxide: (T1) continuous bath - 30ppm and (T2) 50ppm; (T3) short bath - 150ppm, during 1h and (T4) 250ppm during 1h; control group (without any chemical agent). 2. Chlorine dioxide: (T1) continuous bath - 4ppm and (T2) 20ppm; (T3) short bath - 200ppm, during 1min; (T4) short bath - 400ppm, during 1min and control group. Data analysis demonstrated a concentration of 82.54ppm of the commercial product (or 24.76ppm of the active chemical agent) as LC50, 96h of H2O2 and 38.4ppm product (or 2.68ppm of the active chemical agent) for ClO2. Hydrogen peroxide concentration causing 100% mortality rate of theronts in 1h was 25ppm (product, or 7.5ppm of the active chemical agent). At the end of the fourth day of curative experiment, 98% of the animals died by ichthyophthiriasis. No treatment was effective against the parasite.

RESUMO: Os peixes, seja em ambiente natural ou em piscicultura, sofrem efeito do estresse na captura, manejo, transporte ou tratamentos que provocam baixo desempenho zootécnico. Anestésicos, como o eugenol, obtido a partir do óleo de cravo, tem sido usado estrategicamente não só em peixes de água doce, mas também em peixes marinhos e estuarinos para reduzir o estresse. Apesar da indicação de eugenol como anestésico em peixes e da sua baixa toxicidade para estes animais, não se tem clareza da sua ação sobre o ambiente. Bioensaios ou testes de ecotoxicidade com organismos indicadores são usados em para avaliar o modo de ação de poluentes no ambiente. O objetivo deste estudo foi testar a toxicidade aguda do eugenol utilizando o microcrustáceo Daphnia magna e a bactéria Aliivibrio fischeri e sua toxicidade crônica para alga Desmodesmus subspicatus. O eugenol, nas concentrações de 50, 75 e 100mg L-1, foi tóxico para organismos indicadores testados.

ABSTRACT: Fish in both natural and farming conditions are exposed to stress of capture, handling, transport or treatment that provoke low zootechnical performance. Anesthetics like eugenol obtained from clove oil have been used strategically not only in freshwater but also in marine and estuarine fish in order to reduce the stress. Apart from the eugenol indication as anesthetic and its low toxicity for animals, its environment action is not clear. Bioassays or ecotoxicity tests with indicator organisms are used to evaluate the mode of action of the pollutants in the environment. The aim of this study was to test the acute toxicity of eugenol using the microcrustacean Daphnia magna and the bacterium Aliivibrio fischeri, and also its chronic toxicity for the algae Desmodesmus subspicatus. Eugenol in the concentrations of 50, 75 and 100mg L-1 were toxic to tested indicator organisms.
