Objetivou-se avaliar a reação de 55 genótipos de feijão comum da coleção nuclear da Embrapa Arroz e Feijão à raça 3/Tipo HG 0 de H. glycines. RESUMO Vários nematoides têm sido relatados causando danos ao feijoeiro comum, sendo o principal deles o das galhas (Meloidogyne incognita). Objetivou-se avaliar a reação de 20 genótipos de feijão frente à infestação por M. incognita, em experimento conduzido em casa-de-vegetação e delineamento inteiramente casualizado. As plantas foram inoculadas com 4.000 ovos + J2 deM. incognita e, aos 60 dias após a inoculação, retiradas para avaliação da massa fresca da raiz e da parte aérea, nematoides por planta e por grama de raiz e fator de reprodução. Os genótipos BGF0011762, BGF0011854, BGF0011861, BGF0011862, BGF0011987, BGF0012533, BGF0013294, BGF0013355, BGF0013875 e BGF0013955 foram classificados como resistentes. ABSTRACT Several nematodes have been reported to damage common bean, being the main one the root-knot nematode (Meloidogyne incognita). This study aimed to evaluate the reaction of 20 common bean genotypes against the infestation of M. incognita, in an experiment conducted under greenhouse conditions, in a completely randomized design. The plants were inoculated with 4,000 eggs + J2 of M. incognita and, at 60 days after inoculation, they were removed for evaluation of root and shoot fresh mass, nematodes per plant and per gram of root, and reproduction factor. Resumo: Os microrganismos de solo apresentam uma grande diversidade, envolvendo grupos taxonomicamente distintos que desempenham papel na decomposição da matéria orgânica, ciclagem de nutrientes, agregação dos solos, dentre outros. Nesta diversidade, os fungos do gênero Trichoderma tem apresentado sucesso como agentes de biocontrole de fitopatógenos, como promotores de crescimento de plantas e, ainda, como indutores de resistência de plantas a doenças. Além disso, são importantes na sustentabilidade dos ecossistemas naturais. Com o objetivo de verificar a densidade populacional de fungos do gênero Trichoderma em ambientes naturais e agroecossistemas, em área de Cerrado, amostras de solos e raízes oriundas de vegetação nativa e de sistema de produção agroecológica foram coletadas na região do Distrito Federal, Brasil. Os pontos de coleta foram selecionados aleatoriamente, e cada amostra de solo ou raiz foi acondicionada individualmente. O solo aderido às raízes foi removido para as avaliações. Foram realizadas diluições seriadas das amostras e contagem do número de Unidades Formadoras de Colônias (UFCs) de isolados de Trichoderma. Os resultados mostraram que o número de UFC variou dependendo da planta e da localidade avaliada. A substituição da vegetação nativa por sistemas de cultivo orgânicos não resultou em importante redução neste número.

Abstract: Soil microorganisms present a great diversity, involving taxonomically distinct groups that play a role in the decomposition of organic matter, nutrient cycling, soil aggregation, among others. In this diversity, the fungi of the genus Trichoderma have been successful plant pathogen biocontrol agents, as plant growth promoters and as inducers of plant resistance to diseases. In addition, they are important in the sustainability of natural ecosystems. Aiming to verify the population density of Trichoderma fungi in natural environments and agroecosystems, in Cerrado area, samples of soils and roots from native vegetation and agroecological production system were collected in the Federal District, Brazil. The collection points were randomly selected, and each soil or root sample was individually wrapped. The soil adhered to the roots was removed for evaluations. Serial sample dilutions and number of Colony Forming Units (CFUs) of Trichoderma isolates were performed. The results showed that the number of CFU varied depending on the plant and location evaluated. The replacement of native vegetation by organic farming systems did not result in a significant reduction in this number.

Resumo Antibiose é um mecanismo pelo qual certos microrganismos respondem à presença de outros, secretando compostos ou metabólitos capazes de inibir ou impedir o seu desenvolvimento. O extrato bruto de Trichoderma contém uma mistura de compostos secundários e tem sido utilizado na prospecção deste fungo para o controle biológico e demais fins industriais. Diante da crescente demanda da agricultura por alternativas ecologicamente compatíveis para o manejo de doenças, este trabalho teve como objetivo investigar o espectro de ação de Metabólitos Não Voláteis (MNVs), produzidos por isolados de Trichoderma, contra diferentes fungos fitopatogênicos. O potencial antagônico dos MNVs foi avaliado através do método de incorporação do extrato líquido filtrado em meio BDA. Os ensaios mostraram que todos os MNVs produzidos inibiram de forma semelhante o fungo Sclerotinia sclerotiorum. Por outro lado, os isolados CEN1245 e CEN1274, ambos Trichoderma brevicompactum, mostraram um amplo espectro de ação, atuando contra Sclerotium rolfsii, Colletotrichum gloesporioides, Verticillium dahliae, Fusarium oxysporum e Cylindrocladium sp. O presente estudo descreve isolados que produziram metabólitos não voláteis com amplo espectro de ação antifúngico, assim como patógeno-específico. Os MNVs de Trichoderma spp. obtidos de diferentes amostras de solo cultivadas com vegetais, mandioca e milho, foram eficientes na inibição de fungos fitopatogênicos pertencentes a outros patossistemas, como os de espécies florestais e frutíferas, o que poderia aumentar sua potencial aplicação no controle de doenças de plantas. Adicionalmente, estes fungos antagonistas devem ser mais bem estudados para identificação de moléculas bioativas de interesse industrial ou formulação de produtos para o controle biológico de patógenos de plantas.

Abstract Antibiosis is the mechanism by which certain microorganisms respond to the presence of others, secreting compounds or metabolites capable of inhibiting or impeding their development. The crude extract of Trichoderma contains a mixture of secondary compounds, which may show antibiotic effect, and has been used for the prospect of this fungus for biological control and other industrial purposes. Faced with the increasing demand of agriculture for ecologically compatible alternatives for the management of diseases, this work aimed to investigate the spectrum of action of Non-Volatile Metabolites (NVMs) of Trichoderma isolates against different plant pathogenic fungi. The antagonistic potential of NVMs was evaluated through the incorporation method of the filtered liquid extract in PDA medium. The assays showed that all the NVMs produced inhibited the fungus Sclerotinia sclerotiorum similarly. On the other hand, strains CEN1245 and CEN1274, both belonging to the species Trichoderma brevicompactum, showed broad spectrum against Sclerotium rolfsii, Colletotrichum gloesporioides, Verticillium dahliae, Fusarium oxysporum and Cylindrocladium sp. The present study describes isolates producing non-volatile metabolites with broad spectrum of antifungal action, as well as pathogen-specific. The Trichoderma spp. NVMs obtained from different soil samples cultivated with vegetables, cassava and maize were efficient in inhibiting plant pathogenic fungi belonging to other patossystems, such as forest or fruit, which could increase their potential application in biological control of plant diseases. In addition, these antagonistic fungi should be studied in greater depth for the identification of bioactive molecules of industrial interest or in commercial formulations of products for biological control of plant pathogens.

RESUMO A teca é uma espécie florestal que assumiu grande importância no Brasil, onde encontrou excelentes condições de desenvolvimento desde a sua introdução na década de 1960. Entretanto, problemas fitossanitários começam a ameaçar a exploração desta espécie madeireira. Um exemplo é o cancro da teca, causado pelo fungo Lasiodiplodia theobromae (Lt), cuja etiologia foi recentemente elucidada no Brasil e, portanto, ainda não existem métodos de controle recomendados. Sendo assim, o objetivo deste trabalho foi avaliar o controle deste patógeno, investigando o potencial dos agentes de controle biológico (ACB) Trichoderma spp., Bacillus sp. e Enterobacter sp., inicialmente por meio de ensaios in vitro e, posteriormente, com realização de testes in vivo. De acordo com os resultados dos testes in vitro, os isolados de Trichoderma CEN162 e CEN1153 e a estirpe UnB1366 de Bacillus sp., foram os tratamentos que se destacaram, sendo capazes de inibir completamente o crescimento micelial de alguns isolados de Lt. Quando esses isolados foram testados de forma preventiva, observou-se variação nos níveis de controle, dependendo do isolado de Lt e da interação antagonista-clone, onde CEN162 (T. asperellum) e UnB166 (Bacillus sp.) apresentaram 100% de controle. Dessa forma, houve uma correlação positiva entre os testes in vitro e in vivo, uma vez que os mesmos ACB se destacaram. Apesar de ter-se obtido bons níveis de controle com os ACB utilizados, pode-se concluir que existe uma variação no antagonismo a diferentes isolados ou mesmo na interação antagonista-clone, corroborando as informações disponíveis na literatura científica sobre este fungo fitopatogênico.

ABSTRACT Teak is a forest species that has assumed great importance in Brazil, where it has found excellent conditions for development since its introduction into the country in the 1960s. However, phytosanitary problems are beginning to threaten the production of this timber species. An example is teak canker, caused by the fungus Lasiodiplodia theobromae (Lt), which has only recently been reported in Brazil, and for which, therefore, there are no recommended control methods. Thus, this study evaluated the control of this pathogen, investigating the potential of the biocontrol agents (BCAs) Trichoderma spp., Bacillus sp. and Enterobacter sp., initially through in vitro assays and, subsequently, with in vivo tests. According to the in vitro assay results, the Trichoderma isolates CEN162 and CEN1153 and the strain of Bacillus sp. (UnB1366) were the treatments that stood out, as they were able to completely inhibit mycelial growth of some isolates of Lt. When these isolates were tested in a preventive way, the control levels varied depending on the Lt isolate and the antagonist-clone interaction, where CEN162 (T. asperellum) and UnB166 (Bacillus sp.) showed 100% control. Thus, there is a positive correlation between the in vitro and in vivo tests, since the same BCAs stood out. Although good levels of control have been obtained with the BCAs used, it can be concluded that there is a variation in the antagonism to different Lt isolates or even in the antagonist-clone interaction, corroborating the information available in the scientific literature on this plant-pathogenic fungus.

RESUMO: Em 2013, amostras foliares de plantas de gérbera, apresentando sintomas de crestamento, foram coletadas em telados de cultivo de corte, no núcleo rural alexandre gusmão, localizado em brazlândia-df. sete isolados bacterianos obtidos foram submetidos à caracterização fenotípica e molecular, incluindo testes de patogenicidade, testes lopat e sequenciamento parcial do gene 16s rdna. todos os isolados foram gram-negativos, aeróbios, oxidase positivos, produziram pigmento fluorescente, induziram reação de hipersensibilidade em folhas de fumo, utilizaram sorbitol e glutamato, e foram patogênicos a 24 diferentes espécies de plantas. os resultados destes testes e análises de sequências do rdna mostraram 100% de identidade com pseudomonas cichorii. para o nosso conhecimento, este é o primeiro relato de p. cichorii, em gérbera, no distrito federal.

ABSTRACT: In 2013, leaf samples of gerbera plants showing symptoms of bacterial blight were collected in cut-flower high tunnels, in the settlement of núcleo rural alexandre gusmão, located in brazlândia, distrito federal, brazil. seven isolates obtained were subjected to phenotypic and molecular characterization, including pathogenicity tests, lopat, and partial sequencing of the 16s rdna gene. all isolates were gram-negative, aerobic, oxidase-positive, produced fluorescent pigment, induced hypersensitivity in tobacco leaves, used sorbitol and glutamate and were pathogenic to 24 different plant species. results of these tests and analysis of the sequences of rdna showed 100% identity with pseudomonas cichorii. to our knowledge, this is the first report of p. cichorii in gerbera in the federal district.

Resumo Quarenta e nove isolados de Trichoderma obtidos no centro-oeste do Brasil foram avaliados quanto a sua atividade antagônica in vitro contra Sclerotinia sclerotiorum (agente causal do mofo branco) e identificados com base nas sequências ITS do DNA ribossômico nuclear. Os testes de cultivo pareado mostram que todos os isolados exibiram algum antagonismo, com um máximo de 77% de inibiação micelial e inibição total da produção de escleródios. Dois isolados se destacaram como os mais promissores, considerando ambos os parâmetros avaliados (CEN1253 - T. koningiopsis e CEN1265 - T. brevicompactum). Cinco espécies diferentes foram identificadas: T. harzianum (23), T. spirale (9), T. koningiopsis (8), T. brevicompactum (7) and T. asperellum (2). Estes isolados estão armazenados na Coleção de Fungos para Controle Biológico da Embrapa e as informações obtidas nos experimentos serão incorporadas na base de dados de ativos biológicos, no sistema de informações de recursos genéticos, e disponibilizados para estudos futuros.

Abstract Forty-nine isolates of Trichoderma from the Brazilian Midwest were evaluated for their antagonistic activity in vitro against Sclerotinia sclerotiorum (causal agent of white mold), which were then identified based on their nuclear ribosomal ITS sequences. Paired culture tests showed that all isolates exhibited some antagonism, with a maximum of 77% mycelial inhibition and complete inhibition of sclerotia production. Two isolates were found to be the most promising biocontrol agents, considering both antagonistic parameters (CEN1253 - T. koningiopsis and CEN1265 - T. brevicompactum). Five different species were identified: T. harzianum (23), T. spirale (9), T. koningiopsis (8), T. brevicompactum (7) and T. asperellum (2). These isolates are stored in the Embrapa Fungi Collection for Biological Control and the information obtained in the experiments will be incorporated into the database of biological assets within the genetic resources information system (Allele) and be made available for further studies.

Neste estudo, avaliou-se o efeito da microbiolização de sementes com bactérias extremófilas facultativas (Bacillus sp. e Enterobacter sp.), isoladas, em trabalhos anteriores, a partir de condições extremas de pH e NaCl e capazes de levar ao incremento na fitomassa de eucalipto, na germinação de Eucalyptus urophylla S.T. Blake. Para avaliar a germinabilidade, foram mensurados o tempo médio, a velocidade e o coeficiente de velocidade de germinação. O delineamento experimental foi inteiramente casualizado com 11 tratamentos (cinco estirpes de Bacillus sp., cinco de Enterobacter sp. e uma testemunha sem bactéria), formados por oito repetições com 25 sementes cada. Os resultados do estudo indicaram que as estirpes UnB 1366 e UnB 1374 de Bacillus sp. reduziram, significativamente, menor tempo médio e maiores coeficientes e velocidades de germinação em relação às demais estirpes.

In this study the effect of seed microbiolization with facultative extremophile bacteria (Bacillus sp. and Enterobacter sp.), isolated in previous work, from extreme conditions of pH and NaCl and able to increase eucalyptus phytomass, on seed germinability of . Eucalyptus urophylla S.T. Blake was evaluated. Mean germination time, speed of germination and coefficient of speed germination were measured to evaluate germinability. The experiment was conducted in a completely randomized design including 11 treatments (five strains of Bacillus sp., five of Enterobacter sp. and one control without bacteria), formed by 8 replicates with 25 seeds each. The results showed that Bacillus sp. strains UnB 1366 and UnB 1374 significantly reduced the lower mean germination time and higher coefficients and speed of germination in relation to the other strains.

A murcha bacteriana do eucalipto (Raça 3 biovar 2T de Ralstonia solanacearum) foi descrita em 2009 em campo com o híbrido de eucalipto urograndis. O objetivo deste trabalho foi avaliar a virulência de duas estirpes de R. solanacearum a dezessete espécies de Eucalyptus, através do teste de microbiolização de sementes. A análise estatística dentre espécies mostrou que a virulência variou entre as estirpes estudadas. A virulência da R3bv2T foi significativamente maior em: urograndis, E. urophylla, E. grandis x E. camaldulensis, E. grandis, E. microcorys, E. deanei e E. phaeotricha; e maior para R1bv1 somente na espécie E. pellita. Entre espécies de eucalipto, observou-se maior suscetibilidade em "urograndis", E. grandis e E. paniculata para R3bv2T; e em E. cloeziana, E. paniculata, E. botryoides, E. pellita, E. propinqua e E. resinifera devido a R1bv1.

The bacterial wilt of Eucalyptus (Ralstonia solanacearum Race 3 biovar 2T) was reported in field of Eucalyptus hybrid urograndis. The objective of this research was to evaluate the virulence of two R. solanacearum strains to 17 Eucalyptus species, using the seed microbiolization method for pathogen inoculation. The results of the statistical analyses showed that virulence varied between the two R. solanacearum strains. The R3bv2T virulence was higher than the one in R1bv1 in the following Eucalyptus species: urograndis, E. urophylla, E. grandis x E. camaldulensis, E. grandis, E. microcorys, E. deanei, and E. phaeotricha. The virulence of the R1bv1 strain was higher only in E. pellita. Among species of Eucalyptus the following ones were more susceptibles: to R3bv2T strain - urograndis, E. grandis and E. paniculata, and to R1bv1 strain - E. cloeziana, E. paniculata, E. botryoides, E. pellita, E. propinqua and E. resinifera.

Atualmente, a árvore mais plantada no Brasil é o Eucalyptus spp., ocupando 81,6% das florestas plantadas. Sua produtividade em 2009 foi de 44,2 m³ de eucalipto com casca/ha, gerando aproximadamente 46.850 empregos diretos. Estudos de bactérias benéficas, como as Rizobactérias Promotoras de Crescimento de Plantas (RPCPs), vêm sendo desenvolvidos há mais de um século. Este trabalho objetivou avaliar estirpes de bactérias extremófilas facultativas que possuam potencial na promoção de crescimento do eucalipto. As sementes do híbrido "urograndis" foram microbiolizadas com uma suspensão de 10(9) UFC/mL das 10 estirpes bacterianas, através da agitação a 150 rpm, em incubador rotativo a 28 ºC, por 24 h. Em seguida, foram plantadas em sementeiras e mantidas em casa de vegetação. Após 60 dias, avaliou-se o peso de matéria seca da parte aérea e das raízes. O peso de matéria seca da parte aérea revelou que todas as estirpes bacterianas resultaram em ganhos quando comparado com o da testemunha, variando entre 11,3 e 78,0%. Entretanto, as estirpes UnB 1366, UnB 1371, UnB 1375, UnB 1370 e UnB 1373 foram as que se diferiram significativamente. Em contrapartida, a estirpe UnB 1368 (Bacillus sp.) destacou-se individualmente no incremento (130,0%) da biomassa radicular. Tais estirpes devem ser mais bem estudadas, quanto a formas de veiculação, combinações, formulações etc., para que possam ser utilizadas na otimização da produção de mudas.

Currently, Eucalyptus spp. is the tree that is most widely planted in Brazil, occupying 81.6% of planted forests. In 2009 its productivity was 44.2 m³ of eucalyptus with bark/hectare, generating about 46,850 direct jobs. Studies of beneficial bacteria, such as plant growth-promoting rhizobacteria (PGPRs), have been carried out for more than a century. The objective of this work was to evaluated strains of facultative extremophile bacteria that show potential in promoting eucalyptus growth. Seeds from the hybrid "urograndis" were microbiolized with a suspension of 10(9) UFC/mL from 10 bacterial strains, by means of shaking at 150 rpm in a rotating incubator, at 28 ºC for 24 h. Next, they were planted in seed-trays and kept in the greenhouse. After 60 days seed germination and dry weight of the aerial part and roots were evaluated.. The dry weight of the aerial part revealed that all the bacterial strains produced gains when compared to the control, varying from 11.3 to 78.0%. However, strains UnB 1366, UnB 1371, UnB 1375, UnB 1370 and UnB 1373 were the ones that differed significantly. In contrast, the UnB 1368 strain (Bacillus sp.) stood out individually in promoting an increase of 130.0% in root biomass. These strains should be more thoroughly studied with regard to vehicle, combinations, formulations and others, so that they may be used in optimizing seedling production.

The objective of this study was to characterize isolates of biovar 2 of Ralstonia solanacearum pathogenic to eucalyptus "urograndis" hybrid by means of biochemical tests, as well as evaluation of host range and identification by PCR with primers for species, biovar and phylotype. Carbohydrate utilization assays revealed that the eucalyptus isolates belong to phenotype Tropical of biovar 2 (2T). From artificial inoculations it was possible to reproduce symptoms or recover the bacterium from: eucalyptus, potato, tomato, eggplant, datura, geranium, turnip, mustard, nasturtium, beetroot, sunflower, bean, French marigold, horseradish tree, and cashew. The PCR assays confirmed that the isolates belong to biovar 2, phylotype II of the bacterium. The confirmation of new biovar naturally infecting eucalyptus indicates that R. solanacearum easily adapts to new hosts.

O cancro bacteriano, causado por Xanthomonas campestris pv. viticola, afeta o cultivo irrigado de videira no Vale do Submédio São Francisco, nos estados de Pernambuco e Bahia. O manejo da doença tem sido realizado através de um conjunto de práticas que inclui a aplicação de cúpricos. Este tem sido o único método de controle químico disponível e o mais utilizado nas áreas de ocorrência da doença. O objetivo deste estudo foi avaliar a sensibilidade ao cobre de estirpes de X. campestris pv. viticola coletadas em diferentes localidades, entre os anos de 1998 a 2006. Detectou-se variabilidade na sensibilidade ao oxicloreto de cobre e ao sulfato de cobre entre as 21 estirpes testadas. A concentração mínima inibitória variou entre 10 e 60 µg/mL Cu2+, para os dois produtos. De forma geral, observouse uma evolução no crescimento da tolerância ao cobre ao longo dos anos, com as estirpes brasileiras apresentando maior tolerância que a estirpe-tipo, coletada em 1972 na Índia. As diferenças observadas em sensibilidade ao cobre entre as estirpes de X. campestris pv. viticola podem levar, com o uso contínuo de compostos cúpricos na região, à seleção e dominância das estirpes mais tolerantes na população bacteriana.

Bacterial canker, caused by Xanthomonas campestris pv. viticola, affects grapevines in the irrigated areas of the São Francisco river valley, in the states of Pernambuco and Bahia. Several practices for disease management have been adopted including copper sprays. This is the only available chemical control method and most frequently used in the areas affected by the disease. The objective of this work was to determine the sensitivity to copper of strains of X. campestris pv. viticola collected in different locations and over a period of years, from 1998 to 2006. Variation in sensitivity to copper oxychloride and copper sulfate was observed among the 21 strains tested. The minimum inhibitory concentration (MIC) varied from 10 to 60 µg/mL Cu2+, for both compounds. A general increase in copper tolerance over the years was also observed, with the Brazilian strains being more tolerant than the type-strain, collected in 1972 in India. The differences observed in copper sensitivity may lead to the selection and dominance of the more tolerant strains in the bacterial population as copper compounds continue to be used in the region.

Com o objetivo de desenvolver um método molecular para detecção e identificação de Xanthomonas campestris pv. viticola (Xcv), agente causal do cancro bacteriano da videira, oligonucleotídeos (primers) foram desenhados com base na seqüência parcial do gene hrpB. As combinações de primers Xcv1F/Xcv3R e RST2/Xcv3R que amplificaram fragmentos de 243 e 340 pb, respectivamente, foram testadas quanto à especificidade e sensibilidade para detecção do DNA de Xcv. Com os dois pares de primers, amplificação foi positiva com o DNA de 44 isolados de Xcv, mas também com quatro isolados de X.c. pv. mangiferaeindicae e cinco de X. axonopodis pv. passiflorae. Contudo, a digestão dos produtos de PCR permitiu diferenciar Xcv dos isolados desses patovares. Nenhum dos dois pares de primers amplificou o DNA de videira, nem de 20 bactérias não patogênicas isoladas da flora da videira, ou de 10 isolados de outros seis gêneros de bactérias fitopatogênicas. A sensibilidade dos primers Xcv1F/Xcv3R e RST2/Xcv3R foi de 10 pg e 1 pg de DNA purificado de Xcv, respectivamente. O limite de detecção de RST2/Xcv3R foi de 10(4) UFC/ml, mas empregando-se uma segunda rodada de amplificação com o primer interno Xcv1F, esse limite foi de 10² UFC/ml. Não foi possível detectar por PCR a presença de Xcv usando-se, diretamente na reação, o extrato do macerado de pecíolos de videira previamente inoculados. Entretanto, amplificações foram positivas quando se utilizou uma etapa de enriquecimento em meio de cultura antes da PCR. Detectou-se Xcv em 1 µl da suspensão obtida do lavado das placas e em uma suspensão obtida a partir de uma única colônia. A identidade da bactéria foi confirmada pela análise de RFLP dos produtos de amplificação dos primers RST2/Xcv3R com HaeIII.

In order to develop a molecular method for detection and identification of Xanthomonas campestris pv. viticola (Xcv) the causal agent of grapevine bacterial canker, primers were designed based on the partial sequence of the hrpB gene. Primer pairs Xcv1F/Xcv3R and RST2/Xcv3R, which amplified 243- and 340-bp fragments, respectively, were tested for specificity and sensitivity in detecting DNA from Xcv. Amplification was positive with DNA from 44 Xcv strains and with DNA from four strains of X. campestris pv. mangiferaeindicae and five strains of X. axonopodis pv. passiflorae, with both primer pairs. However, the enzymatic digestion of PCR products could differentiate Xcv strains from the others. None of the primer pairs amplified DNA from grapevine, from 20 strains of nonpathogenic bacteria from grape leaves and 10 strains from six representative genera of plant pathogenic bacteria. Sensitivity of primers Xcv1F/Xcv3R and RST2/Xcv3R was 10 pg and 1 pg of purified Xcv DNA, respectively. Detection limit of primers RST2/Xcv3R was 10(4) CFU/ml, but this limit could be lowered to 10² CFU/ml with a second round of amplification using the internal primer Xcv1F. Presence of Xcv in tissues of grapevine petioles previously inoculated with Xcv could not be detected by PCR using macerated extract added directly in the reaction. However, amplification was positive with the introduction of an agar plating step prior to PCR. Xcv could be detected in 1 µl of the plate wash and from a cell suspension obtained from a single colony. Bacterium identity was confirmed by RFLP analysis of the RST2/Xcv3R amplification products digested with Hae III.
