Abstract Interspecific hybridization is required for the development of Jatropha curcas L. improved cultivars, due to its narrow genetic basis. The present study aimed to analyze the parental genomic composition of F1 and BC1F1 generations derived from interspecific crosses (J. curcas/J. integerrima and J. curcas/J. multifida) by GISH (Genomic In Situ Hybridization), and the meiotic index and pollen viability of F1 hybrids. In F1 cells from both hybrids, 11 chromosomes of each parental was observed, as expected, but chromosome rearrangement events could be detected using rDNA chromosome markers, suggesting unbalanced cells. In the BC1F1, both hybrids had 22 chromosomes, suggesting that only n = 11 gametes were viable in the next generation. However, GISH allowed the identification of three and two alien chromosomes in J. curcas//J. integerrima and J. curcas//J. multifida BC1F1 hybrids, respectively, suggesting a preferential transmission of J. curcas chromosomes for both hybrids. Pollen viability in F1 hybrids derived from J. curcas/J. integerrima crosses were higher (82-83%) than those found for J. curcas/J. multifida (68%), showing post-meiotic problems in these last hybrids, with dyads, triads, polyads, and micronuclei as post-meiosis results. The here presented cytogenetic characterization of interspecific hybrids and their backcross progenies can contribute to the selection of the best genotypes for future assisted breeding of J. curcas.

Foram testadas diferentes fontes de nitrogênio na indução e enraizamento de brotações axilares de Chrysanthemum morifolium. Manteve-se constante o teor de nitrogênio total (60 mM) em todos os tratamentos realizados, variando-se apenas as fontes nitrogenadas. O tratamento E1 constituiu-se do meio MS (Murashige & Skoog 1962) completo. Os demais tratamentos foram: E2 = amônio; E3 = nitrato; E4 = nitrato+uréia (1,65 mM); E5 = nitrato+uréia (3,33 mM); E6 = uréia; E7 = uréia+glutamina e E8 = glutamina. Em nenhum tratamento foram adicionados fitorreguladores. No tratamento E1 houve um favorecimento de desenvolvimento de plantas de crisântemo in vitro tanto da parte aérea como de raízes. Porém, a utilização apenas de nitrato como única fonte de nitrogênio (tratamento E3) foi suficiente para sustentar o desenvolvimento das plantas, pois, para a maioria dos parâmetros analisados não foi observada diferença significativa em relação às plantas mantidas no meio básico de MS. A presença de uréia no meio de cultura incrementou o desenvolvimento de raízes de crisântemo, principalmente quando utilizada em adição ao nitrato (tratamentos E4 e E5), proporcionando um sistema radicular bastante denso. Quando utilizadas como única fonte de nitrogênio, tanto a glutamina (tratamento E7) como o íon amônio (tratamento E2) não se mostraram eficientes. O resultado do uso das fontes mencionadas foi um baixo desenvolvimento das plantas, como evidenciado pelos baixos valores de crescimento obtidos em todos os parâmetros analisados.

The effect of different nitrogen sources on the induction and rooting of axillary shoots in Chrysanthemum morifolium was tested. The total nitrogen concentration was maintained constant (60 mM) in all treatments, only the nitrogen source varying. Treatment E1 was a complete MS (Murashige & Skoog 1962) medium. The remaining treatments were: E2 = ammonium; E3 = nitrate; E4 = nitrate+urea (1.65 mM); E5 = nitrate+urea (3.33 mM); E6 = urea; E7 = urea+ glutamine, and E8 = glutamine. No phytohormones were added to the media. In treatment E1, both shoot and root development were promoted. The use of nitrate as the only source of nitrogen (treatment E3) was sufficient to maintain vigorous growth and no significant difference was found in any of the parameters measured when compared with the basic MS medium. The presence of urea promoted root development in Chrysanthemum, especially when used in addition to nitrate (treatments E4 and E5), hereby producing a dense root system. Both glutamine (treatment E7) and ammonium (treatment E2) were inefficient when used as the sole nitrogen source. Both of these sources resulted in a less vigorous plant growth, this being reflected in lower values for all of the parameters measured.
