RESUMO Utilizando um anticorpo monoclonal contra a aflatoxina B1 (AFB1) como ligante, foi identificado um mimotopo específico de aflatoxina B1 após se realizarem quatro ciclos de seleção biológica de 7-peptídeos aleatórios em biblioteca de fago exibida. O mimotopo é denominado P10, e sua sequência de aminoácidos é YRRHEKD. O soro imunológico de ratos Balb/c imunizados com P10 foi especificamente ligado à aflatoxina B1-albumina, indicando que o anticorpo era específico ao AFB1. Esses resultados sugerem que é possível desenvolver a vacina baseada em mimotopo associado à toxina.

The exploitation of shallow coal seams causes severe surface subsidence and triggers the development of numerous ground fissures, which threaten the safety of underground mining and damage the surface eco-environment. We studied the law of surface movement and the development of ground fissures at three panels in the Daliuta coal mine, Shendong mining district of western China. The results showed that mining-induced ground fissures developed in a cycle of 'crack-expand-close', and formed permanent cracks that were arranged in an elliptical shape on the edge of the goaf. There was a positive linear relationship between the fissure width and the surface horizontal deformation, as one would expect, and a positive logarithmic relationship between the fissure angle and the mining rate during coal face advancing. We propose a basis and standard for treating dynamic fissures which recommends that, to ensure the safety of underground mining, the fissures should be treated when the surface cracks extend as far as the fractured zone of rock strata. The results will support predictions of the development of mining-induced ground fissures so that disasters can be prevented in shallow coal seam mining areas.

Abstract Shallow lowland lakes are critical components of the water cycle, providing an essential service function. However, the impacts of microcystin from phytoplankton communities on benthic macroinvertebrate community diversity and structure have seldom been investigated. During 2008-2012, the impacts of water environmental variables on the diversity of macrobenthic communities, including water temperature, pH, dissolved oxygen, transparency, conductivity, the permanganate index, Chlorophyll a, total phosphorus, total nitrogen, and microcystin-LR (L for leucine and R for arginine), were measured in a typical shallow lowland lake, Ge Lake. The results of the present study demonstrated that there were 31 benthic macroinvertebrate taxa in Ge Lake, including 7 oligochaetes, 7 Mollusca, 14 chironomids, and 3 other taxa. Among the macrobenthic taxa in the benthic community, opportunistic taxa such as Limnodrilus hoffmeisteri are present and can occur at greater densities in disturbed habitats. However, a significant reduction/disappearance of sensitive and clean taxa was observed among the benthic macroinvertebrates. Water temperature, dissolved oxygen, conductivity, ammonia nitrogen, transparency and total phosphorus were the main environmental variables influencing macrobenthic community structure, while water temperature, conductivity, ammonia nitrogen, Chlorophyll a, the permanganate index, total phosphorus and total nitrogen were the main factors that influenced macrobenthic community diversity indices (the numbers of taxa, Shannon’s diversity index, Margalef’s richness index, and Pielou’s evenness index). Our results underscore the severity of the effects of human activity on Ge Lake and strongly suggest that restoring the benthic invertebrate community to previous conditions would require the control and reduction of environmental pollutants and nutrients in Ge Lake.

To reduce production costs and increase efficiency in ageing coal mines, a system that utilizes two shearers in a fully mechanized longwall working face is proposed. Using theoretical and engineering experience, three issues pertaining to the system were determined: (a) the two-shearer mining technique; (b) matching and modifying the equipment; (c) and coal-cutting task allocation for each shearer. Two mining processes are proposed, with the two shearers travelling in either the same or opposing directions. To avoid breakage of chains and pan extrusion of the armoured face conveyor (AFC), the length of the AFC 'snake' was controlled. To ensure continuous cutting and transporting of coal, the cross-sectional dimensions of the AFC were checked and the shearer closer to the head drive was modified. Based on the assumption of equal cutting times for each shearer, the meeting position of the two shearers was obtained by using a theoretical model of mining task allocation. Application of this technique at No. 2 Jining Mine, China, has shown remarkable benefits: daily production capacity was increased by 54% and personnel efficiency was improved by 33 t/d per person.

ABSTRACT The bursa of Fabricius (BF) is the central humoral immune organ unique to birds. The present study investigated the possible difference on a molecular level between two duck breeds. The digital gene expression profiling (DGE) technology was used to enrich the differentially expressed genes (DEGs) in BF between the Jianchang and Nonghua-P strains of ducks. DGE data identified 195 DEGs in the bursa. Gene Ontology (GO) analysis suggested that DEGs were mainly enriched in the metabolic pathways and ribosome components. Pathways analysis identified the spliceosome, RNA transport, RNA degradation process, Jak-STAT signaling pathway, TNF signaling pathway and B cell receptor signaling pathway. The results indicated that the main difference in the BF between the two duck strains was in the capabilities of protein formation and B cell development. These data have revealed the main divergence in the BF on a molecular level between genetically different duck breeds and may help to perform molecular breeding programs in poultry in the future.

RESUMO: Carex physodes é uma espécie efêmera no deserto frio de Gurbantunggut, no noroeste da China, que possui padrões reprodutivos tanto sexuais como assexuais. Os objetivos principais deste estudo foram caracterizar os sistemas de reprodução e identificar o papel da dispersão dos frutos na reprodução sexual de C. physodes. Foram medidas a biomassa aérea e de raízes, a razão raiz-parte aérea, a biomassa de inflorescência e a frutificação de C. physodes e estudada a dispersão de estames perigínicos e aquênios em habitat natural e ambiente interno. A biomassa das raízes de C. physodes foi aproximadamente dez vezes maior do que a biomassa aérea. A maior parte da biomassa aérea é alocada para a inflorescência, o que sugere que C. physodes aloca a maioria da biomassa para a parte reprodutiva. Ela produz estames perigínicos com pericarpo contendo um aquênio. Os estames perigínicos se dispersam a uma distância muito maior do que os aquênios na velocidade do vento de 1 e 4 m s-1, e o tempo de flutuação dos estames perigínicos na água foi muito maior que o dos aquênios. Os estames perigínicos podem reter mais água e aderir ao solo muito mais facilmente do que os aquênios, o que sugere que esses estames são apropriados para dispersão por vento; além disso, estão adaptados para dispersão por grande distância quando ocorrem chuvas ocasionais. No entanto, os aquênios podem permanecer perto das plantas-mãe e dispersar-se apenas por distâncias curtas. C. physodes é adaptada morfológica e fisiologicamente ao ambiente do deserto frio por meio de uma combinação de caracteres associados aos rizomas e aos estames perigínicos. Essa adaptação pode aumentar a oportunidade de sobrevivência e expansão da população de C. physodes.

ABSTRACT: Carex physodes is an ephemeral species in the cold desert of Gurbantunggut in Northwest China. It has both asexual and sexual reproductive patterns. The primary aims of this study were to characterize the reproduction systems and identify the role of fruit dispersal in the sexual reproduction of C. physodes. Aboveground and underground biomass, root-shoot ratio, inflorescence biomass, fruit-set of C. physodes were measured and dispersal of perigynia and achenes in the natural habitat and indoor condition were studied. The underground biomass of C. physodes was approximately 10 times more than the aboveground biomass. The most parts of aboveground biomass is allocated to the inflorescence, which suggests that C. physodes allocates most biomass to the reproductive part. C. physodes produces perigynium with a pericarp containing one achene. The perigynia disperse at a much greater distance than achenes at both 1 and 4 m s-1 wind velocity, and the floating time of perigynia in water was much longer than that of achenes. Perigynia can hold more water and adher soil much more easily than achenes, which suggests that perigynia are suitable for wind dispersal, and they also adapt to spread at a long distance by occasionally rainfall. However, achenes may remain near the mother plants and only disperse at short distances. C. physodes is morphologically and physiologically adapted to the cold desert environment via a combination of characters associated with the rhizomatous and perigynium. This adaption may increase the opportunity of survival and expansion of population of C. physodes.

The modes of occurrence of arsenic and the effects of low-intensity leaching-flotation on arsenic removal from coal slime from the Tanggongta Plant, Inner Mongolia, China were investigated. The coal slime was examined using hydride generation inductively coupled plasma optical emission spectrometry and X-ray diffraction to obtain the content of elements and the major minerals. The modes of occurrence of arsenic in minerals were determined using selective leaching, float-and-sink analysis, and polarized light microscopy. The results indicate that pyrite is the dominant carrier of arsenic in the coal slime, which predominantly exists in association with clay. Significant proportion of the arsenic is removed by a low-intensity leaching-flotation process, consistent with the data from selective leaching and flotation of gangue. The results show that low intensity leaching-alkali washing-flotation is more efficient than direct flotation and low-intensity leaching-flotation.

The aim of this study was to investigate whether exogenous retinoic acid (RA) can upregulate the mRNA and protein expression of growth-associated protein 43 (GAP-43), thereby promoting brain functional recovery in a rat distal middle cerebral artery occlusion (MCAO) model of ischemia. A total of 216 male Sprague Dawley rats weighing 300–320 g were divided into 3 groups: sham-operated group, MCAO+vehicle group and MCAO+RA group. Focal cortical infarction was induced with a distal MCAO model. The expression of GAP-43 mRNA and protein in the ipsilateral perifocal region was assessed using qPCR and immunocytochemistry at 1, 3, 7, 14, 21, and 28 days after distal MCAO. In addition, an intraperitoneal injection of RA was given 12 h before MCAO and continued every day until the animal was sacrificed. Following ischemia, the expression of GAP-43 first increased considerably and then decreased. Administration of RA reduced infarction volume, promoted neurological functional recovery and upregulated expression of GAP-43. Administration of RA can ameliorate neuronal damage and promote nerve regeneration by upregulating the expression of GAP-43 in the perifocal region after distal MCAO.

ABSTRACT The effects of the Kluyveromyces marxianus M3 strain, isolated from Tibetan mushrooms, on plasma lipids, egg cholesterol level, egg quality, and intestinal health of laying hens were evaluated. In total, 160 Beijing fatty laying hens (43 weeks old) were divided into four groups and fed a basal diet supplemented with 0%, 0.1%, 0.3%, or 0.5% freeze-dried K. marxianus M3 powder for four weeks. The results showed that yeast supplementation reduced serum total cholesterol (TC), triglyceride (TG), low density lipoprotein-cholesterol (LDL-C), and very low density lipoprotein-cholesterol (VLDL-C) levels (p<0.01), and increased serum high density lipoprotein-cholesterol (HDL-C) level (p<0.05). Moreover, regardless of K. marxianus M3 dietary addition level, the cholesterol content of the eggs decreased by more than 26%. When0.3% yeast was supplemented, significant differences were found in the egg weights, shell strength, albumen height, Haugh unit and nutrient content of the eggs (p<0.01). Finally, 0.3% yeast supplementation improved the intestinal flora conditions of the hens by decreasing the Salmonella and Staphylococcus aureus counts (p<0.01) and increasing the Bifidobacterium count (p<0.01). The results in this work demonstrated that yeast culture supplementation to the diets deceased the serum and egg yolk cholesterol, and increased egg quality.

Zinc deficiency is a worldwide nutritional constraint for crop production in many types of soil, particularly in cereals growing in calcareous soil. Two field trials were conducted to study the effects of zinc fertilizer doses and application method on photosynthetic characteristics and grain yield of summer maize, from June to September of 2013, in Xun County (34º40,12” N, 114º32,24”E) and Yuzhou County (33º59,19”N, 113º33,47”E). Optimal Zn fertilizer application (30 kgZnSO4·7H2O hm-2) increased chlorophyll content in leaves, improved photosynthesis, and increased grain yield of summer maize. At 15 kg ZnSO4·7H2O hm-2, grain yield was higher in Xun County and lower in Yuzhou County when the entire amount was applied as basal fertilization as opposed to being split between the basal and foliar fertilization. The opposite result (grain yield lower in Xun County than in Yuzhou County) was obtained at 30 kg ZnSO4·7H2O hm-2. These results have important implications for guiding the rational application of Zn fertilizer and improving the grain yield of summer maize in Henan province.

BACKGROUND: Termination of pregnancy (TOP) remains a controversial issue, regardless of legislation. Access to services as well as psychological effects may vary across the world. OBJECTIVES AND METHODS: To better understand the psychological effects of TOP, this study describes the circumstances of 102 women who underwent a TOP from two socioeconomic sites in Johannesburg, South Africa, one serving women with few economic resources and the other serving women with adequate resources. The relationship between demographic characteristics, resilience and symptoms of post-traumatic stress disorder (PTSD) and depression before, 1 month after and 3 months after the procedure was also examined. RESULTS: Time since TOP, age, chronic burden, resilience and the interaction of site with religion and site with chronic burden were significant. In addition, site differences were found for religion and chronic burden in predicting depression scores. Women from both sites had significant decreases in depression scores over time. The interaction of time with site was not significant. Higher chronic burden scores correlated with higher depression scores. No variables were significant in the bivariate analysis for PTSD. CONCLUSION: Resilience, religion and chronic burden emerge as significant variables in women undergoing a first-trimester TOP, and warrant further assessment in studies of this nature.

ABSTRACTThis study aimed at examining the effect of overfeeding on the activity of the mTOR pathway in the liver and muscle tissues of Gang geese. Eighty healthy male Gang geese were reared under the same feeding conditions, and were divided at 14 weeks of age into a control group and an overfed group. All birds were slaughtered after three weeks of over feeding. Gene expression and protein content of several genes involved in the mTOR pathway were evaluated. The results showed that the gene expression of mTOR, raptor, and rictor, and the protein contents of mTOR and PI3K were higher in liver, breast muscle, and leg muscle of the overfed group than that of control group. However, the S6K expression level was clearly lower in the liver of the overfed group than that of control group, and there was no evident difference in both breast muscle and leg muscle between the control group and the overfed group. These results suggest that overfeeding induces the activity of raptor, rictor, and mTOR, and that mTOR signaling pathway was closely linked with PI3K pathway in the evaluated geese.

This study aimed to determine the role of mitochondrial adenosine triphosphate-sensitive potassium (mitoKATP) channels and protein kinase C (PKC)-ε in the delayed protective effects of sevoflurane preconditioning using Langendorff isolated heart perfusion models. Fifty-four isolated perfused rat hearts were randomly divided into 6 groups (n=9). The rats were exposed for 60 min to 2.5% sevoflurane (the second window of protection group, SWOP group) or 33% oxygen inhalation (I/R group) 24 h before coronary occlusion. The control group (CON) and the sevoflurane group (SEVO) group were exposed to 33% oxygen and 2.5% sevoflurane for 60 min, respectively, without coronary occlusion. The mitoKATP channel inhibitor 5-hydroxydecanoate (5-HD) was given 30 min before sevoflurane preconditioning (5-HD+SWOP group). Cardiac function indices, infarct sizes, serum cardiac troponin I (cTnI) concentrations, and the expression levels of phosphorylated PKC-ε (p-PKC-ε) and caspase-8 were measured. Cardiac function was unchanged, p-PKC-ε expression was upregulated, caspase-8 expression was downregulated, cTnI concentrations were decreased, and the infarcts were significantly smaller (P<0.05) in the SWOP group compared with the I/R group. Cardiac function was worse, p-PKC-ε expression was downregulated, caspase-8 expression was upregulated, cTnI concentration was increased and infarcts were larger in the 5-HD+SWOP group (P<0.05) compared with the SWOP group. The results suggest that mitoKATP channels are involved in the myocardial protective effects of sevoflurane in preconditioning against I/R injury, by regulating PKC-ε phosphorylation before ischemia, and by downregulating caspase-8 during reperfusion.

Studies have indicated that early-life or early-onset depression is associated with a 2- to 4-fold increased risk of developing Alzheimers disease (AD). In AD, aggregation of an abnormally phosphorylated form of the tau protein may be a key pathological event. Tau is known to play a major role in promoting microtubule assembly and stabilization, and in maintaining the normal morphology of neurons. Several studies have reported that stress may induce tau phosphorylation. The main aim of the present study was to investigate possible alterations in the tau protein in the hippocampus and frontal cortex of 32 male Sprague-Dawley rats exposed to chronic unpredictable mild stress (CUMS) and then re-exposed to CUMS to mimic depression and the recurrence of depression, respectively, in humans. We evaluated the effects of CUMS, fluoxetine, and CUMS re-exposure on tau and phospho-tau. Our results showed that a single exposure to CUMS caused a significant reduction in sucrose preference, indicating a state of anhedonia. The change in behavior was accompanied by specific alterations in phospho-tau protein levels, but fluoxetine treatment reversed the CUMS-induced impairments. Moreover, changes in sucrose preference and phospho-tau were more pronounced in rats re-exposed to CUMS than in those subjected to a single exposure. Our results suggest that changes in tau phosphorylation may contribute to the link between depression and AD.

Based on simulations of the transports of a typically formed Chinese intercity passenger train hauled respectively by major types of the locomotives on a certain railway line in China, this research analyzes the changes of the traction energy cost intensity and passenger transport efficiency of the train with varying its passenger capacity utilization rate and target speed. It is found that if the passenger capacity utilization rate of such a train is higher than 60.00%, the effect of its passenger capacity utilization rate and target speed on its traction energy cost intensity and passenger transport efficiency is relatively very small. In contrast, if the passenger capacity utilization rate of the train is reduced from 40.00%, the efficiencies of its traction energy cost and passenger transport both decrease obviously and such decreases are evidently affected by its target speed. Therefore, strictly preventing the passenger capacity utilization rate of a conventional intercity passenger train from decreasing from 40.00% in particular and meanwhile striving to make its passenger capacity utilization rate higher than 60.00% as new evaluation benchmarking are able to effectively improve the railway passenger transport operation management in China.
