Abstract The situation of native orchids from natural environments becomes more vulnerable each day, especially those not protected by conservation units. Thus, studies on the propagation of species, especially on in vitro germination and initial establishment, are necessary for maintaining species in their habitat and reintroduction in restoration areas. This study verifies the influence of the nutritional composition of the culture media on in vitro germination and initial establishment of Miltonia flavescens. Seeds from natural pollination were sown in four different culture media and put in a growing room under controlled photoperiods and temperature. The germination was evaluated 30 days after sowing, and the initial growth of the seedlings at 120 and 180 days after sowing. The results indicated the Murashige and Skoog medium, complete and with half of the salt concentrations, promoted a higher germination percentage and were more effective in the initial development of M. flavescens seedlings.

RESUMO: O cultivo in vitro tem sido utilizado para propagação de plantas ornamentais. A escolha do meio de cultivo e a concentração ideal de produtos orgânicos utilizados no enriquecimento dos meios, como a sacarose, são decisivos na elaboração de protocolos. Dessa forma, objetivou-se avaliar o crescimento inicial de Miltonia flavescens, cultivada por 90 e 180 dias, em quatro meios de cultivo com diferentes concentrações de sacarose. Plantas com 12 meses e germinadas in vitro foram cultivadas nos meios MS, MS 1/2, K e VW, cada um deles suplementado com 1,5g L-1 de carvão ativado, 6,5g L-1 de ágar bacteriológico e com 25, 30, 35, 40 e 45g L-1 de sacarose. Aos 90 e 180 dias, foram avaliadas quanto à massa fresca, número de raízes, de folhas e de brotos e quanto ao comprimento da parte aérea e da maior raiz. As maiores médias para número de raízes, comprimento da parte aérea e massa fresca total foram obtidas nas plantas cultivadas por 180 dias em meio MS 1/2, suplementado com 25g L-1 de sacarose. O maior comprimento de raiz foi verificado nas plantas cultivadas por 180 dias em meio MS suplementado com 45g L-1 de sacarose. Já o número de brotos e de folhas não foram influenciados pelas concentrações de sacarose. M. flavescens apresentou maior crescimento da parte aérea e raiz, quando cultivada por 180 dias em meio MS 1/2, suplementado com 25g L-1 de sacarose.

ABSTRACT: The in vitro culture has been widely used for the ornamental plants propagation. The choice of the best medium and organic products concentration, such as sucrose, are decisive in the protocols. Thus, the aim of this study was to evaluate the initial growth of Miltonia flavescens, grown for 90 and 180 days, in four culture media supplemented with different concentrations of sucrose. Plants aged 12 months were germinated in vitro and cultured on MS, MS 1/2 K and VW media each one supplemented with 1.5g L-1 of activated charcoal, 6.5g L-1 bacteriological agar and 25, 30 35, 40 and 45g L-1 sucrose. After 90 and 180 days, fresh mass, number of roots, leaves and shoots, length shoot and the largest root were evaluated. The highest average for number of roots, shoot, length and total fresh weight were obtained in plants grown for 180 days on MS 1/2 medium supplemented with 25g L-1 sucrose. The greater length of roots was observed in plants grown for 180 days on MS medium supplemented with 45g L-1 sucrose. M. flavescens showed higher growth of shoots and roots when cultivated for 180 days on MS 1/2 medium supplemented with 25g L-1 sucrose.

O sucesso na micropropagação de Dendrobium phalaenopsis Deang Suree é alto, porém, quando transplantado para o viveiro, sua sobrevivência é mínima. Com o intuito de aumentar a sobrevivência na produção, no presente trabalho, avaliou-se o efeito da aclimatização intermediária, por 30 dias em sala de crescimento, sob as seguintes condições de luminosidade: 1- luz fluorescente branca (18,9µmol m-2 s-1); 2- luz fluorescente branca + luz fluorescente vermelha (14,85µmol m-2 s-1); e 3- luz fluorescente vermelha (9,45µmol m-2 s-1), o controle foi acondicionado diretamente em viveiro coberto (162,0µmol m-2 s-1). Na sequência, as folhas foram caracterizadas anatomicamente e as plantas foram transferidas para o viveiro que continha o controle. Foi avaliada a porcentagem de sobrevivência e o número final de raízes, sendo calculadas também as relações entre os valores finais e iniciais de massa fresca, número de folhas, comprimento e diâmetro do maior pseudobulbo, número de pseudobulbos e comprimento da maior raiz das plantas aclimatizadas. Apenas as plantas submetidas à luz vermelha foram estatisticamente superiores ao controle, em relação à porcentagem de sobrevivência e relação de massa fresca, enquanto que o controle apresentou maior número de raízes que plantas aclimatizadas nessa condição de luminosidade. Aclimatização intermediária, utilizando luz vermelha ou a luz vermelha + branca, é recomendada para D. phalaenopsis Deang Suree.

The success in micropropagation of Dendrobium phalaenopsis Deang Suree is high, but when transplanted into the greenhouse, their survival is minimal. To increase survival in production in the present study it was evaluated the effect of intermediate acclimatization for 30 days in a grow room utilizing the following luminosity conditions: 1- white fluorescent light (B) (18.9µmol m-2 s-1); 2- white fluorescent light + red fluorescent light (GRO-LUX(r)) (BV) (14.85µmol m-2 s-1); 3- red fluorescent light (GRO-LUX(r)) (V) (9.45µmol m-2 s-1) and the control plants were accommodated directly in a greenhouse (162.0µmol m-2 s-1). After this the leaves were characterized anatomically and the plants transferred to the control greenhouse. It was evaluated survival percentage and final number of roots, and calculated the relations between the final and initial values of fresh weight, number of leaves, length and diameter of the largest pseudo bulb, number of pseudo bulbs and longest root length. Only plants submitted to red light, were statistically better than the control in relation to the survival percentage and in relation to fresh weight, while the control showed a higher number of roots that plants acclimatized in this luminosity conditions. Intermediate acclimatization, using red light or red + white light, is recommended for D. phalaenopsis Deang Suree.
