RESUMO: O objetivo deste trabalho foi avaliar o aumento da produção de óleos unicelulares por Aphanothece microscopica Nägeli utilizando água residuária de processamento de laticínios como meio de cultura. O estudo concentrou-se na (i) otimização da temperatura para produção de biomassa, (ii) aplicação de choque frio para induzir a biossíntese de lipídios e (iii) determinação do perfil de ácidos graxos sob diferentes condições. Os resultados indicam que a temperatura de 20°C foi a melhor condição em termos de parâmetros cinéticos, atingindo produtividades de biomassa de 160,25mg/L.h. Nesta condição também foi observado um teor lipídico de 12,65%, o que resultou em uma produtividade lipídica de 20,27mg/L.h. Além disso, o choque frio de 0°C foi o mais eficiente para aumentar o conteúdo lipídico intracelular, que atingiu 28,4% em peso seco. Os choques frios também mostraram influência na saturação da composição de ácidos graxos, em que os ácidos graxos saturados diminuíram, e os ácidos graxos monoinsaturados e poli-insaturados ampliaram com o aumento da aplicação de frio. Assim, o uso de choques frios indica ser uma condição chave para melhorar as perspectivas de produção eficiente de óleos unicelulares.

ABSTRACT: The aim of this research was to evaluate the enhanced single-cell oil production by cold shock in Aphanothece microscopica Nägeli using dairy processing wastewater as culture medium. The study focused on (i) temperature optimization for biomass production, (ii) cold shock application to induce lipids biosynthesis and (iii) determination of fatty acids profile under different conditions. Results indicated that temperature of 20°C was the best condition in terms of kinetics parameter, reaching biomass productivities of 160.25mg/L.h. Under these conditions, a lipid content of 12.65% was also observed, resulting in a lipid productivity of 20.27mg/L.h. Additionally, the 0°C cold shock was the most efficient in increasing intracellular lipid content, reaching 28.4% in dry weight. Cold shocks also showed influence on the saturation of fatty acid composition, where the saturated fatty acids decreased, and the monounsaturated and polyunsaturated fatty acids increased by increasing the cold application. Thus, the use of cold shocks indicates to be a key condition for improving the prospects of efficient single-cell oils production.

ABSTRACT: The systems that use microalgae to obtain bioproducts are considered a promising area for industrial applications. The objective of this work was to evaluate the production of phycobiliproteins and chlorophyll-a by Aphanothece microscopica Nageli, when it is cultivated in autotrophic and heterotrophic systems. Thus, the cultures were grown in heterotrophic (absence of light) and autotrophic (2 klux of light intensity) system, in bioreactors at pH 7.6, 20 °C, 1 vvm aeration, using a dairy industry effluent, with C/N 20 and N/P 10, and BG11 as culture media. Initial inoculum 200 mg/L. The concentrations of chlorophyll-a andphycobiliproteins in logarithmic and stationary phase were monitored. The highest concentrations of pigment were observed in logarithmic phase: The highest concentrations of chlorophyll-a in autotrophic cultures, and predominance of allophycocyanin and phycoerythrin in heterotrophic cultures. The results show the effect of the variables under study for pigment production, bearing in mind the logarithmic growth phase, suggesting the possibility of producing in heterotrophic systems by Aphanothece microscópica Nageli, around 3,185 ton/year of biomass, and 3,127 kg/year of chlorophyll, 232,825 kg/year of phycocyanin, 47,775 kg/year of allophycocyanin and 7,008 kg/year of phycoerythrin, when dairy effluent is used as culture medium.

RESUMEN: Los sistemas basados en el uso de microalgas para la obtención de bioproductos son considerados un área prometedora para aplicaciones industriales. El objetivo del trabajo fue evaluar la producción de ficobiliproteínas y clorofila-a cuando Aphanothece microscópica Nageli es cultivada en sistemas autotróficos y heterotróficos. Para ello, se desarrollaron sistemas de cultivos heterotróficos (en ausencia de luz) y autotróficos (2 klux de intensidad luminosa) en biorreactores a pH 7,6, 20 °C, 1 vvm de aireación, empleando como medios de cultivo un efluente de industria láctea con relación C/N 20 y N/P 10, y BG11. Inóculo inicial 200 mg/L. Se efectuó el monitoreo de concentraciones de clorofila-a y ficobiliproteinas en fase logarítmica y estacionaria. Se observaron las mayores concentraciones de pigmento en la fase logarítmica: mayores concentraciones de clorofila-a en cultivos autotróficos y predominio de aloficocianina y ficoeritrina en cultivos heterotróficos. Los resultados mostraron el efecto de las variables en estudio para la producción de pigmentos, en la fase de crecimiento logarítmico, lo que indica la posibilidad de producir en sistemas heterotróficos de Aphanothece microscopica Nageli, aproximadamente 3.185 ton/ año de biomasa, para 3.127 kg/año clorofila, 232.825 kg/año ficocianina, 47.775 kg/año aloficocianina y 7.008 kg/año ficoeritrina, cuando se utiliza efluente de industria láctea como medio de cultivo.

ABSTRACT This study aimed to evaluate third generation biodiesel production by microalgae Phormidium autumnale using cassava processing wastewater as a platform. Experiments were performed in a heterotrophic bubble column bioreactor. The study focused on the evaluation of the bioreactor (batch and fed-batch) of different operational modes and the analysis of biofuel quality. Results indicate that fed-batch cultivations improved system performance, elevating biomass and oil productions to 12.0 g L−1 and 1.19 g L−1, respectively. The composition of this oil is predominantly saturated (60 %) and monounsaturated (39 %), resulting in a biodiesel that complys with U.S., European and Brazilian standards. The technological route developed indicates potential for sustainable production of bulk oil and biodiesel, through the minimization of water and chemical demands required to support such a process.

O consumo de probióticos está em constante crescimento, devido aos inúmeros benefícios conferidos à saúde dos consumidores. Neste contexto, a microencapsulação é uma tecnologia que favorece a viabilidade das culturas probióticas em produtos alimentícios, principalmente pelas propriedades de proteção contra as condições ambientais adversas e a liberação controlada. Atualmente, existem diversos procedimentos para a microencapsulação, com a utilização de vários tipos de polímeros de origem natural e sintética. O alginato de sódio é um dos polímeros com maior potencial para aplicação na encapsulação de probióticos, devido à sua versatilidade, biocompatibilidade e isenção de toxicidade. O objetivo desta revisão é apresentar técnicas viáveis de encapsulação de probióticos com alginato, enfatizando a gelificação iônica interna e gelificação iônica externa, com a possibilidade de aplicação, bem como as tecnologias promissoras para o melhoramento destas.

The consumption of probiotics is constantly growing due to the numerous benefits conferred on the health of consumers. In this context, Microencapsulation is a technology that favors the viability of probiotic cultures in food products, mainly by the properties of protection against adverse environmental conditions and controlled release. Currently there are different procedures for microencapsulation using polymers of various types of natural and synthetic origin. The use of sodium alginate polymers is one of the largest potential application in the encapsulation of probiotics because of their versatility, biocompatibility and toxicity exemption. The aim of this review is to present viable encapsulation techniques of probiotics with alginate, emphasizing the internal ionic gelation and external ionic gelation, with the possibility of applying, as well as promising for improving these techniques.

O objetivo do estudo foi avaliar o biodiesel de 3 a geração produzido a partir do cultivo heterotrófico da microalga Phormidium sp., empregando amido de mandioca como fonte de carbono orgânico. Um planejamento experimental foi realizado para determinar as condições ótimas de temperatura e razão C/N. A partir da obtenção das melhores condições de cultivo desenvolveram-se cultivos em batelada e batelada alimentada em biorreator e avaliaram-se as características do biodiesel produzido. Os resultados indicaram que a temperatura de 30ºC e a razão C/N de 68 são as condições ideais do processo. As maiores produtividades em biomassa (50,41mgL-1h-1) e lipídica (7,49mgL-1h-1) foram obtidas no cultivo em batelada. Os ácidos graxos com maior representatividade foram os ácidos caproico (65,29%) e oleico (23,88%). As propriedades de combustão do biodiesel: conteúdo de ésteres (99,9%), número de cetano (54,88), índice de iodo (21,47gI2100g-1), grau de instauração (23,88%) e ponto de entupimento de filtro a frio (39,21ºC) se mostraram adequadas às principais normativas nacionais e internacionais

The aim of the study was to evaluate the third generation biodiesel produced from heterotrophic cultivation of the microalgae Phormidium sp. employing cassava starch as source of organic carbon. An experimental design was performed to determine the optimal conditions of temperature and C/N ratio. From the best growing conditions, was developed cultivations in batch and fed-batch in a bioreactor and evaluated the biodiesel quality. The results indicate that the temperature of 30ºC and the C/N ratio of 68 are the ideal conditions of the process. The highest biomass productivity (50.41mgL-1h-1) and lipid productivity (7.49mgL-1h-1) were obtained in batch cultivations. The fatty acids most representative were caproic acid (65.29%) and oleic acid (23.88%). The fuel properties of biodiesel: ester content of 99.9%, cetane number of 54.88, iodine value of 21.47gI100g-1, unsaturation degree of 23.88% and a cold filter plugging point of 39.21ºC, comply with the main international and national standards

Microalgal heterotrophic bioreactors are a potential technological development that can convert organic matter, nitrogen and phosphorus of wastewaters into a biomass suitable for energy production. The aim of this work was to evaluate the performance of microalgal heterotrophic bioreactors in the secondary treatment of cattle-slaughterhouse wastewater and the reuse of microalgal sludge for biodiesel production. The experiments were performed in a bubble column bioreactor using the microalgae Phormidium sp. Heterotrophic microalgal bioreactors removed 90 % of the chemical oxygen demand, 57 % of total nitrogen and 52 % of total phosphorus. Substantial microalgal sludge is produced in the process (substrate yield coefficient of 0.43 mg sludge mg chemical oxygen demand−¹), resulting in a biomass with high potential for producing biodiesel (ester content of more than 99 %, cetane number of 55, iodine value of 73.5 g iodine 100 g−¹, unsaturation degree of ~75 % and a cold filter plugging point of 5 ºC).

As elevadas concentrações de fósforo em águas residuárias industriais é uma das principais causas da eutrofização de corpos hídricos receptores, havendo em virtude disto, uma crescente preocupação com o gerenciamento das cargas de fósforo dos efluentes industriais. Além dos aspectos de contenção ambiental, é estrategicamente desejável que os processos de tratamento permitam a recuperação e o reúso deste elemento que é considerado de elevada demanda biológica. Neste sentido, esta revisão objetiva descrever e analisar criticamente o estado atual das tecnologias de gerenciamento de fósforo em águas residuais industriais, com ênfase no reúso do elemento. As tecnologias consideradas incluem abordagens químicas (precipitação química e adsorção), biológicas (sistemas bacterianos, uso macroalgas e microalgas) além de técnicas de fertirrigação.

High phosphorus concentrations in industrial wastewaters is one of the main causes of the eutrophication of water bodies, leading to a growing concern with the management of phosphorus loads from industrial wastewaters. In addition to the environmental aspects of containment, it is strategically desirable that the treatment processes enable the recovery and reuse of this element, which is considered to be of high biological demand. For these reasons, this review describes and critically analyzes the current state of phosphorus management technologies for industrial wastewaters, with emphasis on the reuse of the element. The technologies considered include chemical approaches (chemical precipitation and adsorption), biological (bacterial systems, microalgae and macroalgae) plus fertigation techniques.

Devido aos seus efeitos benéficos, os probióticos têm sido incorporados nos mais diversos alimentos, incluindo iogurtes, queijos, sorvetes, leites fermentados e sobremesas congeladas. No entanto, existem ainda diversos problemas com relação à viabilidade e resistência das culturas probióticas nesses alimentos. Neste artigo de revisão, são abordados os aspectos tecnológicos utilizados na microencapsulação de probióticos que permitem aumentar a sua viabilidade durante a fermentação, processamento e utilização nos produtos comerciais. A microencapsulação de bactérias probióticas pode ser utilizada para aumentar a viabilidade durante o processamento, como também para liberá-las de maneira controlada no trato gastrointestinal.

Because of their health benefits, probiotics have been incorporated into a range of dairy products, including yogurts, soft-, semi-hard and hard cheeses, ice cream, milk powders and frozen dairy desserts. However, there are still several problems with respect to the low viability of probiotic bacteria in dairy foods. This review focuses mainly on current knowledge and techniques used in the microencapsulation of probiotic microorganisms to enhance their viability during fermentation, processing and utilization in commercial products. Microencapsulation of probiotic bacteria can be used to enhance the viability during processing, and also for the targeted delivery in gastrointestinal tract.

Neste trabalho, a eficiência da cianobactéria Aphanothece microscopica Nägeli em remover heterotroficamente fosfato total dissolvido do efluente do processamento de laticínios foi avaliada. Os experimentos foram conduzidos em biorreatores operados descontinuamente, alimentados com 4,5 L de efluente. As condições experimentais foram: concentração celular inicial de 0,2 g/L, pH ajustado a 7,6, reator isotérmico operando em temperaturas de 10, 20 e 30ºC, ausência de luminosidade e aeração contínua de 1 VVM. Os resultados mostraram que a remoção de fosfato é fortemente dependente da temperatura do processo. A cianobactéria A. microscopica Nägelifoi efetiva na remoção de fosfato,alcançando taxas de remoção de 3,77 mg/L.h, que refletiram em conversões de 98,4% em tempos de detenção hidráulica de 24 h.

The removal of total dissolved phosphate by the cyanobacteria Aphanothece microscopica Nägeli cultivated heterotrophically in dairy processing wastewater was investigated in this work. The experiments were carried out in bioreactors operating in a batch mode, fed with 4.5 L of wastewater. The experimental conditions were as follows: initial cell concentration of 0.2 g/L, pH adjusted to 7.6, isothermal reactor operating at temperatures of 10, 20 and 30ºC with absence of light and continuous aeration of 1 VVM. The results showed that phosphate removal is strongly dependent on process temperature. A. microscopica Nägeli was effective in the removal phosphate achieving removal rates of 3.77 mg/L.h, which reflected in the conversion of 98.4% in hydraulic detention times of 24 h.

Microalgae are a promising source of raw material for biodiesel production. This review discusses the latest developments related to the application of microalgae biomass for biodiesel production. Characterization of fatty acid of microalgae and comparisons with other sources of raw materials and processes are presented. Furthermore, technological perspectives and approaches for growing microalgae in photobioreactors, microalgal oil extraction techniques, and procedures for synthesizing biodiesel are reviewed.

Considerou-se neste trabalho a avaliação funcional de bases proteicas desidratadas de anchoita (Engraulis anchoita) foi considerada neste trabalho. A polpa do pescado foi separada mecanicamente e submetida à lavagem com dois solventes, água e ácido fosfórico. A secagem em camada delgada das bases proteicas foi conduzida em temperaturas de 40, 60 e 70°C e espessuras de amostra de 5mm. O produto foi avaliado considerando a determinação de proteínas solúveis, composição centesimal e propriedades funcionais, expressas pela solubilidade protéica, capacidade de retenção de água, capacidade de retenção de gordura e capacidade emulsificante. Os resultados indicaram que a obtenção da base proteica de anchoita usando ácido fosfórico como solvente de lavagem apresentou melhores características, se consideradas as operações de extração de proteínas solúveis, secagem e propriedades funcionais do produto final. Com relação à secagem, verificou-se que esta operação origina um produto com melhores características funcionais quando são empregadas temperaturas de 40 e 60°C.

The functional evaluation of two dehydrated anchovy (Engraulis anchoita) protein bases was considered in this work. The fish meat was mechanically separated and subjected to washing with two solvents, water and phosphoric acid, to obtain the protein bases. Thin layer drying of the protein bases was carried out at temperatures of 40, 60 and 70 °C, with a sample thickness of 5mm. The products were evaluated considering the determinations of the soluble proteins, proximate composition and the functional properties such as protein solubility, water holding capacity, fat holding capacity and emulsifying capacity. The results indicated that obtaining the anchovy protein base using phosphoric acid as the washing solvent resulted in better characteristics, when considering the operations of extracting and drying the soluble proteins and the functional properties of the final product. With respect to drying, it was found that this operation led to a product with better functional characteristics when temperatures of 40 and 60°C were used.

In this work we described the cultivation of Chlorella vulgaris in a photobioreactor to algal biomass production. The dried biomass was used as feedstock for biodiesel production, it presented 26% lipids and via sonocatalysis stage of the methodology resulted in 60% of fatty acid methyl esters (FAME). The FAME content was confirmed by Gas Chromatography (GC).

O objetivo do presente trabalho foi avaliar a fração protéica da cianobactéria Aphanothece microscopica Nägeli cultivada no efluente da parboilização do arroz quando submetida a diferentes condições de secagem. A produção da biomassa foi realizada a partir da água residuária do processo de parboilização do arroz em bioreatores de coluna de bolhas. A biomassa foi separada do efluente por centrifugação e desidratada em secador descontínuo de bandejas nas condições de 40, 50 e 60ºC e espessuras de bandejas de 5 e 7 mm. O comportamento eletroforético das proteínas da biomassa foi analisado por eletroforese em gel de poliacrilamamida na presença de dodecil sulfato de sódio (SDS-PAGE) e apresentou bandas com peso molecular de 15 kDa a 62,5 kDa. O perfil aminoacídico mostrou teores superiores aos recomendados pelo padrão FAO, com exceção dos aminoácidos lisina e dos sulfurados (MET+CYS). A caracterização funcional, expressa em termos de solubilidade e capacidade emulsificante, demonstrou a influência da condição de secagem na funcionalidade protéica da biomassa.

The objective of the present work was to evaluate the protein fraction of Aphanothece microscopica Nägeli cyanobacteria cultivated in the parboiled rice effluent, submitted to different drying conditions. The production of the biomass was done by using wastewater from the process of rice parboiling in a bubble column bioreactor. The biomass was separated from the effluent by centrifugation and dehydrated in a tray dryer at 40, 50 and 60ºC and thicknesses of 5 and 7 mm. The eletroforetic profile of proteins of the biomass was analysed by SDS-PAGE and presented bands with molecular weight between 62.5-15kDa. The amino acid profile was higher than the concentrations recommended by standard FAO, with the exception of the amino acids (LYS) and the sulphurated (MET+CYS). The functional properties characterisation in terms of protein solubility and emulsification capacity demonstrated the influence of the drying condition in the protein functionality of the biomass.
