RESUMO: Este estudo teve como objetivo avaliar a diversidade genética e a suscetibilidade a antimicrobianos de cepas de Staphylococcus aureus isoladas de vacas leiteiras em Minas Gerais, Brasil. Trinta e sete cepas provenientes de cinco municípios (oito rebanhos) foram genotipadas usando a técnica multi-locus sequence typing (MLST) e a suscetibilidade a 12 antimicrobianos foi avaliada pelo método de difusão em disco. Foram detectadas altas taxas de resistência para penicilina [75,68% (28/37)], ampicilina [70,27% (26/37)] e tetraciclina [70,27% (26/37)] entre os isolados. A multirresistência foi observada em sete [18,92% (7/37)] isolados e dois foram classificados como sugestivos de Staphylococcus aureus resistente à meticilina (MRSA). Entre os 37 isolados, 33 novos sequence types (ST) e dois STs conhecidos (ST126 e ST746) foram identificados pelo MLST. Os complexos clonais mais frequentemente observados foram: CC97 [78,38%; (29/37)], CC1 [8,11%; (3/37)] e CC5 [5,40%; (2/37)]. Foi construída uma minimum‐spanning tree (MSTs) com todos isolados estudados e esta não mostrou padrão de agrupamento quando comparada com dados epidemiológicos como municípios e rebanho, do qual foi isolado, e perfis de resistência a antimicrobianos. Uma segunda MST foi construída comparando os isolados deste estudo e todas as cepas de S. aureus depositadas no banco de dados PubMLST provenientes do Brasil, que mostrou associação entre o genótipo, STs e a origem da cepa. Foi possível observar entre os isolados do estudo que, aqueles que pertenciam ao CC97, eram geneticamente mais próximos das cepas depositadas no PubMLST isoladas de leite e de produtos lácteos, enquanto aqueles que pertenciam aos CC1 e CC5 estavam mais próximos a cepas isoladas de humanos ou do ambiente de fazendas e indústrias de laticínios. Em conclusão, nossos resultados mostraram um alto índice de resistência às penicilinas e tetraciclinas e grande diversidade genética entre as cepas de S. aureus isoladas de casos de mastite bovina.

ABSTRACT: This study evaluated the genetic diversity and antimicrobial susceptibility of Staphylococcus aureus isolates from dairy cows in Minas Gerais, Brazil. Thirty-seven isolates from five municipalities (8 herds) were genotyped using multi-locus sequence typing (MLST) and susceptibility to 12 antimicrobial agents was tested using the disk diffusion method. High resistance rates for penicillin [75.68% (28/37)], ampicillin [70.27% (26/37)], and tetracycline [70.27% (26/37)] were detected. Multidrug resistance was observed in seven [18.92% (7/37)] isolates, and two were suggestive of Methicillin-resistant Staphylococcus aureus (MRSA). Among the 37 isolates, 33 novel sequence types (ST) and two known STs (ST126 and ST746) were identified in MLST. The clonal complexes more frequently observed were: CC97 [78.38%; (29/37)], CC1 [8.11%; (3/37)] and CC5 [5.40%; (2/37)]. Minimum‐spanning tree (MST) analysis according to data from municipalities, herds, and resistance patterns for all isolates did not show any clustering pattern. However, the MST comparing all Brazilian S. aureus isolates deposited in the PubMLST database and from this study depicted an association between the genotype and strain origin (clinical sample). Isolates from this study that belong to CC97 were close to database isolates from milk and dairy products, while those that belong to CC1 and CC5 were close to database isolates from human sources and the environment of dairy farms or industries. In conclusion, our results showed a high rate of resistance to penicillins and tetracyclines and great genetic diversity among the S. aureus isolates from bovine mastitis genotyped in the present study.

ABSTRACT The present study explored the predictive values of milk leukocyte differentials (MLD) as a basis for improving the diagnosis of intramammary infections (IMIs) and subclinical mastitis. Quarter milk samples were collected for bacteriological analysis, quarter somatic cell count (qSCC), and MLD. The MLD were assessed using the cytospin technique, direct microscopic smears, and flow cytometry. The predictive values of each single leukocyte population and useful potential indices that could better reflect immune complexity were also calculated. Changes in the percentage of any leukocyte alone failed to substantially improve the predictive value of qSCC in diagnosing IMIs. Although certain parameters increased the area under the receiver operating characteristic curve (ROC curve) as a result of increased specificity values, a slight reduction in sensitivity was observed. The so-called CD8 complex was a unique parameter which improved both the sensitivity (78.79 %) and the specificity (80.77 %) in IMI diagnosis, resulting in the highest area under the ROC curve (0.87). To diagnose subclinical mastitis, the percentage of macrophages and the sum of the percentage PMNLs and T CD8+ cells divided by the percentage of macrophages showed the highest predictive values (sensitivity = 79.63, specificity = 73.68, and area under the ROC curve = 0.83) in the differentiation of the inflammatory condition status of cows. In conclusion, this study provides further insights into using T CD8+ lymphocytes in diagnosing bovine IMIs, combined with PMNLs and macrophages. The antidromic trend of macrophages vs. PMNLs and T CD8+ lymphocytes due to the increasing qSCCs was crucial to differentiating quarters under both inflammatory and non-inflammatory conditions.

RESUMO: Os objetivos do presente estudo foram (i) genotipar amostras de Corynebacterium pseudotuberculosis, C. silvaticum e C. auriscanis usando Enterobacterial Repetitive Intergenic Consensus (ERIC-PCR), bem como (ii) analisar as relações epidemiológicas entre os isolados de acordo com biovar (Equi e Ovis), espécie, hospedeiro e origem geográfica das amostras de C. pseudotuberculosis. Sessenta e oito isolados de C. pseudotuberculosis, nove C. silvaticum, um C. auriscanis, C. pseudotuberculosis ATCC® 19410 ™ e a amostra vacinal atenuada C. pseudotuberculosis 1002 foram tipificadas por ERIC 1 + 2-PCR. As amostras de campo foram isoladas de diferentes hospedeiros (bovinos, búfalos, ovinos, caprinos, equinos, cães e suínos) em seis países (México, Portugal, Brasil, Guiné Equatorial, Egito e Israel). Uma alta diversidade genética foi observada entre os isolados de Corynebacterium spp., agrupados em vinte e quatro genótipos com um índice de diversidade Hunter & Gaston (HGDI) de 0,937. A análise da minimal spanning tree (MST) de Corynebacterium spp. revelou três complexos clonais, cada um associado a uma espécie bacteriana. Vinte e dois genótipos foram observados entre isolados de C. pseudotuberculosis, com um HGDI de 0,934. Na análise da MST, três grandes complexos clonais foram formados, agrupando-se em torno da origem geográfica dos isolados de C. pseudotuberculosis. Esses resultados reforçam a alta tipabilidade, concordância epidemiológica e poder discriminatório do ERIC-PCR como método consistente de genotipagem para C. pseudotuberculosis, podendo ser útil como ferramenta epidemiológica no controle da linfadenite caseosa. Além disso, os resultados também indicam o grande potencial de ERIC 1 + 2-PCR para genotipagem de espécies do gênero Corynebacterium além de C. pseudotuberculosis.

ABSTRACT: The aims of the present study were (i) to genotype Corynebacterium pseudotuberculosis, C. silvaticum, and C. auriscanis strains using enterobacterial repetitive intergenic consensus (ERIC-PCR), and (ii) to analyze the epidemiological relationships among isolates according to biovar (Equi and Ovis), species, host, and geographical origin of the C. pseudotuberculosis strains. Sixty-eight C. pseudotuberculosis, nine C. silvaticum, and one C. auriscanis, C. pseudotuberculosis ATCC® 19410™ strain and the attenuated C. pseudotuberculosis 1002 vaccinal strain were fingerprinted by ERIC 1+2-PCR. Field strains were isolated from various hosts (cattle, buffaloes, sheep, goats, horses, dogs, and pigs) in six countries (Mexico, Portugal, Brazil, Equatorial Guinea, Egypt, and Israel). High genetic diversity was found among the studied Corynebacterium spp. isolates, clustering in 24 genotypes with a Hunter & Gaston diversity index (HGDI) of 0.937. The minimal spanning tree of Corynebacterium spp. revealed three clonal complexes, each associated with one bacterial species. Twenty-two genotypes were observed among C. pseudotuberculosis isolates, with an HGDI of 0.934. Three major clonal complexes were formed at the minimal spanning tree, grouped around the geographic origin of C. pseudotuberculosis isolates. These results reinforce the high typeability, epidemiological concordance, and discriminatory power of ERIC-PCR as a consistent genotyping method for C. pseudotuberculosis, which could be useful as an epidemiological tool to control caseous lymphadenitis. Moreover, our results also indicate the potential of ERIC 1+2-PCR for the genotyping of other species of Corynebacterium other than C. pseudotuberculosis.

RESUMO: Campilobacteriose Genital Bovina (CGB) é uma doença venérea e subclínica que causa problemas reprodutivos em rebanhos, causada por Campylobacter fetus subsp. venerealis. Este trabalho teve como objetivo selecionar peptídeos miméticos ao agente da CGB de uma biblioteca de fagos. Phage display é uma técnica que aplica bibliotecas de bacteriófagos que expõem peptídeos fusionados ao capsídeo viral em seleções biológicas contra proteínas alvo. Biopannings foram realizados para seleção biológica na biblioteca de fagos por meio de soro hiperimune de coelho e extrato proteico de C. fetus subsp. venerealis. Cinco heptapeptídeos selecionados foram considerados miméticos para Cfv-NCTC 10354 a partir de análises de bioinformática e ensaios com soro hiperimune e muco cérvico-vaginal de novilhas. ALASLPL e LSYLFPP foram os peptídeos mais reativos e considerados promissores como possíveis imunógenos miméticos para C. fetus subsp. venerealis.

ABSTRACT: Bovine genital campylobacteriosis (BGC) is a venereal and subclinical disease that affects the fertility of cattle herds, and it is caused by Campylobacter fetus subsp. venerealis . This study selected peptides mimetic to the BGC-causing agent from a phage library. Phage display is a technique that applies bacteriophage libraries that reveal peptides fused to the viral capsid in biological selections against target proteins. Biopannings were performed for biological selection in the phage library using rabbit hyperimmune serum and C. fetus subsp. venerealis protein extract. Five selected heptapeptides were considered mimetic to Cfv-NCTC 10354 based on the results of bioinformatics analysis and assays with hyperimmune serum and cervicovaginal mucus obtained from heifers. ALASLPL and LSYLFPP were the most reactive peptides and considered promising as possible mimetic immunogens for C. fetus subsp. venerealis.

RESUMO: A leptospirose é uma zoonose causada pelo agente etiológico Leptospira. Cães domésticos atuam como hospedeiro de diversos sorovares deste agente. Com intuito de analisar a dinâmica da leptospirose em uma população canina, com e sem imunização, um estudo longitudinal foi realizado avaliando a resposta sorológica destes animais e identificando seus sorovares. Foram coletadas amostras de 331, 373 e 347 cães em três anos consecutivos (2015 a 2017). As soroprevalências foram de 11%, 7% e 14%, respectivamente, e a incidência em 2016 foi de 5% e em 2017 de 14%. Os sorovares mais frequentes foram Cynopteri e Butembo em 2015, Cynopteri, Butembo e Hardjoprajitno em 2016, e Canicola e Butembo em 2017. Estes cães estão atuando como bio-indicadores da presença de Leptospira na região do estudo, incluindo sorovares zoonóticos, e contribuindo com a sua manutenção no ambiente. A soropositividade para sorovares protegidos pela vacina foi mais alta do que nos anos anteriores à imunização, enquanto para os sorovares não protegidos pela vacina diminuiu, demonstrando a importância da imunização para essa população de cães. Medidas de prevenção e controle para a leptospirose, como imunização e controle populacional canino, são recomendadas no local para inibir a transmissão do agente entre as populações de cães e humanos envolvidas.

ABSTRACT: Leptospirosis is a zoonotic disease caused by Leptospira and domestic dogs can act as host of some serovars. In order to analyze the transmission dynamics in a dog population, with and without immunization, a longitudinal study was carried out with a focus to evaluate antibody response and to identify serovars. Blood samples were collected in three consecutive years (2015 to 2017) from 331, 373 and 347 dogs respectively. The dog seroprevalence in each year was 11%, 7% and 14%, respectively, and the incidence in 2016 was 5% and in 2017, 14%. The most frequent serovars were Cynopteri and Butembo in 2015, Cynopteri, Butembo and Hardjoprajitno in 2016, and Canicola and Butembo in 2017. Dogs can play a role as sentinel animals and hosts of Leptospira serovars. The percentage of seropositive dogs due to vaccination was higher than the previous years without immunization and lower than in previous years for other serovars, which we interpret as evidence for the importance of immunization. These parameters associated with active canine population control are important for prevention and control of leptospirosis not only in dogs but alsoto inhibit the transmission between dogs and humans.

RESUMO: O objetivo foi determinar os fatores de virulência, os grupos filogenéticos e as possíveis relações entre os patovares e os grupos filogenéticos identificados de cepas de Escherichia coli isoladas de fezes de bezerros bubalinos. Um total de 217 amostras de E. coli foram identificadas a partir de cultura das fezes e submetidas a reação em cadeia da polimerase (PCR) para detecção dos fatores de virulência EAST-1, enterohemolisina, Saa, CNF2, F41, F5, STa, intimina, Stx1 e Stx2. Foram identificadas 134 cepas positivas para um ou mais fatores de virulência: 84isoladas de bezerros bubalinos diarreicos e 50 de bezerros bubalinos saudáveis. Os patovares de E. coli obtidos de fezes diarreicas foram ETEC (F5+) (2/84), NTEC (16/84), STEC (20/84), EPEC (3/84), EHEC (3/84), e EAEC (EAST-1+) (33/84). Os patovares isolados de fezes não diarreicas foram NTEC (21/50), STEC (17/50), EHEC (1/50) e EAEC (7/50). Cepas de E. coli positivas para EAST-1 (P = 0,008) e filogrupo C (P = 0,05) foram associadas com a presença de diarreia. A análise de filogrupos revelou que 58,95% dos isolados pertencem ao filogrupo B1, seguido por E (9,70%), B2 (5,90%), C (5,90%), D (5,22%), A (2,24%) e F (1,50%). O filogrupo B1 predomina em cepas de E. coli patogênicas isoladas de bezerros búfalos e o filogrupo C constitui um filogrupo de E. coli patogênica entérica para bezerros.

ABSTRACT: This study aimed to determine the virulence factors, phylogenetic groups, and the relationships between pathovars and phylogenetic groups of E. coli strains isolated from feces of buffalo calves. A total of 217 E. coli strains were obtained from feces after culture and were screened by PCR for detection of virulence factors EAST-1, enterohemolysin, Saa, CNF2, F41, F5, STa, intimin, Stx1 and Stx2. One hundred and thirty-four isolates were positive for one or more virulence factors: eighty-four from diarrheic animals, and fifty from non-diarrheic calves. The pathovars of E. coli identified in diarrheic feces were ETEC (F5+) (2/84), NTEC (16/84), STEC (20/84), EPEC (3/84), EHEC (3/84), and EAEC (EAST-1+) (33/84). Pathovars identified in non-diarrheic animals were NTEC (21/50), STEC (17/50), EHEC (1/50) and EAEC (7/50). E. coli strains positive for EAST-1 (P=0.008) and phylogroup C (P = 0.05) were associated with the presence of diarrhea. Phylogenetic analysis showed that 58.95% of the isolates belonged to phylogroup B1, followed by E (9.70%), B2 (5.90%), C (5.90%), D (5.22%), A (2.24%), and F (1.50%). Phylogroup B1 predominated in pathogenic E. coli isolated from water buffalo, and phylogroup C constituted an enteropathogenic E. coli for water buffalo calves.

Abstract A modified TaqMan real-time polymerase chain reaction targeting a 138 bp fragment within the lipl32 gene was developed to identify exclusively pathogenic Leptospira spp. in dog urine samples. Thirty-five samples from dogs with suspected clinical leptospirosis and 116 samples from apparently healthy dogs were tested for presence of leptospiral DNA using the TaqMan-based assay. The results were compared with those from a well-established conventional PCR targeting the 16S RNA encoding gene associated with nucleotide sequencing analysis. The overall agreement between the assays was 94.8% (confidence interval [CI] 95% 88-100%). The newly developed assay presented 91.6% (CI 95% 71.5-98.5%) relative sensitivity (22[+] lipl32 PCR/24[+] 16S RNA and sequencing), 100% (CI 95% 96.3-100%) relative specificity and 98.7% accuracy (CI 95% 94.8-100%). The lipl32 assay was able to detect and quantify at least 10 genome equivalents/reaction. DNA extracted from 17 pathogenic Leptospira spp., 8 intermediate/saprophytic strains and 21 different pathogenic microorganisms were also tested using the lipl32 assay, resulting in amplification exclusively for pathogenic leptospiral strains. The results also demonstrated high intra and inter-assay reproducibility (coefficient of variation 1.50 and 1.12, respectively), thereby qualifying the newly developed assay as a highly sensitive, specific and reliable diagnostic tool for leptospiral infection in dogs using urine specimens.

ABSTRACT Although the tuberculin test represents the main in vivo diagnostic method used in the control and eradication of bovine tuberculosis, few studies have focused on the identification of mycobacteria in the milk from cows positive to the tuberculin test. The aim of this study was to identify Mycobacterium species in milk samples from cows positive to the comparative intradermal test. Milk samples from 142 cows positive to the comparative intradermal test carried out in 4,766 animals were aseptically collected, cultivated on Lowenstein-Jensen and Stonebrink media and incubated for up to 90 days. Colonies compatible with mycobacteria were stained by Ziehl-Neelsen to detect acid-fast bacilli, while to confirm the Mycobacterium genus, conventional PCR was performed. Fourteen mycobacterial strains were isolated from 12 cows (8.4%). The hsp65 gene sequencing identified M. engbaekii (n=5), M. arupense (n=4), M. nonchromogenicum (n=3), and M. heraklionense (n=2) species belong to the Mycobacterium terrae complex. Despite the absence of M. tuberculosis complex species in the milk samples, identification of these mycobacteria highlights the risk of pathogen transmission from bovines to humans throughout milk or dairy products, since many of mycobacterial species described here have been reported in pulmonary and extrapulmonary diseases both in immunocompetent and immunocompromised people.

RESUMO: Infecções por Staphyloccocus pseudintermedius (SP) na pele de cães tem sido objeto de estudos, uma vez que esses microrganismos geralmente apresentam resistência múltipla à antibióticos. Este estudo teve como objetivo identificar e avaliar a susceptibilidade antimicrobiana das cepas de SP isoladas de cães com otite. Amostras de ambas orelhas de 52 cães com otite (n=104) foram incluídas. As bactérias foram cultivadas em ágar Muller-Hinton suplementado com 5% de sangue equino e incubadas a 37ºC por 24 a 48h. Todas as colônias foram submetidas à avaliação bioquímica para identificação de estafilococos. A identificação das colônias como SP foi confirmada pela reação em cadeia da polimerase. A susceptibilidade antimicrobiana das cepas SP foi avaliada pela técnica de difusão em disco. A presença do gene mecA foi avaliada em todos os isolados de SP por PCR. 44 estirpes de SP foram isoladas de 31 cães (31/52, 59,6%). 75% das cepas foram suscetíveis a cefalexina e 93,2% à amoxicilina mais ácido clavulânico. Menos de 23% das estirpes eram suscetíveis à penicilina. Para antimicrobianos não beta-lactâmicos, 63,6% apresentaram resistência ao sulfametoxazol-trimetoprim, 61,4% à tetraciclina e 38,64% à enrofloxacina. A frequência de resistência a aminoglicosídeos foi de 27,3% para gentamicina. A resistência a oxacilina in vitro foi detectada em 13 das 44 estirpes (29,55%). Um total de 12 amostras (27,3%) foram positivas para mecA, cinco das quais foram suscetíveis à oxacilina na difusão em disco. 26 amostras (59,1%) foram resistentes a três ou mais classes de antimicrobianos e classificadas como multirresistentes. Nossos resultados mostram elevada frequência de isolados SP e multiresistentes para os antimicrobianos comumente usados em veterinária.

ABSTRACT: Infections by Staphyloccocus pseudintermedius in the skin of dogs have been object of studies, since such microorganisms often present multiple resistance to antibiotics. This study aimed to identify and evaluate the antimicrobial susceptibility of Staphylococcus pseudintermedius (SP) strains isolated from dogs with otitis. Swabs from both ears of 52 dogs with otitis (n=104) were included. Bacteria were cultured using Muller-Hinton agar (supplemented with 5% equine blood and incubated at 37°C for 24 to 48 hours. All colonies underwent biochemical evaluation for identification of staphylococci. The identity of colonies as SP was confirmed by polymerase chain reaction. The antimicrobial susceptibility of SP strains was evaluated by disk diffusion. The presence of the gene mecA was evaluated in all SP isolates by PCR. Forty-four SP strains were isolated from swabs of 31 dogs (31/52, 59.6%). Seventy-five percent of the strains were susceptible to cephalexin and 93.2% to amoxicillin plus clavulanic acid. Less than 23% of the strains were susceptible to penicillin. For non-beta-lactam antimicrobials, 63.6% of the strains showed resistance to sulfamethoxazole-trimethoprim, 61.4% to tetracycline, and 38, 64% to enrofloxacin. Aminoglycoside resistance rate was 27.3% for gentamicin. Resistance to oxacillin in vitro was detected in 13 of the 44 strains (29, 55%). A total of 12 strains (27.3%) were positive for mecA gene and five of these 12 strains were susceptible to in vitro oxacillin. Twenty-six (59, 1%) strains were resistant to three or more classes of antimicrobials, and classified as multi resistant. Our results showed high frequency of SP and multi resistant isolates to antimicrobials commonly used in veterinary.

RESUMO: Em áreas onde a tuberculose humana e a tuberculose bovina coexistem, a diferenciação entre M. bovis e M. tuberculosis é importante para monitorar a disseminação de M. bovis entre bovinos e destes para os seres humanos. Objetivou-se neste estudo isolar e identificar M. bovis em bovinos com diagnóstico positivo pelo teste de tuberculinização no estado da Paraíba, nordeste do Brasil. Foram submetidos 32 bovinos positivos ao teste de tuberculinização comparativa, dos quais foram colhidas amostras de qualquer órgão com lesões sugestivas de tuberculose, e, nos casos em que não foram observadas lesões sugestivas, foram colhidas amostras de linfonodos. As amostras foram submetidas a exame histopatológico, cultivo micobacteriológico, coloração de Ziehl-Neelsen e diagnóstico molecular. Apresentaram lesões sugestivas de tuberculose 21 animais (65,6%). Com relação à distribuição das lesões de acordo com a região corporal, 77,7% localizavam-se na cavidade torácica, 12,4% na cabeça e 9,9% na cavidade abdominal. De 55 amostras submetidas ao cultivo de micobactérias, em 31 (56,4%) foram isoladas micobactérias, sendo que em 13 (41,9%) foi identificado M. bovis, e nas 18 restantes (58,1%) foi identificado Mycobacterium spp. Conclui-se que o isolamento e a identificação de M. bovis e Mycobacterium spp. em bovinos indicam que os seres humanos estão expostos ao risco de infecção. Isso reforça a necessidade de intensificação e otimização de medidas de prevenção e controle previstas no Programa Nacional de Controle e Erradicação da Brucelose e Tuberculose Bovina. Sugere-se a realização de estudos de isolamento e identificação de micobactérias em outros estados do Nordeste.

ABSTRACT: In areas where human tuberculosis and bovine tuberculosis coexist, differentiation between M. bovis and M. tuberculosis is important for monitoring the spread of M. bovis among cattle and from cattle to humans. The objective of this study was to isolate and identify M. bovis in bovines with positive diagnosis identified on tuberculin test in the State of Paraíba, Northeastern Brazil. Thirty-two bovines that tested positive in the comparative tuberculin test were used, from which samples of any organ with lesions suggestive of tuberculosis were collected, as well as lymph nodes, when no gross lesions were observed. Samples were submitted to histopathological exam, mycobacterial culture, Ziehl-Neelsen staining and molecular diagnosis. Twenty-one (65.6%) animals presented lesions suggestive of tuberculosis. As to body region 77.7% of lesions were found in the thoracic cavity, 12.4% in the head and 9.9% in the abdominal cavity. Among 55 samples submitted to mycobacterial culture, mycobacteria were isolated in 31 (56.4%), being 13 (41.9%) identified as M. bovis and 18 (58.1%) as Mycobacterium spp. Conclusion is that isolation and identification of M. bovis and Mycobacterium spp. in cattle suggests that humans are exposed to the risk of infection. This reinforces the need for intensification and optimization of prevention and control measures foreseen in the Brazilian National Program for the Control and Eradication of Bovine Brucellosis and Tuberculosis. Mycobacteria isolation and identification surveys are, therefore, encouraged in other Northeastern states.

RESUMO: Este estudo pesquisou genes de virulência, patovares e grupos filogenéticos de amostras de E. coli isoladas de fezes de cães com e sem diarreia. Os genes de virulência e a identificação de grupos filogenéticos foram estudados pela técnica de reação em cadeia da polimerase (PCR). 37 isolados de E. coli foram positivos para pelo menos um fator de virulência na análise de PCR. Destes, 21 (57,8%) foram isolados de fezes de cães com diarreia e 16 (43,2%) de fezes de cães não diarreicos. E. coli enteropatogênica (EPEC) (23/37, 62,2%) foi o patovar mais frequente detectado em fezes de cães e foram classificados principalmente como filogrupos B1 e E. E. coli necrotoxigênica (NTEC) positivos para CNF1 foram detectados (6/37, 16,2%) e classificados como B2 e D. Todas as amostras de E. coli foram negativas quanto à presença de genes de enterotoxinas de E. coli enterotoxigênica (ETEC), mas quatro amostras foram positivas para fimbrias relacionadas ao ETEC, 987P (2) e F18 (2). As amostras de E. coli (STEC) produtora de toxina Shiga foram positivas para a toxina Stx2 (1/37) e Stx2e (1/37), ambas do filogrupo B1. Nossos resultados indicaram que EPEC foi o patovar mais frequente e B1 e E foram os filogrupos mais comuns detectados em amostras E. coli isoladas de fezes de cães diarreicos e não diarreicos.

ABSTRACT: This study identified the virulence genes, pathovars, and phylogenetic groups of Escherichia coli strains obtained from the feces of dogs with and without diarrhea. Virulence genes and phylogenetic group identification were studied using polymerase chain reaction. Thirty-seven E. coli isolates were positive for at least one virulence factor gene. Twenty-one (57.8%) of the positive isolates were isolated from diarrheal feces and sixteen (43.2%) were from the feces of non-diarrheic dogs. Enteropathogenic E. coli (EPEC) were the most frequently (62.2%) detected pathovar in dog feces and were mainly from phylogroup B1 and E. Necrotoxigenic E. coli were detected in 16.2% of the virulence-positive isolates and these contained the cytotoxic necrotizing factor 1 (cnf1) gene and were classified into phylogroups B2 and D. All E. coli strains were negative for the presence of enterotoxigenic E. coli (ETEC) enterotoxin genes, but four strains were positive for ETEC-related fimbriae 987P and F18. Two isolates were Shiga toxin-producing E. coli strains and contained the toxin genesStx2 or Stx2e, both from phylogroup B1. Our data showed that EPEC was the most frequent pathovar and B1 and E were the most common phylogroups detected in E. coli isolated from the feces of diarrheic and non-diarrheic dogs.

RESUMO: Pseudorraiva (PR) é uma doença viral altamente contagiosa de grande importância sanitária e econômica na indústria suína. O objetivo deste estudo foi avaliar diferentes regiões genômicas, químicas de PCR em tempo real e equipamentos para o diagnóstico molecular de PR. Foram avaliados quatro genes (GB, GC, GE, gD), três diferentes produtos químicos (SybrGreen, sondas de hidrólise e plexor) e dois equipamentos (ABI7500, Rotorgene 3000). Oligonucleotídeos com alvo para gB baseados na química de sondas de hidrólise apresentaram o melhor desempenho nos testes de eficiência (99%), de limite de detecção (10-1.5 TCID50 mL-1) e sensibilidade diagnóstica. Portanto, estes foram selecionados para fatores de verificação de desempenho, tais como a repetibilidade, reprodutibilidade e robustez (1,39% de variância entre dias, 24% variância entre analistas e 4,07% de variância por erro de análise). A qPCR, padronizada e validada neste trabalho, mostrou-se confiável para o diagnóstico de PR e pode ser utilizada em laboratórios de diagnóstico que se seguem normas internacionais como ISO 17025 e ISO 16140.

ABSTRACT: Pseudorabies (PR) is a highly contagious viral disease of great animal health and economic importance in swine industry. The aim of this study was to evaluate different genomic regions, real-time PCR chemistries and equipment for the molecular diagnosis of PR. Eight primer pairs targeting four genes (gB, gC, gE, gD), three different qPCR chemistries (SybrGreen, hydrolysis probes and plexor) and two equipment (ABI7500, Rotorgene 3000) were evaluated. Oligonucleotides targeting gB using hydrolysis probes showed the best performance after evaluating efficiency (99%), the detection limit (10-1.5 TCID50 mL-1) and diagnostic sensitivity and; therefore, those primers were selected for performance verification factors such as repeatability, reproducibility and robustness (1.39% variance between days, 24% variance between analysts and 4.07% variance in analysis error). The qPCR standardized and validated in this research proved to be reliable for the diagnosis of PR and may be used in diagnostic laboratories that follow ISO 17025 and ISO 16140.

RESUMO: Amostras de leite de 16 vacas positivas no teste da tuberculinização no Estado da Paraíba, Nordeste do Brasil, foram utilizadas para isolamento e identificação de micobactérias. Foram isoladas micobactérias em cinco (31,25%) das 16 amostras de leite: três amostras foram classificadas como M. bovis e duas como pertencentes ao gênero Mycobacterium. De acordo com nosso conhecimento, este é o primeiro estudo de isolamento e identificação de M. bovis em leite de vacas no Nordeste do Brasil, o que sugere que os seres humanos estão em risco de contaminação por ingestão.

ABSTRACT: Milk samples from 16 cows that tested positive on the tuberculin test in the state of Paraíba, northeastern Brazil, were used for mycobacteria isolation and identification. Mycobacteria were isolated from five (31.25%) of the 16 milk samples; three samples were classified as M. bovis, and two as belonging to the Mycobacterium genus. This is probably the first study of isolation and identification of M. bovis in milk from cows in Northeastern Brazil, which suggests that humans are at risk of contamination by ingestion.

Resumo: O objetivo deste trabalho foi investigar a presença do herpesvírus bovino tipo 1 (BoHV-1) no fluido folicular e em complexos cumulus-oócito (COC) coletados de vacas naturalmente infectadas mas sem sinais clínicos da doença. Vacas soropositivas (n=38) ou soronegativas (n=8, controle) para o vírus da rinotraqueíte infecciosa bovina foram selecionadas por prova de soroneutralização em microplacas. A presença do vírus foi investigada por PCR em COC e fluido folicular. O DNA viral não foi encontrado em qualquer das amostras. Os resultados obtidos são indicativos de que vacas soropositivas, mas sem sinais clínicos da doença, oferecem risco negligível de transmissão do BoHV-1 por COC ou fluido folicular.

Abstract: The objective of this work was to investigate the presence of bovine herpesvirus type 1 (BoHV-1) in follicular fluid and in cumulus-oocyte complexes (COC) recovered from naturally infected cows but with no clinical signs of the disease. Cows that were seropositive (n=38) or seronegative (n=8, control) to infectious bovine rhinotracheitis were selected after a serum neutralization test in microplates. The presence of the virus was investigated by PCR in COC and in follicular fluid. Viral DNA was not found in any of the samples. The obtained results suggest that serologically positive cows with no clinical signs of the disease offer negligible risk of transmitting BoHV-1 by COC or follicular fluid.

Complexo de Doenças Respiratórias de Suínos (CDRS ) é um grupo de doenças que causam grandes perdas na indústria suína. Vários agentes infecciosos estão relacionados com a CDRS , entre eles o circovírus suíno 2 (PCV -2), vírus da pseudo-raiva (SuHV -1) , Haemophilus parasuis (HP) ,Mycoplasma hypneumoniae (MH ) e Pasteurela multocida (PM). O objetivo com este estudo foi desenvolver PCR em tempo real (qPCR) para a detecção destes agentes infecciosos. Os oligonucleotídeos foram concebidos para cada agente infeccioso específico e marcado com fluoróforos diferentes para amplificar partes específicas do genoma em dois grupos de reacções , uma qPCR dúplex em SuHV -1 e PCV- 2 e uma qPCR multiplex para detectar as três bactérias simultaneamente. As reações foram testadas em 142 amostras de pools de linfonodos e pulmões de suínos com sinais clínicos de CDRS. Foram detectadas 135 amostras positivas para PCV- 2 , 61 para a HP, 29 para PM e 30 para MH e zero para SuHV -1, dentre esses foram registrados 76 casos de co-infecção . As qPCRs desenvolvidas neste estudo são ferramentas úteis no diagnóstico da CDRS.

Porcine Respiratory Disease Complex (PRDC) is a group of diseases that cause high losses in the swine industry. Several infectious agents are related to PRDC including porcine circovirus 2 (PCV-2), pseudorabies virus (SuHV-1),Haemophilus parasuis (HP), Mycoplasma hypneumoniae (MH) and Pasteurela multocida (PM). The aim of this study was to develop real-time PCRs (qPCR) for the detection of these infectious agents. Oligonucleotides were designed for each specific infectious agent and labeled with different fluorophores to amplify specific parts of the genome. This was done in two groups of reactions—a duplex qPCR for SuHV-1 and PCV-2 and a multiplex qPCR to detect the three bacteria simultaneously. The reactions were tested in 142 pooled samples of swine lymph nodes and lungs with clinical signs of PRDC. There were 135 samples that tested positive for PCV-2, 61 for HP, 29 for PM, 30 for MH and zero for SuHV-1. We recorded 76 cases of co-infection. The qPCRs developed in this study are useful tools in the diagnosis of PRDC.
