Resumo O objetivo deste estudo foi otimizar e estabelecer um protocolo visando elevar a qualidade e quantidade do ácido desoxirribonucleico (DNA) extraído de folhas, em diferentes estágios de desenvolvimento e conservação, coletadas em plantas adultas e plantas jovens. Além disso, almejou-se padronizar a amplificação via reação de polimerase em cadeia (PCR) e selecionar os marcadores de inter sequência simples repetida (ISSR) de Elaeis guineensis (dendezeiro). O protocolo brometo de cetiltrimetilamônio (CTAB) modificado, devido à suplementação com betamercapto etanol (0,3%) e polivinilpirrolidone (PVP) (3%) no tampão de extração e CTAB 10% com 1,4 M de NaCl a 20 min de incubação, a 650C, resultou em melhorias qualitativas e quantitativas na extração do DNA, mas com variações entre amostras, provavelmente, devido às variações na degradação e estágio de desenvolvimento das folhas. Foram utilizados dois protocolos de PCR (I e II) para a amplificação do DNA, os quais diferiram principalmente com relação à presença de albumina de soro bovino (BSA) e concentração de primer. Não foi realizada a correlação entre amplificação via PCR e qualidade de DNA, obtidos de folhas danificadas ou sadias, mas se observou que a adição de BSA (0,075 mg/mL) e o aumento na concentração de primer (0,5 pcmol) (protocolo II) resultou em bandas mais intensas e distinguíveis, porém se ressalta que a qualidade do DNA foi essencial para uma boa amplificação, considerando-se todas as amostras. A amplificação do DNA com o protocolo II possibilitou a seleção de cinco primers: UBC 807, 810, 812, 834 e 848; os quais foram utilizados para a amplificação de13 famílias, compostas de uma planta parental e oito progênies cada.

Abstract The objective of this study was to optimize and establish a protocol to enhance the quality and quantity of deoxyribonucleic acid (DNA) extracted from leaves under different development stages and conservation conditions collected from mature plants and seedlings. Furthermore, we aimed to standardize the polymerase chain reaction (PCR) and select an inter simple sequence repeat (ISSR) marker for Elaeis guineensis (oil palm). The modified cetyltrimethylammonium bromide (CTAB) protocol, which used β-mercaptoethanol (0.3%) and polyvinylpyrrolidone (PVP 3%) supplementation in the extraction buffer and 10 % CTAB with 1.4 M NaCl for a 20-min incubation at 65ºC, resulted in improved DNA quality and quantity. However, this protocol presented variable results among samples, probably due to variations in leaf degradation levels and development stages. The two PCR protocols (I and II) for amplifying the DNA, differed mainly in the presence or absence of bovine serum albumin (BSA) and primer concentration. Although, a correlation between PCR amplification and the quality of the DNA extracted from leaves was not established, the addition of BSA (0.075 mg/mL) and the highest primer concentration (0.5 pmol) (protocol II) resulted in more intense and distinguishable bands on gel electrophoresis. DNA quality was essential for a satisfying amplification, considering all the samples. The use of protocol II allowed the selection of five primers: UBC 807, 810, 812, 834, and 848; these were used in the amplification of the DNA extracted from 13 families consisting of one parental plant and eight progenies each.

Abstract Forest loss and fragmentation are the main threats to the maintenance of the Atlantic Forest, an important global biodiversity hotspot. Because of the current critical level of deforestation, ecological corridors are needed to facilitate species dispersion and gene flow among fragments. This study was conducted to investigate the genetic variability and gene pool sharing of Eschweilera ovata in five forest remnants in southern Bahia, Brazil using nuclear simple sequence repeat (nSSR) and plastid simple sequence repeat (cpSSR) microsatellite markers. cpSSR marker analysis revealed the domains of four haplotypes, showing that 80% of the individuals had only four maternal origins, reflecting a founder effect and/or genetic bottleneck. The results of cpSSR and nSSR analyses indicated moderate genetic diversity, particularly in conservation units with full protection, which showed the best parameters of all areas evaluated. Another indication of the susceptibility of these populations to forest loss and fragmentation was the strong genetic bottleneck observed. In contrast, genetic structure analyses (FST and discriminant analysis of principal components) revealed gene pool sharing between the subpopulations, which may reflect the historical gene flow that occurred before forest fragmentation.

Studies investigating rickettsial infections in ticks parasitizing wild animals in the Northeast region of Brazil have been confined to the detection of Rickettsia amblyommii in immature stages of Amblyomma longirostre collected from birds in the state of Bahia, and in immatures and females of Amblyomma auriculariumcollected from the striped hog-nosed skunk (Conepatus semistriatus) and armadillos (Euphractus sexcinctus) in the state of Pernambuco. The current study extends the distribution of R. amblyommii (strain Aranha), which was detected in A. longirostre collected from the thin-spined porcupine Chaetomys subspinosus and the hairy dwarf porcupine Coendou insidiosus. In addition, we report the first detection of Rickettsia bellii in adults of A. longirostre collected from C. insidiosus in the state of Bahia.

Poucos estudos ictiofaunísticos estão disponíveis em rios típicos do semi-árido, apesar da constante ameaça à diversidade local devido a influências antrópicas, com destaque para o represamento e construção de barragens. O presente trabalho teve como objetivo avaliar, por meio de diferentes metodologias, a estrutura genética de populações de uma espécie de caracídeo, Astyanax aff. bimaculatus, amplamente distribuída em bacias hidrográficas da Bahia, Nordeste do Brasil. Análises morfológicas (dados merísticos e morfométricos), citogenéticas (cariótipo e Ag-RONs) e moleculares (RAPD e SPAR) foram realizadas em espécimes coletados à montante e à jusante da Barragem da Pedra, na calha principal do médio rio de Contas (bacia do Rio de Contas) e no ribeirão Mineiro, pertencente à bacia adjacente do Recôncavo Sul. Poucas diferenças externas foram detectadas entre as populações, sendo os indivíduos originários do reservatório, à montante da barragem, ligeiramente maiores. Os dados citogenéticos também mostraram padrões cariotípicos semelhantes (2n=50; 6m+28sm+12st+4a; FN= 96) e RONs situadas nos braços curtos de até dois pares cromossômicos, com variação numérica inter- e intra-populacional. Contudo, as análises por RAPD e SPAR diferenciaram as três populações de forma eficiente, revelando freqüências alélicas significativamente diferentes entre as localidades amostradas e índices significativos de estruturação populacional (Fst=0.1868, P<0.0001). As diferenças entre populações do mesmo rio foram tão significativas quanto entre bacias hidrográficas distintas, indicando que a represa constitui uma barreira eficiente ao fluxo gênico. Além disso, acredita-se que peculiaridades ambientais de cada localidade possam também influenciar os padrões genéticos encontrados. Por outro lado, a similaridade entre amostras das bacias do Rio de Contas e Recôncavo Sul pode estar relacionada a uma história evolutiva comum, já que ambas estão geograficamente próximas. Por fim, o presente estudo demonstra que a realização de estudos envolvendo diferentes marcadores é extremamente útil para a identificação de estruturas de populações em espécies amplamente distribuídas e para avaliação dos impactos das atividades humanas sobre as populações naturais de peixes.

Few reports are available about the ichthyofauna of typical semi-arid rivers, although the regional diversity has been constantly threatened by human activities, mainly related to impoundment and construction of dams. The goal of the present work was to evaluate using different methods, the population genetic structure of a characin fish, Astyanax aff. bimaculatus, widespread throughout hydrographic basins of Bahia, Northeastern Brazil. Morphological (meristic and morphometric data), cytogenetic (karyotype and Ag-NOR), and molecular (RAPD and SPAR) analyses were carried out in specimens collected upstream and downstream of Pedra Dam, in the main channel of Contas River (Contas River Basin), and in the Mineiro stream, which belongs to the adjacent Recôncavo Sul basin. Few external differences were detected among populations, where the individuals collected upstream of Pedra Dam were slightly larger than the others. Cytogenetic data also showed a similar karyotypic pattern (2n=50; 6m+28sm+12st+4a; FN= 96) and NORs located on the short arms of up to two chromosome pairs, with numerical inter- and intra-populational variation. Nonetheless, RAPD and SPAR analyses differentiated reliably the three populations, revealing striking differences in the allele frequencies among the localities studied and a significant difference in population structure index (Fst=0.1868, P<0.0001). The differences between populations within a same river were as significant as those between distinct hydrographic basins, indicating that the dam/reservoir represents an effective barrier to gene flow. Additionally, environmental peculiarities from each locality are also believed to influence the genetic patterns detected herein. On the other hand, the similarity between samples from Contas River and Recôncavo Sul basins could be related to a common evolutionary history, since both basins are geographically close to each other. Finally, the present study shows that a multi-approach analysis is particularly useful in identifying the population structure of widely distributed species and to evaluate the impacts of human activities on natural fish populations.

O Pontal do Paranapanema é a região mais recentemente desflorestada do Estado de São Paulo. Os efeitos da fragmentação florestal na estrutura genética de Copaifera langsdorffii Desf. foram avaliados por meio da genotipagem, com seis locos microssatélites, de pelo menos 30 indivíduos regenerantes e 30 adultos em três áreas do Pontal do Paranapanema. A amostragem compreendeu dois fragmentos localizados em assentamentos rurais, Tucano (800 ha) e Madre Cristina (300 ha); e uma população localizada em floresta contínua, no Parque Estadual do Morro do Diabo (35.000 ha). Níveis elevados de diversidade genética foram detectados tanto nos regenerantes como nos adultos. A diversidade gênica (He) variou de 0,718 a 0,835 e o número médio de alelos por loco (Â) variou de 8,67 a 11,83, mostrando que a fragmentação ainda não causou perda de alelos. As estimativas elevadas dos índices de fixação (0,183 a 0,387) foram conseqüentes da ocorrência de alelos nulos em alguns locos e de endogamia nas populações. A divergência genética foi pequena entre os adultos (R ST = 0,035), mas aumentou entre os regenerantes (R ST = 0,075). Como esperado, a fragmentação florestal está causando redução do fluxo gênico interpopulacional, especialmente em relação ao fragmento mais distante da floresta contínua (Tucano - cerca de 8 km). Uma alternativa para promover o fluxo gênico entre os remanescentes florestais do Pontal do Paranapanema seria o estabelecimento de sistemas agroflorestais pelos agricultores assentados, os quais atuariam como trampolins ecológicos, favorecendo a dispersão de pólen e sementes entre fragmentos.

The " Pontal do Paranapanema" is the most recently deforested region of the state of São Paulo in Brazil. The effects of forest fragmentation on the population genetic structure of Copaifera langsdorffii Desf. were evaluated by genotyping with six microsatellite loci at least 30 adult and 30 sapling individuals of three populations in " Pontal do Paranapanema" . The sampling areas comprised two fragments located in the rural settlements Tucano (800 ha) and Madre Cristina (300 ha); and one population located in a continuous forest, the " Parque Estadual do Morro do Diabo" (35,000 ha). High levels of genetic diversity were detected in saplings and adults, gene diversity (He) ranged from 0.718 to 0.835 and average number of alleles per locus (Â) ranged from 8.67 to 11.83, showing that the fragmentation has not caused loss of alleles yet. The high estimated fixation indexes (from 0.183 to 0.387) are due to null alleles in some loci and the occurrence of inbreeding within populations. Genetic divergence was low among adults (R ST = 0.035) but increased among saplings (R ST = 0.075). As expected, forest fragmentation has caused a decrease in gene flow among the populations, especially between the most distant fragment (Tucano - about 8 km from the " Parque Estadual do Morro do Diabo" ). An alternative to promote gene flow among forest fragments of " Pontal do Paranapanema" would be the settle of agroforests, which would act as stepping-stones, favouring pollen and seed dispersion among fragments.

O objetivo deste trabalho foi avaliar a variabilidade genética de estoques comerciais do camarão Litopenaeus vannamei, por meio de marcadores RAPD e diferentes métodos estatísticos de análises, em Canavieiras, BA. Vinte primers foram utilizados para a obtenção de 59 marcadores polimórficos. A análise com o programa AMOVA evidenciou diferenciação genética significativa entre os estoques, com fiST = 0,186 (p<0,001). Entretanto, as análises obtidas com o programa HICKORY sugeriram não existir estruturação (<FONT FACE=Symbol>q b</FONT > ou = 0,002), mas considerável taxa de endogamia dentro dos estoques, em razão, provavelmente, do cruzamento entre indivíduos geneticamente mais similares (f = 0,729). Testes com bootstrap mostraram 48 como o número mínimo de marcadores RAPD adequados para estudos de variabilidade genética nessa espécie. Pelo dendrograma, gerado a partir da matriz de similaridade de Jaccard (Sj), observou-se que nos três estoques comerciais estudados existem animais geneticamente distintos.

The objective of this work was to evaluate the genetic variability of commercial stocks of shrimp Litopenaeus vannamei, using RAPD markers and different statistical methods of analysis in Canavieiras, BA, Brazil. Twenty primers were used to obtain 59 polymorphic markers. A significant differentiation among stocks, with phiST = 0.186 (p<0.001), was observed by analysis with the software AMOVA. However, an analysis with the software HICKORY suggested no genetic structure (<FONT FACE=Symbol>q b</FONT > or = 0.002), but considerable inbreeding within stocks, possibly due to consanguineous crossings (f = 0.729). Statistical tests with bootstrap indicated 48 as the minimum number of RAPD markers suitable for studies on genetic variability of this species. By the dendrogram produced from the Jaccard similarity matrix, it was observed that the analyzed stocks are composed of genetically different individuals.
