Abstract Background: Conopeptides from cone snail venom have aroused great interest related to the discovery of novel bioactive candidates, due to their excellent prospects for the treatment of various health problems such as pain, addiction, psychosis and epilepsy. In order to explore novel biopeptides, we investigated the structure and function of five novel conopeptides isolated from the venom of Conus marmoreus from South China Sea. Methods: C. marmoreus crude venom was prepared, fractionated and purified by HPLC system. The primary sequences of the five novel disulfide-poor conopeptides Mr-1 to Mr-5 were identified by comprehensive analysis of de novo MALDI-TOF tandem mass spectrometry and Edman degradation data. In order to investigate their function, these five conopeptides were synthesized by Fmoc-SPPS chemistry, and their biological effects at several heterologous rat nicotinic acetylcholine receptor (nAChR) subtypes (α1β1δε, α3β2, α3β4, α4β2) were determined by electrophysiological technique. Results: Five novel disulfide-poor conopeptides were identified and named as follows: Mr-1 (DWEYHAHPKPNSFWT), Mr-2 (YPTRAYPSNKFG), Mr-3 (NVIQAPAQSVAPP NTST), Mr-4 [KENVLNKLKSK(L/I)] and Mr-5 [NAVAAAN(L/I)PG(L/I)V]. None of them contains a disulfide bond. The sequences of conopeptides Mr-2 to Mr-5 do not belong to any category of the known disulfide-poor conopeptides. No significant activity against the above nAChR subtypes were observed for the five conopeptides at 100 µM. Conclusion: We purified and structurally characterized five novel disulfide-poor conopeptides from C. marmoreus crude venom and first investigated their nAChR inhibitory effects. This work expanded our knowledge on the structure and function of disulfide-poor conopeptides from this cone snail venom.

Abstract In this study, we investigated the preconsolidation pressure (P C) of sedimentary soft rock and its relationship with brittle-ductile transition (BDT) pressure. We proposed a generalized preconsolidation pressure (P G) suitable for both soil and sedimentary soft rock, which is defined in terms of the yield stress of the initial sediment structure under confined lateral compression. We then explored the digenetic effect on P G using 24 sediment types. Next, P G and BDT pressure were verified using confined compression and conventional triaxial tests, respectively, on red-bed soft rock in the Dingxi region of China. Finally, we discussed the BDT mechanism and analyzed the relationship between the two characteristic pressures for a given initial yield surface considered structural strength. We found that the relationship was dependent on the strength parameters, the initial structural strength, and the coefficient of earth pressure at rest. The findings of this study will have a significant impact on the determination of BDT pressure of sedimentary rock in future studies.

Post-stroke depression (PSD) is a very common complication that leads to increased physical disability, poor functional outcome, and higher mortality. Therefore, early detection and treatment are very important. Since there are currently no specific guidelines for this disorder in China, the purpose of this study was to develop PSD guidelines and provide suggestions for clinicians and related workers.

Safeners are an important tool used to ensure the safe using of herbicide. The objective of this paper was to investigate the protective effect of four 3-dichloroacetyl oxazolidine safeners (3-dichloroacetyl-2,2-dimethyl-1,3-oxazolidine [R-28725], racemate of 3-dichloroacetyl-2,2-dimethyl-4- ethyl-1,3-oxazolidine, and its two chiral stereoisomers) in reducing the injury caused by imazethapyr. Physiological and biochemical tests were conducted under laboratory condition, by using seed treatment with safeners and soil treatment with imazethapyr, respectively. The interaction of two safeners (R-28725 and R-stereoisomer) and imazethapyr reduced the injury of maize significantly, and also increased glutathione content, activity of glutathione S-transferases, and activity of acetolactate synthase in maize. When induced by R-stereoisomer, the GSH content in root and in shoot increased 100.7% and 73.6%, respectively. When induced by R-28725, the GST activity in vivo increased threefold and the GST activity in vitro more than doubled. The kinetic parameter Vmax of GST in the maize treated with R-28725 and R-stereoisomer increased by 102.2% and 81.9%, respectively, compared with the control. The results also showed that R-28725 and R-stereoisomer induced glutathione S-transferases affinity for the substrate of conjugation reaction significantly.

INTRODUÇÃO: Radiotraçadores para neuroimagem de transportador de dopamina (TDA) foram desenvolvidos para estimar a perda de neurônios dopaminérgicos in vivo na doença de Parkinson (DP). OBJETIVO: Avaliar a densidade de TDA in vivo utilizando [99mTc]-TRODAT-1 (INER-Taiwan) e SPECT em uma população de pacientes brasileiros com DP. MÉTODO: Quinze pacientes com DP e 15 controles saudáveis pareados realizaram exames de SPECT com [99mTc]-TRODAT-1 (INER-Taiwan). Estimativas da densidade de TDA estriatal foram calculadas usando potencial de ligação (PL). Pacientes foram avaliados com escalas para PD. RESULTADOS: Pacientes com DP apresentaram redução significativa do PL-TDA (0,38±0,12) comparado aos controles (0,84±0,16, p<0,01). Foi possível discriminar casos de DP de controles com uma sensibilidade de 100% e especificidade de 100%. Foram obtidas correlações negativas entre PL-TDA e escalas de severidade da DP (rho= -0,7, p<0,001) e disfunção motora (rho= -0,8, p<0,001). CONCLUSÃO: Exames de SPECT com [99mTc]-TRODAT-1 foram capazes de discriminar pacientes com DP de controles. Esta técnica é um instrumento útil para medir a densidade de TDA e pode ser utilizado para clínica e pesquisa no Brasil.

BACKGROUND: Dopamine transporter (DAT) neuroimaging radiotracers were developed to estimate dopamine neuronal loss in vivo in Parkinson’s disease (PD). OBJECTIVE: To evaluate DAT density in vivo using [99mTc]-TRODAT-1 and single photon computerized tomography (SPECT) in a population of Brazilian PD. METHOD: Fifteen PD patients and 15 matched healthy controls scanned with [99mTc]-TRODAT-1 (INER-Taiwan) and SPECT. Estimates of striatum DAT density were calculated using binding potential (BP). Patients were assessed with PD scales. RESULTS: PD patients had significantly lower striatal DAT-BP (mean±SD) (0.38±0.12) compared to controls (BP=0.84±0.16; p<0.01). A 100% sensitivity and 100% specificity was obtained to discriminate PD cases from controls. Negative correlations between striatal DAT-BP and PD severity (rho= -0.7, p<0.001) and motor scales (rho= -0.80, p<0.001) were found. CONCLUSION: [99mTc]TRODAT-1 SPECTs scanning was able to discriminate PD patients from controls. The technique is a powerful instrument to measure DAT density that can be used in clinical and research settings in Brazil.
