Abstract Water deficit is an important climatic problem that can impair agriculture yield and economy. Genetically modified soybean plants containing the AtNCED3 gene were obtained aiming drought-tolerance improvement. The NCED3 gene encodes a 9-cis-epoxycarotenoid dioxygenase (NCED, EC 1.13.11.51), an important enzyme in abscisic acid biosynthesis. ABA activates the expression of drought-responsive genes, in water-deficit conditions, targeting defense mechanisms and enabling plants to survive under low water availability. Results from greenhouse experiments showed that the transgene AtNCED3 and the endogenous genes GmAREB1, GmPP2C, GmSnRK2 and GmAAO3 presented higher expression under water deficit (WD) in the event 2Ha11 than in WT-plants. No significant correlation was observed between the plant materials and WD conditions for growth parameters; however, gas exchange measurements decreased in the GM event, which also showed 80% higher intrinsic water use when compared to WT plants. In crop season 2015/16, event 2Ha11 showed higher total number of pods, higher number of pods with seeds and yield than WT plants. ABA concentration was also higher in GM plants under WD. These results obtained in field screenings suggest that AtNCED3 soybean plants might outperform under drought, reducing economic and yield losses, thus being a good candidate line to be incorporated in the soybean-breeding program to develop drought-tolerant cultivars.

RESUMO A uva fina de mesa ‘Rubi’ destaca-se pela grande aceitação do mercado consumidor devido ao preço acessível e à sua cor atrativa, consequência do acúmulo de antocianinas. No entanto, nem sempre as condições ambientais favorecem o adequado acúmulo desses pigmentos, acarretando na desvalorização comercial e nutricional desses frutos. Desta forma, o presente estudo objetivou investigar o efeito da aplicação de diferentes concentrações de ethephon, associadas ou não à aplicação de CaCl2, na cor e qualidade de bagas de uva ‘Rubi’. Primeiramente, foram avaliadas as características específicas de bagas tratadas com 0; 200; 400; 600 e 800 mg L d-1e ethephon associado à solução de CaCl2 1,5% - pH, acidez titulável (AT), sólidos solúveis (SS), firmeza das bagas, açúcares solúveis totais e redutores e terores de antocianinas e flavonóis. Esta primeira etapa de avaliação não revelou mudanças nos teores de açúcares totais, firmeza, SS, AT, ratio (SS/AA) e flavonóis, enquanto mudanças foram detectadas para teores de açúcares redutores e antocianinas. Desta forma, numa segunda etapa, foram investigadas (ao longo do tempo) algumas características (SS, AT, teores de açúcares redutores e de antocianinas e atividade das enzimas fenilalanina amônia liase (PAL) e glutationa S-transferase (GST)) de bagas de uva ‘Rubi’ tratadas com 0; 200; 400; 600 e 800 mg L-1 de ethephon associado ou não à solução de CaCl2 1.5%. Os resultados do presente estudo indicam que o ethephon associado ao CaCl2 pode contribuir para uma alta qualidade pós-colheita de uvas ‘Rubi’ em função do maior acúmulo de antocianinas em consequência da maior atividade das enzimas PAL e GST, relacionadas à biossíntese e ao armazenamento de antocianinas, respectivamente, e aos maiores teores de açúcares redutores (sob baixas concentrações de ethephon), sem alterar outros aspectos de qualidade. Portanto, uma única aplicação de ethephon, a partir da concentração de 200 mg L-1, associada à aplicação de CaCl2 1,5% no período final de maturação, quando as bagas apresentam de 30 a 50% de cobertura de cor, é suficiente para produzir resultados satisfatórios na melhoria de cor de bagas da uva ‘Rubi’.

ABSTRACT ‘Rubi’ table grape has wide acceptance of the consumer market due to its affordable price and attractive color as a function of the accumulation of anthocyanins. However, environmental conditions do not always favor the suitable accumulation of these pigments, resulting in commercial devaluation and nutritional depreciation of fruits. Thus, the present study aimed to investigate the effect of application of different ethephon concentrations, associated or not with CaCl2 application, on the skin color and quality of ‘Rubi’ grape berries. First, specific traits of berries treated with 0, 200, 400, 600, 800 mg L-1 ethephon associated with 1.5% CaCl2 solution were evaluated and pH, titratable acidity (TA), soluble solids (SS), berry firmness, total and reducing soluble sugars, and anthocyanin and flavonol levels were analyzed. This first stage of evaluation revealed no changes in total sugar levels, firmness, SS, TA, SS/TA ratio, and flavonol levels, while changes were detected regarding reducing sugar and anthocyanin levels. Thus, in the second stage, the following variables were evaluated: SS, TA, reducing sugar levels, anthocyanin levels, phenylalanine ammonia lyase (PAL) and glutathione S-transferase (GST) activity of ‘Rubi’ grape berries treated with 0, 200, 400, 600, 800 mg L-1 ethephon, associated or not with 1.5% CaCl2 solution. The results of the present study indicate that ethephon associated with CaCl2 can contribute to improve the post-harvest quality of ‘Rubi’ grape, since this association increased the accumulation of anthocyanins due to the higher activity of PAL and GST, related to biosynthesis and storage of antocyanins, respectively, and increased the levels of reducing sugars (at low ethephon concentrations), not changing other quality aspects. Therefore, a single ethephon application from 200 mg L-1 associated with the application of 1.5% CaCl2 at the final ripening stage, when bunches present from 30 to 50% skin color coverage, was sufficient to produce satisfactory results regarding skin color improvement of ‘Rubi’ grape berries.

Diversos estudos vêm sendo realizados com a finalidade de aumentar o conhecimento sobre a ocorrência e a atividade do óxido nítrico (ON) nas plantas. Nesse sentido, a presente revisão objetivou abordar alguns aspectos referentes ao ON nas plantas, tais como propriedades químicas, vias de síntese, efeitos fisiológicos, ação antioxidante, transdução do sinal, interação com hormônios vegetais e expressão gênica. Nos últimos anos, muitos avanços têm sido obtidos em relação à síntese de ON e seus efeitos fisiológicos nas plantas. Porém, os mecanismos moleculares que fundamentam seus efeitos permanecem pouco compreendidos. É sinalizada uma investigação em detalhes sobre as estreitas interações entre ON, Ca2+, ADP-ribose cíclica (cADPR) e proteínas quinases. Além disso, ainda não foi possível identificar uma enzima vegetal que apresente atividade semelhante à da óxido nítrico sintase (NOS). A elucidação de tais aspectos representa um desafio para futuros trabalhos.

Several studies were carried out in order to improve the knowledge about the occurrence and activity of nitric oxide (NO) in plants. Thus, this review discusses some aspects related to NO in plants such as chemical properties, synthesis pathways, physiological effects, antioxidant action, signal transduction, interaction with plant hormones and gene expression. In the last years, many advances have been obtained regarding NO synthesis and its physiological effects in plants. However, the molecular mechanisms underlying its effects remain poorly understood. It is signalized that tight interplays among NO, Ca2+, cyclic ADP ribose (cADPR), and protein kinases need to be investigated in details. In addition, it has not yet been possible to identify a plant enzyme displaying a nitric oxide synthase (NOS)-like activity. The elucidation of such aspects represents a challenge to future studies.

Níveis de biossólido equivalentes a 0, 28, 56 e 112 t ha-1 foram avaliados no desenvolvimento de Mentha piperita L. Determinaram-se área foliar e matéria seca total e dos diferentes órgãos, os índices fisiológicos razão de área foliar, área foliar específica, taxa assimilatória líquida e taxa de crescimento relativo, aos 30, 44, 58, 72 e 86 dias após plantio (DAP) e o rendimento de óleo essencial aos 90, 110 e 120 DAP. Os índices fisiológicos revelaram que o biossólido prolongou a fase vegetativa das plantas, que se adaptaram com o tempo e apresentaram comportamentos inversos em relação à produtividade, resultado de seu metabolismo primário e representada pela produção de matéria seca da parte aérea e produção de óleo, resultado do metabolismo secundário. A referida adaptação das plantas de menta à presença do biossólido pode ser devido à função fitorremediadora dessa espécie, cujos mecanismos intrínsecos poderão ser melhor compreendidos na avaliação dos efeitos residuais em sua parte aérea.

Development of Mentha piperita L . on biosolids amended soil with levels equivalent to 0, 28, 56 and 112 t ha-1 was evaluated. In order to measure the productivity and its relation with mint essential oil yield, different indices were determined: leaf area, total and several organ dry matter, leaf area ratio, specific leaf area, net assimilation rate and relative growth rate at 30, 44, 58, 72 and 86 days after planting (DAP), and essential oil yield at 90, 110 and 120 DAP. Physiological indices revealed that biosolids prolonged the vegetative phase of the plants, which adapted themselves to the presence of biosolids with time. Plants showed inverse behaviors in relation to productivity, resulting from the primary metabolism, represented by the shoot dry matter yield, and oil yield, resulting from the secondary metabolism. Adaptation of the mint plants to the growth on biosolids could be due to a phytoremediation function of this species. The intrinsic mechanisms of these processes could be better understood in a further evaluation of residual effects in mint plant shoots.

Esse estudo avaliou os efeitos dos níveis de biossólido no rendimento e na composição química do óleo essencial de Mentha piperita L. Plantas de menta foram cultivadas em casa de vegetação em vasos contendo o equivalente a 0, 28, 56 e 112 t.ha-1 de biossólido. Três avaliações foram realizadas aos 90, 110 e 120 dias após plantio (DAP). O óleo foi extraído da matéria seca da parte aérea por hidrodestilação e sua composição determinada por CG/EM. Sua produção foi afetada de modo discreto pelo biossólido, aumentando quando as plantas foram cultivadas com 28 t.ha-1, condição que não resultou em sua melhora de qualidade. O acetato de mentila foi o componente encontrado em maior porcentagem em todos os tratamentos. As plantas apresentaram aos 90 DAP maior percentagem de mentol, segundo maior constituinte do óleo, com conteúdo de 42,3% naquelas cultivadas sem biossólido. A presença do biossólido favoreceu a formação de mentofurano. Como o mentol, a mentona diminuiu com o desenvolvimento das plantas. Nessas condições, recomenda-se sua colheita aos 90 DAP, época em que o nível de mentol foi maior. Com o aumento de produção de lodo de esgoto, há necessidade de sua adequada destinação, havendo restrições para seu uso em relação ao ambiente e à planta. Assim, apesar de o cultivo com 28 t.ha-1 estar dentro do permitido pela legislação, essa condição, que aumentou o rendimento do óleo, não melhorou sua qualidade. Assim, o biossólido da Estação de Barueri não foi recomendável para o cultivo da espécie.

This research evaluated the effects of biosolid levels on yield and chemical composition of Mentha piperita L. essential oil. Mint plants were grown in a greenhouse in pots containing the equivalent to 0, 28, 56, and 112 t.ha-1 biosolid. Three evaluations were made at 90, 110, and 120 days after planting (DAP). The oil was extracted from the dry matter of shoots by hydrodistillation, and composition was determined by GC/MS. Oil production was slightly affected by the biosolid, increasing when plants were grown with 28 t.ha-1, a condition which did not result in quality improvement. Menthyl acetate was the component obtained at the highest percentage in all treatments. At 90 DAP, plants showed a higher percentage of menthol, the second-highest oil constituent, with a content of 42.3% in plants grown without biosolid. The presence of biosolid favored menthofuran formation. As with menthol, menthone decreased with plant development. Under these conditions, plant harvesting is recommended at 90 DAP, period in which the menthol level was higher. Since the production of biosolid is on the rise, a suitable destination must be given to it, and restrictions exist for its use in relation to the environment and plants. Thus, although cultivation with 28 t.ha-1 is within the limits allowed by law, such a rate, which increased oil yield, did not improve oil quality. Therefore, biosolid from the Barueri Station is not recommended for cultivation of this specie.
