A ocratoxina A é um contaminante que pode estar presente em vários alimentos e ser prejudicial tanto para a saúde quanto para a economia, então deve ser medida. Mas, para bem quantificar um analito, mesmo com o uso de métodos oficiais, é necessário verificar o sistema de medição; nisto se empregam requisitos de validação. Assim, procedeu-se a uma verificação intralaboratorial de método por purificação em coluna de imunoafinidade e cromatografia líquida de alta eficiência em amostras de café verde artificialmente contaminadas e medidas. Foi obtido o comportamento de medição esperado ao longo do analito, a faixa de trabalho teve o limite inferior entre 0,489 e 1,59 e o superior entre 220 e 300 μg.kg-1. A linearidade não foi rejeitada nesta faixa e não houve interferência externa significativa no modelo. O intercepto não diferiu significativamente da origem e o coeficiente linear não diferiu significativamente de 1,00. Os níveis de desvios padrão dependeram das concentrações estudadas, como ocorrido em diversas publicações, e os desvios padrão relativos não demonstraram inconformidade nos estudos colaborativos localizados. Concluiu-se pela competência do laboratório no método pesquisado, especialmente para café verde.

Ochratoxin A is a food contaminant which can appear in several foods and can be harmful not only to human health, but also to the economy. In order to quantify properly an analite, even when using standard methods, it is necessary to check the validation method requirements. Therefore, a single laboratory validation of the method using immunoaffinity column and high performance liquid chromatography in spiked samples of ground green coffee beans was conducted. The evolution of the measurements along the analyte was sigmoid, as expected, and the range showed botton between 0.489 and 1.59, and top between 220 and 300 μg.kg-1, top. Linear regression was not rejected in this range and there was no external significant interference in the model. The intercept was in the origin and the linear coefficient did not differ significantly from 1.00 (p < 0.05). The standard deviations depended on the concentrations of ochratoxin A, similarly to studies in the literature, and the estimated values of the relative standard deviation obtained were in accordance with the values from those studies. Therefore, the laboratory proved competence in this measurement process, especially for green coffee beans.

Sessenta amostras de milho pós-colheita foram avaliadas quanto à contaminação fúngica endógena e o potencial toxígeno de espécies do gênero Aspergillus e seus teleomorfos. Quarenta grãos aparentemente sadios de cada amostra foram desinfestados em NaClO e incubados em câmara úmida a 25±1ºC para exteriorização dos fungos, que posteriormente foram isolados em ágar Czapek-Dox. Foram identificadas as espécies Aspergillus flavus, A. parasiticus, Eurotium amstelodami e E. chevalieri. O potencial toxígeno dos fungos A. flavus e A. parasiticus foi avaliado quanto à síntese de aflatoxinas em meio ágar-coco. Espécies do gênero Eurotium foram avaliadas quanto à síntese de esterigmatocistina, nos meios ágar-amendoim e trigo triturado. A porcentagem de grãos contaminados variou entre 0 e 100%, prevalecendo os gêneros Aspergillus, Penicillium e Fusarium. A espécie predominante foi a A. flavus (64%), seguida por E. amstelodami (19%), E. chevalieri (10%) e A. parasiticus (7%). A partir de 109 isolados de A. flavus, evidenciou-se que 73 isolados sintetizaram aflatoxinas B1 e B2, 20 sintetizaram B1, sete sintetizaram B1 e G1, três sintetizaram B1, B2 e G1 e em seis não foi detectada a síntese de aflatoxina. A síntese de esterigmatocistina pelas espécies E. amstelodami e E. chevalieri não foi detectada.

Sixty post-harvested samples of maize kernels from three regions of the Paraná State, Brazil, were evaluated concerning endogenous fungi contamination and toxigenic potential of Aspergillus spp. and some of their teleomorfs. Forty apparently healthy kernels were selected from each sample, disinfested with NaClO and incubated at 25±1ºC for fungal growth. Fungus species were isolated in Czapek-Dox agar plates. The species Aspergillus flavus, A. parasiticus, Eurotium amstelodami and E. chevalieri were identified. The toxigenic potential of Aspergillus species were analyzed in coconut agar medium. Eurotium spp. were evaluated for their metabolics in peanut agar medium and in wheat grits. The kernel contamination varied from 0 to 100% and the prevalent genera detected were Aspergillus, Penicillium and Fusarium. A. flavus was the predominant species (64%), followed by E. amstelodami (19%), E. chevalieri (10%) and A. parasiticus (7%). From 109 A. flavus species isolated, 73 strains synthesized aflatoxins B1 and B2, 20 synthesized B1, seven synthesized B1 and G1, three synthesized B1, B2 and G1 and in six strains aflatoxin production was not detected. All A. parasiticus species produced, simultaneously, B1, B2, G1 and G2. Sterigmatocystin synthesis was not detected in any condition by E. amstelodami and E. chevalieri.
