ABSTRACT Background: The determination of seed moisture content is of utmost importance for the evaluation of its quality. Near-infrared (NIR) spectroscopy has been successfully applied to estimate properties of biological materials. However, studies using this technique on forest seeds are still scarce. Thus, the aim of this study was to explore the potential of NIR spectroscopy for estimating the moisture content of E. uniflora seeds. NIR spectra were obtained using optical fiber from 100 individual seeds that were dehydrated in silica gel for 2, 7, 14, and 21 days. Subsequently, the seeds were subjected to the moisture test by the oven method, and had moisture contents of 52.4%, 41.7%, 33.1%, 21.8%, and 18.1%, respectively. Results: Principal component analysis (PCA) of the spectral signatures explained 100% of the data variability. The moisture content was associated with the spectra by partial least squares regression (PLS-R) and the predictive model presented a coefficient of determination in cross-validation (R²cv) of 0.88 and the root mean square error of cross-validation (RMSEcv) was 5.43%. Conclusion: The statistics associated with the models indicate that NIR spectroscopy has potential for estimating the moisture content of E. uniflora seeds. RESUMO: A sensibilidade à dessecação é uma condição presente em sementes de diversas espécies arbóreas de importância econômica e ecológica, o que torna tais sementes incapazes de tolerar a secagem e o armazenamento. Assim, são necessários estudos que busquem estratégias para manter a viabilidade dessas sementes após serem submetidas à secagem. Uma alternativa para esse fim é a utilização de priming em soluções de potencial osmótico conhecido. Soluções de polietilenoglicol, glicerol, bem como fitohormônios como ácido abscísico e ácido salicílico estão relacionados à tolerância ao estresse em sementes. O objetivo deste trabalho foi estudar sementes sensíveis à dessecação de Inga vera, Eugenia uniflora e Tapirira guianensis, utilizando soluções de PEG, SA, sacarose, glicerol e combinações de SA+PEG na tentativa de reduzir a sensibilidade à dessecação das sementes. ABSTRACT: Desiccation sensitivity is a condition present in seeds of several tree species of economic and ecological importance, which makes such seeds unable to tolerate drying and storage. Thus, studies that seek strategies to maintain the viability of such seeds after being subjected to drying are necessary. An alternative for this purpose is the use of priming in solutions of known osmotic potential. Solutions of polyethylene glycol (PEG), glycerol, as well as phytohormones such as abscisic acid (ABA) and salicylic acid (SA) are related to stress tolerance in seeds. RESUMO: Diversas técnicas são utilizadas para avaliar a qualidade das sementes; entretanto, a maioria deles é destrutiva e requer um tempo de execução considerável. O teste de raios X tem se mostrado uma técnica não destrutiva eficiente para a avaliação da qualidade de sementes. Este estudo teve como objetivo avaliar danos internos em sementes de ipê-roxo (Handroanthus impetiginosus) por meio de testes de raios X e investigar como esses danos afetam a germinação das sementes. Amostras de quatro lotes de sementes de ipê-roxo foram expostas à radiação e, posteriormente, as radiografias foram analisadas no software ImageJ, sendo as sementes divididas em quatro classes de acordo com sua morfologia interna: 1) sementes com cavidade embrionária preenchida com mais de 50%; 2) cavidade embrionária preenchida até 50%; 3) cavidade embrionária atacada por insetos; e 4) cavidade embrionária vazia. Em seguida, as sementes foram submetidas ao teste de germinação e, após 21 dias, foram classificadas como germinadas, não germinadas, mortas e plântulas normais e anormais. ABSTRACT: Several techniques are used to evaluate seed quality; however, most of them are destructive and require considerable execution time. The X-ray test has proven to be an efficient nondestructive technique for evaluating the quality of seeds. This study aimed to evaluate internal damage in ipê-roxo (Handroanthus impetiginosus) seeds through X-ray tests and to investigate how it affects seed germination. Samples from four lots of ipê-roxo seeds were exposed to radiation and, subsequently, the radiographs were analyzed using ImageJ software and the seeds divided into four classes based on their internal morphology: 1) seeds with an embryonic cavity filled with more than 50%; 2) an embryonic cavity filled up to 50%; 3) an embryonic cavity attacked by insects; and 4) an empty embryonic cavity. After that, the seeds were subjected to a germination test and, after 21 days, scored as germinated, nongerminated, dead, and normal and abnormal seedlings. The X-ray test proved to be effective in evaluating the internal morphology of ipê-roxo seeds and their relationship with physiological quality. Radiographic analysis enabled the precise identification of damage, such as embryonic malformation and damage by insects. ABSTRACT Background: The aim of this research was to investigate the efficiency of methods to overcome primary dormancy and biochemical processes associated with germination of Erythrina falcata Benth seeds. Seeds were submitted to dormancy overcoming treatments and the water uptake pattern was analysed. We then evaluated the activity of the antioxidant enzymes SOD, CAT, and APX, and quantified lipid peroxidation levels, hydrogen peroxide content and total protein content. The experiments were performed in a completely randomized design and the statistical analysis used was Scott-Knott test at 5% probability. Results: Mechanical scarification with sandpaper leads to approximately 94% of germination in E. falcata, while control non-scarified seeds show only approximately 37% of germination. Treatments with hot water led to high mortality in E. falcata seeds. Rapid water absorption was observed in the first 18 hours of imbition when the seeds were scarified with sandpaper, and radicle emergence was observed after 36 hours. During imbibition there was an increase of antioxidant enzyme activity and a decrease in lipid peroxidation and H2O2 production, suggesting an efficient mechanism for regulating imbibition damage. A decrease in the total protein content was observed during germination of E. falcata seeds. Conclusions: Mechanical scarification is an efficient method for breaking dormancy of E. falcata seeds. During germination of E. falcata seeds there is a reduction in the production of H2O2 and lipid peroxidation, and an increase in the activity of antioxidant enzymes. Resumo: Vários estudos têm utilizado com sucesso o polietilenoglicol (PEG) e/ou ácido abscísico (ABA) para induzir a tolerância à dessecação em sementes germinadas de espécies ortodoxas, melhorando nosso entendimento dos mecanismos que fundamentam essa característica. Nesse contexto, tratamentos com PEG e ABA foram testados para avaliar sua eficiência em reduzir a sensibilidade à dessecação e aumentar o período de armazenamento das sementes de Magnolia ovata. A secagem das sementes a 10% de teor de água não afetou significativamente a germinação, independentemente de terem sido previamente tratadas ou não com ABA. No entanto, sementes tratadas com PEG apresentaram menor viabilidade. Após o armazenamento, as sementes tratadas com ABA e dessecadas a 5% de teor de água, apresentaram maiores taxas de germinação, sendo que o tratamento com ABA 100 µM proporcionou a maior capacidade de armazenamento. Não foram observadas alterações na atividade da catalase, peroxidase ou superóxido dismutase ou na abundância de proteínas totais ou resistentes ao calor.

Abstract: Several studies have successfully used polyethylene glycol (PEG) and/or abscisic acid (ABA) to reinduce desiccation tolerance in germinated seeds of orthodox species, improving our understanding of the mechanisms that underlie this trait. In this context, treatments with PEG and ABA were tested to assess their efficiency in reducing the sensitivity to desiccation and increasing the storage period of Magnolia ovata seeds. The drying of seeds to 10% water did not significantly affect germination, nor was germination impaired in seeds that were dried and treated with ABA. However, seeds treated with PEG had lower viability. After storage, ABA-treated seeds with as low as 5% water content (WC) had higher germination rates, the treatment with 100 µM ABA providing the greatest storage capacity. No changes were observed in the activity of catalase, peroxidase, or superoxide dismutase or in the abundance of total or heat-resistance proteins.

ABSTRACT Background: The germination ecology of P. guajava seeds collected in two regions of Brazil was investigated aiming to (1) characterize the seed batches; (2) determine the best temperature for germination; and (3) assess variations in seed desiccation tolerance (DT). For each batch, we determined seed water content and initial germination. To evaluate temperature effects, we tested seed germination. After, we generated the imbibition curves for the two provenance. To evaluate the loss of DT, the seeds were left to germinate along different time intervals. We monitored water loss by regularly weighing the seeds until achieving stabilization, keeping them under these conditions for 72 hours. After drying, the seeds were pre-hydrated and submitted to the germination test. Results:We identified variations in physiological characteristics between the seeds from north and southeast batches. At the temperature extremes, the seeds from both provenance failed to germinate. The highest germination percentages were observed at 20 and 25 °C (constant) and between 20-30 °C (alternating) for the two batches and, for the southeast batch specifically, at 30 °C. The imbibition curve displayed a triphasic pattern with a rapid increase in seed fresh weight in the first 48 hours. Conclusion: The southeast provenance seeds lost DT abruptly, until its complete loss after radicle protrusion, which occurred in both batches. The highest values of germination speed index (GSI) of both provenance were observed after a 9-day imbibition period, suggesting seed hydration memory. Drying resulted in necrosis of the root tissue and death of the seeds with visible roots.

Resumo Devido à baixa germinação de sementes de Bixa orellana L., há a necessidade de se testar diferentes técnicas que viabilizem sua reprodução, como o condicionamento fisiológico. O objetivo do presente trabalho foi caracterizar a germinação de dois lotes de sementes de urucum sob efeito do condicionamento fisiológico. Para isso, sementes de dois lotes (recém-colhido e armazenado) foram submetidas ao condicionamento fisiológico em solução aerada de: 1) água deionizada; 2) água catódica; 3) nitrato de potássio a 0,3%; 4) nitroprussiato de sódio a 100 µM; por 24 e 48h, e um tratamento adicional (controle) sem condicionamento. Após o teste de germinação, as sementes que não germinaram foram submetidas ao teste de tetrazólio a 0,075% a 30 °C, no escuro, durante 24 h. O delineamento experimental utilizado foi o inteiramente casualizado, em esquema fatorial (4 x 2 + 1) com quatro repetições de 25 sementes por tratamento. Apesar da baixa germinação já relatada para a espécie, foi possível obter germinação sem nenhum tipo de pré-tratamento como quebra de dormência, mesmo em lote armazenado por 10 anos e com teor de água de 8%. Os agentes condicionantes não foram eficientes no aumento da germinação de sementes de urucum. São necessários novos trabalhos para classificação da espécie quanto ao seu armazenamento.

Abstract Due to the low germination of annatto seeds (Bixa orellana L.), it is necessary to test different techniques that facilitate and enable its production, such as the use of the physiological conditioning method. Here, we aimed to characterize the germination of two annatto seed lots under physiological conditioning. For this, two lots of seeds (newly-collected and stored) were submitted to physiological conditioning using an aerated solution of 1) deionized water; 2) cathodic water; 3) potassium nitrate 0.3%; 4) sodium nitroprusside 100 µM; for 24h and 48h, and an additional treatment (control) without conditioning. After the germination test, ungerminated seeds were submitted to a tetrazolium test at 0.075% at 30 °C, in the dark for 24 h. We used a completely randomized factorial design (4 x 2 + 1) with four replications of 25 seeds per treatment. Despite the low germination already mentioned in the literature for this species, we obtained high germination without any type of dormancy-break pretreatment, even in lots stored for over 10 years with a water content of 8%, characterizing this species as non-dormant. The germination and viability of Bixa orellana were influenced by seed water content decrease. The conditioning agents were not efficient to increase annatto seed germination. Further studies are needed to classify these seeds according to their storability.

Abstract Some species of the Lauraceae family produce seeds that are generally sensitive to desiccation, which makes them difficult to store. The objective of this study was to characterize changes in seed quality of C. aschersoniana from two lots, as well in the physiological and cellular aspects during 12 months of storage. Seeds were stored with their original moisture content (MC) or after pre-drying to 35% MC in a cold chamber (5 °C) at a relative humidity of 40%. Seeds were sampled and tested at time 0, 3, 6 and 12 months of storage regarding to moisture content, germination and ultrastructural features. The seeds were dispersed with dormancy that was overcame by the cold storage condition and the reserves in undried seeds were partly consumed during storage. Both undried and pre-dried seeds remained viable for at least 12 months.

Resumo: A desidratação é um procedimento necessário antes de expor as sementes ao armazenamento a longo prazo, mas isso está associado à lesão ligada ao metabolismo mediada por lesão celular. A fim de avaliar as alterações celulares durante o restabelecimento da tolerância à dessecação (TD) em sementes germinadas de C. fissilis e sua relação com danos no DNA, verificamos a ocorrência de fragmentação do DNA através do teste de TUNEL e suas evidências por meio da análise citológica. Para restabelecer o TD, as sementes germinadas foram incubadas por 72 h em polietilenoglicol (PEG, -2,04 MPa) antes da desidratação em sílica gel (a 10% de umidade) seguida de reidratação. O teor de água foi alterado durante o restabelecimento da tolerância à dessecação. Foi utilizado o rotulador TDT-dUPT (TUNEL) para avaliar as taxas de morte celular. A coloração TUNEL foi realizada usando o teste de imagem Click-iT-TUNEL Alexa Flour. O teste TUNEL mostrou uma fragmentação de DNA consistente nas radículas com 2 e 5 mm de comprimento. Além disso, alterações nucleares e cromossômicas foram observadas no ciclo celular meristemático de 5 mm, contribuindo para a identificação de diagnósticos de marcadores de morte celular.

Abstract: Dehydration is a necessary procedure prior to exposing seeds to long term storage, but this is associated with metabolism-linked injury mediated by cell injury. In order to assess cellular alterations during re-establishment of desiccation tolerance (DT) in C. fissilis germinated seeds and their relation to DNA damage, we verified the occurrence of DNA fragmentation through the TUNEL test and its evidence through the cytological analyses. To re-establish DT, germinated seeds were incubated for 72 h in polyethylene glycol (PEG, -2.04 MPa) before dehydration in silica gel (at 10% moisture content) followed by rehydration. The moisture content changes during the reestablishment of the desiccation tolerance was accomplished. (DT)TdT-dUPT terminal nick-end labeling (TUNEL) was used to assess rates of cell death. TUNEL staining was performed using Click-iT-TUNEL Alexa Flour imaging assay. The TUNEL test showed a consistent DNA fragmentation in the 2 and 5 mm long radicles. Moreover, nuclear and chromosomal alterations were observed in the 5 mm meristematic root cell cycle, contributing to the identification of diagnostic markers of cell death.

RESUMO A germinação é um processo fisiológico que começa com a hidratação. Canais específicos conhecidos como aquaporinas são responsáveis pela absorção de água através das membranas celulares biológicas. O cloreto mercúrico (HgCl2) inibe reversivelmente o transporte de água pelas aquaporinas durante a germinação e pode alterar a integridade do ácido desoxirribonucleico (DNA), o que pode resultar na deficiência das aquaporinas. O objetivo deste estudo foi avaliar o efeito de HgCl2 e ditiotreitol (DTT) sobre a atividade das aquaporinas e sobre a integridade/degradação do (DNA) em eixos embrionários de Schizolobium parahyba (Vell.) Blake durante o processo de germinação. Os eixos isolados foram expostos a diferentes concentrações de HgCl2 ou DTT; e a atividade de aquaporinas, hidratação do embrião e a integridade do DNA foram avaliadas durante a embebição do eixo embrionário. O crescimento e as mudanças no peso fresco do eixo foram quantificados. Descobrimos que o metal pesado afeta a hidratação das sementes por inibição da atividade das aquaporinas e não causa mudanças na integridade do DNA.

ABSTRACT Germination is a physiological process that begins with hydration. Specific channels known as aquaporins are responsible for water uptake through biological cell membranes. The mercuric chloride (HgCl2) reversibly inhibits water transport during germination through the aquaporins and can changes deoxyribonucleic acid (DNA) integrity, which can result in aquaporins deficiency. The aim of this study was to evaluate the effect of HgCl2 and dithiothreitol (DTT) on aquaporins activity and on the integrity/degradation of DNA in embryonic axes of Schizolobium parahyba (Vell.) Blake during the germination process. Isolated axes were exposed to different concentrations of HgCl2 or DTT; and aquaporins activity, embryo hydration, and DNA integrity were evaluated during embryonic axes imbibition. Growth and changes in axis fresh weight were quantified. We found that the heavy metal affects seed hydration by inhibition of aquaporins activity and does not cause changes in DNA integrity.

ABSTRACT The maturation process of Sesbania virgata seeds was investigated through fruits and seeds at different maturation stages, where they were described morphologically. For the seeds, fresh and dry mass and moisture were determined and structural analysis of the tegument under a light microscope was performed. Germination tests in the presence and absence of drying and the electrophoretic profile of heat resistant proteins were also performed. The results showed that the seeds reached physiological maturity at 44 days after anthesis (DAA), evidencing a secondary cell wall. The acquisition of desiccation tolerance was observed at 44 DAA, with the seeds responding to the drying treatment. Protein bands were observed from 32 DAA, which remained constant until the end of maturation. These results allowed the characterization of the development and acquisition of desiccation tolerance in S. virgata seeds.

RESUMO A compreensão da tolerância à dessecação é de grande interesse para a tecnologia de sementes, visto que tem aplicação prática no estabelecimento de estratégias para a conservação de sementes e produção de mudas. O presente trabalho objetivou estudar a perda da tolerância à dessecação durante a germinação de sementes das espécies arbóreas sucupira (Bowdichia virgilioides), pau-ferro (Libidibia ferrea), cedro (Cedrela fissilis), tamboril (Enterolobium contortisiliquun), ipê-roxo (Handroanthus impetiginosus) e pau-jacaré (Piptadenia gonoacantha). Sementes coletadas na região de Lavras - MG foram usadas para os experimentos, sendo estes realizados isoladamente para cada espécie. Primeiramente foi estabelecida a curva de embebição por meio da pesagem das sementes nas condições de germinação a intervalos regulares. Baseado nessa informação, as sementes foram postas para embeber por tempos determinados pela curva de embebição e submetidas à secagem até atingir o mesmo percentual de umidade encontrado antes da semeadura. Em seguida as sementes foram reidratadas e aquelas que continuaram a germinação e originaram plântulas normais foram consideradas tolerantes à dessecação. Cada espécie apresentou um padrão próprio quanto à perda da tolerância à dessecação, sendo que em todos os casos, não houve tolerância à dessecação após a protrusão da raiz primária. Contudo, foi observada em sementes de pau-jacaré uma redução mais drástica logo no início do processo germinativo. Nas condições do presente trabalho, observou-se perda da tolerância à dessecação nas primeiras etapas do processo germinativo em todas as espécies, o que pode ter implicações em trabalhos que envolvam o condicionamento fisiológico das sementes.

ABSTRACT Understanding the loss of desiccation tolerance is of great importance in seed technology for its implications in the development of strategies for seed conservation and seedling production. In the present work, the loss of desiccation tolerance was studied in seeds of tree species Bowdichia virgilioides, Libidibia ferrea, Cedrela fissilis, Enterolobium contortisiliquum, Handroanthus impetiginosus, and Piptadenia gonoacantha. The seeds were collected in the region of Lavras, MG, and subjected to desiccation experiments individually for each species. Imbibition curve was created for each species by measuring seed weight at regular intervals under germination conditions. Based on this information, the seeds were allowed to imbibe for a time inferred from the imbibition curve, and then allowed to dry until reaching the initial water content measured before the imbibition begun. Then, the seeds were rehydrated, and those that developed into normal seedlings were considered desiccation-tolerant. The loss of seed desiccation tolerance followed a specific pattern in each studied species, but none of the species was tolerant to desiccation after radicle protrusion. A drastic decrease in desiccation tolerance was observed in P. gonoacantha seeds at the beginning of the germination process. Under the conditions implemented in this study, the loss of desiccation tolerance in all species started at the beginning of the germination process. These results have implications in both theoretical studies (seed priming) and the practice (in seedling production).

Resumo: O conhecimento sobre estratégias de regeneração natural é importante devido a sua influência no ecossistema local. Objetivou-se estudar o comportamento do banco de sementes aéreo de P. dubium. Os frutos foram colhidos mensalmente antes e após a maturação das sementes em matrizes localizadas no município de Lavras - MG. Mensalmente, foram determinados o conteúdo de água, aquisição da tolerância a dessecação, viabilidade das sementes, manutenção da dormência e tempo médio de germinação. Verificou-se que a maturidade fisiológica da espécie ocorreu no mês de junho, quando foi observado baixo conteúdo de água, máxima matéria seca, e alta porcentagem de germinação. Verificou-se que não houve alteração na dormência, porcentagem de germinação e no perfil proteico de sementes durante o período em que permanecem no banco de sementes aéreo. A espécie P. dubium forma banco de sementes aéreo de curta serotinosidade, com frutos permanecendo presos à planta por 9 meses.

Abstract: Knowledge concerning natural regeneration strategies is important due to the effect of natural regeneration on the local ecosystem. The aim of this study was to examine the ecophysiological aspects of the Peltophorum dubium aerial seed bank. Fruits were harvested monthly, before and after seed maturation, from selected trees in the municipality of Lavras, MG, Brazil, for determination of moisture content, acquisition of tolerance to desiccation, seed viability, maintenance of dormancy, and mean germination time. Physiological maturity of the species was attained in June, when low moisture content, maximum dry matter, and high germination percentage were observed. We found that there was no change in dormancy, in germination percentage, or in the protein profile of seeds during the period they remained in the aerial seed bank. The P. dubium species creates an aerial seed bank of short serotiny, with fruits remaining attached to the tree for 9 months.

RESUMO Objetivou-se com esse trabalho avaliar a integridade do DNA e identificar mudanças ultra-estruturais em raiz primária de sementes de Peltophorum dubium submetidas a secagem. Para tanto, sementes foram posta a germinar e ao atingirem 1 mm de comprimento da raiz primária, foram desidratadas, até atingirem diferentes graus de umidade, sendo em seguida reidratadas e avaliadas quanto à sobrevivência. Após esse procedimento foram selecionados diferentes pontos de interesse (controle, 20% e 10% de umidade) para avaliação da integridade do DNA e da ultraestrutura celular no intuito de detectar possíveis mudanças celulares nas raízes primárias após desidratação. Quando secas até 32% de umidade não houve influência na retomada do crescimento das sementes germinadas, mas ao serem desidratadas a 20% de umidade, a sobrevivência foi reduzida a 67% e a secagem a 8% de umidade concorreu para nulidade na sobrevivência. Pela avaliação de mudanças ultraestruturais em células da raiz observou-se que secagem a aproximadamente 20% de umidade marcou o início dos danos por dessecação, tanto fisiológica quanto ultraestruturalmente. Secagem a níveis próximos a 10% de umidade concorreu não somente para total desconfiguração e ruptura das membranas como também promoveu colapso intracelular. Foi observada perda da integridade do DNA genômico no decorrer da secagem.

ABSTRACT The aim of this study was to evaluate DNA integrity and to identify ultrastructural changes in the primary root of Peltophorum dubium seeds subjected to drying, since these changes may reflect the germination capacity. Seeds were germinated and were dehydrated when reached 1 mm in length of the primary root, until reaching different moisture contents, being then rehydrated and evaluated in relation to survival. After this procedure, different points of interest (control, 20% and 10% moisture content) were selected to evaluate DNA and cell ultrastructure integrity in order to detect possible cellular changes in primary roots after dehydration. When dried down to 32% moisture, there was no influence on the growth resumption of the germinated seeds, but when they were dehydrated to 20% moisture, the survival was reduced to 67%, reaching nullity in the survival with drying to 8% moisture. By the evaluation of ultrastructural changes in root cells, it was observed that drying to approximately 20% moisture content marked the onset of damages by desiccation, both physiologically and ultrastructurally. Drying to levels close to 10% moisture content was not only responsible for total disorganization and rupture of membranes, but also promoted intracellular collapse. Loss of genomic DNA integrity was observed during drying.

Resumo: Este trabalho teve-se como objetivo avaliar o efeito do condicionamento osmótico na germinação de sementes de Eucalyptus urophylla e do híbrido de E. grandis x E. urophylla em condições de estresse salino. Foram testados dois potenciais de condicionamento induzidos por PEG 6000 (-1,0 e -1,5 MPa) em dois períodos (1 e 3 dias). Após o condicionamento osmótico, as sementes foram colocadas para germinar sob estresse salino em solução de NaCl nos potenciais 0,0 (controle) -0,5, -0,75 e -1,0 MPa a 25 °C. Os parâmetros avaliados foram a germinação e o índice de velocidade de germinação (IVG). Houve diminuição da germinação e do IVG à medida que o potencial hídrico do meio germinativo tornou-se mais negativo, tanto para E. urophylla quanto para o híbrido, entretanto, o E. urophylla foi mais tolerante ao estresse salino, em comparação ao híbrido, apresentando maior germinação em todos os potenciais hídricos testados. O osmocondicionamento no potencial de -1,0 MPa por três dias foi o mais indicado tanto para E. urophylla quanto para E. urophylla x E. grandis, quando submetidos à germinação em solução salina a -1,0 MPa, demonstrando que esse tratamento foi eficiente em favorecer a tolerância ao estresse salino.

Abstract: The objective of this experiment was to evaluate the effect of priming on the germination of Eucalyptus urophylla and of hybrid E. urophylla × E. grandis seeds under salt stress. Two osmotic potentials (-1.0 and -1.5 MPa) were tested, using PEG 6000 for 1 and 3 days. After priming, seeds were germinated under salt stress in a NaCl solution at 0.0 (control), -0.5, -0.75 and -1.0 MPa potentials, at 25 °C. Seed germination and germination speed index decreased as the water potential of the germination medium decreased. However, E. urophylla was more tolerant to salt stress; it showed a higher germination percentage under all tested potentials, when compared to the hybrid. The osmotic conditioning at -1.0 MPa for three days was more effective when E. urophylla x E. grandis was germinated in a salt solution at -1.0 MPa, indicating that this treatment was more effective in inducing tolerance to salt stress.
