RESUMO: Foram utilizados 12 lagartos Teiús (Tupinambis merianae) do Nordeste do Brasil. Encontramos nove animais positivos (75%) para Leptospira sp. na PCR e seis (50%) soropositivos. O sequenciamento de DNA de Leptospira sp. revelou 99% de semelhança com L. interrogans. Os resultados indicam que esta espécie pode desempenhar um papel importante na transmissão da leptospirose humana.

ABSTRACT: We used 12 tegu lizards (Tupinambis merianae) from northeastern Brazil, and we reported nine (75%) Leptospira sp. PCR-positive animals and six (50%) seropositive. Leptospira sp. DNA sequencing revealed 99% similarity with L. interrogans. Our findings indicated that this species may play a role in the transmission of human leptospirosis.

Abstract In swine and bovines, leptospirosis prevention and control is carried out via vaccination of susceptible animals using bacterins. However, the efficiency of leptospirosis vaccines has been questioned. This work aimed to investigate the potency of five leptospirosis vaccines sold commercially in Brazil, challenging the animals with one autochthonous strain of Leptospira, Canicola serovar, denoted LO4, isolated from swine. The standard protocol was followed, and renal carriers of Leptospira were identified among the surviving animals by culture and PCR. Of the five vaccines tested, only two proved effective. None of the surviving animals was positive by culture; however, one animal was positive by PCR. Three of the five vaccines sold commercially in Brazil for the immunization of swine or bovines failed the test of the efficacy to protect the vaccinated animals following challenge with an autochthonous Leptospira strain, Canicola serovar. The two vaccines provided protection against the renal carrier state in the surviving animals. The criteria used to produce leptospirosis bacterins sold commercially in Brazil must be reviewed. The industry should support researches on leptospiral vaccinology to improve the quality of the present vaccines and discover new immunogenic strains, because it is known that vaccination is one of the most important tools to increase the reproduction rates in livestock.

RESUMO: A criação de ovinos é uma atividade econômica de grande valor para a indústria pecuária brasileira. As doenças infecciosas podem comprometer significativamente a sua produtividade. O objetivo deste estudo foi detectar Leptospira spp. em ovinos sem raça definida a partir de uma região semiárida do estado da Paraíba, Nordeste do Brasil. Quarenta e nove amostras de sangue e tecido renal, bem como amostras de urina, de 25 das 49 ovelhas, foram coletados na linha de abate. A presença de anticorpos anti -Leptospira spp. foi verificada com o teste de aglutinação microscópica (SAM). As amostras de urina e rins foram processadas por PCR e cultura bacteriológica. Sororreatividade foi observada em 8,2% (4/49) dos animais. Em relação à PCR, três amostras renais foram positivas, no entanto, todas as amostras de urina foram negativas. Nenhuma cultura pura de leptospiras foi recuperada a partir das amostras testadas. Levando em consideração a baixa frequência de positividade na sorologia e PCR, é possível sugerir que a rusticidade das ovelhas de raças mestiças colaborou para a baixa ocorrência de leptospirose na região semiárida investigada.

ABSTRACT: Sheep breeding is an economic activity of great value for the Brazilian livestock industry, and infectious diseases can significantly compromise its productivity. The aim of this study was to detect Leptospira spp. in mixed breed sheep from a semiarid region of the State of Paraíba, Northeast Brazil. In total, 49 blood and renal tissue samples, as well as 25 urine samples, were collected from 49 sheep in the slaughter line. The microscopic agglutination test (MAT) was used to verify the presence of anti -Leptospira spp. antibodies in blood samples. Renal tissue and urine samples were analyzed by both PCR and bacterial culture. Sero reactivity was observed in 8.2% (4/49) of animals. Three renal samples were positive for leptospiral DNA by PCR although all urine samples were negative. No leptospires in pure culture were recovered from the samples tested. Low prevalence of Leptospira spp. observed by serology and PCR suggested that the rusticity of the mixed-breed sheep may have contributed to the low occurrence of leptospirosis in the semiarid region investigated.

RESUMO: Foi realizado um inquérito epidemiológico objetivando determinar a ocorrência de anticorpos anti-Leptospira e elencar os fatores de risco em equinos da microrregião do Pajeú, estado de Pernambuco. Foram examinados 100 soros de equinos provenientes de 38 propriedades de 4 municípios da região, pela prova de soroaglutinação microscópica (SAM), utilizando 24 sorovares de Leptospira spp. como antígeno. Das 100 amostras testadas, 28 (28%) foram reagentes. Os sorovares mais frequentes foram Patoc (35,71%), Butembo (32,14%) e Sentot (14,30%). O uso de pasto como volumoso (odds ratio =7,80; intervalo de confiança de 95% - IC95% 1,71-35,50; p=0,008) foi identificado como fator de risco. Sugere-se a necessidade de estudos acerca do isolamento do agente e da caracterização de sua patogenicidade, bem como se recomenda o controle de roedores em depósitos de ração e no ambiente.

ABSTRACT: An epidemiological survey aiming to determine the occurrence of anti-Leptospira antibodies and to identify risk factors was carried out in horses of the Pajeú microregion, state of Pernambuco. Sera from 100 horses from 38 properties in four counties in the region were examined by the microscopic agglutination test (MAT), using 24 Leptospira spp. serovars as antigens. Of the 100 samples tested, 28 (28%) were positive. Most prevalent serovars were Patoc (35.71%), Butembo (32.14%) and Sentot (14.30%). Supply of pasture as forage (odds ratio =7.80; 95% confidence interval - 95%CI 1.71-35.50; p=0.008) was identify as risk factor. It is suggested the need for studies on agent isolation and characterization of its pathogenicity, as well as it is recommended the rodent control in deposits of feed and environment.

Este trabalho teve como objetivo determinar a prevalência de rebanhos ovinos positivos (focos) e de animais soropositivos para leptospirose na região semiárida do Estado da Paraíba, bem como identificar fatores de risco. Foram colhidas amostras de sangue de 1.275 animais procedentes de 117 rebanhos em 19 municípios da mesorregião do Sertão, Estado da Paraíba. Para o diagnóstico sorológico da leptospirose foi utilizado o teste de soroaglutinação microscópica (SAM), com 24 sorovares de Leptospira spp. como antígenos. Um rebanho foi considerado positivo quando apresentou pelo menos um animal soropositivo. Das 117 propriedades utilizadas 33 (28,20%) apresentaram pelo menos um animal soropositivo, e dos 1.275 animais 69 (5,41%) foram soropositivos, com títulos variando de 100 a 3200. O sorovares reagentes foram Autumnalis (49,30%), Andamana (27,53%), Sentot (17,39%), Whitcomb (4,34%) e Australis (1,44%). Possuir mais de 48 animais no rebanho (odds ratio = 2,26; IC 95% = 1,33 -5,07; p = 0,021) e participação em exposições (odds ratio = 9,05; IC 95% = 0,96 - 85,71; p = 0,055) foram identificados como fatores de risco. Sugere-se a necessidade de estudos acerca do isolamento do agente, da caracterização de sua patogenicidade e do seu impacto econômico nos rebanhos ovinos da região, bem como recomenda-se maior controle sanitário nas aglomerações de animais.

The aim of this investigation was to determine the herd-level and animal-level prevalence of leptospirosis in sheep from the semiarid of Paraíba State, Northeast Brazil, as well as to identify risk factors. Blood samples were collected from 1,275 sheep from 117 flocks in 19 counties in the Sertão mesoregion, Paraíba. For the serological diagnosis of leptospirosis the microscopic agglutination test (MAT) using 24 Leptospira spp. serovars as antigens was carried out. Of the 117 flocks studied 33 (28.20%) presented at least one seropositive sheep, and of the 1,275 animals 69 (5.41%) were seropositive with titers ranging from 100 to 3,200. Reactant serovars were Autumnalis (49.30%), Andamana (27.53%), Sentot (17.39%), Whitcomb (4.34%) and Australis (1.44%). Herd size > 48 sheep (odds ratio =2.26; 95% CI =1.33-5.07; p=0.021) and participation in animal exhibits (odds ratio =9.05; 95% CI =0.96-85.71; p=0.055) were identified as risk factors. The need was suggested for studies on the isolation of the agent, characterization of its pathogenicity and its economic impact on sheep flocks of the region, and sanitary control in sheep agglomerations was recommended.
