ABSTRACT: Seafood is one of the most important sources of nutrients. However, they have a short shelf-life and the traditional preservation methods may generate losses in their natural flavour and nutrients. Thisstudy evaluated and optimize the High Pressure Processing (HPP) regarding pressure level (200-400 MPa) and holding time (0-5 min) applied to lion’s paw scallop (Nodipecten nodosus) to reduce microbial contamination while maintaining desirable characteristics. Response surface methodology with a Box-Behnken design and Desirability function were employed to simultaneously optimize these quality attributes. HPP enhanced microbial quality at 200 MPa/5 min, despite promoting inadequate physical-chemical modifications in the adductor muscle of the scallop. In such processing condition, in spite of a slight increase in muscle humidity which could be of benefit, pH increase was also verified, as well as a decrease in water holding capacity (WHC). At more severe level (400 MPa/5 min), a decrease in the shear force related to instrumental texture and in Whiteness (W) and Luminosity (L*) related to color was observed. Simultaneous optimization provided a value of 365MPa / 2min where physicochemical characteristics would be the more similar to the scallop without facing a preservation process. RESUMO: Os frutos do mar são uma das fontes mais importantes de nutrientes. No entanto, possuem vida de prateleira curta e os métodos tradicionais de conservação podem gerar perdas em seu sabor natural e nutrientes. O objetivo deste estudo foi avaliar e otimizar o Processamento por Alta Pressão (APH) em relação ao nível de pressão (200-400 MPa) e tempo de espera (0-5 min) aplicado à vieira pata de leão (Nodipecten nodosus) para reduzir a contaminação microbiana, mantendo características desejáveis. Metodologia de superfície de resposta com design Box-Behnken e função de Desejabilidade foram empregadas para otimizar simultaneamente esses atributos de qualidade. A APH melhorou a qualidade microbiana a 200 MPa/5 min, apesar de promover modificações físico-químicas inadequadas no músculo adutor da vieira. Resumo O objetivo deste estudo foi avaliar a hidrólise da lactose em relação a duas diferentes temperaturas iniciais de ação enzimática e fermentação, a fim de melhorar o período de fermentação, verificando a formação de componentes e o nível de hidrolise por CLAE (HPLC). Os resultados das análises de pH e acidez titulável entre os tratamentos com maior temperatura inicial (HIT), 42 ºC, e menor temperatura inicial (LIT), 30 ºC, foram diferentes (P<0,05) durante a fermentação. HIT obteve menores valores de pH e maiores valores de acidez titulável como 4,6±0,04 e 0,73±0,01 g acido láctico.100 mL-1, respectivamente, contra 4,82±0,01 e 0,64±0,01 g acido láctico.100 mL-1 de LIT. O teor de lactose difereriu (P <0,05) entre os tratamentos durante a fermentação; entretanto, no final do processo alcançaram 4,40±0,14 mg.mL-1 e 4,32±0,14 mg.mL-1 para LIT e HIT (P>0,05), respectivamente. Teores de glicose e galactose mantiveram-se dinâmicos. A concentração de ácido láctico em LIT (P<0,05) foi maior (18,64±0,62 mg.mL-1) que em HIT (17,56±0,53 mg.mL-1), embora o teor dos ácidos cítrico e acético tenha diminuído durante o processo. Assim, pode-se concluir que a enzima lactase contribuiu para reduzir o teor de lactose, sem influenciar o processo de fermentação. Além disso, ambos os tratamentos obtiveram valores mais baixos de lactose, o que é suficiente para o consumo por pessoas com intolerância a lactose.

Abstract The aim of this study was to evaluate lactose hydrolysis between two different initial temperatures of enzymatic and fermentation action to improve the fermentation period by checking the components formed and hydrolysis levels of lactose by HPLC. pH and titratable acidity analysis between treatments - higher initial temperature (HIT), 42 ºC, and lower initial temperature (LIT), 30 ºC - were different (P<0.05) during the fermentation process. HIT obtained lower pH and higher titratable acidity values as 4.6±0.04 and 0.73±0.01 g lactic acid.100 mL-1, respectively, against 4.82±0.01 and 0.64±0.01 g lactic acid.100 mL-1 from LIT. Lactose content was different (P<0.05) between treatments while fermenting; however, at the end of the fermentation, it reached 4.565±0.34 mg.mL-1 and 4.398±0.18 mg.mL-1 for LIT and HIT (P>0,05), respectively. Glucose and galactose remained buoyant during the fermentation period, which indicates its production and consumption at the same time by the lactic acid bacteria involved. The lactic acid concentration in LIT was higher (P<0.05) (18.64±0.62 mg.mL-1) than in HIT (17.56±0.53 mg.mL-1) although citric and acetic acids content decreased throughout the process. In conclusion, the lactase enzyme contributed to reduce the lactose content without affecting the fermentation process. In addition, both treatments obtained lower values of lactose, which is sufficient for the consumption by those who have lactose malabsorption.

Resumo O objetivo deste estudo foi investigar a qualidade nutricional bem como a estabilidade lipídica de filés de Arapaima gigas. Um total de 27,32 kg de filés de A. gigas foi obtido e a composição centesimal e o perfil de ácidos graxos foram imediatamente determinados. Além disso, os parâmetros de oxidação lipídica foram analisados durante 15 e 90 dias, a 4 °C e -20 °C, respectivamente. Os filés de A. gigas apresentaram elevado teor de proteína (>15%) e baixa quantidade de lipídeos (<2%), com elevado conteúdo de ácidos graxos poli-insaturados (AGPI) (43,97%). Os índices de qualidade nutricional foram: índice de aterogenicidade (0,35); índice de trombogenicidade (0,28) e razão de ácidos graxos hipocolesterolêmico/hipercolesterolêmico (2,37). Em geral, um aumento seguido por uma diminuição foi observado nos resultados de índice de peróxidos (IP) e malonaldeído (MDA), em ambas as temperaturas de estocagem (p < 0,05). O perfil lipídico apresentou uma elevada qualidade nutricional, entretanto novos métodos de conservação devem ser investigados nesta matriz, devido ao aumento de oxidação lipídica durante a refrigeração e o congelamento.

Abstract The nutritional quality and lipid stability of Arapaima gigas fillets were investigated. A total of 27.32 kg of A. gigas fillets were obtained and the proximate composition and fatty acid profile immediately determined. In addition, the lipid oxidation parameters were analyzed during 15 and 90 days at 4 °C and –20 °C, respectively. The A. gigas fillets presented high protein (> 15%) and low lipid (< 2%) contents with elevated polyunsaturated fatty acid (PUFAs) contents (43.97%). The nutritional quality indices were the atherogenicity index (0.35), thrombogenicity index (0.28) and the hypocholesterolemic acid/ hypercholesterolemic acid ratio (2.37). Overall, an increase followed by a decrease was observed in the peroxide index (PI) and malondialdehyde content (MDA) results at both storage temperatures ( p < 0.05). The lipid profile exhibited great nutritional quality, however new conservation methods should be investigated for this matrix due to increased lipid oxidation during refrigerated and frozen storage.

RESUMO: O objetivo desse estudo foi investigar a influência do tipo muscular na oxidação proteica, perfil de textura (dureza, elasticidade, coesividade e mastigabilidade) e composição centesimal da carne de bovinos Bos indicus (Nelore) terminados com grãos no Brasil. Os músculos longissimus lumborum (LL) e psoas major (PM) foram obtidos 24 horas post mortem de oito (n=8) carcaças de touros Nelore, cortados em cinco bifes (1,5 cm), e dispostos em condições aeróbicas por nove dias a 4 °C. Composição centesimal e perfil de textura foram analisados no dia 0, enquanto que a oxidação proteica foi analisada durante 9 dias de estocagem. Os bifes LL demonstraram maior (P<0.05) concentração de proteína em relação aos bifes PM, enquanto que PM apresentou maior (P<0.05) conteúdo de lipídeos e cinzas que LL. Além disso, os bifes LL exibiram maior (P<0,05) dureza, elasticidade, coesividade e mastigabilidade em relação aos bifes PM. Em contraste, a oxidação proteica foi maior (P<0,05) nos bifes PM do que nos bifes LL em todos os dias de estocagem. Os resultados sugerem que as diferenças no metabolismo e na composição muscular contribuíram para a variação nos atributos bioquímicos e perfil de textura entre os bifes LL e PM. Estratégias músculo-específicas são indicadas a fim de promover a estabilidade de cor nos bifes PM oriundos de bovinos Bos indicus terminados com grãos.

ABSTRACT: The aim of this research was to investigate the influence of muscle type on protein oxidation, texture profile (hardness, springiness, cohesiveness and chewiness) and proximate composition of beef from grain-finished Bos indicus (Nellore) cattle in Brazil. The muscles longissimus lumborum (LL) and psoas major (PM) were collected 24 h post mortem from eight (n=8) Nellore bull carcasses, fabricated into five steaks (1.5-cm) and displayed under aerobic conditions for nine days at 4 °C. Proximate composition and texture profile were analyzed on day 0, whereas protein oxidation was analyzed during 9 days of storage. LL exhibited greater (P<0.05) protein concentration than PM steaks, whereas PM demonstrated greater (P<0.05) lipid and ash content than their correlative LL. In addition, LL steaks exhibited greater (P<0.05) hardness, springiness, cohesiveness and chewiness than PM steaks. In contrast, protein oxidation was greater (P<0.05) in PM than in LL steaks throughout the storage. The results suggest that the differences of muscle metabolism and composition contributed to the variation on biochemical attributes and texture profile of LL and PM steaks. Muscle-specific strategies are indicated to improve the color stability of PM steaks from grain-finished Bos indicus cattle.

Resumo Foi avaliado o efeito do estresse produzido pelo tempo de espera pré-abate sobre a produção de aminas biogênicas, pH e cor pelo sistema CIELab (onde três variáveis L*, a*, b* foram determinadas: L* indica a luminosidade, a* indica o vermelho e b* determina o amarelo) na carne de peito de frango. Antes do abate 25 frangos de carne foram selecionados e divididos em cinco grupos conforme o tempo de espera pré-abate (30 min ou 3 h), programação dia/noite (diurno ou noturno) e controle (sem tempo de espera). Após abate, os peitos (M. Pectoralis major) foram separados, estocados por 9 dias a 4°C, e analisados cada 3 dias. Os resultados evidenciaram uma redução do pH nas aves que aguardaram o abate na noite. Além disso durante a estocagem incrementou-se os valores de aminas biogênicas e da luminosidade (L*) e houve redução dos teores de vermelhos (a*) no tempo de espera de três horas (diurno ou noturno). As concentrações da cadaverina incrementaram-se a partir do dia 6 de estocagem. O tempo e horário de espera em frangos de carne pode é um fator de estresse pré-abate que afeta a qualidade da carne. Contudo, é recomendável realizar o abate dos frangos imediatamente após de sua chegada ao centro de abate.

Abstract The effect of pre-slaughter lairage stress on biogenic amines, pH, and color with the CIELab system (where three variables L*, a*, b* are estimated: L* for lightness, a* defines redness, and b* defines yellowness) in chicken breast meat was investigated. Before slaughtering, 25 broilers were selected and divided into five groups according to lairage holding time (30 min or 3 h), day/night schedule (daylight or nighttime) and control (no lairage). After slaughtering, breasts (M. Pectoralis major) were removed, stored for 9 days at 4 °C, and analyzed every 3 days. The results showed a pH decrease during nighttime holding. Also, a three-hour pre-slaughter holding (daytime or night) resulted in high biogenic amine concentration, increase in lightness (L*), and reduction in redness (a*) during storage. Cadaverine concentration showed a rapid increase from day 6 onward. The time and schedule of chicken lairage is a pre-slaughter stress factor that affects meat quality. Based on these observations, it is recommended to slaughter chickens immediately on arrival at the processing plants.

Resumo O objetivo do presente estudo foi investigar relevantes parâmetros físico-químicos e sensoriais de três diferentes porções musculares da espécie Arapaima gigas. Porções cranial, medial e caudal foram analisadas em relação a composição centesimal, parâmetros instrumentais de cor e textura e atributos sensoriais. As porções medial e caudal apresentaram o maior conteúdo de lipídeos e valor energético (P < 0,05); no entanto, os menores teores de umidade e carboidrato ( P < 0,05). O conteúdo de proteína foi similar ( P > 0,05) entre as diferentes porções musculares. Antes do cozimento, as porções medial e caudal demonstraram os maiores valores de L* , a*, dureza e mastigabilidade (P < 0,05). Após o cozimento, não se observou diferença (P > 0,05) nos parâmetros instrumentais de cor entre as diferentes porções musculares. A porção medial apresentou menor dureza e mastigabilidade ( P < 0,05), quando comparada à porção caudal. A porção cranial apresentou o menor escore de sabor e impressão global (P < 0,05). Portanto, as porções musculares medial e caudal de Arapaima gigas demonstraram potencial superior para satisfazer as exigências dos consumidores.

Abstract The objective of the present study was to investigate the relevant physicochemical and sensory parameters of three different Arapaima gigas muscle portions. Cranial, medial and caudal portions were analysed regarding their proximate compositions, instrumental colour and texture parameters, and sensory evaluations. The medial and caudal portions exhibited the greatest (P < 0.05) lipid contents and energy values and the lowest (P < 0.05) moisture and carbohydrate levels. The protein contents were similar (P > 0.05) for the different muscle portions. Before cooking, the medial and caudal portions displayed the greatest (P < 0.05) values for lightness, redness, hardness and chewiness. After cooking, no differences ( P > 0.05) were observed between the different muscle portions for the instrumental colour parameters, while the medial portion exhibited lower (P < 0.05) values for hardness and chewiness as compared to the caudal portion. The cranial portion received the lowest (P < 0.05) scores for flavour and overall liking. Thus the Arapaima gigas medial and caudal muscle portions presented the greatest potentials to satisfy the consumer requirements.

Os metais traço apresentam grande toxicidade quando presentes em teores elevados nos organismos humano e animal. No ambiente aquático, alguns elementos formam compostos orgânicos que bioacumulam nos tecidos de organismos vivos. Os moluscos bivalves se destacam, pois possuem grande participação na dinâmica de contaminantes metálicos no ambiente marinho devido à capacidade de concentrá-los em níveis mais elevados que na água. Os mexilhões são moluscos bivalves que constituem bons indicadores de biodisponibilidade de metais, sendo amplamente utilizados como biomonitores em ecossistemas aquáticos. No Brasil, a espécie de mexilhão Perna perna é o mais cultivado devido à importância na alimentação de grande parte da população das regiões costeiras. Portanto, o consumo de mexilhões pode representar um risco à saúde humana quando proveniente de locais contaminados com metais traço, o que reforça a importância de se conhecer os teores desses elementos nos ambientes que fornecem alimento ao Homem. Devido à constante busca pela qualidade ambiental associada à utilização do mexilhão P. perna como biomonitor e como alimento, é importante conhecer o panorama de contaminação do território nacional a fim de auxiliar na prevenção de intoxicação por metais traço pelo consumo desse molusco por grande parte da população brasileira.

Trace elements are considered toxic in high levels for human and animal organisms. In aquatic environment, some elements can form organic compounds and tend to bioaccumulate in organisms tissues. Bivalve mollusks participate on the metallic contaminants dynamics in aquatic environment due to capacity in concentrate trace elements in their soft tissues in higher levels than in water. Mussels are mollusks widely used as bioindicators of trace metal pollution in coastal areas because they can accumulate various elements as filter-feeders. In Brazil, Perna perna is one of the most commonly consumed bivalve mollusks and the most cultivated in coast regions. Therefore, the consumption of mussels may be a potential risk for human health, which enhances the importance to know trace metal levels in environment that provides food to humans. Due to constantly search for environmental quality and the use of mussels as food and often employed to monitor metal pollution in sea, it is of great interest and importance an overview of metallic contamination in national territory to assist in prevention of intoxication by trace elements.

O teor de hidroximetilfurfural (HMF) é reconhecido internacionalmente pela capacidade de indicar o frescor do mel, podendo ser usado para avaliar o processamento e condições de armazenamento do mel. Neste trabalho objetivou-se avaliar a evolução dos níveis de HMF em mel recém-obtido, submetidas a diferentes temperaturas que variaram de 30 a 100º C, por intervalos de tempo pré-definidos (30, 45, 60, 180 e 720 minutos). O ensaio foi realizado em mel floral, cujo valor inicial de HMF foi de 2,2 mg/Kg. O valor máximo preconizado pela legislação brasileira não foi ultrapassado nas amostras submetidas ao aquecimento por 30, 45 e 60 minutos independente das temperaturas. Por um período de 180 minutos, o limite legal de 60 mg/Kg foi extrapolado nas amostras tratadas a 90º C. Os valores mais elevados foram observados nas amostras submetidas ao aquecimento por 720 minutos, tendo o limite legal ultrapassado em temperatura mais branda (70º C). Com base nos resultados obtidos, pôde-se concluir que o conteúdo de HMF aumenta gradativamente quando o mel é exposto a altas temperaturas, por tempo prolongado. Sendo assim, sugere-se uma otimização nos processos de beneficiamento, considerando a quantidade inicial de HMF no produto, com processamento padronizado em temperaturas e tempo controlados para garantia da qualidade do produto final.

Hydroxymethylfurfural (HMF) content is internationally recognized for its ability to indicate the freshness or lack of freshness of honey and can be used to judge the processing and storage condition of honey. The objective of the present study was to evaluate the evolution of HMF levels in fresh extracted honeys submitted to different temperatures ranging from 30 to 100º C, during pre-established time intervals (30, 45, 60, 180 and 720 minutes).The test was conducted in floral honey with an initial HMF content of 2.2 mg/Kg. The maximum value recommended by the Brazilian law (60 mg/Kg) was not exceeded in the samples subjected to heating for 30, 45 and 60 minutes regardless of the temperatures used. When the samples were heat treated during 180 minutes at 90º C the official value was surpassed. The highest values were observed in samples subjected to heating over 720 minutes, and the limit was exceeded at 70º C. The results obtained indicate that the HMF content gradually increases when the honey is heated at high temperatures for long periods. Therefore, we suggest a process optimization, considering the initial HMF content of the product and standardizing times and temperatures to ensure a good quality of the final product.

The aim of this study was to standardize a methodology to obtain two-dimensional (2D) maps of whey proteins from bovine colostrum and mature milk, using two-dimensional electrophoresis, in order to identify the minor proteins by matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometry (MALDI TOF/TOF MS). A total of 38 proteins were identified, 20 spots in the colostrum whey and 18 in the mature milk whey; 5 of them were identified for the first time.
