ABSTRACT Objective: To evaluate the possible renal and hepatic alteration by root canal filling pastes in mice. Material and Methods: Fifty-four mice were divided into nine groups and received one polyethylene tube implant containing two filling pastes (CTZ or calcium hydroxide pastes). Empty polyethylene tubes were used as a negative control. All tubes were implanted subcutaneously in the back of the mice. After time intervals of 7, 21, and 63 days, 1.5 mL of blood was collected by cardiac puncture, and serum samples were used for serological testing. Urea, creatinine, aspartate transferase (AST), alanine transferase (ALT), alkaline phosphatase (ALP), and gamma-glutamyltransferase (GGT) were evaluated. Data were analyzed by 2-way ANOVA (p<0.05). Results: When comparing CTZ and calcium hydroxide pastes and empty tubes and experimental time intervals, no significant differences in the results were found for any of the biochemical parameters analyzed (p>0.05). No differences were observed in the interactions (material*experimental time intervals) and the biochemical parameters analyzed (p>0.05). Conclusion: CTZ and calcium hydroxide pastes did not cause hepatic and renal alterations in mice, demonstrating the pastes' safety. OBJECTIVES: To evaluate the effect of Carbopol gel formulations containing pilocarpine on the morphology and morphometry of the vaginal epithelium of castrated rats. METHODS: Thirty-one female Wistar-Hannover rats were randomly divided into four groups: the control Groups I (n=7, rats in persistent estrus; positive controls) and II (n=7, castrated rats, negative controls) and the experimental Groups, III (n=8) and IV (n=9). Persistent estrus (Group I) was achieved with a subcutaneous injection of testosterone propionate on the second postnatal day. At 90 days postnatal, rats in Groups II, III and IV were castrated and treated vaginally for 14 days with Carbopol gel (vehicle alone) or Carbopol gel containing 5% and 15% pilocarpine, respectively. Next, all of the animals were euthanized and their vaginas were removed for histological evaluation. A non-parametric test with a weighted linear regression model was used for data analysis (p<0.05). RESULTS: The morphological evaluation showed maturation of the vaginal epithelium with keratinization in Group I, whereas signs of vaginal atrophy were present in the rats of the other groups. Morphometric examinations showed mean thickness values of the vaginal epithelium of 195.10±12.23 μm, 30.90±1.14 μm, 28.16±2.98 μm and 29.84±2.30 μm in Groups I, II, III and IV, respectively, with statistically significant differences between Group I and the other three groups (p<0.0001) and no differences between Groups II, III and IV (p=0.0809). CONCLUSION: Topical gel formulations containing pilocarpine had no effect on atrophy of the vaginal epithelium in the castrated female rats.

OBJECTIVES: Vaginal atrophy and breast cancer are common conditions in postmenopausal women and tamoxifen is the standard endocrine treatment for hormone-sensitive tumors. The present study aimed to assess the effect of tamoxifen on Ki-67 protein expression in the vaginal epithelium of castrated rats. MATERIAL AND METHODS: Forty Wistar-Hannover adult, virgin, castrated rats were randomly divided into two groups, group I (control, n=20) and group II (tamoxifen, n=20), receiving 0.5 ml of propylene glycol and 250 µg of tamoxifen diluted in 0.5 ml of propylene glycol, respectively, daily by gavage for 30 days. On the 31st day, the rats were euthanized and their vaginas were removed and fixed in 10% buffered formalin for the immunohistochemical study of Ki-67 protein expression. Data were analyzed by the Levene and Student’s t tests (p<0.05). RESULTS: The mean index of Ki-67 expression in the rat vagina of groups I and II was 4.04±0.96 and 26.86±2.19, respectively (p<0.001). CONCLUSIONS: According to the results of the present study, tamoxifen, at the dose and treatment length used, induced a significant increase in the cell proliferation of the vaginal mucosa in castrated rats, as evaluated by Ki-67 protein expression.

Esta pesquisa teve por objetivo avaliar as características histológicas do escroto de caprinos com diferentes graus de divisão escrotal. Foram utilizados 15 caprinos distribuídos em três grupos (GI, escroto não bipartido; GII, escroto bipartido até 50% do comprimento testicular; e GIII, escroto bipartido superior a 50% do cumprimento do testículo). Fragmentos do escroto foram submetidos a processamento histológico e analisados em microscópio de luz. Na pele do escroto, a epiderme apresentava-se composta por epitélio pavimentoso estratificado queratinizado, organizado nos estratos basal, espinhoso, granuloso e córneo. Sua espessura, independentemente da divisão escrotal, aumentou gradativamente da região proximal à distal, sendo mais espessa nos caprinos do GIII (68,91µm) e mais fina na região da rafe escrotal do grupo GI. A derme apresentava-se escassa em tecido adiposo e possuía uma maior quantidade de glândulas sudoríparas apócrinas nos animais do GIII (18,12GS mm-2), quando comparado com os do GI (16,14GS mm-2) e GII (14,82GS mm-2). Com relação às glândulas sebáceas, não foi encontrada diferença numérica significativa entre os animais pesquisados. Concluiu-se que os caprinos que apresentam bipartição escrotal mais acentuada (GIII) podem apresentar uma maior produção de suor, pois contém maior quantidade de glândulas sudoríparas no escroto, fato que pode favorecer a perda de calor por evaporação, colaborando no processo de termorregulação testicular.

Among male goats in tropical regions, a bipartition in scrotum happens frequently Studies have shown that goats with bigger accented division in scrotum have reproductive advantages in relation to those that had not presented this characteristic, giving them a better thermoregulation for testis in the animals with bipartite scrotum. This research aimed to evaluate histological characteristics of the scrotum, in 15 goats, distributed in three groups, according to scrotal configuration (GI, unique scrotum; GII, scrotum bipartite up to 50% of the testicular length; GIII, scrotum bipartite upper than 50%). It was removed fragments of the organ and they were processed, according to histological routine and analyzed in light microscope. The scrotum skin is constituted of epidermis and dermis, and the epidermis is formed by stratified keratinizing epithelium, organized in basale, spinosum, granulosum, and corneum stratums. The epidermis width, independent of the scrotum configuration, increased gradually from the proximal to the distal region, being wider (average = 68,91µm) in GIII's animals. In the medial region of bipartite scrotum the epidermis showed to be thinner than on the raphe in animals of unique scrotum. The dermis, rare in fat tissue, showed a greater quantity of sweat apocrine glands in the animal of GIII, average of 18,12GS mm-2, while in the GI's, 16,14GS mm-2 and GII, 14,82GS mm-2, suggesting a greater production of sweat that could favor the lost of heat by evaporation. About the number of sebaceous glands, there was no statistic difference among the groups.

O objetivo deste trabalho foi analisar a distribuição dos vasos arteriais nos testículos em caprinos com diferentes graus de divisão escrotal. A configuração escrotal foi classificada da seguinte forma: Grupo I: constituído por caprinos com escroto único, Grupo II: com escroto separado até a metade do testículo e Grupo III: com separação escrotal estendendo-se além da metade do testículos. As artérias foram injetadas e coradas com solução de acetado de vinil, sendo os orgãos (30 pares) submetidos à corrossão para obtenção dos moldes vasculares. As artérias testiculares emergem da aorta abdominal, com trajeto retilíneo, atravessam o canal inguinal, apresentam-se espiraladas e envolvidas parcialmente pelo plexo pampiniforme. Próximo à extremidade caudada do testículo, dividem-se mais freqüentemente nos ramos cranial e caudal, os quais emitem vasos colaterais, de onde emergem ramos penetrantes. Os testículos dos animais com nível intermediário de divisão escrotal (Grupo II) apresentam menor quantidade destes ramos, sendo os quadrantes mais povoados o ventrolateral e o dorsolateral. Conclui-se que a origem, o trajeto e a distribuição das artérias testiculares não apresentam variações relacionadas ao grau de divisão escrotal em caprinos.

The objective of this study was to describe the distribution of testicular arterial vessels in caprines with different degrees of scrotal division. Scrotal configurations were classified as follows; Group I: scrotum without bipartition; Group II: scrotum showing a ventral division until the middle of the testis; and, Group III: scrotal separation extended beyond the middle of the testis. A colored vinyl acetate solution was injected into 60 testicular arteries (30 pairs). After, the samples were submitted to corrosion cast method to obtain tubular vascular models for macroscopic analysis. We observed that the testicular arteries emerge from the abdominal aorta, cross the inguinal canal to become coiled and involved by the pampiniform plexus. Close to the caudal extremity of the testis, the arteries become divided in cranial and caudal branches that emit collateral vessels to originate emerging branches. Testicles with intermediate level of scrotal division (group II) showed a smaller amount of those branches than the others, being the most populated the ventrolateral and the dorsolateral quadrants. The origin, distribution and localization of the testicular arteries did not show any differences related to the degree of scrotal division in caprines.
