ABSTRACT The objective of the present study was to evaluate the stability of candidate reference genes and select the genes that can be used for normalizing real-time polymerase chain reaction (PCR) in the liver, skeletal muscle, and jejunum tissues of Nellore or Nellore × Angus steers fed different diets. Fourteen purebred and 14 crossbred steers were used, in which half of the animals of each genetic group received a diet containing whole shelled corn (WSC) and the other half whole shelled corn and sugarcane bagasse (WSCB). Stability was analyzed by the RefFinder program. To validate the selection of candidate reference genes, the expression of target genes was evaluated using the different groups of reference genes. The most stable genes were 18S, ACTB, and CASC3 for skeletal muscle; HMBS, ACTB, and 18S for the liver; and GAPDH, ACTB, and CASC3 for the jejunum, regardless of breed and diet provided. Possible errors caused in data analyses were clarified comparing the more and less stable genes as reference for normalization of the target genes FASN, ACOX, SCD1, MGAM, and SLC2A1. The use of the more stable and less stable sets of reference genes may lead to different conclusions in respect to the expression profile of the target studied gene. The selection of more suitable reference genes for each experiment is of utmost importance to ensure the reliability of gene expression studies so that they can be applied in practice.

O Brasil é o maior produtor de citros do mundo, sendo responsável por mais de 20% de sua produção. No entanto, a produção ainda é baixa, em decorrência de problemas fitossanitários, como o Declínio do Citros que é uma anomalia cuja causa ainda não foi determinada e, consequentemente, não existem medidas de controle para minimizar as perdas na produção. O uso de variedades resistentes é considerado como a medida de controle mais adequada. Contudo, pouco se conhece sobre os genes envolvidos na resposta de defesa das plantas a essa anomalia. Considerando que muitas alterações fisiológicas associadas com respostas a estresses em plantas são controladas em nível transcripcional, neste estudo objetivou-se a identificação e caracterização dos produtos de expressão gênica diferencialmente expressos na resposta ao Declínio dos Citros. Por meio da técnica de hibridação subtrativa supressiva, bibliotecas de cDNAs expressos foram construídas utilizando mRNAs isolados de raízes de limoeiro "Cravo" (Citrus limonia L. Osbeck) sob laranja "Pera" (Citrus sinensis L. Osbeck) de plantas sadias e doentes. Cento e vinte e nove clones foram obtidos por subtração e suas sequências foram comparadas em bancos de dados. Trinta e quatro delas relacionaram-se a proteínas associadas a processos de estresses, enquanto as outras foram similares a sequências de funções desconhecidas ou não apresentaram similaridade com sequências depositadas nos bancos de dados. Três genes foram selecionados e suas expressões foram estudadas por RT- qPCR em tempo real. Plantas com Declínio dos Citros apresentaram um aumento no nível de expressão em dois desses genes, sugerindo que estes podem estar diretamente envolvidos com essa anomalia.

Brazil is the largest citrus producer in the world, being responsible for more than 20% of its production, which is, however still low due to phytosanitary issues such as citrus blight. Citrus blight is an anomaly whose causes still have not yet been determined, therefore there are no efficient control measures to minimize the production losses with the use of resistant varieties being considered the most appropriate method. However, little is known about the genes involved in the defense response of the plants to this anomaly. Considering that many physiological alterations associated with plant stress responses are controlled at a transcriptional level, in this study we sought the identification and characterization of the gene expression products differentially expressed in the response to the citrus blight. Through the suppressive subtractive hybridization technique, expressed cDNA libraries were built using mRNAs isolated from "Cravo" lemon tree roots (Citrus limonia L. Osbeck) under "Pera" orange (Citrus sinensis L. Osbeck) of healthy and sick plants. 129 clones were obtained by subtraction and their sequences were compared in databases. 34 of them linked to proteins associated to stress processes, while the others were similar to sequences of unknown functions or did not present similarity with sequences deposited in the databases. 3 genes were selected and their expressions were studied by RT - qPCR in real-time. Plants with citrus blight presented an increase of the expression level in two of those genes, suggesting that these can be directly involved with this anomaly.

Zinc deficiency is a global problem of considerable importance for agriculture and human health. Under zinc deficiency conditions, many essential zinc-dependent physiological functions are unable to operate normally, and the cellular homeostasis is adversely affected. This paper described the potential damages that low-zinc bioavailability in soil can have for plants, humans, and animals. In addition, current knowledge on physiological and molecular aspects of zinc homeostasis in plants and strategies used to increase zinc deficiency tolerance were discussed.

No processo de produção comercial de mudas de gérbera, a cor da flor é uma das principais características morfológicas de interesse agronômico, sendo uma característica importante em programas de melhoramento genético. A utilização de marcadores moleculares pode servir para direcionar cruzamentos, confirmar novos híbridos ou genótipos mutantes e identificar novos genótipos para fins comerciais. Nesse contexto, o objetivo deste trabalho foi analisar a divergência genética entre seis cultivares de Gerbera jamesonii ('Jaguar Yellow', 'Jaguar Cream', 'Jaguar Lemon', 'Jaguar Salmon Pastel', 'Jaguar Red', 'Jaguar Deep Rose'). A análise de divergência genética entre as cultivares de gérbera foi realizada utilizando-se 21 primers, os quais amplificaram 37 fragmentos polimórficos de DNA, que foram usados para estimar o coeficiente de Jaccard, o qual apresentou uma média de 0,38, variando de 0,28 a 0,56. A estrutura genética entre as cultivares foi estimada pelo UPGMA, revelando dois grupos distintos, a 38% de similaridade genética. A maior similaridade genética encontrada (56%) foi entre as cultivares 'Jaguar Yellow' e 'Jaguar Lemon'. Os resultados demonstram que a técnica RAPD oferece uma maneira rápida, relativamente barata e útil para a caracterização da divergência genética entre as diferentes cultivares de Gerbera jamesonii com relação à cor da flor.

During the commercial production of gerbera seedlings, flower color is one of the main morphological aspects that have an agronomic interest and becoming an important feature in genetic breeding programs. The use of molecular markers may serve to direct crossings, new hybrids and mutants, besides confirm and identify new genotypes for commercial purposes. In that context, this work aimed to analyze the genetic divergence among six cultivars of Gerbera jamesonii ('Jaguar Yellow', 'Jaguar Cream', 'Jaguar Lemon', 'Jaguar Salmon Pastel', 'Jaguar Red', 'Jaguar Deep Rose'). The genetic divergence among cultivars of gerbera was carried out with 21 primers, which amplified 37 DNA polymorphic fragments, used to estimate the Jaccard index and presented an average of 0,38, ranging from 0,28 to 0,56. The genetic structure among cultivars was estimated by UPGMA and revealed two distinct groups, at 38% genetic similarity. The largest genetic similarity found (56%) was between cultivars 'Jaguar Yellow' and 'Jaguar Lemon'. The results showed that the RAPD is a fast, relatively inexpensive and useful technique for genetic divergence characterization between different cultivars of Gerbera jamesonii.

Activation tagging is a powerful tool to identify new mutants and to obtain information about possible biological functions of the overexpressed genes. The quadruple cauliflower mosaic virus (CaMV) 35S enhancer fragment is a strong enhancer, which is most commonly used for this purpose. However, the constitutive nature of this enhancer may generate lethal mutations or aberrations in different plant organs by the same overexpressed gene. A tissue-specific activation tagging approach may overcome these drawbacks and may also lead more efficiently to the desired phenotype. For this reason the SHATTERPROOF2 (SHP2) promoter fragment was analysed for enhancer activity. The SHP2 gene is involved in dehiscence zone development and expressed during silique development. The aim of the experiments described here was to identify a dehiscence zone specific enhancer that could be used for tissue-specific activation tagging. The chosen SHP2 enhancer fragment was found to be expressed predominantly in the dehiscence zone and showed enhancer activity as well as ectopic expression activity. This activity was not influenced by its orientation towards the promoter and it was still functional at the largest tested distance of 2.0 kb. Based on these results, the SHP2 enhancer fragment can potentially be used in a tissue-specific activation tagging approach to identify new Arabidopsis mutants with an altered dehiscence zone formation.
