Muitas espécies das abelhas-sem-ferrão (Hymenoptera: Meliponini) como Tetragonisca angustula coletam resina para a defesa dos seus ninhos contra invasores como formigas ou abelhas cleptobióticas. Pequenas porções de resina são aderidas nos corpos e extremidades dos invasores causando a sua imobilização. Observa-se que a resina é removida com facilidade das mandíbulas das abelhas, mas cola fortemente na cutícula dos invasores. Foi testada a hipótese de que a resina adere menos nas mandíbulas das abelhas-sem-ferrão do que na superfície dos invasores devido a estruturas especiais de superfície ou a propriedades adesivas dessas estruturas. As superfícies da mandíbula de T. angustula e do trocanter de Camponotus sericeiventris foram estudadas por microscopia eletrônica de varredura. Para medir suas propriedades de adesão, superfícies selecionadas foram montadas em uma alfinete de vidro fino e retiradas de uma ponta de vidro coberta com resina. A deformação do alfinete de vidro indica forças de adesão operando entre a resina e a superfície selecionada. O valor absoluto da força é calculado através da rigidez do alfinete. Foi observado que a resina adere mais na mandíbula da abelha com superfície lisa do que no trocanter estruturado da formiga. Uma nova hipótese a ser testada diz que as abelhas lubrificam as mandíbulas com néctar ou mel para reduzir a adesão da resina temporariamente.

Many Stingless Bees (Hymenoptera: Meliponini) like Tetragonisca angustula collect resin to defend their nests against intruders like ants or Robber Bees. Small portions of resin are attached to intruders bodies and extremities causing their immobilization. It has been observed that resin is removed easily from the bee's mandible but adheres strongly to the intruder's cuticle. We tested the hypothesis that resin sticks lesser to the mandibles of Stingless Bees than to the surface of intruders due to special surface structures or adhesive properties of these structures. The surface structures of the mandible of T. angustula and the trochanter of Camponotus sericeiventris were studied by scanning electron microscopy. To measure adhesion properties, selected surfaces were fixed on a fine glass pin and withdrawn from a glass tip covered with resin. The deformation of the glass pin indicates adhesion forces operating between the resin and the selective surface. The absolute value of the forces is computed from the glass pin's stiffness. It has been shown that resin sticks more to the smooth mandible of the bee than to the structured trochanter of the ant. A new hypothesis to be tested says that the bees might lubricate their mandibles with nectar or honey to reduce the resin's adhesion temporarily.

Análises fisico-quimicas foram realizadas para avaliar vinte e sete amostras de mel de três espécies do gênero Melipona (M. capixaba, M. rufiventris e M. mondury) coletadas nos Estados do Espírito Santo e de Minas Gerais. Os parâmetros atividade de água (aW), porcentagem de sólidos solúveis (Brix %), pH, acidez (meq.Kg-1) e umidade (%) foram avaliados. As características do mel dessas amostras foram muito similares às do mel de outras espécies de Melipona. Entretanto, para o mel de Apis apenas os valores de pH foram similares. O baixo valor de pH e a elevada acidez detectados no mel de Melipona são fatores potenciais para uma maior vida útil do mesmo por não oferecerem condições favoráveis ao desenvolvimento microbiano. Por outro lado, o alto teor de atividade de água favorece o crescimento de microrganismos, especialmente de leveduras o que requer maior cuidado no manuseio e armazenamento do mel. As diferenças observadas entre o mel de Melipona e o de Apis reforçam a necessidade de estabelecimento de padrões de qualidade específicos para o mel de abelhas sem ferrão.

Physicochemical analyses were carried out to evaluate 27 samples of honeys from three species of the Brazilian genus Melipona (M. capixaba, M. rufiventris and M. mondury) from Espírito Santo and Minas Gerais States. The parameters water activity (Aw), percentage of soluble solids (Brix %), pH, acidity (meq/Kg) and moisture (%) were evaluated. The honey characteristics obtained from these samples were very similar to the ones from other Melipona species. However, regarding the honey from Apis (honey bee), only the pH values were similar. The low pH value and the high acidity detected in Melipona honey are potential factors for increasing the honey shelf life because they do not provide favorable conditions for the microbial development. On the other hand, the high level of water activity favors the growth of microorganisms, especially yeast, which demands a more careful handled and storage. The observed differences between Melipona and Apis honey reinforce the need for specific quality settings for stingless bee honey.

For a study of diversity and genetic structuring in Melipona quadrifasciata, 61 colonies were collected in eight locations in the state of Minas Gerais, Brazil. By means of PCR analysis, 119 ISSR bands were obtained, 80 (68%) being polymorphic. He and H B were 0.20 and 0.16, respectively. Two large groups were obtained by the UPGMA method, one formed by individuals from Januária, Urucuia, Rio Vermelho and Caeté and the other by individuals from São João Del Rei, Barbacena, Ressaquinha and Cristiano Otoni. The Φst and θB values were 0.65 and 0.58, respectively, thereby indicating high population structuring. UPGMA grouping did not reveal genetic structuring of M. quadrifasciata in function of the tergite stripe pattern. The significant correlation between dissimilarity values and geographic distances (r = 0.3998; p < 0.05) implies possible geographic isolation. The genetic differentiation in population grouping was probably the result of an interruption in gene flow, brought about by geographic barriers between mutually close geographical locations. Our results also demonstrate the potential of ISSR markers in the study of Melipona quadrifasciata population structuring, possibly applicable to the studies of other bee species.

O gênero Melittobia Westwood compreende várias espécies de microparasitóides, das quais apenas duas foram registradas no Brasil até agora: M. australica Girault e M. hawaiiensis Perkins. Entretanto, essas espécies são de difícil identificação através de métodos taxonômicos tradicionais. No presente trabalho, marcadores moleculares amplificados ao acaso (PCR-RAPD) foram utilizados com o objetivo de se discriminar as duas espécies e, ao mesmo tempo, analisar a variabilidade genética em populações de M. australica. A maioria dos fragmentos gerados foi espécie-específicos estando presente em todos os indivíduos de uma espécie e ausente nos indivíduos da outra espécie, demonstrando a adequação dessa técnica na distinção das espécies de Melittobia estudadas. A técnica de PCR-RAPD também demonstrou que os indivíduos das diferentes populações estudadas são muito semelhantes entre si, o que pode ser atribuído ao efeito fundador e/ou a grande capacidade de dispersão dessas populações. As distâncias genéticas dentro (D = 1,19-3,54%) e entre as populações (D = 1,93-5,28%) de M. australica apresentaram valores muito baixos, refletindo a reduzida variação genética existente nas populações dessa espécie.

The genus Melittobia Westwood comprises several species of microparasitoids and only two of them are know to occur in Brazil up to now: M. australica Girault and M. hawaiiensis Perkins. Nevertheless, the differentiation between these two species using traditional taxonomy is very difficult. In the present study, we used random amplified polymorphic DNA chain reaction (RAPD-PCR) to test for its ability to discriminate between these two species and to examine the genetic variation among the studied populations of M. australica. Most of the generated fragments were species-specific, occurring in all individuals of one species and absent in the individuals of the other species demonstrating the appropriateness of such technique to distinguish between both of the Melittobia species occurring in Brazil. RAPD-PCR also demonstrated low variability among different populations of M. australica, which may be due to a founder effect and/or high dispersion capacity of these populations. Genetic distances within (D = 1.19-3.54%) and among populations (D = 1.93-5.28%) presented very low values, reflecting the reduced genetic variation that exists among populations of M. australica.

A espécie de abelha sem ferrão Melipona quadrifasciata apresenta duas subespécies, Melipona quadrifasciata quadrifasciata Lep. e Melipona quadrifasciata anthidioides Lep. A diferença morfológica entre as duas subespécies é a presença de três a cinco bandas tergais amarelas do 3º ao 6º segmentos em operárias e machos de M. q. quadrifasciata e duas a cinco bandas interrompidas em M. q. anthidioides. Nós identificamos um marcador de DNA que está presente em M. q. quadrifasciata e ausente em M. q. anthidioides. Este marcador está ausente em abelhas coletadas no norte do Estado de Minas Gerais (Brasil), embora esses indivíduos apresentem morfologia similar à de M. q. quadrifasciata. Este marcador poderá ser utilizado em estudos da zona de hibridação entre as subespécies.

The stingless bee species Melipona quadrifasciata includes two subspecies, Melipona quadrifasciata anthidioids and Melipona quadrifasciata quadrifasciata. The morphological difference between the two subspecies is the presence of three to five continuous yellow stripes on the terga on the 3rd to 6th segments in workers and males of M. q. quadrifasciata, and two to five interrupted bands in M. q. anthidioides. We identified a DNA marker which is present in M. q. quadrifasciata and absent in M. q. anthidioides. Only one among the M. q. quadrifasciata colonies did not present the marker. It was also absent in bees collected in northern Minas Gerais State (Brazil), despite their morphological resemblance to M. q. quadrifasciata. The marker can be used for studying the genetic structure of the hybridization zone formed by the intercrossing of the two subspecies.

Three methods of division of Melipona quadrifasciata Lep. colonies were evaluated: fisogastric queen naturally mated; orphans colonies, and queen mated in laboratory. Out of six colonies, five had grade 7 in 221 days, reaching 83,3% of success, in the 1st method. Out of four, 100% were formed using the 2nd method, in 221 days, and 71,4% out of seven colonies were formed with grade 7 in 195 days using the 3rd method. All colonies were treated with Syrup-A; the same management and same system of evaluation. Colonies obtained were described performance by the following mathematic models: Grade1 = 1.202 + 0.262 x reading; Grade2 = 1.474 + 0.250 x reading and Grade3 = 1.406 + 0.287 x reading. The 3rd method was the best considering time of the colonies development; however, it consumed more labor and laboratory equipments.
