Abstract: The global medicine market is about 1.1 trillion US dollars. About 35 percent of medicines have originated from natural products. Brazil presents the largest biodiversity in the world, with more than 50,000 species of higher plants. However, few innovative products have been developed in Brazil from active constituents derived from the Brazilian biodiversity. Scientific evidences on plants and venoms have been internationally published by Brazilian scientists over the last 4 decades; but few examples of innovative products are commercially available. Few examples include the anti-hypertensive drug captopril first identified in the venom of the Brazilian viper Bothrops jararaca by Professor Sergio Ferreira; and some phytotherapeutic agents such as Acheflan®, Syntocalmy® and Melagrião® produced by standardized plant extracts with scientific proof of safety, efficacy and quality. Still, only Acheflan® and Melagrião® are obtained from native Brazilian plants. Several issues contribute to the lack of innovative products from the Brazilian biodiversity, but in my opinion, the most challenging ones are i) the lack of specific regulations to allow researchers and companies to access biodiversity for the purposes of scientific and technological innovation; and ii) the absence of a long-term government program to support research and innovation in this field.

The anti-inflammatory and antiallodynic effects of spray dried powders starting from leaves, stems, roots, the mixture of leaves and stems, as well as the whole plant aqueous solutions of Phyllanthus niruri L., Phyllanthaceae, were assessed. Gallic acid, used as chemical marker, was quantified by HPLC in the spray dried powders. Carrageenan-induced inflammatory and allodynic responses in the mouse paw were used as pharmacological models. Quantitative and qualitative differences among chemical composition of different herb parts were observed. The oral administration of leaves or leaves plus stems spray dried powders (100 mg/kg) significantly inhibited the carrageenan-induced allodynic effect (42±5 and 54±3%, respectively). Additionally, the spray dried powders of leaves significantly reduced carrageenan-induced paw oedema (35±6%). The spray dried powders of roots, stems, or the mixture of leaves, stems and roots (100 mg/kg, p.o.) did not exhibit antiallodynic or antioedematogenic effect in the same model. In conclusion, differences in the chemical composition of spray dried powders from P. niruri are reflected in their in vivo pharmacological actions. Despite of a direct relationship of anti-inflammatory and antiallodynic effects with the gallic acid content had been observed, especially in the spray dried powders of leaves, the use of spray dried powders of leaves plus stems showed to be more effective, suggesting a synergic effect between their constituents.

Três triterpenos pentacíclicos inéditos foram isolados da resina de Protium kleinii. Suas estruturas, 3-oxo-11beta,16beta-diidróxi-urs-12-eno (1), 3-oxo-11beta-hidróxi-urs-12-eno (2) e 3-oxo-11beta-hidróxi-olean-12-eno (3), foram elucidadas por espectroscopia de RMN. A administração sistêmica destes compostos (10-100 mg kg-1, i.p.) foi capaz de inibir tanto a primeira quanto a segunda fase da nocicepção induzida pela formalina em camundongos. Em adição, foram isolados os triterpenos conhecidos 3beta,16beta-dihidróxi-urs-12-eno (breina) (4) e uma mistura de alfa (5)- e beta-amirina (6).

Three novel pentacyclic triterpenes have been isolated from the resin of Protium kleinii and their structures elucidated by NMR spectroscopy as 3-oxo-11beta,16beta-dihydroxy-urs-12-ene (1), 3-oxo-11beta-hydroxy-urs-12-ene (2) and 3-oxo-11beta-hydroxy-olean-12-ene (3). The systemic administration of the above compounds (10-100 mg kg-1, i.p.) was able to inhibited both first and second phase of the formalin-induced nociception in mice. The known triterpenes 3beta,16beta-dihydroxy-olean-12-ene (brein) (4) and a mixture of alpha (5)- and beta-amyrin (6) were also found.

Bradykinin (BK) a nonapeptide generated in plasma during tissue injury, is involved in many physiological and pathological states. Kinin actions are mediated by specific membrane receptors and involve a complex signal transducer and also second messager mechanisms. Due to its inequivocal relevance, an intensive effort has been focused in recent years to develop selective and competitive BK antagonists. Thus, the development of a new series of peptide BK antagonists has made an important contribution to the understanding of the pharmacological, physiological and pathophysiological role of BK, and this is certain to provide a firm basis for developing new drugs to relieve pain and inflammation. However, BK antagonists derived from peptide origin reported to date have limited clinical use due to their poor oral absortion and short duration of effect. Thus, considerable effort has also been made in developing stable nonpeptide BK antagonists. Up to now, most nonpeptide compounds reported to exhibit BK antagonistic activity have been derived from plants, including many flavonoids, terpenes, and also synthetic substances with various molecular structures. Amongst them, the pregnane glycoside compounds isolated from the plant Mandevilla velutina are the most promising. These compounds are effective in antognizing BK responses in a variety of preparations, and they also exhibit potent and long-lasting analgesic and anti-inflammatory activities. The exact mechanism underlying their action however, is not yet completely understood.

Estudos de nosso laboratório surgerem que o ácido tânico (AT) exerce diversos efeitos "in vitro" ou "in vivo", podendo ser responsável por várias ações inespecíficas freqüentemente observadas com extratos brutos de plantas medicinais. O presente trabalho amplia estes estudos em outos modelos biológicos. A administração crônica ou aguda, por vida oral (v.o.), de AT em ratos não produziu alterações gástricas ou no processo inflamatório. Em ratos não-diabéticos observou-se modificações variáveis na glicemia de acordo com a metodologia empregada. O AT, cronicamente (v.o.), não interferiu na reprodução de fêmeas, embora constatou-se redução do número de filhotes de machos tratados. O AT (3mg/kg, v.o.) provocou aumento da diurese, porém doses mais elevadas não modificaram o volume urinário. Os testes hematológicos demonstraram que o AT possui efeito anticoagulante "in vitro", sem modificar a fragilidade de hemácias ou na homeastase sanguínea "in vivo". No músculo esquelético, o AT reduziu a resposta contrátil à acetilcolina, enquanto no músculo cardíaco (átrio esquerdo) observou-se efeito inotrópico positivo ou negativo. Estes resultados estendem os estudos anteriores e reforçam a sugestão de que parte significativa dos efeitos inespecíficos observados com extratos brutos de plantas medicinais podem estar relacionados com a presença de taninos nos mesmos.

Extending previous studies the present report analyses the pharmacological effects of tannic acid (TA) in several tests. TA prevented carragenin-induced edema when administered by i.p. but not p.o. route. Long term p.o. treatment with TA did not induce gastric lesions and showed inconsistent effects on glicemia in non-diabetic rats. Rats chronically exposed to TA during the fetal stage or following birth did not exhbit changes in gain of or organ weights, but adult male rats showed a lower fertility ratio. Urinary output was slightly increased by administration of a small dose of TA (3mg/kg, p.o.), but not by higher doses. The addition of TA "in vitro" to blood samples did not influence osmotic fragility but prevented clotting for at least 30 min. In isolated tissues, TA non competitively antagonized acetylcholineinduced contractions of the frog rectus-abdominal muscle and induced, depending of the dose, both positive and negative inotropic effects in the electrically-stimulated left rat atrium. These results confirm and extend our previous observations and suggest that a great number of non-specific effects detected in studies on the pharmacology of different folk medicinal plants may be related to the presence of tannins.
