ABSTRACT Due to the supposed involvement of minerals in cases of ruminant periodontitis, this study aimed to analyze the concentrations of phosphorus (P) in bone, and cobalt (Co), copper (Co), iron (Fe), zinc (Zn) and selenium (Se) in liver of a cohort of sheep affected or not by periodontitis. From an outbreak of the disease in 2011 in Pará state, Brazil, rib and liver samples were obtained from 22 sheep with periodontitis and seven samples from healthy animals. Based on the concentrations of the different minerals in the tissues, we concluded that there was no relationship between periodontal disease in sheep with any mineral deficiency status. In contrast, most of the minerals in the tissues were above or within the recommended concentrations in bone and liver. Within the various aspects which until now have been studied regarding periodontitis in ruminants, the results obtained here corroborate the fact that periodontal disease in sheep is an infectious disease and it is not a consequence of the deficiency or excess of mineral elements in the diet.

RESUMO Devido ao suposto envolvimento de minerais em casos de periodontite em ruminantes, este trabalho teve como objetivo analisar as concentrações de fósforo (P) nos ossos, além de cobalto (Co), cobre (Co), ferro (Fe), zinco (Zn) e selênio (Se), no fígado de ovelhas afetadas ou não por periodontite. De um surto da doença em 2011, no estado do Pará, Brasil, foram obtidas amostras de costelas e fígado de 22 ovelhas com periodontite e sete amostras de animais saudáveis. Com base nas concentrações dos diferentes minerais nos tecidos, conclui-se que não houve relação entre a doença periodontal em ovinos com qualquer estado de deficiência mineral. A maioria dos minerais estava acima ou dentro das concentrações recomendadas, seja no tecido ósseo, seja no hepático. Entre os diversos aspectos até agora estudados sobre a periodontite em ruminantes, os resultados obtidos corroboram o fato de a doença periodontal em ovinos ser uma doença infecciosa e não decorrente da deficiência ou do excesso de elementos minerais na dieta.

RESUMO Este estudo avaliou os aspectos clínicos e produtivos de matrizes Nelore, suplementadas ou não com P. Noventa, e cinco nulíparas prenhes foram distribuídas em dois grupos (G40 e G0). Os animais do G40 receberam uma mistura mineral com 224g de Na e 40g de P/kg, até a data do parto, e o G0 recebeu apenas NaCl. Após o parto, as primíparas foram divididas em quatro grupos: do G40, 28 continuaram a receber a mistura contendo 224g de Na e 40g de P/kg (grupo 40/40), e 12 passaram a receber apenas NaCl (grupo 40/0); do G0, 26 continuaram recebendo apenas NaCl (grupo 0/0), e 15 passaram a ser suplementadas com a mistura contendo 224g de Na e 40g de P/kg (grupo 0/40). Na segunda fase experimental, 205 vacas foram divididas em dois grupos: G1 (40/40), composto por 40 vacas, suplementadas com a mistura mineral contendo 244g de Na e 40g de P/kg, durante a gestação e a lactação, e o G2 (0/0) foi formado por 165 vacas, as quais foram suplementadas apenas com NaCl, durante a gestação e a lactação. Em ambos os experimentos, seja nas matrizes em gestação ou naquelas em lactação, não houve qualquer sinal da deficiência de P, e isso se refletiu na ausência de diferenças nos pesos dos bezerros aos 120 e 210 dias.

ABSTRACT The aim of this case study was to evaluate the clinical and productive data of Nellore cows during pregnancy and lactation, supplemented or not with P. Ninety-five pregnant heifers were divided into two groups (G40 and G0). Until the date of calving animals from G40 received a mineral mixture composed of 224g of Na and 40g of P/kg, and G0 received only NaCl. After calving heifers were divided into four treatments: from G40, 28 first-calf cows continued to receive the mineral mixture containing 224g of Na and 40g of P/kg (group 40/40) and 12 started to receive only NaCl (group 40/0); from G0, 26 continued to receive only NaCl (group 0/0) and 15 started to receive a mixture containing 224g of Na and 40g of P/kg (group 0/40). In the second experimental phase, 205 multiparous cows were divided into two groups: G1 (40/40) consisting of 40 cows, receiving a mineral mixture containing 244g of Na and 40g/P per kg, during pregnancy and lactation, and group 2 (0/0) formed by 165 cows, which were supplemented only with NaCl. In both experiments, whether in pregnant or lactating breeders, there was not any sign of P deficiency or differences in calves LW at 120 and 210 days.

RESUMO A eficácia do tratamento da deficiência de cobre, por via parenteral, em bezerros bubalinos criados na Ilha de Marajó (PA), foi investigada em 14 animais, distribuídos em dois grupos. O grupo 1 recebeu três doses de 120mg de glicinato de cobre (20mgCu/mL), por via intramuscular, nos dias um, 45 e 90, e o grupo 2 (controle) foi formado por animais que não receberam o cobre, seja suplementado por via oral ou por via parenteral. Em ambos os grupos, foram realizadas biópsias hepáticas nos dias um e 135. O diagnóstico da deficiência de cobre foi estabelecido pelo histórico, pelos sinais clínicos e também pelos teores de cobre hepático. No grupo 1, a média do teor de cobre hepático aumentou de 17,4±5,84mg/kgMS (antes da administração) para 311,2±133,5mg/kgMS (após 135 dias) (P<0,05). Ao final do experimento (135 dias), nos animais do grupo controle, as médias dos teores de cobre diminuíram de 17,1±3,4 para 9,11±1,1mg/kgMS, (P<0,05). Diante dos dados, ficou demonstrado que a administração de cobre, por via parenteral, foi eficiente no tratamento da deficiência de cobre nos bezerros búfalos e pode se tornar uma opção muito promissora para a realidade dos criadores de búfalos da Ilha de Marajó.

ABSTRACT The objective of this study was to investigate the effectiveness of the treatment of copper deficiency, by parenteral route, in buffalo calves raised on the Marajó Island (PA). Fourteen animals were divided into two groups. Group 1 received three doses of 120 mg of copper glycinate (20mgCu/mL), intramuscularly, on days 1, 45 and 90; in the group 2 (control) the animals were Not supplemented with copper. In both groups liver biopsies were performed on days 1 and 135. The diagnosis of copper deficiency was established through history, clinical signs, and by the analysis of hepatic copper content. In group 1, the mean hepatic copper content increased from 17.4±5.84mg/kgDM (before administration) to 311.2±133.5mg/kgDM (after 135 days) (P<0.05). In the control group, the mean of hepatic copper contents decreased from 17.1±3.4 to 9.11±1.1mg/kgDM at the end of the experiment (P<0.05). Based on the results, it was demonstrated that the administration of copper, by parenteral route, was efficient in the treatment of copper deficiency in buffalo calves and could become a very promising option for the reality of buffalo producers on the Island of Marajó.

RESUMO O objetivo deste estudo foi comparar a eficiência reprodutiva de búfalas leiteiras submetidas a protocolos de inseminação artificial em tempo fixo (IATF) à base de progesterona/estrogênio (P4/E2) e eCG, durante a estação reprodutiva desfavorável, usando-se sêmen resfriado (SR) e congelado (SC) Um total de 446 búfalas (> 40 dias após o parto) foi distribuído aleatoriamente em quatro blocos (anos): B1-2014 (n = 143), B2-2015 (n = 34), B3-2016 (n = 90) e B4-2017 (n = 179). Cada bloco foi subdividido em dois (IA com SR e SC utilizando-se a mesma ejaculação de cada touro). Assim, a subdivisão do bloco foi a seguinte: B1 (SR = 71 e SC = 72); B2 (SR = 18 e SC = 16); B3 (SR = 47 e SC = 43); e B4 (SR = 90 e SC = 89). Os ejaculados de oito touros Murrah coletados com vagina artificial foram divididos em duas alíquotas: uma alíquota diluída em diluente comercial Botu-Bov® e resfriada (BB-SR), e a outra diluída no mesmo diluente e congelada (BB-SC). As alíquotas de BB-SR foram resfriados a 5°C/24h usando-se um refrigerador. As alíquotas do grupo BB-SC também foram resfriadas e, após equilíbrio a 5°C por 4h, foram colocadas em uma caixa de isopor de 21L, 5 cm acima da superfície do nitrogênio líquido. À tarde (A), no D0 (14h), os animais receberam BE 2,0 mg IM (Estrogin®) e um implante auricular (Crestar® 3,0 mg P4). No D9 (A), o implante foi retirado e os animais receberam eCG 400 UI IM (Folligon® 5000) + cloprostenol PGF2α 0,530 mg IM (Sincrocio®). No D10 (A), os animais receberam BE 1,0mg IM (Estrogin®), e, no D12 (8h da manhã), foram realizadas as IAs. No D42, a gestação foi diagnosticada por ultrassonografia. As taxas de concepção (TC) totais foram 48,2% SR e 34,6% SC para os anos de 2014 a 2017, com uma diferença significativa de 13,7% (P<0,05). Em conclusão, o sêmen resfriado resultou em maior TC do que o sêmen congelado em bubalinos leiteiros sob P4/E2 e eCG FTAI durante a estação reprodutiva desfavorável.

ABSTRACT The objective of this study was to compare the reproductive efficiency of dairy buffaloes undergoing fixed-time artificial insemination (FTAI) protocols based on progesterone/estrogen (P4/E2) and eCG during unfavorable breeding season using cooled (CS) and frozen semen (FS). A total of 446 buffaloes (> 40 days postpartum) were randomly distributed into four blocks (years): B1-2014 (n = 143), B2-2015 (n = 34), B3-2016 (n = 90), and B4-2017 (n = 179). Each block was subdivided into two (AI with CS and FS using the same ejaculate of each bull). Thus, the block subdivision was as follows: B1 (CS = 71 and FS = 72); B2 (CS = 18 and FS = 16); B3 (CS = 47 and FS = 43); and B4 (CS = 90 and FS = 89). The ejaculates of eight Murrah bulls collected using an artificial vagina were divided into two aliquots: one aliquot was diluted in Botu-Bov® commercial extender and cooled (BB-CS), and the other was diluted in the same extender and frozen (BB-FS). BB-CS aliquots were cooled at 5 °C/24 h using a refrigerator. BB-FS group aliquots were also cooled, and after equilibrating at 5 °C for 4 h, were placed in a 21-L Styrofoam box, 5 cm above the surface of liquid nitrogen. In the afternoon (A) on D0 (2:00 p.m.) the animals received EB 2.0 mg IM (Estrogin®) and an ear implant (CRESTAR® 3.0 mg P4). At D9 (A), the implant was removed, and the animals received eCG 400 IU IM (Folligon® 5000) + Cloprostenol PGF2α 0.530 mg IM (Sincrocio®). At D10 (A), the animals received EB 1.0 mg IM (Estrogin®), and at D12 (8:00 a.m.), AI was performed. At D42, pregnancy was diagnosed via ultrasonography. Total CRs were 48.2% CS and 34.6% FS for years 2014 to 2017, with a significant difference of 13.7% (P<0.05). In conclusion, cooled semen resulted in higher CR than frozen semen in dairy buffaloes under the P4/E2 and eCG FTAI during the unfavorable reproductive season.

RESUMO Este estudo investigou in vitro a eficácia de quatro diferentes extensores (TES-TRIS e TRIS com lipoproteína de baixa densidade - LDL, nas concentrações de 10 ou 5%) sobre a longevidade espermática de búfalos no processo de refrigeração a 5ºC. A motilidade espermática foi avaliada a cada 24 horas até 72 horas de incubação, por sistema computadorizado “CASA”, e a integridade de membrana espermática foi examinada pelo teste hiposmótico (HOST) em T1, T24, T48 e T72 horas. Foram utilizados 11 búfalos (um ejaculado por búfalo) da raça Murrah, com idade variando de quatro a cinco anos. Imediatamente após a coleta, cada ejaculado foi fracionado em quatro alíquotas, e cada alíquota foi diluída em um dos quatro diluidores para a obtenção de 50x106 SPTZ/mL. As amostras foram envasadas em palhetas de 0,5 mL, refrigeradas (-0,25oC/minuto) até 5oC e mantidas nessa temperatura até a avaliação. Previamente à avaliação, as amostras foram aquecidas a 37oC por 30 segundos. O pacote estatístico utilizado para as análises foi o STATA 12.0 “Statistical Analysis Software”, e as médias foram comparadas pelo teste de Friedman (P<0,05). Os resultados de cinética e HOST até o tempo de 48 horas indicam que o diluidor TRIS com 10% LDL seria uma alternativa promissora para a refrigeração do sêmen a 5ºC, a ser utilizado na inseminação artificial e na inseminação artificial em tempo fixo.

ABSTRACT This study investigated in vitro the efficacy of four different extenders (TES-TRIS and TRIS with LDL low-density lipoprotein at concentrations of 10 or 5%) on the longevity of buffalo sperm in the refrigeration process at 5ºC. Sperm motility was assessed every 24 hours up to 72 hours of incubation using computer assisted sperm analysis and sperm membrane integrity was examined by the hypoosmotic test (HOST) at T1, T24, T48 and T72 hours. Eleven buffaloes (1 ejaculate per buffalo) of the Murrah breed were used, ranging in age from 4 to 5 years. Immediately after collection, each ejaculate was fractionated into 4 aliquots, and each aliquot was diluted in one of four diluents to obtain 50x106SPTZ/mL. The samples were packed in 0.5mL straws and refrigerated (-0.25°C/min) to 5°C and maintained at this temperature until evaluation. Prior to evaluation the samples were heated at 37°C for 30 seconds. The statistical package used for analysis was STATA 12.0 "Statistical Analysis Software" and means were compared by the Friedman test (P<0.05). The results of sperm kinetics and HOST indicate that the TRIS diluent with 10% LDL could be a promising alternative for semen refrigeration at 5ºC, to be used in conventional and fixed time artificial insemination.

O objetivo geral deste trabalho foi analisar o efeito da radiação vermelha de baixa intensidade sobre alguns parâmetros cinéticos do espermatozoide canino criopreservado. Ejaculados de oito cães adultos foram centrifugados, rediluídos em meio tris-gema de ovo com 6% de glicerol, e, posteriormente, fracionados em: T1: incidência de radiação vermelha (660 NM) (Fisioled - Mmoptics - 100mW) por 60 segundos, antes do resfriamento e após a descongelação; T2: incidência somente antes do resfriamento; T3: incidência somente após a descongelação; e T4: sem incidência. Após a descongelação, as amostras foram submetidas ao TTR utilizando-se Sperm Class Analyzer(r). No TTR0, TTR60 e TTR90, não houve diferença entre as variáveis analisadas pelo CASA. Somente no TTR30 os efeitos da incidência da radiação vermelha foram evidentes e significativos em T1 e T2; T1 resultou em baixa MT (12,5 + 10,6%) e T2 determinou o melhor resultado de MT 40,3 + 26,1%. De forma similar T1 apresentou maior número de espermatozoides estáticos (77,5±28,9%) em relação ao T2 (50,6±28%). Concluiu-se que a dupla incidência de radiação vermelha de baixa intensidade antes do resfriamento e após a descongelação teve efeito deletério sobre a motilidade do espermatozoide canino, expressa principalmente aos 30 minutos após descongelação.

The aim of this study was to analyze the effect of low intensity red light on some kinetic parameters of cryopreserved canine sperm. Ejaculates from 08 adult dogs were centrifuged, diluted in Tris-egg yolk with 6% glycerol, and subsequently separated into: T1: incidence of red light (660 nm) (Fisioled -MMOptics - 100mW) for 60 seconds before the cooling and after thawing, T2: just before cooling, T3: only after thawing, and T4: no incidence. After thawing the samples were subjected to TTR using Sperm analyzer(r). In TTR0, TTR60 and TTR90 there were no differences between the variables analyzed by CASA. Only in TTR30 the effects of the incidence of red light were visible and significant in T1 and T2. T1 resulted in low MT (12.5 ±10.6%) and T2 determined the best result of MT 26.1%±40.3. Similarly T1 showed a higher number of static spermatozoa (77.5±28.9%) compared to T2 (50.6±28%). We concluded that the double incidence of low intensity red light, before cooling and after thawing, had a deleterious effect on canine sperm motility expressed at 30 minutes after thawing.

RESUMO Para estudar as lesões anátomo-histopatológicas e suas variações na infecção experimental de cobaias pela Burkholderia mallei, foram utilizados 60 animais incluídos em 5 grupos formados por 12 animais cada. Cada grupo foi inoculado com uma amostra de campo de B. mallei isolada do conteúdo purulento de nódulos cutâneos fechados de eqüídeos com mormo. Para o estudo anátomohistopatológico foram eutanasiados 2 animais por grupo a cada 48h após a inoculação. As lesões macroscópicas mais freqüentes foram congestão e hemorragia pulmonar, abscessos hepáticos, aderência da túnica vaginal, túnica vaginal hiperêmica e congesta, vaginalite purulenta e testículos hemorrágicos. O exame histológico revelou congestão, vacuolização difusa e necrose dos hepatócitos; congestão cortical e medular e vacuolização das células epiteliais; edema, congestão e hemorragias nas túnicas vaginal e albugínea, além de formação de piogranulomas; processo degenerativo tubular com descamação celular, presença de macrófagos e debris celulares, além de células sinciciais na luz dos túbulos do testículo e epidídimo. Os achados macro e microscópicos observados neste estudo indicam uma possível variação na virulência das amostras de B. mallei isoladas e que a prova de Strauss é um teste bastante sensível e que pode ser empregado no diagnóstico do mormo.

ABSTRACT A histopatological study was conducted in guinea pigs inoculated experimentally with a Burkholderia mallei strain isolated from equides from farms located in Pernambuco State, Brazil. The microbiological aspects of diagnosis of glanders in horses from these farms also were studied. The B. mallei strain was obtained from nasal discharge and aspirated fluids from closed subcutaneous nodules from horses with clinical signs of glanders. The material was collected and transferred to a sterile-screw-capped tube for transport to the laboratory where bacterial isolation and biochemical tests were carried out. For the clinical and histopathological study 5 groups with 12 male guinea pigs were used to verify the injuries caused by B. mallei. During the course of the study (1-10 days post-infection) clinical evaluation was carried out in all animals. At each 48h 2 inoculated animals were euthanaized and samples from lungs, liver, spleen, kidney and testis were collected to verify gross or histologic lesions. Strauss test was positive in all animals 48h. Gross examination showed pulmonary, liver and testis injuries characterized by congestion and hemorrhages of lungs, liver abscess and changes of vaginal layer varied from vaginalitis to hemorrhages in this tissue. Microscopic lesions were observed in liver, kidney and especially in testis with the pyogranulom located not only in the albuhin but also in vaginal layer degenerative changes.
