ABSTRACT: Surface contamination by pathogenic and deteriorating microorganisms is a constant concern in the food industry. Therefore, in this study we evaluated the antimicrobial activity of silver nanoparticles (AgNPs), produced using the essential oil of Lippia origanoides Kunth and ethanolic extract of Hymenaea martiana Hayne, against bacteria used in milk processing. For this, AgNPs were synthesized and their antimicrobial activity was evaluated for minimum inhibitory and bactericidal concentrations, time-kill, interference on the biofilm, and application on the surfaces of different materials. AgNPs showed bacteriostatic and bactericidal activity against Staphylococcus aureus (ATCC 25923), Staphylococcus aureus (ATCC 33591), Escherichia coli (ATCC 25922), Salmonella enterica subsp. enterica serovar Choleraesuis (ATCC 10708), Listeria monocytogenes, Escherichia coli, Salmonella spp.,andPseudomonas aeruginosa,and interfered with biofilm formation more than with consolidated biofilms. A 1hour period was sufficient to reduce the bacterial cells, whereas a Minimum Duration for Killing (MDK) of 99% was reached after approximately 30 min. AgNPs were effective against bacteria attached to stainless steel and polyethylene, but ineffective on tile surfaces. Thus, owing to the growing microbial resistance and the need to develop new products based on the concepts of green chemistry, these AgNPs are presented as a new possibility for cleaning processes in the food industry.

RESUMO: A contaminação de superfícies por micro-organismos patogênicos e deteriorantes é uma preocupação constante na indústria de alimentos. Dessa forma, o objetivo deste trabalho foi avaliar a atividade antimicrobiana de nanopartículas de prata (AgNPs) produzidas utilizando óleo essencial de Lippia origanoides Kunth e extrato etanólico da Hymenaea martiana Hayne, frente a bactérias de importância na contaminação em áreas de processamento do leite. Para isso, as AgNPs foram sintetizadas e sua atividade antimicrobiana foi avaliada através das concentrações inibitória e bactericida mínimas, tempo de morte, interferência sobre o biofilme, e por fim, a sua aplicação sobre superfícies de diferentes materiais. As AgNPs apresentaram atividade bacteriostática e/ou bactericida frente à Staphylococcus aureus (ATCC 25923), Staphylococcus aureus (ATCC 33591), Escherichia coli (ATCC 25922), Salmonella enterica subsp. enterica serovar Choleraesuis (ATCC 10708), Listeria monocytogenes, Escherichia coli, Salmonella spp. e Pseudomonas aeruginosa, e interferiram na formação do biofilme mais do que sobre o biofilme consolidado. O tempo de uma hora foi o suficiente para reduzir as populações bacterianas, enquanto o Minimum Duration for Killing MDK 99% foi atingido próximo a 30 minutos. As AgNPs foram eficientes contra bactérias aderidas ao inox e ao polietileno. Assim, tendo em vista a crescente resistência microbiana e o desenvolvimento de novos produtos baseados nos conceitos da química verde, essas nanopartículas de prata apresentam-se como uma nova possibilidade de uso nos processos de higienização da indústria de alimentos.

RESUMO No presente estudo, investigamos a contaminação do leite cru refrigerado produzido na região oeste do Paraná, sul do Brasil, com micro-organismos psicrotróficos, visando testar a atividade proteolítica dos isolados e identificar Pseudomonas fluorescens, a principal espécie proteolítica associada à deterioração de produtos lácteos. Amostras de leite cru de 50 fazendas leiteiras foram submetidas à contagem de micro-organismos psicrotróficos, caracterizando-se a microbiota por seu comportamento mesofílico e sua capacidade proteolítica, além de identificação molecular de P. fluorescens. Entre as amostras avaliadas, 94% apresentaram contagem psicrotrófica variando de 3 a 7,1 log UFC mL-1 e 48,5% destas apresentaram comportamento mesofílico. Entre os isolados, 48,0% apresentaram atividade proteolítica em pelo menos uma das temperaturas testadas (21 e 30°C) e 39,3% apresentaram atividade proteolítica em ambas as temperaturas. Entre os 61 isolados sub-metidos à identificação molecular por reação em cadeia da polimerase (PCR), 86,8% continham expressão do gene 16S característico de P. fluorescens. Neste estudo, demonstramos que P. fluorescens é a espécie de bactérias psicrotróficas mais prevalente em leite refrigerado cru e sua capacidade proteolítica promove elevados riscos de deterioração para a indústria de laticínios.

ABSTRACT In this study, we investigated the contamination of refrigerated raw milk produced in the western region of Paraná, southern Brazil, with psychrotrophic microorganisms, aiming to assay the proteolytic activity of the isolates and to identify Pseudomonas fluorescens, the main proteolytic species associated with the spoilage of milk products. Raw milk samples from 50 dairy farms were submitted to the counting of psychrotrophic microorganisms, being the microbiota characterized by its mesophilic behavior and proteolytic capacity, besides molecular identification of P. fluorescens. Of the samples evaluated, 94% had psychrotrophic counts ranging from 3 to 7.1 log CFU mL-1, and 48.5% of these showed mesophilic behavior. Of the isolates, 48.0% had proteolytic activity in at least one evaluated temperature (21 and 30°C), and 39.3% had proteolytic activity in both temperatures. Among the 61 isolates submitted to molecular identification by polymerase chain reaction (PCR), 86.8% contained the expression of the 16S gene characteristic for P. fluorescens. In this study, we demonstrated that P. fluorescens is the most prevalent psychrotrophic bacteria species in raw refrigerated milk and their proteolytic ability poses high risks to the dairy industry.

ABSTRACT. In order to evaluate how beef buyers in the State of Paraná, Brazil south value meat quality indicators, and attributes of credibility, 519 interviews were conducted in three cities with different sizes (Big - Curitiba, Medium - Campo Mourão and Small - Palotina). The interviews were applied after the meat was placed in the shopping cart and were composed of five questions, including question of spontaneous response: (i) factors that the buyer considers at the purchasing time; (ii) information which the buyer considers important to appear on the meat label; and three questions of stimulated response: (i) preference for meat preparation and exposure for sale, (ii) factors that the buyer considers when purchasing the meat, and (iii) preference for marbling (by photograph) according to the beef preparation. The results showed that beef buyers are more concerned with extrinsic meat quality indicators than the intrinsic ones, especially the meat presentation and hygiene of the sale point; have little knowledge about the factors that positively influence the eating experience of beef and do not cares about attributes of credibility, except for the meat expiration date. Beef buyers from medium (Campo Mourão) and small (Palotina) cities have a similar behavior, and differ from the buyers of a big (Curitiba) city among the attributes surveyed, in relation to the greater appreciation of inspection, tenderness and marbling, and less importance given to the expiration date. Beef buyers prefer meat cut by the butcher; however, this preference is higher in the small city.

RESUMO: Os objetivos desta pesquisa foram verificar o perfil, identificar os principais motivos para o consumo e avaliar laboratorialmente as amostras de leite cru informal de cinco municípios do oeste do estado do Paraná, Brasil. Os tipos de leite mais frequentemente consumidos foram: 42,3% ultra-high temperature (UHT), 38,3% pasteurizado, 17,6% leite cru informal e 1,7% em pó. As frequências de consumidores que preferiram leite cru informal, por cidade, foram: 17,6% em Palotina, 29,2% em Marechal Cândido Rondon, 18,9% em Assis Chateaubriand, 17,6% em Palotina e 10% em Toledo. Sabor foi o principal motivo que influenciou o consumo de leite cru informal, e famílias com renda familiar de um a quatro salários mínimos foram as que mais consumiram esse tipo de leite. A comercialização informal de leite foi mais vantajosa financeiramente para o produtor em relação à comercialização formal. Todas as amostras analisadas apresentaram-se em desacordo em pelo menos um parâmetro, sendo 60,9% para mesófilos, 56,6% para estrato seco desengordurado, 52,1% para índice crioscópico, 43,5% para acidez, 23,9% para densidade, 23,9% para índice de caseíno macropeptídeo, 17,4% para teor de gordura, 8,7% reagente ao teste do anel em leite (TAL), e 2,2% reagente para presença de inibidor de crescimento bacteriano. A adulteração encontrada foi a adição de água em 20% das amostras analisadas. O leite cru informal avaliado nesta pesquisa envolveu a comercialização de um produto sem qualidade e que constitui um perigo financeiro, em razão do consumo de produtos adulterados, além do risco à saúde do consumidor.

ABSTRACT: The objectives of this study were to analyze consumer profile; to identify the main reasons for raw milk consumption; and to analyze in laboratory samples of uninspected raw milk from five towns in the western region of the state of Paraná, Brazil. The types of milk most frequently consumed were: 42.3% ultra-high temperature (UHT), 38.3% pasteurized milk, 17.6% uninspected raw milk, and 1.7% powered milk. The frequencies of households that preferred uninspected raw milk were, according to the town, 32.7% in Iporã, 29.2% in Marechal Cândido Rondon, 18.9% in Assis Chateaubriand, 17.6% in Palotina, and 10% in Toledo. Flavor was the main reason for uninspected raw milk consumption, and the purchase of this product was more frequent in households whose income was between one to four minimum wages. It was observed that the sales of uninspected milk are more financially advantageous to the producer than sales of inspected milk. All samples analyzed showed lack of compliance with at least one parameter, 60.9% for mesophilic counts, 56.6% for non-fat dry matter, 52.1% for freezing point, 43.5% for acidity, 23.9% for density, 23.9% for the casein macropeptide, 17.4% for fat content, 8.7% were reactors in the milk ring test, and 2.2% were reactors in microbial growth inhibitor test. Fraud by addition of water was observed in 20% of the samples. Uninspected raw milk analyzed in this study involved a low-quality product that is a financial hazard as it may be adulterated, and that poses risk to consumer health.

Abstract The aim of this study was to investigate the occurrence of multidrug-resistant, extended spectrum beta-lactamase (ESBL) producing Salmonella spp. isolated from conveyor belts of broiler cutting rooms in Brazilian broiler processing plants. Ninety-eight strains of Salmonella spp. were analyzed. Multidrug resistance was determined by the disk diffusion test and the susceptibility of the isolated bacteria was evaluated against 18 antimicrobials from seven different classes. The double disk diffusion test was used to evaluate ESBL production. Of the 98 strains tested, 84 were multidrug resistant. The highest rates of resistance were against nalidixic acid (95%), tetracycline (91%), and the beta-lactams: ampicillin and cefachlor (45%), followed by streptomycin and gentamicin with 19% and 15% of strain resistance, respectively. By contrast, 97% of the strains were sensitive to chloramphenicol. 45% of the strains were positive for the presence of ESBL activity. In this study, high rates of multidrug resistance and ESBL production were observed in Salmonella spp.

A linguiça frescal é um embutido curado que, devido à manipulação durante as etapas de produção, pode se tornar contaminado com S. aureus, patógeno frequentemente relacionado com práticas inadequadas de higiene durante a produção de alimentos. A utilização de nitrito em embutidos tem sua eficácia comprovada contra C. botulinum e, para o S. aureus, estudos indicam que a ação bactericida depende de fatores extrínsecos e intrínsecos. O objetivo do presente estudo foi avaliar o efeito da concentração de nitrito e do pH frente à contaminação por S. aureus e microbiota autóctone em linguiças frescais estocadas em diferente tempos e temperaturas. Linguiças foram produzidas com concentrações de 50, 150 e 200ppm de nitrito e contaminadas com S. aureus. As linguiças foram armazenadas sob refrigeração (7 e 12°C) e a quantificação de S. aureus e psicrotróficos foi realizada nos dias 0, 2, 4, 7 e 10. Os resultados demonstraram que a influência das concentrações de nitrito e temperaturas utilizadas sobre a multiplicação de S. aureus e da microbiota autóctone foi mínima, sendo dependente apenas do período de estocagem. Entretanto, ao final de dez dias, as contagens foram estatisticamente iguais nos grupos analisados, mostrando que as condições de temperatura e concentrações de nitrito utilizadas não exerceram controle efetivo no desenvolvimento destes micro-organismos. Sugere-se que este produto seja armazenado sob temperaturas inferiores a 7°C ou sob congelamento para maior estabilidade microbiológica

Fresh sausages are cured meat products that may be contaminated with Staphylococcus aureus during the manufacturing procedure, which is frequently related with inadequate handling practices. The use of nitrite in meat products has proven efficacy against Clostridium botulinum, and studies indicate that bactericidal action against S. aureus depends on factors that are intrinsic and extrinsic to the product. The objective of the present study was to evaluate the effect of nitrite concentration, and pH on S. aureus and psychrotrophic autochthone microbiota in fresh sausages stored at different times and temperatures. Fresh sausage were produced at nitrite concentrations 50, 150 and 200ppm and contaminated with S. aureus. The sausages were storage at refrigeration (7 and 12°C) and the quantification of S. aureus and psychrotrophic microorganisms was carried out on days 0, 2, 4, 7, and 10. Results showed that nitrite concentrations and the temperatures used had minimal effect on the multiplication of S. aureus and psychrotrophic autochthone microbiota. Final counts depended only on the length of storage: at the end of 10 days, counts were statistically similar in the different groups, showing that temperature and nitrite concentrations used did not control microbial growth effectively. It is suggested that the product should be stored below 7°C or at freezing temperatures for greater microbiological stability

A estabilidade dos óleos vegetais à oxidação depende do equilíbrio entre a composição e a presença de pró-oxidantes e antioxidantes. O objetivo deste estudo consistiu em avaliar o efeito da presença de antioxidantes sintéticos durante o processo de foto-oxidação dos óleos de canola e milho. As amostras dos óleos de canola e milho em presença e em ausência dos antioxidantes butil-hidroxitolueno (BHA), propil galato (PG) e terc butil-hidroquinona (TBHQ), foram submetidas ao estresse foto-oxidativo. A reação de foto-oxidação seguiu uma cinética de primeira ordem. A constante de velocidade no período de 20 dias de foto-oxidação evidenciou que o antioxidante PG apresentou maior efeito protetor para o óleo de canola e o TBHQ para o óleo de milho. A partir dos dados de UV e RMN de ¹H, no período de 60 dias, constatou-se que os três antioxidantes apresentaram um efeito protetor a foto-oxidação. Os dados de UV evidenciaram aumento de absorção da banda em 232 nm devido à foto-oxidação e formação de dienos conjugados. A redução da intensidade de absorção desta banda na foto-oxidação com o tempo de exposição revelou que a proteção, tanto para o óleo de canola como para o de milho, foi mais efetiva com o antioxidante PG. Os índices de oxidação Roa e fração residual de hidrogênios (H) dialílicos, alílicos e vinílicos também evidenciaram que o antioxidante PG apresentou o melhor desempenho na proteção dos dois tipos de óleo.

The stability of vegetable oils in relation to oxidation depends on the balance between the composition and the presence of antioxidants and pro-oxidants. The objective of this study was to evaluate the effect of the presence of antioxidants in vegetable oils in the protection of photo-oxidation. Samples of canola oil and corn oil in the presence and absence of the antioxidants, butylated hydroxytoluene (BHA), propyl gallate (PG) and tert-butylhydroquinone (TBHQ), were subjected to photo-oxidative stress at room temperature for 60 days. The photo-oxidation reaction followed a first order kinetics. The rate constant for the period of 20 days of photo-oxidation showed that the PG antioxidant showed greater protective effect for canola oil and TBHQ showed a greater protective effect for corn oil. The UV and ¹H NMR data at 60 days showed that the three antioxidants had a protective effect on photo-oxidation. The UV data showed increased absorption at the 232nm band due to photo-oxidation and the formation of conjugated dienes. The reduction in intensity of this absorption band in photo-oxidation with time of exposure revealed that the protection, both for the canola oil and for the corn oil, was more effective with the PG antioxidant. The rates of Roa oxidation and residual fraction of diallyl, allyl and vinyl hydrogens also showed that the PG antioxidant showed the best performance in protecting both oils.

O crescimento da produção e consumo de carne têm sido acompanhados da diversificação de produtos processados, com maior elaboração de itens de conveniência, praticidade e valor agregado, em detrimento da comercialização de cortes in natura. Fosfatos e proteínas não cárneas, sobretudo as da soja e do soro de leite, figuram entre os ingredientes mais usados para marinar carnes, melhorando sua textura e promovendo a retenção de líquidos que podem trazer aumento do rendimento de fabricação e benefícios sensoriais para o consumidor. Nessa tecnologia, os principais processos empregados são a injeção e o massageamento da carne. No entanto, práticas abusivas devem ser coibidas pela fiscalização, impondo limites à incorporação de água pelo produto final e à utilização de ingredientes não cárneos. No presente artigo, são revisados aspectos da utilização desses ingredientes, ressaltando a importância da disponibilidade de técnicas analíticas adequadas ao controle de produtos injetados e marinados.

Meat production and consumption growth has been accompanied by a wide diversification of processed products, while the trading of non-processed meat has decreased. Phosphates and non-meat proteins, mainly soy and whey proteins, are widely used to marinate meat, improving its texture and promoting the fluids retention that can bring sensorial benefits to the consumers and improve producers' yield. On this technology, the main processes employed are the injection and the tumbling of raw materials. However, some fraudulent practices must be avoided by the meat inspection service, setting limits to the replacement of meat proteins by water and non-meat ingredients. The present article reviewed the main aspects of the use of such ingredients enlightening the importance of appropriate analytical techniques to control injected, and marinated products.

No presente trabalho, a utilização de 1,5% de proteína isolada de soja em lombo de suíno foi detectada por meio do teor de isoflavonas, utilizando-se cromatografia líquida de alta eficiência. Elegeu-se o pico de genistina para quantificação, construindo-se uma curva-padrão com a proteína isolada de soja utilizada nas salmouras para injeção das carnes. A técnica revelou-se como uma alternativa rápida e eficaz para ser implantada no controle da utilização desse ingrediente em produtos cárneos.

The present research aimed to detect the use of soy protein isolate (1.5%) in enhanced pork loins through isoflavones identification, using high performance liquid chromatography (HPLC). The genistine peak has been chosen for the soy quantification, and it has been correlated with a standard curve built with the soy protein isolate used for the preparation of brines which have been injected into meat. Identification of isoflavones with HPLC is an efficient technique for the detection of soy in meat products.

A legislação brasileira permite a adição intencional de proteínas não cárneas em alguns produtos cárneos; entretanto, há poucas técnicas adaptadas e próprias para controle desses ingredientes. Na presente pesquisa, empregou-se a técnica de eletroforese em gel de poliacrilamida na presença de dodecil-sulfato de sódio (SDS-PAGE) para comparar amostras de carne suína (lombo), adicionadas de 1,5% de proteínas não cárneas (proteína isolada de soja e concentrado protéico de soro de leite). Mediante a padronização de dois protocolos distintos de extração (uréia 6M e Tris-HCl-SDS-mercaptoetanol) e a utilização de controles positivos e negativos, obtiveram-se perfis eletroforéticos típicos do músculo e das proteínas utilizadas. Ambas as extrações utilizadas foram adequadas para separação das proteínas do soro de leite das proteínas da carne, mas a visualização dos marcadores da soja não foi possível em razão da sobreposição de bandas.

Brazilian meat regulations allow the use of non-meat extenders in certain meat products. Thus, the need for controlling the use of these ingredients with proper analytical methods is evident, in order to avoid illegal addition. In the present research, SDS-PAGE electrophoresis has been applied to pork enhanced with 1.5% of soy protein isolate and whey protein concentrate, through two different methods of protein extraction (urea 6M and Tris-HCl-SDS-mercaptoethanol), in the presence of positive and negative controls. Electrophoresis has revealed typical profiles, for whole-muscle and non-meat proteins, showing to be adequate to detect the addition of whey proteins in meat products. However, the soy proteins could not be detected, due to the overlapping of bands with meat proteins.

O presente trabalho avaliou a multiplicação de L. monocytogenes naturalmente presente em mortadelas fatiadas, embaladas a vácuo e estocadas a 5ºC durante sua vida de prateleira. O teste Tukey indicou que as populações de L. monocytogenes nos tempos 10, 20, 30 e 40 dias diferiram significativamente (p<0,05) indicando multiplicação durante o armazenamento. Em três repetições, o aumento médio foi de 1,80 ciclos log. A embalagem a vácuo e estocagem sob refrigeração não foram suficientes para o controle da multiplicação de L. monocytogenes em mortadelas fatiadas, indicando que as boas práticas de fabricação e um sistema HACCP implantado são fundamentais para assegurar a segurança desse produto.

This study evaluated the growth of naturally occurring L. monocytogenes in sliced, vacuum-packed mortadella samples during storage at 5ºC until the expiration date. Tukey's test indicated that counts of L. monocytogenes on 0, 10, 20, 30 and 40 days of storage were significantly different (p<0.05), indicating growth during shelf life. In three trials, the mean increase was 1.72 log cycles. Vacuum packing and storage under refrigeration were not effective in controlling the growth of L. monocytogenes in sliced mortadella, indicating that good manufacturing practices and implemented HACCP programs are essential to assure safety of this product.
