Abstract Despite major advances in the study of the brain, investigations on neurochemistry in vivo still lack the solid ground of more established methods, such as structural and functional magnetic resonance imaging. Proton magnetic resonance spectroscopy (MRS) is a technique that might potentially fill in this gap. Nevertheless, studies using this approach feature great methodological heterogeneity, such as varying voxel of choice, differences on emphasized metabolites, and absence of a standardized unit. In this study, we present a methodology for creating a systematic review and meta-analysis for this kind of scientific evidence using the prototypical case of attention-deficit/hyperactivity disorder. Abstract The FOXP subfamily is probably the most extensively characterized subfamily of the forkhead superfamily, playing important roles in development and homeostasis in vertebrates. Intrinsically disorder protein regions (IDRs) are protein segments that exhibit multiple physical interactions and play critical roles in various biological processes, including regulation and signaling. IDRs in proteins may play an important role in the evolvability of genetic systems. In this study, we analyzed 77 orthologous FOXP genes/proteins from Tetrapoda, regarding protein disorder content and evolutionary rate. We also predicted the number and type of short linear motifs (SLIMs) in the IDRs. Similar levels of protein disorder (approximately 70%) were found for FOXP1, FOXP2, and FOXP4. However, for FOXP3, which is shorter in length and has a more specific function, the disordered content was lower (30%). Mammals showed higher protein disorders for FOXP1 and FOXP4 than non-mammals. Specific analyses related to linear motifs in the four genes showed also a clear differentiation between FOXPs in mammals and non-mammals. We predicted for the first time the role of IDRs and SLIMs in the FOXP gene family associated with possible adaptive novelties within Tetrapoda. For instance, we found gain and loss of important phosphorylation sites in the Homo sapiens FOXP2 IDR regions, with possible implication for the evolution of human speech.

Genetic studies have long suggested the important role of the DRD2 gene in psychiatric disorders and behavior. Further research has shown a conjoined effect of genes in the Chr11q22-23 region, which includes the NCAM1, TTC12, ANKK1 and DRD2 genes, or NTAD cluster. Despite a growing need to unravel the role of this cluster, few studies have taken into account interspecies and evolutionary approaches. This study shows that behaviorally relevant SNPs from the NTAD cluster, such as rs1800497 (Taq1A) and rs6277, are ancient polymorphisms that date back to the common ancestor between modern humans and Neanderthals/Denisovans. Conserved synteny and neighborhood indicate the NTAD cluster seems to have been established at least 400 million years ago, when the first Sarcopterygians emerged. The NTAD genes are apparently co-regulated and this could be attributed to adaptive functional properties, including those that emerged when the central nervous system became more complex. Finally, our findings indicate that NTAD genes, which are related to neurogenesis and dopaminergic neurotransmission, should be approached as a unit in behavioral and psychiatric genetic studies.

O diagnóstico do transtorno de déficit de atenção e hiperatividade (TDAH) e do transtorno de oposição desafiante (TOD) em adultos carece de instrumentos adequados. Esta investigação visa avaliar a confiabilidade entre observadores das seções do K-SADS-E adaptadas para os diagnósticos destes transtornos em adultos. As adaptações realizadas se limitaram ao direcionamento das perguntas ao próprio paciente, investigação do início dos sintomas em um período mais tardio e abordagem de sintomas próprios de adultos. Seis avaliadores treinados pontuaram 16 entrevistas gravadas. Os coeficientes Kappa foram de 1,00 para o diagnóstico de TDAH no passado, 0,91 para o subtipo no passado, 1,00 para diagnóstico de TDAH atual e 0,95 para o subtipo atual. Os coeficientes Kappa para o TOD foram de 1,00 para o diagnóstico no passado e 0,89 para o diagnóstico atual. Todos os coeficientes tiveram um p<0,001. Os resultados demonstram excelente concordância para os diagnósticos de TDAH e TOD em adultos utilizando a forma adaptada da versão em português do K-SADS-E.

The diagnosis of attention deficit hyperactivity disorder (ADHD) and oppositional defiant disorder (ODD) in adults lacks adequate assessment instruments. The aim of this investigation is to evaluate the interrater reliability of the sections of K-SADS-E adapted for the diagnosis of these disorders among adults. The adaptations were limited to asking questions directly to the patient, investigation of a later onset of symptoms, and asking for adult specific symptoms. Six trained raters scored 16 recorded interviews. Kappa coefficients were 1.00 for ADHD diagnosis in the past, 0.91 for subtype in the past, 1.00 for ADHD diagnosis in the present, 0.95 for subtype in the present, 1.00 and 0.89 for ODD diagnosis in the past and present, respectively (all with p<0.001). These results reveal excellent agreement for the ADHD and ODD diagnosis in adults using the adapted form of the Portuguese version of K-SADS-E.

Os avanços no conhecimento da neurobiologia da dependência permitiram delimitar uma série de genes candidatos para a predisposição. Atualmente, iniciam-se os estudos sobre o papel de polimorfismos genéticos na resposta ao tratamento. A integração de abordagens clínicas, epidemiológicas e de genética molecular pode identificar grupos clínicos mais responsivos a abordagens terapêuticas específicas.
