RESUMEN La uchuva (Physalisperuviana L.) es una fruta de gran interés por su alto contenido nutricional y potencial medicinal. En el cultivo, la marchitez vascular, enfermedad ocasionada por el hongo Fusarium oxysporum f. sp. physali (Foph), genera pérdidas hasta del 100%, siendo necesario identificar cultivares resistentes. Para contribuir con procesos de fitomejoramiento, se caracterizaron fisicoquímicamente frutos de 33 genotipos de uchuva con 18 descriptores cuantitativos. Los genotipos fueron sembrados en campo, bajo condiciones de alta presión del patógeno Foph y sin presencia de este. El test de Student detectó diferencias estadísticas (P<0.05) entre las dos condiciones para rendimiento, rajado del fruto y pH. El análisis de componentes principales explicó con cinco factores el 84.96% de la varianza total, donde las variables físicas del fruto fueron las de mayor contribución para el primer componente (41.65%). Mediante un análisis de conglomerados, se clasificaron genotipos con y sin presencia del patógeno, en siete y ocho grupos, respectivamente. Sobre dos genotipos constrastantes por su respuesta diferencial al patógeno, se realizó un análisis de capacidad antioxidante de frutos, en donde los métodos DPPH y ORAC presentaron diferencias (P<0.05) entre los dos genotipos siendo mayor la actividad antioxidante en el material suceptible.

ABSTRACT Cape gooseberry (Physalis peruviana L.) is a fruit of great interest, due to its high nutritional and potential medicinal value. Vascular wilt disease caused by the fungus Fusarium oxysporum f. sp. Physali (Foph) is responsible for crop losses of up to 100% which makes necessary to identify resistant cultivars. To contribute to crop improvement processes, a physicochemical characterization was performed on fruits of 33 cape gooseberry genotypes using 18 quantitative descriptors. The genotypes were planted in the field under high and no pressure of Foph. The Student's t test detected statistically significant differences (P<0.05) between the two conditions for yield, fruit cracking (%) and fruit juice pH. The principal component analysis explained in five factors 84.96% of the total variance, in which the fruit physical variables were the major contributor to the first component (41.65%). Cluster analysis grouped the genotypes under high and no pressure in seven and eight clusters, respectively. Two contrasting genotypes showing differential resistance response to the pathogen were analyzed for fruit antioxidant capacity, in which DPPH and ORAC methods presented significant differences (P<0.05) between the two genotypes with greater antioxidant activity in the susceptible material.

La resistencia sistémica inducida (ISR) es un mecanismo mediante el cual las plantas aumentan sus defensas frente a cualquier condición de estrés. El objetivo de este trabajo fue localizar en el mapa genético de tomate, genes candidatos involucrados en ISR y promoción de crecimiento inducidos por la cepa colombiana nativa Th003 de Trichoderma koningiopsis. Se realizó una búsqueda de polimorfismos en cuarenta y nueve genes candidatos previamente identificados en plantas de tomate inoculadas con Th003 y la cepa T382 de T. hamatum. Diez y seis de estos genes candidatos fueron integrados en el mapa genético de tomate altamente saturado, llamado “TOMATO EXPEN 2000”. La ubicación de seis unigenes fue similar a la localización de genes análogos de resistencia (RGAs), ESTs relacionados con defensa y QTLs de resistencia previamente identificados, sugiriendo posibles nuevos candidatos para estas regiones de QTLs. Los genes candidatos o marcadores pueden ser usados en futuros programas de selección asistida relacionados con ISR o promoción de crecimiento en tomate.

Induced systemic resistance (ISR) is a mechanism by which plants enhance defenses against any stress condition. ISR and growth promotion are enhanced when tomato (Solanum lycopersicum) is inoculated with several strains of Trichoderma ssp. This study aims to genetically map tomato candidate genes involved in ISR and growth promotion induced by the Colombian native isolate Trichoderma koningiopsis Th003. Forty-nine candidate genes previously identified on tomato plants treated with Th003 and T. hamatum T382 strains were evaluated for polymorphisms and 16 of them were integrated on the highly saturated genetic linkage map named “TOMATO EXPEN 2000”. The location of six unigenes was similar to the location of resistance gene analogs (RGAs), defense related ESTs and resistance QTLs previously reported, suggesting new possible candidates for these quantitative trait loci (QTL) regions. The candidate gene-markers may be used for future ISR or growth promotion assisted selection in tomato.

La mora de Castilla (Rubus glaucus Benth.) es un cultivo promisorio para Colombia por su potencial comercial internacional derivado de su alto valor nutricional y nutracéutico. De la colección nacional de mora se seleccionaron genotipos provenientes de ocho accesiones sobresalientes por sus características nutricionales y agronómicas, para ser distribuidos entre los productores. El objetivo de este trabajo fue evaluar la huella genómica y las relaciones genéticas de estos genotipos élite mediante el análisis de marcadores AFLP generados con el uso de tres combinaciones de cebadores. De un total de 179 loci amplificados por las tres combinaciones, 20% resultaron polimórficos. La combinación EAGG/MCTT fue la más informativa con un 32% de polimorfismo y un mayor poder de diferenciación. Los genotipos presentaron una alta similitud promedio (96%) y las accesiones San Antonio e ILS-1863 formaron grupos independientes con buen soporte estadístico en el análisis de agrupamiento. El resto de accesiones no formaron grupos discretos con buen soporte (<50%), probablemente debido a la homogeneidad genética entre ellas y/o al bajo poder de diferenciación de los marcadores. El estudio representa una primera aproximación al conocimiento de la huella genómica de estos genotipos de agricultor sobresalientes para protección, certificación de semilla y posterior apoyo a programas de mejoramiento.

The Andean blackberry (Rubus glaucus Benth.) is a promissory fruit crop for Colombia with potential to become an international commodity due to its high nutritional and nutraceutical value. Farmer genotypes from the national R. glaucus collection were selected from eight outstanding accessions according to their nutritional and agronomic value, for distribution among local producers. The goal of this work is to evaluate the genomic fingerprint by AFLP analysis of these elite genotypes using three primer combinations. From 179 total amplified loci produced by the three combinations, 20% resulted polymorphic. The EAGG/MCTT combination was the most informative with a 32% polymorphism and greater discrimination power. The genotypes tested showed a high average similarity (96%) and the accessions San Antonio and ILS-1863 formed independent groups with good statistical support in the clustering analysis. The remaining accessions did not form discrete groups with good support (<50%), probably due to genetic homogeneity among them and/or low resolving power of markers. This study is one of the first attempts to generate a genomic fingerprint of these farmer elite genotypes for protection, seed certification and future support to breeding programs.

The Lulo or naranjilla (Solanum quitoense Lam.) and the tree tomato or tamarillo (Solanum betaceum Cav. Sendt.) are both Andean tropical fruit species with high nutritional value and the potential for becoming premium products in local and export markets. Herein, we present a report on the genetic characterization of 62 accessions of lulos (n = 32) and tree tomatoes (n = 30) through the use of PCR-based markers developed from single-copy conserved orthologous genes (COSII) in other Solanaceae (Asterid) species. We successfully PCR amplified a set of these markers for lulos (34 out of 46 initially tested) and tree tomatoes (26 out of 41) for molecular studies. Six polymorphic COSII markers were found in lulo with a total of 47 alleles and five polymorphic markers in tree tomato with a total of 39 alleles in the two populations. Further genetic analyses indicated a high population structure (with F ST > 0.90), which may be a result of low migration between populations, adaptation to various niches and the number of markers evaluated. We propose COSII markers as sound tools for molecular studies, conservation and the breeding of these two fruit species.
