ABSTRACT Microalgae hold great potential for producing purified high-value products (e.g. pigments and polyunsaturated fatty acids) and represent a source of bioavailable nutrients in aquaculture feeds; however, its production is limited by the high costs of harvesting. Recently, microalgae autoflocculation has been considered a useful solution due to the easy recovery of cell aggregates and natural increase in pH that prevents cell lysis. This work evaluates some cultivation conditions that could contribute to autoflocculation, such as natural pH increase and precipitate formation over the productivity and flocculation of 16 microalgae consortia. Results showed a biomass production of 1.4 to 4.4 dry weight (g L-1) and high flocculation of 81 to ~100%, probably due to Ca3(PO4)2 formation. Moreover, the alkaline pH could have facilitated the assimilation of CO2 and explained the increase of inorganic carbon in the solution obtained. Likewise, results showed a marked descent in electrical conductivity (EC) in solution and high mineral content in the biomass (21.4-35.9%). Finally, this study suggests that the studied culture conditions facilitated microalgae harvest, and the resulting biomass could be a source of bioavailable dietary minerals. Therefore, some of these results might be addressed in future studies with individual microalgae species.

Abstract: Recent research has shown that the use of biological extracts, which contain a wide variety of nutrients and phytohormones are potentially useful in the production of plants of agricultural importance. Seaweed extracts are particularly rich in minerals and micronutrients necessary for the growth of microalgae. The effect of three commercial extracts of Ascophyllum nodosum as plant growth inducers (Stimplex®, Acadian Soils® and Liquid Seaweed Concentrate®), provided by Acadian Sea Plants Ltd., were evaluated in relation to the growth and production of biomass and carotenoids in Haematococcus pluvialis. The results were evaluated by one-way ANOVA and post-hoc analysis utilizing Tukey's test with a significance level of 95% (α= 0.05). The reference medium used was the Bold basal medium, to which 250 ppm of each extract was added. The Stimplex® extract had an increase of 22.79% in cell density and 17% in dry matter, higher than the control culture. The content of carotenoids and antioxidant capacity, respectively, with Stimplex® was 30.05% and 141.76% higher than the control culture.

RESUMO Objetivo: Avaliar o efeito anti-hiperglicêmico do extrato hidroalcoólico de Plathymenia reticulata, alterações no peso, lipídeos e efeito sobre o pâncreas. Métodos: O diabetes foi induzido pela administração de estreptozotocina 65mg/kg, em 75 ratos Wistar adultos machos, divididos em 8 grupos diferentes: ratos diabéticos e controle + água, ratos diabéticos e controle + 100mg/kg ou 200mg/kg de extrato, ratos diabéticos e controle + gliburida. O tratamento foi realizado por gavagem (oral) por 30 dias. Níveis de glicose e peso foram verificados semanalmente. Os animais foram sacrificados, e amostras de lipídeos e do pâncreas foram analisadas. A análise estatística incluiu ANOVA, post-hoc Tukey-Kramer, teste t de Student pareado e teste do χ2, com nível de significância de 5%. Resultados: O extrato 100mg/kg promoveu redução nos níveis de glicose sanguínea em ratos diabéticos na segunda, terceira (198,71±65,27 versus 428,00±15,25) e quarta semanas (253,29±47,37 versus 443,22±42,72), perda de peso (13,22±5,70 versus 109,60±9,95) e diminuição do colesterol (58,75±3,13 versus 80,11±4,01) em ratos controle. Com extrato de 200mg/kg, houve redução dos níveis de glicose na quarta semana, nos ratos diabéticos; de peso na segunda, terceira e quarta semanas, nos ratos controle; e de colesterol nos animais diabéticos e controle. Ocorreram hiperplasia de ilhotas (p=0,005) e dilatação dos ductos pancreáticos (p=0,047) em ratos diabéticos e controles. Conclusão: O extrato de Plathymenia reduziu os níveis de glicose em ratos diabéticos e de peso em ratos controle, além de ter promovido hiperplasia de ilhotas pancreáticas em diabéticos e controles.

ABSTRACT Objective: To investigate the anti-hyperglycemic effects of Plathymenia reticulata hydroalcoholic extract and related changes in body weight, lipid profile and the pancreas. Methods: Diabetes was induced in 75 adult male Wistar rats via oral gavage of 65mg/Kg of streptozotocin. Rats were allocated to one of 8 groups, as follows: diabetic and control rats treated with water, diabetic and control rats treated with 100mg/kg or 200mg/kg of plant extract, and diabetic and control rats treated with glyburide. Treatment consisted of oral gavage for 30 days. Blood glucose levels and body weight were measured weekly. Animals were sacrificed and lipid profile and pancreatic tissue samples analyzed. Statistical analysis consisted of ANOVA, post-hoc Tukey-Kramer, paired Student's t and χ2 tests; the level of significance was set at 5%. Results: Extract gavage at 100mg/kg led to a decrease in blood glucose levels in diabetic rats in the second, third (198.71±65.27 versus 428.00±15.25) and fourth weeks (253.29±47.37 versus 443.22±42.72), body weight loss (13.22±5.70 versus 109.60±9.95) and lower cholesterol levels (58.75±3.13 versus 80.11±4.01) in control rats. Extract gavage at 200mg/Kg led to a decrease in glucose levels on the fourth week in diabetic rats, body weight loss in the second, third and fourth weeks in control rats, and lower cholesterol levels in diabetic and control rats. Islet hyperplasia (p=0.005) and pancreatic duct dilation (p=0.047) were observed in diabetic and control rats. Conclusion: Plathymenia extract reduced blood glucose levels in diabetic rats, and body weight in control rats, and promoted pancreatic islet hyperplasia in diabetic and control rats.

This review discusses microalgae assimilation of C, N and P as affected by pH-dependent factors (solubility, ionization state and bioavailability) and CO2 enrichment in tertiary water treatment during microalgae biomass production. This study indicates the importance of monitoring critical conditions such as: 1) pH and inorganic carbon species interaction; 2) P bioavailability; 3) nitrogen removal through NH3 volatilization and 4) NH3 toxicity. Also, optimal CO2 enriched air considerations, such as: 1) combining intermittent and variable CO2 enrichment to avoid losses into the atmosphere; 2) preventing microalgae growth inhibition due to H2CO3 formation and pH acidification at early stages of the culture and 3) maintenance of optimal pH (≈ 8.5) through the buffer system (CO2 / HCO3_). Finally, we suggest that biological technologies based on microalgae culture to remove excessive nutrient loads and compliance with environmental laws from water, represent a viable alternative attached to recycling and revaluation of resources through the generation and potential uses of biomass (e.g., fertilizer, pigments and antioxidants).

Esta revisión analiza la asimilación de C, N y P por microalgas y la influencia de los factores dependientes del pH (solubilidad, estado de ionización y biodisponibilidad), así como la adición de CO2 y su interacción en el tratamiento terciario de aguas municipales durante la producción de biomasa de microalgas. Este estudio destaca la importancia de monitorizar condiciones críticas como: 1) la interacción entre el pH y las especies de carbono inorgánico; 2) la biodisponibilidad de P; 3) reducción de N por formación de NH3 y 4) toxicidad de NH3. Por otra parte, para el uso óptimo de aire enriquecido con CO2 se recomienda considerar: 1) el suministro intermitente y variable de CO2 para evitar pérdidas a la atmósfera; 2) optimizar el consumo de CO2 evitando la acidificación del pH por formación de H2CO3 e inhibición de las microalgas en etapas tempranas de cultivo; y 3) conservación de pH óptimo (≈ 8,5) a través del sistema de amortiguamiento CO2 / HCO3_. Finalmente, se sugiere que las tecnologías biológicas basadas en el cultivo de microalgas para la eliminación de nutrientes y cumplimiento de normativas ambientales, representan una alternativa viable, que une el reciclaje y revalorización de recursos a través de la generación y usos potenciales de la biomasa (e.g., fertilizantes, pigmentos y antioxidantes).

RESUMO: O vírus Vaccinia (VACV) é o agente etiológico da vaccínia bovina, uma doença zoonótica re-emergente e de importância sanitária e econômica para rebanhos leiteiros. O VACV também pode ocasionalmente infectar outras espécies, incluindo equinos. Nesse sentido, um surto de VACV em equinos foi descrito em Pelotas, RS, em 2008, no qual uma coinfecção com dois isolados de VACV (denominados Pelotas 1 [P1V] e Pelotas 2 [P2V]) foi detectada. Considerando a rara ocorrência da infecção pelo VACV em equinos, o objetivo deste estudo foi investigar a susceptibilidade e a patogenia experimental da infecção pelo VACV nessa espécie. Para isso, seis equinos adultos foram inoculados com P1V ou P2V (106,3DICC50/ml) após escarificação na superfície nasolabial e monitorados para os aspectos virológicos, clínicos e patológicos por 28 dias. Quatro animais inoculados (4/6) desenvolveram lesões macroscópicas leves nos locais da inoculação, consistentes com aquelas causadas por VACV, caracterizadas por úlceras e crostas focais restritas à área inoculada, entre os dias 2 e 8 pós-inoculação (pi). Microscopicamente, havia acantose, degeneração hidrópica do estrato espinhoso e necrose da epiderme, com bactérias intralesionais. Infiltração moderada de neutrófilos, macrófagos e linfócitos foi observada na derme superficial. Inclusões intracitoplasmáticas eosinofílicas foram infrequentemente observadas nos queratinócitos degenerados adjacentes às áreas necróticas. Excreção viral foi detectada por PCR e isolamento viral das lesões de um equino inoculado com o P2V, entre os dias 4 e 8pi. Não foi detectada produção de anticorpos específicos para o VACV em nenhum animal inoculado. Em resumo, a inoculação de cavalos com os isolados P1V e P2V resultou em um baixo nível de replicação viral e em baixa frequência, resultando em lesões cutâneas mais discretas em comparação com outras espécies susceptíveis ao VACV. Apesar dos equinos serem susceptíveis à infecção pelo VACV, possivelmente apresentam baixo potencial de manutenção e de transmissão do vírus para outras espécies.

ABSTRACT: Vaccinia virus (VACV) is the etiologic agent of bovine vaccinia, an emerging zoonotic disease with potential health issues for dairy herds and humans. VACV may occasionally infect other species, including horses. In this sense, an outbreak of VACV disease in horses was described in Pelotas, RS, in 2008, where a co-infection with two VACV strains (named Pelotas Virus 1 [P1V] and Pelotas Virus 2 [P2V]) was detected. Considering the rare occurrence of VACV infection in horses, the objective of this study was to investigate the susceptibility and pathogenesis of VACV infection in this species. Six adult horses were inoculated with VACV P1V or P2V (106.3TCID50/ml) through scarification of the nasolabial surface and monitored for virological and clinical aspects during 28 days. Four inoculated horses (4/6) developed mild lesions in the site of inoculation. Ulcers and scabs restricted to inoculated areas were observed between days 2 and 8 post-inoculation (pi). Microscopically there were acanthosis, ballooning degeneration of the stratum spinosum, necrosis and loss of the epidermis. Infiltration of neutrophils, macrophages and lymphocytes were observed in the dermis. Intracytoplasmic eosinophilic inclusions were infrequently observed in degenerate keratinocytes from adjacent necrotic areas. Virus shedding was detected between days 4 and 8 pi by PCR and virus isolation (infectious virus) from the lesions of one horse inoculated with P2V. No neutralizing antibodies were detected in inoculated animals at day 28 pi. In summary, inoculation of horses with VACV P1V and P2V isolates resulted in a low level of replication and at low frequency, with mild cutaneous lesions, when compared with the course of infection of other susceptible species to VACV. Therefore, horses possibly have a low potential for viral maintenance and transmission to other species, albeit being susceptible to VACV infection.

Este trabalho propõe a criação de um índice de estado trófico baseado na estimativa da concentração de clorofila através de sensoriamento remoto. Para atingir este objetivo, medidas do fator de reflectância bidirecional (BRF) adquiridas no reservatório de Ibitinga foram reamostradas para serem combinadas com bandas espectrais simuladas do Landsat/TM (TM_sim) e foram usadas para o cálculo da correlação linear com medidas de concentração de clorofila-a. A simulação de Monte Carlo foi aplicada para selecionar o modelo mais adequado para relacionar concentração de clorofila e reflectância simulada do TM/Landsat. A razão TM4_sim/TM3_sim forneceu o melhor modelo, com um valor de 0.78 para R². O modelo foi então invertido para criar uma tabela (LUT) com as relações entre os intervalos das razões TM4_sim/TM3_sim e as classes de estado trófico, abrangendo todo o intervalo de valores medidos no reservatório. Imagens do Landsat TM, corrigidas atmosfericamente e convertidas para reflectância de superfície, foram usadas para gerar uma imagem para a razão TM4/TM3. A distribuição de frequência da imagem da razão englobou todo o intervalo da razão TM4_sim/TM3_sim, indicando concordância entre os dados orbitais e os dados medidos in situ e corrobora o uso de dados de satélite para mapear a distribuição do estado trófico no reservatório. Com base nestas observações, a LUT foi aplicada à imagem razão Landsat/TM para mapear a distribuição espacial das classes de estado trófico no Reservatório de Ibitinga. Apesar da seleção estocástica da razão TM4_sim/TM3_sim como melhor variável de entrada para modelagem da concentração de clorofila-a, esta escolha foi baseada em fatores físicos: altas concentrações de fitoplâncton aumentam a reflectância na região do infra-vermelho próximo (TM4) e diminuem a reflectância na região espectral do vermelho (TM3). A razão entre bandas, portanto, realça a relação entre a concentração de clorofila-a e a reflectância medida por sensoriamento remoto.

This work proposes a trophic state index based on the remote sensing retrieval of chlorophyll-α concentration. For that, in situ Bidirectional Reflectance Factor (BRF) data acquired in the Ibitinga reservoir were resampled to match Landsat/TM spectral simulated bands (TM_sim bands) and used to run linear correlation with concurrent measurements of chlorophyll-α concentration. Monte Carlo simulation was then applied to select the most suitable model relating chlorophyll-α concentration and simulated TM/Landsat reflectance. TM4_sim/TM3_sim ratio provided the best model with a R² value of 0.78. The model was then inverted to create a look-up-table (LUT) relating TM4_sim/TM3_sim ratio intervals to chlorophyll-α concentration trophic state classes covering the entire range measured in the reservoir. Atmospheric corrected Landsat TM images converted to surface reflectance were then used to generate a TM4/TM3 ratio image. The ratio image frequency distribution encompassed the range of TM4_sim/TM3_sim ratio indicating agreement between in situ and satellite data and supporting the use of satellite data to map chlorophyll-α concentration trophic state distribution in the reservoir. Based on that, the LUT was applied to a Landsat/TM ratio image to map the spatial distribution of chlorophyll-α trophic state classes in Ibitinga reservoir. Despite the stochastic selection of TM4_sim/TM3_sim ratio as the best input variable for modeling the chlorophyll-α concentration, it has a physical basis: high concentration of phytoplankton increases the reflectance in the near-infrared (TM4) and decreases the reflectance in the red (TM3). The band ratio, therefore, enhances the relationship between chlorophyll-α concentration and remotely sensed reflectance.

O objetivo deste trabalho foi avaliar o crescimento e a esporulação de dois isolados (JAB 02 e JAB 45) de Verticillium lecanii (Zimm.) Viégas, submetidos a diferentes valores de pH (4, 5, 6, 7, 8, 9) inicial do meio de cultivo sólido, de temperatura (19, 22, 25, 28 e 31ºC) e fotoperíodo (0, 12 e 24 horas). Após 20 dias de cultivo, não se observou efeito do pH inicial do meio no crescimento e esporulação dos isolados, exceto no pH 4,0 que reduziu o crescimento. A ausência de iluminação proporcionou a melhor condição para o crescimento de JAB 02 (32,70 mm) e JAB 45 (32,67 mm). Não houve efeito do fotoperíodo na produção de esporos. As melhores temperaturas para o crescimento de JAB 02 foram 19, 22 e 25ºC e para JAB 45 foram 19 e 25ºC. Este último isolado apresentou melhor esporulação a 19 e 22ºC, mas a temperatura não influenciou a esporulação de JAB 02. Não houve desenvolvimento dos isolados a 31ºC.

This research aimed to evaluate the growth and sporulation of two Verticillium lecanii (Zimm.) Viégas isolates cultivated on solid medium with different initial pH values (4, 5, 6, 7, 8 and 9), exposed to different temperatures (19, 22, 25, 28 and 31ºC) and photoperiods (0, 12 and 24 hours). There was no substantial effect of the initial pH values on growth and sporulation of both isolates twenty days after inoculation, except at pH 4.0 in which the growth was significantly reduced. Absence of light promoted the best growth of JAB 02 (32.70 mm) and JAB 45 (32.67 mm) isolates but there was no effect of photoperiod on sporulation. The best temperatures for growth of JAB 02 were 19, 22 and 25ºC and for JAB 45, 19 and 25ºC. The latter isolate sporulated most at 19 and 22ºC and there was no effect of temperature on sporulation of JAB 02. None of the isolates developed at 31ºC.

O objetivo deste trabalho foi determinar as melhores condições de crescimento e esporulação de dois isolados de Verticillium lecanii (Zimm.) Viégas, cultivados em vários meios de cultura, fontes de C, fontes orgânicas e inorgânicas de N e relações C:N. Após 20 dias de cultivo, BDA e Meio Completo mostraram ser os melhores meios de cultura, e a lactose e o amido foram as fontes de C que mais favoreceram o crescimento dos isolados JAB 02 e JAB 45, respectivamente; baixa esporulação foi apresentada por JAB 02, enquanto JAB 45 obteve boa esporulação em glicose e maltose. JAB 02 desenvolveu melhor quando a fonte orgânica de N era casitona; JAB 45 obteve bom desenvolvimento em presença de casitona e peptona, mas houve pouca esporulação em presença de triptona e caseína hidrolisada. As fontes inorgânicas de N que favoreceram o melhor desenvolvimento dos isolados foram (NH4)2HPO4 e NaNO3, porém, JAB 02 apresentou baixa esporulação. Entre as relações C:N analisadas, JAB 02 obteve melhor crescimento na relação 60:1 e JAB 45 nas relações 5:1, 10:1 e 20:1; baixa esporulação foi obtida por JAB 02 nas relações testadas, mas não se verificou efeito da relação C:N do meio sobre a esporulação dos isolados.

The objective of this work was to determine the best growth and sporulation conditions of two Verticillium lecanii (Zimm.) Viégas, isolates cultivated in different culture medium, C sources, organic and inorganic N sources and C:N ratio. Twenty days after inoculation, PDA and Complete Medium provided the best development, and lactose and starch were the best C sources to promote the growth of JAB 02 and JAB 45 isolates, respectively; JAB 02 showed low sporulation while JAB 45 sporulated well on glucose and maltose. JAB 02 isolate developed well when the organic N source was casitone; JAB 45 obtained good development when the tested sources were casitone and peptone, although a low sporulation occurred in presence of tryptone and hydrolysed casein. The inorganic N sources that favoured the best development of isolates were (NH4)2HPO4 and NaNO3, although JAB 02 isolate showed low sporulation. In the C:N ratio analysed, JAB 02 isolate developed better on the 60:1 ratio, and the JAB 45 isolate on the 5:1, 10:1 and 20:1 ratio. The sporulation of JAB 02 isolate was low on all the ratios tested, but there was no effect of the C:N ratio on the sporulation of both isolates.
