RESUMO: A adubação com Selênio (Se) em pastagem para a biofortificação de alimentos de origem animal tem sido realizada em solos com baixo Se. O objetivo foi avaliar o aumento no conteúdo de Se na biomassa de Brachiaria spp. com aplicação de ureia revestida com Se. O experimento foi conduzido em casa de vegetação em Latossolo Amarelo. O delineamento foi em blocos casualizados com quatro repetições em esquema fatorial com dois cortes e seis doses de Se (0, 10, 20, 40, 80 e 160gha-1). A Brachiaria brizantha demonstrou ausência de mudanças em altas e perigosas doses de Se sem alterar a atividade da enzima antioxidante glutationa peroxidase. Com isso, concluiu-se que seu enriquecimento com ureia revestida é obtida com 34,5gha-1 de Se.

ABSTRACT: Selenium (Se) fertilization in grazing to biofortification of animal products have been carried out in low Se soils. The objective of this study was to increase the Se content in the biomass of Brachiaria spp.with urea coated with Se. The experiment was performed in a typical Hapludox soil under greenhouse conditions. A completely randomized block design with four replicates in a factorial structure with two cuts and six Se doses (0, 10, 20, 40, 80, and 160gha-1) was used. The Brachiaria brizantha demonstrated the absence of changes in higher harmful doses without modifying the activity of glutathione peroxidase enzyme. We concluded that enrichment of Brachiaria brizantha with coated urea is obtained with 34.5gha-1of Se.

'Golden' papayas at maturity stage 1 (15% yellow skin) were chosen to study selected oxidative processes, the activity of antioxidant enzymes and lipid peroxidation in storage at 22°C, during the ripening of the fruit. An increase in ethylene production was observed on the second day of storage and it was followed by an increase in respiration. An increased activity of catalase, glutathione reductase and ascorbate peroxidase was observed concurrently or soon after this increase in ethylene production and respiration. The increased activity of these enzymes near the peaks of ethylene production and respiration is related to the production of oxidants accompanying the onset of ripening. On the fourth day of storage, there was an increased lipid peroxidation and decreased activities of catalase, glutathione reductase and superoxide dismutase. Lipid peroxidation induces the increase of antioxidant enzymes, which can be verified by further increases in the activities of catalase, glutathione reductase, superoxide dismutase and ascorbate peroxidase. Unlike the other antioxidant enzymes, the ascorbate peroxidase activity in the pulp increased continuously during ripening, suggesting its important role in combating reactive oxygen species during papaya ripening. With regard to physical-chemical characteristics, the soluble solids did not vary significantly, the acidity and ascorbic acid contents increased, and hue angle and firmness decreased during storage. The results revealed that there was variation in the activity of antioxidant enzymes, with peaks of lipid peroxidation during the ripening of 'Golden' papaya. These results provide a basis for future research, especially with regard to the relationships among the climacteric stage, the activation of antioxidant enzymes and the role of ascorbate peroxidase in papaya ripening.

Foram aplicados tratamentos térmicos em laranja "Valência", tangor "Murcott" e lima ácida "Tahiti" armazenadas em baixa temperatura e avaliados os efeitos na redução de injúrias pelo frio e sobre outras características bioquímicas. Os seguintes tratamentos foram aplicados: T1: frutas armazenadas a 1°C (Controle); T2: aquecimento rápido das frutas em água quente a 53°C, durante 3 minutos, sendo, em seguida, armazenadas a 1°C; T3: aquecimento lento das frutas em câmara regulada para 37°C, por 2 dias, com posterior armazenamento a 1°C; e T4: aquecimento intermitente em ciclos de 6 dias a 1°C + 1 dia a 25°C. As frutas foram armazenadas durante 90 dias a 1°C e 90-95% de Umidade Relativa (UR), sendo avaliadas a cada 15 dias. Além da incidência dos danos, foram avaliadas também as atividades das enzimas antioxidativas (catalase, glutationa redutase e ascorbato peroxidase). A lima ácida "Tahiti" e o tangor "Murcott" suportaram até 90 dias de armazenamento a 1°C com aquecimento intermitente, não apresentando danos pelo frio. No tratamento controle (armazenamento contínuo a 1°C), os danos pelo frio surgiram aos 30 dias de armazenamento para a lima "Tahiti" e aos 45 dias para o tangor "Murcott". Em laranjas "Valência", as injúrias pelo frio surgiram aos 45 dias de armazenamento, sendo significativamente menores no condicionamento térmico. Os efeitos dos tratamentos térmicos no aumento da resistência das frutas ao frio podem estar relacionados com a atividade das enzimas antioxidativas. Para a laranja "Valência", os tratamentos térmicos, aplicados na forma de condicionamento térmico ou aquecimento intermitente, reduzem as injúrias pelo frio e podem prolongar a conservação das frutas a 1°C e 90-95% UR. Para tangor "Murcott" e lima "Tahiti", o aquecimento intermitente é o tratamento mais eficiente para a redução de injúrias pelo frio e para o aumento na capacidade de armazenamento.

In the present study thermal treatments on,'Valência' orange, 'Murcott' tangor and 'Tahiti' (temperature conditioning and intermittent warming) were applied lime. The following treatments were applied: T1: Fruits stored at 1°C (Controls); T2: Fast heating of the fruits in hot water at 53°C for 3 minutes and stored at 1°C; T3: Slow heating of the fruits in camera regulated at 37°C for 2 days, with subsequent storage at 1°C; and T4: Intermittent heating in cycles of 6 days at 1°C + 1 day at 25°C. Fruits were stored under low temperature (1°C) and 90-95% relative humidity (RH) during 90 days. Chilling injury incidence and changes in physical-chemical characteristics were evaluated every 15 days. Activity of catalase, glutathione reductase and ascorbate peroxidase was also determined. Intermittent warming was more efficient than temperature conditioning to reduce chilling injury. 'Tahiti' lime and 'Murcott' tangor supported up to 90 days of intermittent warming stored at 1°C, while control fruits showed chilling injury after 30 days ('Tahiti' lime) and 45 days ('Murcott' tangor). 'Valência' orange was affected by chilling injury after 45 days of cold storage, and temperature conditioning has caused lower incidence of this disorder. The effect of thermal treatments can be related to the activity of antioxidant enzymes. For ´Valência" orange, thermal treatments, applied as thermal conditioning or intermittent heating, reduce injuries caused by cold and can lengthen the conservation of fruits at 1°C and 90-95% RH. The intermittent heating is the most efficient treatment to reduce injuries caused by cold and increase at storage capacity of the 'Murcott' tangor and 'Tahiti' lime.

O objetivo da presente pesquisa foi analisar os graus de polimorfismos protéicos entre isolados de C. albicans provenientes de diversos sítios anatômicos de quarenta e dois pacientes clínicos, através do emprego da eletroforese em gel de poliacrilamida (SDS-PAGE) e análise numérica, a fim de se identificar subespécies e suas similaridades nos diversos nichos infecciosos. Culturas celulares foram desenvolvidas em meio YEPD, coletadas por centrifugação e lavadas com solução salina gelada. As proteínas celulares totais, foram extraídas por rompimento celular, usando pérolas de vidro e submetidas à técnica de SDS-PAGE. Após a eletroforese, as bandas de proteínas foram coradas com coomassie-blue e analisadas pelo conjunto de programas estatístico NTSYS-pc versão 1,70. Matrizes de similaridade e dendrogramas foram gerados pela aplicação do coeficiente de similaridade simple-matching e do algoritmo UPGMA, respectivamente. Os resultados obtidos revelaram vários subtipos de C. albicans e seus graus de similaridade (80% a 100%). Tais dados permitiram demonstrar que, certos pacientes podem estar infectados com dois ou mais subtipos de C. albicans em determinados sítios anatômicos (i.e. apenas na cavidade oral de pacientes imunocomprometidos, sangue ou secreção traqueal), ou ainda, dois ou mais pacientes podem estar infectados em sítios anatômicos idênticos (i.e. apenas em lavagem brônquica, urina, cavidade oral, secreção traqueal, secreção vaginal ou saliva saudável) com um mesmo subtipo de C. albicans. No entanto, dois ou mais pacientes também podem apresentar infecções em sítios correspondentes (i.e. apenas na cavidade oral de pacientes imunocomprometidos, sangue, secreção orofaríngea, cavidade oral, secreção traqueal, secreção vaginal e saliva saudável) por diferentes subtipos de C. albicans. Além disso, dois ou mais pacientes também podem estar infectados com subtipos idênticos ou não de C. albicans em diferentes sítios anatômicos (i.e.1. idênticos subtipos na secreção vaginal, secreção traqueal e urina; secreção abdominal e escarro; drenagem e cavidade oral; cateter e saliva saudável - i.e.2. diferentes subtipos em lavagem brônquica, secreção orofaríngea, secreção pulmonar, cavidade oral de pacientes imunocomprometidos e sangue). Dados complementares envolvendo amostras de C. albicans isoladas de vários sítios anatômicos de pacientes imunocompetentes ou imunocomprometidos (antes, durante e após terapias específicas) e seus familiares ou trabalhadores hospitalares, deverão ser obtidos a fim de se estabelecer as possíveis fontes de colonização por esses microrganismos. De modo geral, os perfis de proteínas totais obtidos por SDS-PAGE associados com análise numérica computadorizada, permitem a obtenção de critérios adicionais para os estudos epidemiológicos e taxonômicos de C. albicans.

The aim of the present research was to evaluate the protein polymorphism degrees among forty-eight C. albicans isolates from fourteen anatomical sites of clinical patients by polyacrylamide gel electrophoresis (SDS-PAGE) and numerical analyzes, in order to identify subspecies and their similarities in some infectious niches. Cell cultures were grown in YEPD medium, collected by centrifugation, and washed in cold saline solution. The whole-cell proteins were extracted by cell disruption using glass beads and submitted to SDS-PAGE technique. After electrophoresis, the protein bands were stained with coomassie-blue and analyzed by statistics package NTSYS-pc version 1.70 software. Similarity matrixes and dendrograms were generated by application of similarity coefficient of simple matching and UPGMA algorithm, respectively. The results obtained showed several C. albicans subtypes and their similarity degrees (80% to 100%). Such data showed that same patients may be infected by two or more C. albicans subtypes in certain anatomical sites (i.e. only in oral cavity of immunocompromised patients, blood, or tracheal secretion), or yet, two or more patients can be infected in identical anatomical sites (i.e. bronchial washing, urine, oral cavity, tracheal secretion, vaginal secretion, and healthy saliva) with a same C. albicans subtype. However, two or more patients also can show infections in corresponding sites (i.e. oral cavity of immunocompromised patients, blood, oropharyngeal secretion, oral cavity, tracheal secretion, vaginal secretion, and healthy saliva) by different C. albicans subtypes. Besides, two or more patients also can be infected with identical or different C. albicans subtypes in different anatomical sites (i.e.1. identical subtypes in vaginal secretion, tracheal secretion, and urine; abdominal secretion and spittle; drainage and oral cavity; catheter and healthy saliva - i.e.2. subtypes different in bronchial washing, oropharyngeal secretion, pulmonary secretion, oral cavity of immunocompromised patients, and blood). Complementary studies involving C. albicans sample isolated from several anatomical sites of immunocompetent or immunocompromised patients (before, during and after specifics therapies) and their families or hospital workers must be done in order to establish the sources of C. albicans colonization. The whole-cell proteins profile performed by SDS-PAGE associated with computer-assisted numerical analysis may provide preliminary criteria for taxonomic and epidemiological studies of such microorganisms.
