Resumo A conservação de sementes recalcitrantes de seringueira é pouco duradoura, quando comparada com espécies que possuem sementes ortodoxas. Dessa forma, a preservação da espécie ocorre especialmente em campo na forma da planta inteira, o que requer muito espaço físico. A biotecnologia, por meio da técnica da criopreservação pode ser uma ferramenta auxiliadora na manutenção da viabilidade das sementes durante o seu armazenamento. Assim, o objetivo deste trabalho foi avaliar a viabilidade de embriões zigóticos de seringueira, contendo o tecido de reserva, submetidos a distintas formas de vitrificação, seguido da conservação por meio da criopreservação. Para realização da pesquisa foram utilizadas sementes de seringueira, produzidas no estado de São Paulo. Primeiramente, foram estudadas distintas formas de vitrificação dos embriões zigóticos contendo o tecido de reserva. Os embriões foram avaliados logo após a vitrificação e aos sete dias de conservação em nitrogênio líquido a uma temperatura de -196ºC. As avaliações realizadas foram: determinação do teor de água, viabilidade dos embriões pelo teste de tetrazólio, germinação in vitro e condutividade elétrica. Os resultados foram analisados em delineamento inteiramente casualizados, em esquema fatorial 2 x 5, sendo o primeiro fator época de avaliação (antes e após a criopreservação) e o segundo fator, as formas de vitrificação. Conclui-se que é necessário testar um maior período de exposição dos embriões nas soluções de vitrificação, especialmente nas soluções que apresentaram melhores resultados de viabilidade e germinação (soluções: • 50% de glicerol; • 30% (p/v) glicerol, 15% (p/v) de etilenoglicol, 15% (p/v) dimetilsulfóxido (DMSO) e 0,4 M de sacarose; e • 2 M glicerol seguidos de PVS). Adicionalmente, deve ser analisado um maior nível de desidratação dos embriões, o qual a espécie possa suportar, para que possam ser expostos a criopreservação.

Abstract The conservation of recalcitrant rubber seeds is not very durable when compared to species that have orthodox seeds. Thus, the preservation of the species occurs especially in the field in the form of the entire plant, which requires a lot of physical space. Biotechnology, through the technique of cryopreservation, can be an auxiliary tool in maintaining the viability of seeds during their storage. Thus, the objective of this work was to evaluate the viability of rubber tree zygotic embryos, containing the reserve tissue, submitted to different forms of vitrification, followed by conservation through cryopreservation. To carry out the research, rubber seeds, produced in the state of São Paulo, were used. First, different forms of vitrification of the zygotic embryos containing the reserve tissue were studied. The embryos were evaluated immediately after vitrification and after seven days of storage in liquid nitrogen at a temperature of -196 ºC. The evaluations carried out were: determination of the water content, viability of the embryos by the tetrazolium test, in vitro germination and electrical conductivity. The results were analyzed in a completely randomized design, in a 2x5 factorial scheme, the first factor being the time of assessment (before and after cryopreservation) and the second factor, the forms of vitrification. It is concluded that it is necessary to test a longer period of exposure of embryos in the vitrification solutions, especially in the solutions that presented better results of viability and germination (solutions: • 50% glycerol; • 30% (w/v) glycerol, 15% (w/v) ethylene glycol, 15% (w/v) dimethyl sulfoxide (DMSO) and 0.4 M sucrose; and • 2 M glycerol followed by PVS). In addition, a higher level of dehydration of the embryos, which the species can support, must be analyzed, so that they can be exposed to cryopreservation.

RESUMO O uso de micropropagação de pequenos frutos como a amoreira-preta tem sido empregado devido à necessidade de obtenção de plantas com alta qualidade fitossanitária. A tecnologia de biorreatores tem sido usada para melhorar a eficiência na produção de mudas. Assim, o objetivo deste trabalho foi avaliar a melhor quantidade de meio de cultura e a concentração ideal de sacarose para micropropagação de amoreira-preta em um biorreator de imersão temporária. Brotações in vitro de amoreira-preta foram segmentadas contendo duas gemas com internódio (1.0cm) e colocadas em meio MS suplementado com inositol (0.1 g L-1), BAP (1 mg L-1), sacarose (10, 20, 30 ou 40 g L-1) e volume de meio (150, 175 e 200 mL). Foram avaliados aos 56 dias, o comprimento total, número de folhas, número de brotações e brotações hiper-hídricas. O volume de 175 mL e a concentração de 20 g L-1 apresentaram os melhores resultados e são os mais recomendados para o desenvolvimento e propagação de amora-preta em um sistema de biorreatores.

ABSTRACT Micropropagation of small fruits such as blackberry has been employed due to the need to obtain plants with high phytosanitary quality. Bioreactor technology has been used to improve efficiency in seedling production. Thus, the objective of this work was to evaluate the best culture medium volume and sucrose concentration for blackberry micropropagation in a temporary immersion bioreactor. In vitro blackberry shoots were segmented containing two buds and an internode (1.0 cm) and placed into MS medium supplemented with inositol (0.1 g L-1), BAP (1 mg L-1) and sucrose (10, 20, 30 or 40 g L-1) at different medium volumes (150, 175 and 200 mL). The total length, number of leaves, number of shoots, and number of hyperhydric shoots were evaluated 56 days after start of the project. For blackberry development and propagation in a bioreactor system, the best results were shown at a medium volume of 175 ml and a sucrose concentration of 20 g L-1.

O raleio promove equilíbrio entre o crescimento vegetativo e produtivo, aumento no tamanho dos frutos e evita alternância da produção. O objetivo deste trabalho foi avaliar o efeito de diferentes concentrações de ethephon no raleio químico de frutos das ameixeiras cultivares 'Irati' e 'Reubennel', na região de Arapoti-PR. Os tratamentos foram cinco concentrações de ethephon (0, 60, 80, 100, 120 μL L-1) para cultivar 'Irati' e quatro concentrações (0, 60, 80, 120 μL L-1) para a cultivar 'Reubennel', aplicadas quando os frutos apresentavam 5 mm de diâmetro. Foram avaliados: produção por planta, massa dos frutos, calibre, teor de sólidos solúveis, firmeza, acidez titulável, pH e coloração dos frutos. Na concentração de 120 μL L-1 de ethephon a cultivar 'Irati' produziu 20 kg planta-1 e a cultivar 'Reubennel' 56 kg planta-1, apresentando quantidade satisfatória de frutos com calibre III e IV. Não houve diferença significativa entre os tratamentos para as variáveis firmeza, sólidos solúveis, pH, acidez e coloração, avaliadas na pós-colheita para as duas cultivares de ameixeira

Thinning of plum trees promotes balance between vegetative growth and production, increases the fruit size and avoids alternation of production. The objective of this study was to evaluate the effect of different ethephon concentrations as chemical thinning of plum cv. 'Irati' and cv. 'Reubennel', in Arapoti-PR. Five treatments with ethephon were evaluated at dosages (0, 60, 80, 100, 120 μL L-1) in cv. 'Irati' and four treatments at dosages; 0; 60; 80; 120 μL L-1 in cv. 'Reubennel', all applied when fruits diameter was 5 mm followed by measurements plant yield, weight, size, soluble solids, firmness, titratable acidity, pH and color of fruits. At a concentration of 120 μL L-1 ethephon, cv. 'Irati' produced 20 kg plant-1 and the cv. 'Reubennel' 56 kg plant-1 presenting satisfactory quantity of fruit size III and IV. There was no significant difference between treatments for variable firmness, soluble solids, titratable acidity, pH and color of fruits evaluated in the post-harvest for the two plum cultivars

A resistência genética às doenças é um progresso na produção de todas as culturas e fontes de resistência devem ser reportadas, auxiliando os programas de melhoramento. O presente trabalho objetivou avaliar a resistência de genótipos de nectarina à ferrugem da folha e ao chumbinho, assim como, correlacionar a intensidade das doenças com a desfolha prematura. Foram avaliados três genótipos de nectarina em delineamento de blocos ao acaso com dez repeticões. Com base nos resultados obtidos, o genótipo Bruna mostrou-se resistente ao chumbinho e suscetível à ferrugem, ao passo que o genótipo Sunripe foi resistente à ferrugem; FLA 9-15N foi moderadamente suscetível à ferrugem. Houve correlação apenas entre severidade da ferrugem e desfolha.

Genetic resistance to disease can be seen as progress in the production of all crops, and sources of resistance have to be reported, thus helping improvement programs. This study aimed to evaluate the resistance of nectarine genotypes to leaf rust and shot hole, as well as to correlate the severity of the diseases with premature defoliation. Three genotypes of the nectarine were evaluated in a randomized block design with ten replications. Based on the results, the genotype Bruna proved resistant to shot hole and susceptible to leaf rust, while the genotype Sunripe was resistant to leaf rust. FLA 9-15N was moderately susceptible to leaf rust. There was correlation between leaf-rust severity and defoliation only.

In this work, glucoamylase was produced by Aspergillus niger in solid-state fermentation. The enzyme was partially purified by ammonium sulphate precipitation and ion exchange and gel filtration chromatographies. Its molecular mass was estimated as 118.17 kDa by electrophoresis. The partially purified enzyme had an optimum pH range of 4.5-5.0 and an optimum temperature of 60 °C, with average activity 152.85 U mL-1. Thermal and pH stability assays with the crude extract showed that more than 60 % of the activity remained at pH 4.6 and 60 °C, even after an exposition to these conditions longer than 24 h. Yet, after purification, the enzyme was stable at these for at least 4 h, which indicated that its purification for use in starch saccharification was inadvisable. K M and Vmax were 0.34 mg mL-1 and 160.22 U mL-1, respectively.

A propagação vegetativa da videira favorece infecções virais múltiplas, com expressão diferencial de sintomas em função da combinação da cultivar ou espécie da hospedeira com a espécie viral. Os objetivos deste trabalho foram detectar e identificar as espécies virais presentes em duas espécies/cultivares de videira: uma sintomática e outra assintomática. DsRNA de ambas as amostras foi submetido à RT-PCR com 17 pares de oligonucleotídeos específicos para a detecção de Grapevine virus A (GVA), Grapevine virus B (GVB), Grapevine virus D (GVD), Grapevine fleck virus (GFkV), Grapevine fanleaf virus (GFLV), Arabis mosaic virus (ArMV), Grapevine chrome mosaic virus (GCMV), Rupestris stem pitting-associated virus (RSPaV), Grapevine leafroll-associated virus 1 a 4 (GLRaV-1 a -4), além de três pares de oligonucleotídeos degenerados. Pelo menos um fragmento amplificado, por par de oligonucleotídeos, foi clonado e sequenciado. Plantas sintomáticas e assintomáticas mostraram infecções múltiplas por RSPaV, GLRaV-2 e/ou GLRaV-3. As sequências de nucleotídeos obtidas para sete isolados de RSPaV, três de GLRaV-2 e dois de GLRaV-3 apresentaram identidades superiores a 90% com espécies homólogas e permitiram a definição das possíveis estirpes presentes nas amostras infectadas. Esses resultados evidenciam a necessidade da diagnose viral baseada em testes específicos para determinar a condição sanitária da videira.

The vegetative propagation of grapevine facilitates multiple viral infections, with different symptoms which vary according to combinations of cultivar or host species with viral species. The aims of this research were to detect and identify the viral species infecting two grapevine species/cultivars: one symptomatic and one symptomless. DsRNA from both samples was assayed by RT-PCR using 17 pairs of specific primers for detection of the Grapevine virus A (GVA), Grapevine virus B (GVB), Grapevine virus D (GVD), Grapevine fleck virus (GFkV), Grapevine fanleaf virus (GFLV), Arabis mosaic virus (ArMV), Grapevine chrome mosaic virus (GCMV), Rupestris stem pitting-associated virus (RSPaV) and Grapevine leafroll-associated virus 1-4 (GLRaV-1 to -4), besides three degenerate primer pairs. For each primer pair at least one amplicon was cloned and sequenced. Symtomatic and symptomless plants were multiple infected by RSPaV, GLRaV-2 and/or GLRaV-3. The nucleotide sequences of seven isolates of RSPaV, three of GLRaV-2 and two of GLRaV-3 showed identities higher than 90% with the homologous viral species and allowed to identify possible viral strains in infected samples. These results highlight the necessity of viral diagnosis based on specific assays to determine grapevine sanitary status.

O Rupestris stem pitting-associated virus (RSPaV) é o agente causal das caneluras do lenho da videira. Este trabalho teve como objetivo produzir antissoro policlonal a partir da proteína capsidial (CP) recombinante do RSPaV e avaliar a sua especificidade e sensibilidade. O gene da CP do RSPaV, com 780pb, foi previamente caracterizado. Esse gene foi subclonado no sítio de restrição EcoRI, no vetor de expressão pRSET-B e o plasmídeo recombinante foi utilizado para induzir a expressão da CP em Escherichia coli. A CP, ligada a uma cauda de seis histidinas, foi purificada por meio de cromatografia de afinidade em coluna de Ni-NTA a partir do extrato de proteínas totais extraídas de E. coli. A identidade da proteína purificada foi confirmada em SDS-PAGE e Western blot, utilizando-se anticorpos comerciais contra a cauda de seis histidinas. A CP recombinante expressada in vitro apresentou massa molecular de cerca de 31kDa. A proteína purificada foi quantificada e 2,55mg foram utilizados para a imunização de um coelho. O antissoro policlonal obtido reagiu com diferentes isolados deste vírus, extraídos de videiras em ELISA indireto.

RSPaV is the causal agent of pitting in the grapevine woody cylinder. The aim of this research was to produce polyclonal antiserum against recombinant RSPaV coat protein (CP) and evaluate its specificity and sensibility. The CP gene (780bp) of RSPaV was previously characterized. This gene was subcloned into the EcoRI site of the pRSET-B expression vector and the recombinant plasmid was used to induce the expression of the CP in E. coli cells. The CP, fused to a 6-His-tag, was purified from E. coli total protein extract by affinity chromatography using Ni-NTA resin. Identity of the purified protein was confirmed by SDS-PAGE and Western blot, using antibodies against the histidine tail. The in vitro-expressed recombinant CP presented a MW of ca. 31kDa. The purified protein was quantified and 2.55mg used for the immunization of a rabbit. The obtained polyclonal antiserum reacted with different RSPaV isolates extracted from grapevines in indirect ELISA.

O comércio de caqui fresco resulta em grande quantidade de frutos não classificados. Assim, considerando também que a desidratação prolonga a vida-de-prateleira de alimentos perecíveis, a pesquisa se propôs a fazer uma contribuição para o desenvolvimento do processo de fabricação de passa a partir de frutos de caqui 'Fuyu'. Foi utilizado um secador contendo circulação forçada de ar e cinco bandejas. O fruto descascado foi fatiado em quatro formas diferentes: fatias em metades, de corte transversal; fatias em ¼, de cortes longitudinais; fatias em ¼, de corte longitudinal seguido de corte transversal; fatias em 1/8, de dois cortes longitudinais seguidos de transversais. A condição de secagem que aplicou temperatura de 80ºC por 1h seguida de 60ºC por cerca de 23h produziu passas de caqui 'Fuyu' com a melhor aparência. A forma da fatia passa que teve a preferência dos provadores foi aquela em que o fruto foi cortado em oito fatias (fatias de 1/8). As passas de caqui 'Fuyu' contendo umidades residuais baixas, 19,7 e 27,4%, foram apreciadas, quanto ao paladar, igualmente pelos provadores. Os frutos de caqui proporcionaram, em peso, 16,7 e 18,1% de produtos em passas e perdas estimadas em 25,5%. Os resultados indicaram que a passa de caqui 'Fuyu' constitui-se numa excelente oferta de fruta desidratada.

Because kakis are usually sold fresh, this results in a high number of fruits that do not meet the marketing standard. Thus, considering that dehydration extends the shelf life of perishable foods, this research was aimed at providing a contribution for the development of a product referred to as dehydrated 'Fuyu' kaki. A forced air circulation dehydrator was used, equipped with five trays. The peeled fruit was sliced in four different ways: sliced into halves, transversally; sliced into quarters, longitudinally; sliced into quarters, once longitudinally and once transversally; sliced into eighths, by cutting twice longitudinally and then once transversally. The drying conditions adjusted at 80ºC for 1h, followed by 60ºC for 23h resulted in a dehydrated 'Fuyu' kaki of better pleasant in aspect. The slicing method preferred by the samplers was that in which the fruit was sliced into eight slices (eighths). The dehydrated 'Fuyu' kaki, at low residual moistures of 19.7 or 27.4%, were pleasant in aspect and palate, and had good acceptance by samplers. The kaki fruit yielded 16.7 and 18.1% in weight of dehydrated products. Fruit losses during preparation into slices were estimated at 25.5%. The results demonstrated that the dehydrated 'Fuyu' kaki product is an excellent supply of dehydrated fruit.
