RESUMO Visando à construção de uma ferramenta de trabalho que permitisse expressar a grandeza da saúde animal de propriedades rurais, foi construído um indicador resultante da qualidade da saúde do rebanho denominado “status de saúde animal”. Como exemplo para a construção do indicador, tomaram-se os soros de 923 caprinos oriundos de 17 capris do Estado de São Paulo, nos quais se avaliaram três enfermidades e seus respectivos impactos percentuais: a artrite-encefalite caprina, a neosporose e a toxoplasmose - todas de caráter infeccioso e transmissível e que ocasionam consideráveis perdas econômicas. A lógica da construção matemática do “status de saúde animal” classifica a propriedade numa variação de 0 a 100%, conforme a positividade das enfermidades no rebanho: quanto maior a frequência acumulativa de ocorrência das doenças nos animais, menor o valor do indicador, chegando ao limite de 0% - quando todos os animais estiverem positivos às três enfermidades e de 100% - quando todo o rebanho for negativo. Para detecção de anticorpos anti-T. gondii, anti-N. caninum e anti-CAEV, foram utilizados, respectivamente, reação de imunofluorescência indireta (RIFI ≥ 16), teste de aglutinação para Neospora (NAT ≥ 25) e imunodifusão em gel de agarose (IDGA positivo ou negativo). O “status de saúde animal” das propriedades rurais variou de 32,38% a 96,40%, de acordo com a ocorrência de positividade para as enfermidades. A característica do valor do “status de saúde animal” terá reflexo direto na cadeia de produção dentro e fora da fazenda, proporcionando não somente enormes vantagens mercadológicas, devido à expressão que a saúde animal terá para agregar valor às propriedades rurais de produção animal, mas também no consumidor final que poderá verificar a qualidade da saúde animal no início da cadeia de produção.

ABSTRACT Aiming at developing a tool for assessing the magnitude of farm animal health, an indicator named herd “animal health status” was built. To illustrate the construction of the indicator, serum samples were taken from 923 goats on 17 goat farms in the state of Sao Paulo, Brazil, where three diseases were evaluated along with their impact percentages: caprine arthritis encephalitis, toxoplasmosis and neosporose – all of an infectious and transmissible nature and the cause of considerable economic losses. The mathematical rationale underlying the building of the “animal health status” indicator ranks properties on a 0 - 100% scale in terms of disease positivity in the herd, with the lowest indicator value indicating the highest cumulative disease frequency (0% = all animals tested positive for the three diseases, 100% = all animals tested negative for all diseases). Anti-T. gondii, anti-N. caninum and anti-CAEV antibodies were tested using indirect immunofluorescence reaction (IIFR ≥ 16), Neospora agglutination test (NAT ≥ 25) and agarose gel immunodiffusion (AGID positive or negative), respectively. The animal health status of the farms ranged from 32.38% to 96.40% according to disease positivity. The animal health status value will have a direct reflection on the production chain both on and off the farm, providing not only great market advantages, due to how the expression of animal health will add value to livestock farms, as well as on the consumer who can check the quality of animal health at the beginning of the production chain.

We have shown that myocardial dysfunction induced by food restriction is related to calcium handling. Although cardiac function is depressed in food-restricted animals, there is limited information about the molecular mechanisms that lead to this abnormality. The present study evaluated the effects of food restriction on calcium cycling, focusing on sarcoplasmic Ca2+-ATPase (SERCA2), phospholamban (PLB), and ryanodine channel (RYR2) mRNA expressions in rat myocardium. Male Wistar-Kyoto rats, 60 days old, were submitted to ad libitum feeding (control rats) or 50% diet restriction for 90 days. The levels of left ventricle SERCA2, PLB, and RYR2 were measured using semi-quantitative RT-PCR. Body and ventricular weights were reduced in 50% food-restricted animals. RYR2 mRNA was significantly decreased in the left ventricle of the food-restricted group (control = 5.92 ± 0.48 vs food-restricted group = 4.84 ± 0.33, P < 0.01). The levels of SERCA2 and PLB mRNA were similar between groups (control = 8.38 ± 0.44 vs food-restricted group = 7.96 ± 0.45, and control = 1.52 ± 0.06 vs food-restricted group = 1.53 ± 0.10, respectively). Down-regulation of RYR2 mRNA expressions suggests that chronic food restriction promotes abnormalities in sarcoplasmic reticulum Ca2+ release.

Diets rich in saturated fatty acids are one of the most important causes of atherosclerosis in men, and have been replaced with diets rich in unsaturated fatty acids (UFA) for the prevention of this disorder. However, the effect of UFA on myocardial performance, metabolism and morphology has not been completely characterized. The objective of the present investigation was to evaluate the effects of a UFA-rich diet on cardiac muscle function, oxidative stress, and morphology. Sixty-day-old male Wistar rats were fed a control (N = 8) or a UFA-rich diet (N = 8) for 60 days. Myocardial performance was studied in isolated papillary muscle by isometric and isotonic contractions under basal conditions after calcium chloride (5.2 mM) and ß-adrenergic stimulation with 1.0 µM isoproterenol. Fragments of the left ventricle free wall were used to study oxidative stress and were analyzed by light microscopy, and the myocardial ultrastructure was examined in left ventricle papillary muscle. After 60 days the UFA-rich diet did not change myocardial function. However, it caused high lipid hydroperoxide (176 ± 5 vs 158 ± 5, P < 0.0005) and low catalase (7 ± 1 vs 9 ± 1, P < 0.005) and superoxide-dismutase (18 ± 2 vs 27 ± 5, P < 0.005) levels, and discrete morphological changes in UFA-rich diet hearts such as lipid deposits and mitochondrial membrane alterations compared to control rats. These data show that a UFA-rich diet caused myocardial oxidative stress and mild structural alterations, but did not change mechanical function.

Cardiac structures, function, and myocardial contractility are affected by food restriction (FR). There are few experiments associating undernutrition with hypertension. The aim of the present study was to analyze the effects of FR on the cardiac response to hypertension in a genetic model of hypertension, the spontaneously hypertensive rat (SHR). Five-month-old SHR were fed a control or a calorie-restricted diet for 90 days. Global left ventricle (LV) systolic function was evaluated in vivo by transthoracic echocardiogram and myocardial contractility and diastolic function were assessed in vitro in an isovolumetrically beating isolated heart (Langendorff preparation). FR reduced LV systolic function (control (mean ± SD): 58.9 ± 8.2; FR: 50.8 ± 4.8%, N = 14, P < 0.05). Myocardial contractility was preserved when assessed by the +dP/dt (control: 3493 ± 379; FR: 3555 ± 211 mmHg/s, P > 0.05), and developed pressure (in vitro) at diastolic pressure of zero (control: 152 ± 16; FR: 149 ± 15 mmHg, N = 9, P > 0.05) and 25 mmHg (control: 155 ± 9; FR: 150 ± 10 mmHg, N = 9, P > 0.05). FR also induced eccentric ventricular remodeling, and reduced myocardial elasticity (control: 10.9 ± 1.6; FR: 9.2 ± 0.9%, N = 9, P < 0.05) and LV compliance (control: 82.6 ± 16.5; FR: 68.2 ± 9.1%, N = 9, P < 0.05). We conclude that FR causes systolic ventricular dysfunction without in vitro change in myocardial contractility and diastolic dysfunction probably due to a reduction in myocardial elasticity.

To identify early metabolic abnormalities in type 2 diabetes mellitus, we measured insulin secretion, sensitivity to insulin, and hepatic insulin extraction in 48 healthy normal glucose-tolerant Brazilians, first-degree relatives of type 2 diabetic patients (FH+). Each individual was matched for sex, age, weight, and body fat distribution with a person without history of type 2 diabetes (FH-). Both groups were submitted to a hyperglycemic clamp procedure (180 mg/dl). Insulin release was evaluated in its two phases. The first was calculated as the sum of plasma insulin at 2.5, 5.0, 7.5, and 10.0 min after the beginning of glucose infusion, and the second as the mean plasma insulin level in the third hour of the clamp procedure. Insulin sensitivity index (ISI) was the mean glucose infusion rate in the third hour of the clamp experiment divided by the mean plasma insulin concentration during the same period of time. Hepatic insulin extraction was determined under fasting conditions and in the third hour of the clamp procedure as the ratio between C-peptide and plasma insulin levels. FH+ individuals did not differ from FH- individuals in terms of the following parameters [median (range)]: a) first-phase insulin secretion, 174 (116-221) vs 207 (108-277) µU/ml, b) second-phase insulin secretion, 64 (41-86) vs 53 (37-83) µU/ml, and c) ISI, 14.8 (9.0-20.8) vs 16.8 (9.0-27.0) mg kg-1 min-1/µU ml-1. Hepatic insulin extraction in FH+ subjects was similar to that of FH- ones at basal conditions (median, 0.27 vs 0.27 ng/µU) and during glucose infusion (0.15 vs 0.15 ng/µU). Normal glucose-tolerant Brazilian FH+ individuals well-matched with FH- ones did not show defects of insulin secretion, insulin sensitivity, or hepatic insulin extraction as tested by hyperglycemic clamp procedures.

Non-absorbable microgranular hydroxyapatite was infiltrated into the subepidermal abdominal region of guinea pigs in order to assess the possibility of using this material to correct deficiencies in orbital volume. Microgranular hydroxyapatite (2.0 ml) was subepidermally infiltrated into the abdominal region of 20 guinea pigs. The animals were divided into four experimental groups of 5 animals each, which were killed 7 (G1), 15 (G2), 30 (G3) and 60 (G4) days after infiltration. The area and the largest and smallest diameters of the nodules formed by infiltration were evaluated at the site of infiltration and histological examination was performed. The mean granuloma area was similar in all groups. Histopathological examination showed that the material remained isolated from surrounding tissues by a pseudocapsule that became denser throughout the experiment. A host reaction started with young fibroblastic tissue that evolved to dense tissue until cartilaginous tissue was formed in G4, progressively advancing towards the center of the granuloma from G1 to G4. Non-absorbable microgranular hydroxyapatite is an inert material that was well tolerated by the animals studied, with maintenance of the infiltrated volume, and may perhaps be useful to fill anophthalmic cavities.

Cento e noventa e seis fetos de Zebu mestiço, entre 106 e 246mm de comprimento, coletados em matadouros do Estado de São Paulo e divididos a cada 10mm, em 15 classes, foram medidos e pesados. Alguns órgãos como fígado, coração e pulmões foram removidos e pesados. Para selecionar um método de avaliação do crescimento nesse período fetal, várias funções matemáticas foram examinadas. Entre as equações de regressão significativas, a função monologarítmica foi a escolhida. Além disso, os fetos em condições semelhantes de comprimento e peso foram comparados com fetos de raça pura.

A total of 196 fetuses of crossbred Zebu between 106 to 246mm in length collected from a slaughterhouse in São Paulo, Brazil, and divided into groups of 10mm size differences intervals into 15 classes were measured and weighed. Liver, heart and lungs were also removed and weighed. In order to select the method of evaluation of growth several mathematical functions were applied. Among significant equations the monologarithmic function was selected. Furthermore, the fetuses in similar conditions of length and weight were compared with racially pure fetuses.
