Resumo A leishmaniose visceral é uma zoonose parasitária que afeta principalmente populações mais pobres e vulneráveis, e os cães domésticos são considerados as principais fontes de infecção para o vetor e, portanto, para os humanos. Porém diversos estudos têm pesquisado o papel de outros hospedeiros vertebrados no ciclo da doença. Neste contexto, objetivou-se realizar um levantamento da infecção por Leishmania infantum em asininos e muares, vivendo em região semiárida do Brasil. Foi utilizado sangue total de 72 equídeos (65 asininos e 7 muares) para a realização de diagnóstico molecular por meio da técnica de Reação em Cadeia de Polimerase em Tempo Real (qPCR). Um total de 25% das amostras (18/72) resultaram positivas na qPCR, porém não houve diferença significativa entre as espécies (asininos e muares), sexo (macho e fêmea) e situação de abandono dos animais (sim ou não). Asininos e muares, vivendo em condições semiáridas, apresentam alta frequência de infecção por L. infantum, sendo válido atribuir importância a essas espécies no ciclo epidemiológico da leishmaniose visceral, seja como um reservatório em potencial, seja apenas como uma fonte alimentar abundante para os vetores.

Abstract Visceral leishmaniasis is a parasitic zoonosis that mainly affects poorest and most vulnerable populations, and domestic dogs are considered to be the main source of infection to the vector and therefore humans. However, several studies have investigated the role of other vertebrate hosts in the disease cycle. In this context, the aim of the present study was to conduct a survey of Leishmania infantum infection in donkeys and mules living in a semiarid region of Brazil. Whole blood sampled from 72 equids (65 donkeys and 7 mules) was used to perform molecular diagnosis using the real-time polymerase chain reaction (qPCR) technique. A total of 25% of the samples (18/72) were positive through qPCR, but there were no significant differences between the species (donkeys or mules), sex (male or female) and abandonment situation of the animals (yes or no). Donkeys and mules living under semiarid conditions have high frequency of L. infantum infection. It is therefore worth assigning importance to these species in the epidemiological cycle of visceral leishmaniasis, either as potential reservoirs or just as an abundant food source for vectors.

RESUMO: Foram utilizados 12 lagartos Teiús (Tupinambis merianae) do Nordeste do Brasil. Encontramos nove animais positivos (75%) para Leptospira sp. na PCR e seis (50%) soropositivos. O sequenciamento de DNA de Leptospira sp. revelou 99% de semelhança com L. interrogans. Os resultados indicam que esta espécie pode desempenhar um papel importante na transmissão da leptospirose humana.

ABSTRACT: We used 12 tegu lizards (Tupinambis merianae) from northeastern Brazil, and we reported nine (75%) Leptospira sp. PCR-positive animals and six (50%) seropositive. Leptospira sp. DNA sequencing revealed 99% similarity with L. interrogans. Our findings indicated that this species may play a role in the transmission of human leptospirosis.

ABSTRACT Many countries in the Americas have detected local transmission of multiple arboviruses that cause febrile illnesses. Therefore, laboratory testing has become an important tool for confirming the etiology of these diseases. The present study aimed to compare the sensitivity and specificity of three different Zika virus detection assays. One hundred serum samples from patients presenting with acute febrile symptoms were tested using a previously reported TaqMan® RT-qPCR assay. We used a SYBR® Green RT-qPCR and a conventional PCR methodologies to compare the results. Of the samples that were determined to be negative by the TaqMan® RT-qPCR assay, 100% (Kappa = 0.670) were also found to be negative by SYBR® Green RT-qPCR based on Tm comparison; however, 14% (Kappa = 0.035) were found to be positive by conventional PCR followed by agarose gel electrophoresis. The differences between the ZIKV strains circulating worldwide and the low viremia period can compromise diagnostic accuracy and thereby the accuracy of outbreak data. Therefore, improved assays are required to improve the diagnosis and surveillance of arbovirus.

RESUMO: Felis catus gammaherpesvirus 1 (FcaGHV1) pode causar uma infecção assintomática, que resulta em uma transmissão eficiente e consequente disseminação do virus na população felina. Este estudo utilizou a detecção molecular por qPCR (PCR quantitativa) baseado na amplificação de um fragmento do gene da DNA polimerase para avaliar a ocorrência de FcaGHV1, sendo correlacionado a outros patógenos virais felinos como Carnivore protoparvovirus 1 (CPPV-1), Felid alphaherpesvirus 1 (FeHV-1) e aos retrovírus felinos como vírus da imunodeficiência felina (FIV) e vírus da leucemia felina (FeLV). Das 182 amostras de sangue avaliadas, 23,6% (43/182) foram positivas para FcaGHV1. Aproximadamente 37,9% (33/87) das amostras positivas para retrovirus também foram positivas para FcaGHV1 (P<0,0001). Entre as amostras FIV-infectadas, 49% (24/49) foram positivas para FcaGHV1 (P<0,0001). A infecção por FcaGHV1 não foi associada à coinfecção por FeLV (P>0,66) e CPPV-1 (P>0,46). Todas as amostras foram negativas para FeHV-1. Felinos machos foram significativativamente associados à infecção por FcaHV1 (P <0,0001) e o risco de infecção com FcaGHV1 foi aproximadamente 7,74 vezes maior comparados às femeas. Os filhotes (≤1 ano) foram os menos acometidos pela infecção por FcaGHV1 sendo verificado uma proporção de 7.7% (4/52). Assim, gatos machos com mais de um ano de idade e infectados por FIV foram, consideravelmente, mais susceptíveis a serem infectados com FcaGHV1. Para nosso conhecimento, este é o primeiro estudo que relata a ocorrência de infecção e detecção molecular de FcaGHV1 em gatos domésticos no Brasil e na América do Sul.

ABSTRACT: Felis catus gammaherpesvirus 1 (FcaGHV1) may causes an asymptomatic infection that result in an efficient transmission and subsequently dissemination of the virus in feline population. This study used molecular detection by qPCR (quantitative PCR) based on DNA polymerase gene fragment amplification to evaluate the occurrence of FcaGHV1 and its correlation with other feline viral pathogens, such as Carnivore protoparvovirus 1 (CPPV-1), Felid alphaherpesvirus 1 (FeHV-1), and feline retroviruses such as feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV). Of the 182 blood samples evaluated 23.6% (43/182) were positives for FcaGHV1. Approximately 37.9% (33/87) of the samples that tested positive for retrovirus were also were positive for FcaGHV1 infection (P<0.0001). Among FIV-infected samples, 49% (24/49) were positive for FcaGHV1 (P<0.0001). FcaGHV1 infection was not associated with FeLV (P>0.66) or CPPV-1 (P>0.46) coinfection. All samples were negative for FeHV-1. Male felines were significantly associated to FcaGHV1 (P<0.0001) and their risk of infection with FcaGHV1 was about of 7.74 times greater compared to females. Kittens (≤ 1year) were the least affected by FcaGHV1 infection, being verified a rate of 7.7% (4/52). Therefore, male cats over one year old and infected with FIV were considerably more likely to be infected with FcaGHV1. To our knowledge, this is the first study to report the occurrence and molecular detection of FcaGHV1 infection in domestic cats in Brazil and in South America.

RESUMO: O herpesvírus canino (CaHV-1) afeta os canídeos em todo o mundo, causando morte em neonatos e hospedeiros imunossuprimidos. A infecção aguda por CaHV-1 pode causar problemas reprodutivos, respiratórios e neurológicos em animais adultos. A patogênese viral e expressão de genes hospedeiros durante a infecção por CaHV-1 ainda não são bem compreendidos. No presente estudo, o transcriptoma de células de rim canino Madin-Darby (MDCK) infectadas in vitro com herpesvirus canino foi explorado. Para isso, foi realizado o sequenciamento de RNA (RNA-seq) de amostras coletadas em diferentes momentos durante a infecção. Subsequentemente, a análise transcriptômica dos genes relacionados à atividade celular e aos processos envolvidos no ciclo de infecção do vírus foram avaliadas até 32 horas após a inoculação (pi). Dentre os genes avaliados, constatamos uma elevação significativa e gradativa da expressão da Prostaglandina-endoperoxide sintase 2 (PTGS2) ou ciclooxigenase 2 (COX2), ao longo da infecção viral, foi verificada por análise de expressão gênica diferencial e validada por resultados de transcrição reversa por PCR quantitativo (RT-qPCR). O aumento da expressão de COX2 geralmente é induzida em resposta a inflamação, patógenos ou ativação do sistema imune, mas também pode ser um mecanismo para favorecer a replicação viral. Assim, o aumento do nível de COX2 pode ser um fator favorável à infecção viral pelo vírus CaHV-1 e o uso de inibidores seletivos da COX2 pode ser benéfico para limitar a infecção ou os sinais clínicos, causando a interrupção do ciclo de replicação viral durante a infecção ativa. Além disso, a regulação diferencial da expressão dos genes verificados neste estudo podem contribuir para determinar alvos importantes para inibir a infecção por herpesvírus canino, seja por mecanismos celulares ou virais.

ABSTRACT: Canine herpesvirus (CaHV-1) affects canids worldwide, causing death in neonates and immunosuppressed hosts. Acute infection by CaHV-1 can cause reproductive, respiratory, and neurological problems in adult animals. Viral pathogenesis and host genes expressions during of CaHV-1infection are not clearly understood. In the present study, the transcriptome of canine kidney cell Mardin-Darby (MDCK) infected in vitro with canine herpesvirus was explored. For this, RNA sequencing (RNA-seq) of the samples in different moments during infection was carried out. Subsequently, the transcriptomic analysis genes related to cell activities and process involved to viral cycle infection were evaluated until 32h post-inoculation (pi). Among evaluated genes, was verified a significant and gradative increase of the prostaglandin-endoperoxide synthase 2 (PTGS2) or cyclooxygenase 2 (COX2) gene expression, throughout of infection, though differential gene expression analysis and validated by quantitative reverse transcription PCR (RT-qPCR). High COX2 expression is usually induced in response to inflammation, pathogens or activation of the immune system but can be a viral mechanism to favor viral replication. Thus, COX2 level increase can be a favorable factor for viral infection with Cahv-1 virus and the use of selective COX2 inhibitors may be beneficial for limiting the infection or clinical signs by causing interruption of the viral replication cycle during active infection. Additionally, the regulation genes expression differential verified in this study can contribute to determining important targets for inhibiting canine herpesvirus infection either by cellular or viral mechanisms.

Abstract Background The present study evaluated the effect of treatment with benznidazole on mRNA expression of IFN-γ, IL-17, IL-10, TGF-β and FoxP3 in spleen and heart tissue of BALB/c mice in the acute phase of an experimental infection with Trypanosoma cruzi, strains JLP or Y. Methods The mRNA expression of cytokines and parasite load were assessed by q-PCR. Dependent groups were compared using Student's paired t-test and independent groups were compared using Student's unpaired t-test. Results Infection with the JLP or Y strains increased expression of IFN-γ in the heart and of IL-10 and IL-17 in the spleen and heart compared to uninfected animals. Treatment increased the expression of IFN-γ and decreased the expression of IL-17, IL-10, TGF- β and Foxp3 in spleen and heart tissue compared to untreated infected animals. Conclusion Benznidazole can induce Th1 profile in the initial of the acute phase. The treatment decreased the parasite load in both organs, although the number of parasites in Y-strain-infected mice remained high. The data suggest that benznidazole may modulate cytokine expression in infection and can be dependent of the strain. However, treatment was not fully effective in the infection provoked by Y strain, probably due to the characteristics of the strain itself.

O colapso induzido pelo exercício (EIC) é uma enfermidade hereditária caracterizada por fraqueza muscular, dificuldade de locomoção e colapso após atividade física intensa. Esta enfermidade autossômica recessiva afeta principalmente cães jovens da raça Labrador Retriever e decorre da mutação c.767G>T no gene codificador da proteína dinamina 1 (DNM1). O objetivo deste trabalho é relatar o primeiro caso de EIC em Labrador Retriever no Brasil. O teste molecular para detectar a mutação responsável pela EIC confirmou o diagnóstico clínico em um Labrador Retriever com histórico de fraqueza muscular e colapso após exercício. Uma vez diagnosticada no Brasil, ressalta-se a importância de inserir a EIC entre os diagnósticos diferenciais das enfermidades neuromusculares em cães da raça Labrador Retriever e utilizar o diagnóstico molecular para orientar os acasalamentos.

The exercise-induced collapse (EIC) is a hereditary disease characterized by muscle weakness, impaired locomotion and collapse after intense exercise. This autossomic recessive disorder affects mainly Labrador Retriever presenting the mutation c.767G>T in the dynamin 1 (DNM1) gene. The objective of this study is to report the first case of exercise-induced collapse in Labrador Retriever in Brazil. The molecular test detected the specific genetic mutation and confirmed the clinical diagnosis in a Labrador Retriever with clinical history of weakness and collapse after exercise. It is important to include this disease as part of the differential diagnosis of neuromuscular diseases in Labrador Retriever and use the molecular test to guide matings.

O objetivo do trabalho é descrever uma estratégia para a obtenção de animais negativos para o PCV2 oriundos de uma granja positiva para este vírus. Dezesseis leitões foram obtidos de fêmeas que tiveram os títulos de IgG anti-PCV2 e o DNA viral testados durante a gestação. Esses leitões, aos sete e dez dias de idade, foram transferidos para a unidade de pesquisa. Durante o período de 7 e 10 aos 49 e 52 dias de idade, amostras de soro, suabes nasal e fecal foram coletadas, a cada sete dias. Após esse período, três animais permaneceram na unidade de pesquisa e foram acompanhados dos 49 aos 114 dias de idade, com coletas realizadas a cada 28 dias. Não houve diferença significativa (p = 0,317) de viremia entre marrãs (n = 6) e porcas (n = 10). Com relação aos níveis de IgG, observou-se diferença significativa (p = 0,0213) entre porcas e marrãs. Os leitões (n = 16), obtidos de duas fêmeas, foram transferidos para a unidade de pesquisa. Os animais entre 7 e 10 dias e aos 49 e 52 dias de idade apresentaram queda de IgG e ausência de IgM anti-PCV2; e as amostras de soro, suabe nasal e fecal foram negativos para o DNA de PCV2. Após os 49 dias, nos três animais mantidos isolados, a detecção de IgG, IgM e DNA para PCV2 permaneceu negativa. Concluindo, a estratégia de manejo utilizada permitiu obter suínos negativos para PCV2 oriundo de granjas positivas para o agente.

The present study describes a strategy to obtaining PCV2-negative animals from a PCV2-positive herd. Sixteen piglets were selected from females that had their IgG anti-PCV2 and viral circulation followed during pregnancy. These 7-days old piglets were transferred to the research unit. During the period from 7 to 49 days of age, serum, nasal and fecal swabs were collected every seven days. After this period, three animals remained in the research unit and were followed from 49 to 114 days of age, with samples taken each 28 days. No difference (p = 0.317) in viremia between gilts (n = 6) and sows (n = 10) were observed. Regarding the IgG levels, a significant difference (p = 0.0213) were found between gilts and sows. The piglets (n = 16), obtained from the two females, were transferred to the research unit. The animals between 7 and 10 and 49 and 52 days of age showed a decreased of the IgG title and absence of IgM; the serum and fecal and nasal swabs were negative for PCV2 DNA. After 49 days of age, the three remained animals negative for IgG, IgM and viral DNA for PCV2. In conclusion, the strategy of handling used herein allowed the obtention of PCV-2 negative pigs from PCV2-positive herds.

O Vírus da leucemia felina (FeLV) pertence à família Retroviridae, gênero Gammaretrovirus. Diferentemente de outras retroviroses, uma parcela dos gatos jovens e adultos exposta ao FeLV não apresenta antigenemia/viremia, de acordo com as técnicas convencionais de detecção viral, como isolamento em cultivo celular, imunofluorescência direta e ELISA. O emprego de técnicas de maior sensibilidade para detecção e quantificação viral, como o PCR quantitativo, permitiu a identificação de animais positivos para a presença de DNA proviral e RNA na ausência de antigenemia/viremia e, com isso, um refinamento da análise das diferentes evoluções da infecção. Assim, reclassificou-se a patogenia do FeLV em 4 categorias: infecção abortiva, regressiva, latente e progressiva. Foi possível também detectar DNA proviral e RNA em animais considerados imunes ao FeLV após vacinação. Diante disso, os objetivos desta revisão de literatura foram demonstrar as implicações da utilização de técnicas sensíveis de detecção viral na interpretação e classificação da infecção do FeLV e rever as técnicas de detecção do vírus para fins de diagnóstico. Além disso, apresentar os resultados referentes à eficácia da vacinação contra o FeLV com a utilização dessas técnicas.

Feline leukemia virus (FeLV) belongs to the Retroviridae family, genus Gammaretrovirus. Unlike other retroviruses, a portion of FeLV exposed animals eliminates antigenemia/viremia, according to convectional techniques of virus detection, such as isolation in cell culture, direct fluorescent antibody test and ELISA. The use of more sensitive techniques to detect and quantify viruses enabled the detection of proviral DNA and RNA in cats with undetectable antigenemia/viremia, and thus the refinement of the different infection outcomes analysis. As a result, FeLV pathogenesis was reclassified in 4 categories: abortive, regressive, latent and progressive infections. It was also demonstrated the detection of proviral DNA and RNA in cats believed to be immune to infection after vaccination. Therefore, the objectives of this review were to demonstrate the implications of the use of sensitive techniques for viral detection in the interpretation and classification of FeLV infection and reconsider the techniques for FeLV diagnostic purposes. In addition, it was presented the results concerning the effectiveness of FeLV vaccination with the use of these techniques.

No Brasil, a ocorrência da infecção pelo vírus da imunodeficiência felina (FIV) já foi descrita. Neste estudo, objetivou-se investigar a freqüência da infecção pelo FIV em 454 amostras de sangue de gatos domésticos doentes e sadios, oriundos de 13 cidades do Estado de São Paulo, assim como avaliar os fatores de risco associados à infecção pelo FIV. Os resultados demonstraram que 14,7% (67/454) dos gatos estavam infectados pelo FIV. A avaliação clínica dos animais investigados mostrou que 29,2% dos animais soropositivos para FIV estavam doentes, enquanto 7,3% não apresentavam nenhuma manifestação clínica. Além disso, a vasta maioria dos animais positivos (23,1%) vivia em residências e tinha livre acesso à rua. A incidência da infecção pelo FIV foi maior nos gatos machos (20,3%) do que nas fêmeas (9,7%). Os resultados sugerem que certas características como sexo, estilo de vida e estado de saúde podem estar associadas ao risco de contrair a infecção pelo FIV na população de gatos estudada.

The occurrence of feline immunodeficiency virus (FIV) in Brazil has been previously described. This study aimed to investigate the frequency of FIV infection in 454 blood samples from healthy and sick domestic cats from 13 cities of São Paulo State, Brazil as well as to evaluate the risk factors associated with the infection. The results showed that 14.7% (67/454) of the cats were infected with FIV. The clinical evaluation showed that 29.2% of the FIV-positive animals were sick, while 7.3% did not show any type of clinical manifestation. In addition, the vast majority (23.1%) of positive cases corresponded to free-roaming owned cats. The incidence of FIV infection was higher in males (20.3%) than in females (9.7%). The results suggest that certain characteristics such as gender, health status and lifestyle may be associated with the risk of being infected with FIV in the population of cats studied.

A intoxicação alimentar estafilocócica ocorre devido à ingestão de alimentos contaminados com enterotoxinas. Essa contaminação tem sido oriunda, principalmente, da manipulação humana, ou de matérias-primas procedentes de animais portadores. Embora Staphylococcus aureus coagulase positiva, seja o principal agente de intoxicação alimentar, alguns pesquisadores enfatizam que os estafilococos coagulase-negativa (ECN) podem produzir as enterotoxinas estafilocócicas, podendo contribuir para a intoxicação alimentar. Este estudo teve como objetivos isolar os ECN de alimentos e verificar a capacidade enterotoxigênica dessas linhagens. Foram estudadas 88 amostras de alimentos, sendo que 22,7% foram positivas para ECN com crescimento entre 10² e 10(6) UFC/g or mL. A espécie predominante dentre as linhagens isoladas foi S. epidermidis (40%), seguido por S. warneri (20%), S. xylosus (20%), S. saccharolyticus (15%) e S. hominis (5%). Entre as linhagens isoladas, quatro apresentaram genes para produção de enterotoxinas pelo método de Reação da Polimerase em Cadeia (PCR), com predominância do gene sea. Não se detectou a produção de enterotoxina pelo método de aglutinação em látex (RPLA). Através dos resultados obtidos, observou-se que os ECN isolados de alimentos não devem ser ignorados quanto à sua capacidade toxigênica, necessitando de maior estudo e atenção para melhor caracterização desse grupo de microrganismos em alimentos.

Staphylococcal food poisoning is caused by ingestion of enterotoxins preformed in the food contaminated essentially through human manipulation or raw material obtained from animals. Although coagulase-positive Staphylococcus aureus is the main agent responsible for food intoxication, some researches emphasise that coagulase-negative staphylococci (CNS) are able to produce staphylococcal enterotoxins and may be a potential cause of food poisoning. In the present study CNS were isolated from foods and the toxigenic capacity of the strains determined. A total of 88 food samples were analysed and 22.7% were positive for CNS strains. Staphylococcal counts ranged from 3.0 x 10² to 1.4 x 10(6) CFU/g or mL of food examined. S. epidermidis predominated among the isolates (40%). Further isolates included S. xylosus (20%), S. warneri (20%), S. saccharolyticus (15%), and S. hominis (5%). Four isolates were positive for enterotoxin genes, as detected by polymerase chain reaction, with sea being the predominant gene. Although no enterotoxin production was detected by the reverse passive latex agglutination method, the data showed that the toxigenic capacity of CNS should not be ignored, requiring investigation of this group of microorganisms in food.
