ABSTRACT Background: Global publications on Q fever have increased after the 2007 epidemic in the Netherlands. However, the epidemiology of Q fever/coxiellosis in Brazil is still poorly understood. Accordingly, there have been few studies investigating the presence of Coxiella burnetii in dairy products around the world, especially in Brazil, where consumption of fresh cheese made from raw-milk is very high. Objective: This study was a random survey to assess the prevalence of C. burnetii by PCR in traditional Minas artisanal cheese from the Serro microregion, Brazil, which is manufactured from bovine raw-milk. Methods: DNA extracted from 53 cheese samples were analyzed by nested PCR with C. burnetii-specific primers and the products confirmed by DNA sequencing. Results: Out of the 53 cheese samples five (9.43%) were C. burnetii DNA-positive, each coming from one of the respective randomly selected manufacturing agroindustries.Based on our results, it is estimated that 1.62 tons/day of ready-to-eat cheese made from raw-milk from a total of 16.2 tons produced daily in the study region are contaminated with C. burnetii. Conclusion: To our knowledge, this is the first report of highly heat-resistant zoonotic pathogen in raw-milk Brazilian artisanal cheese. This food safety hazard has been completely neglected in ready-to-eat raw-milk Brazilian artisanal cheese and could imply potential threats to consumers, since C. burnetii survives in artisanal cheese submitted to long ripening periods. Thus, this work established random and representative baseline prevalence of C. burnetii in this food product in Brazil. Further epidemiological studies, monitoring trends and setting control targets are warranted. Finally, these results point out the importance of including C. burnetii in animal and public health surveillance programs.

Abstract This study investigated the presence of generic and verotoxin-producing E. coli as well as enumerated faecal coliforms in 30 beef carcasses in different parts of the slaughter process (after skinning, washing and cooling) at each of three slaughterhouses of the state of Mato Grosso do Sul, Brazil. Among the total number of carcasses examined (n = 90), 39 (43.3%) had generic E. coli. Among the 270 samples analysed, 25 (9.3%) were positive after skinning, 14 (5.2%) were positive after washing and nine (3.3%) were positive after cooling. The majority of isolates of E. coli was collected from samples after skinning, which is considered a critical point of the microbial contamination of carcasses. However, the highest concentration of faecal coliforms was found after the washing step. The cooling step proved to be important to reducing the amount of hygiene-indicator microorganisms. The E. coli isolates had no stx1 or stx2 genes associated with virulence.

BACKGROUND The World Health Organization (WHO) has classified human zoonotic tuberculosis (TB) due to Mycobacterium bovis as a neglected issue in the developing world. In a recent cross-sectional study in Brazil, three of 189 TB patients presented with a coinfection of M. bovis and M. tuberculosis and were selected as cases for this study. OBJECTIVE The aim was to evaluate risk factors (RF) for zoonotic TB in an urban area of Brazil in order to guide preventive programmes. METHODS A matched case-control study was carried out nested within a cross-sectional study. For each of the three cases, 14 age- and sex-matched controls (TB due to M. tuberculosis) were selected. FINDINGS Zoonotic potential exposures (ZE) and extrapulmonary TB (EPTB) were independently associated with zoonotic TB in multivariate analyses. CONCLUSIONS ZE by occupation and consumption of raw milk and derivative products that place individuals in direct and indirect contact with animals and their excretions/secretions increase the risk for zoonotic TB in Brazil, especially among those with EPTB. Therefore, measures such as efficient control of bovine TB, distribution of pasteurised milk and its derivative products, and the diagnosis and monitoring of zoonotic TB in humans are essential steps, especially in developing countries where bovine TB is enzootic, and further studies are necessary.

RESUMO: Considerando a falta de estudos sobre lesões nos órgãos genitais de cadelas naturalmente infectadas por Leishmania infantum , o objetivo do presente estudo foi investigar a correlação entre reação inflamatória crônica, imunomarcação e carga parasitária, no sistema genital. Dez animais foram fornecidos pelo Departamento de Controle de Vetores e Vigilância Animal do município de Caruaru, Estado de Pernambuco, Brasil. Fragmentos de vulva, vagina, cérvix, corpo do útero, corno do útero e ovários foram avaliados por descrição histopatológica, imuno-histoquímica (IHQ) e detecção de DNA de formas amastigotas por qPCR. A relação entre IHQ e infiltrado inflamatório crônico relacionado com L. infantum foi observada apenas na vulva e vagina, enquanto as mesmas reações sem imunomarcação foram observadas em todos os órgãos, exceto nos ovários. DNA de L. infantum foi detectado em todos os órgãos do sistema genital, porém, sem diferença de carga parasitária entre eles. Não houve correlação entre a carga parasitária e lesões inflamatórias nos órgãos avaliados, com exceção do corpo do útero, em que foi encontrada uma correlação inversa. Em conclusão, a vulva e a vagina foram os principais locais de lesões e imunomarcação para formas amastigotas L. infantum no sistema genital de cadelas. A carga parasitária não influenciou a intensidade das lesões nos órgãos avaliados.

ABSTRACT: The aim of the present study was to investigate the correlations among chronic inflammatory reaction, immunostaining and parasite load in the genital system of female dogs naturally infected with Leishmania infantum . Animals (n = 10) used in this study were from the Department of Vector Control and Animal Surveillance of the municipality of Caruaru, state of Pernambuco, Brazil. Fragments of the vulva, vagina, cervix, uterine body, uterine horns and ovaries were submitted to histopathological analysis, immunohistochemistry (IHC) and DNA detection of amastigotes by qPCR. Correlations were found between the IHC findings and chronic inflammatory infiltrate related to L. Infantum only in the vulva and vagina; whereas, the same inflammatory reactions without immunostaining were observed in all organs, except the ovaries. L. Infantum DNA was detected in all organs of genital system, with no difference in parasite load observed among the different organs. No correlation was reported between parasite load and inflammatory lesions in the organs evaluated, except for the uterine body, in which an inverse correlation was detected. In conclusion, the vulva and vagina were the major sites of lesions and immunostaining for L. Infantum amastigotes in the genital system of female dogs. Moreover, parasite load exerted no influence on the intensity of the lesions in the organs evaluated.

RESUMO: A Leishmaniose Visceral Canina é uma importante enfermidade causada pelo protozoário Leishmania infantum que afeta diversos órgãos e sistemas, incluindo o trato genital. No entanto, o tropismo da L. infantum pelo sistema genital masculino e a correlação entre carga parasitária, detecção imunohistoquímica e alterações estruturais nesses órgãos ainda é uma questão controversa. Sendo assim, o objetivo deste estudo foi avaliar essa correlação em órgãos genitais de cães naturalmente infectados por L. infantum. Para tal, amostras de testículos, epidídimos, próstata, glande, prepúcio e escroto foram coletadas de 19 cães adultos positivos. Lesões microscópicas associadas com imunomarcação de formas amastigotas do parasito foram observadas nos testículos (5,2%), epidídimos (2,6%), prepúcio (5,2%) e escroto (5,2%). Por outro lado, alterações estruturais sem imunomarcação foram observadas em todos os órgãos. Além disso, o DNA de L. infantum foi encontrado em todos os órgãos com maior carga parasitária nos testículos, epidídimos e próstata. Curiosamente, os testículos apresentaram a maior carga parasitária, porém apresentaram o menor grau de lesões inflamatórias. Essas lesões inflamatórias foram observadas em todos os órgãos do sistema reprodutor de cães com L. infantum. Entretanto, não houve correlação entre carga parasitária e lesões inflamatórias nestes órgãos.

ABSTRACT: Canine visceral leishmaniasisis an important disease caused by the protozoon Leishmania infantum which affects several organs and systems, including the genital tract. The L. infantum tropism to the male genital system and correlation among parasite load, immunohistochemistry detection and structural changes in these organs is controversy. Therefore, the aim of this study was to evaluate this correlation in the genital organs of the male dogs naturally infected with L. infantum. Samples from testicles, epididymis, prostate, glans penis, prepuce and scrotum were collected from 19 positive adult dogs. Structural changes were observed in the testicles (5.2%), epididymis (2.6%), prepuce (5.2%) and scrotum (5.2%) of the samples positive at immunohistochemistry examination. Conversely, similar structural changes were observed in all tissues negative at immunohistochemistry analysis. Moreover, L. infantum DNA was detected in all organs with the highest parasite load found in testicles, epididymis, and prostate gland. , Testicles had the highest parasite load but the lowest number of inflammatory lesions. These inflammatory lesions were observed in all organs of reproductive system of dogs; however, no correlation was observed between the parasite load and inflammatory lesions of dogs naturally infected with L. infantum.

O objetivo do estudo foi testar a prevalência sorológica e molecular de Anaplasma marginale em búfalos do municipio de Soure, Ilha de Marajó, estado do Pará, Brasil. Para a pesquisa sorologica foram selecionados randomicamente 800 animais e para a pesquisa molecular 50 destes animais foram aleatoriamente escolhidos. Para quantificar a prevalência sorológica utilizou-se o ensaio de imunoadsorção enzimático indireto (iELISA) com antígeno total contendo proteínas de superfície externa e para quantificar a prevalência molecular utilizou-se a reação em cadeia da polimerase (PCR), envolvendo a amplificação de fragmento gênico da proteína de superfície maior 5 (MSP5). A prevalência de animais positivos no ELISA para A. marginale foi de 25% (200/800). Na PCR foi detectada a presença de A. marginale em 2% (1/50) dos animais. Embora apenas um animal tenha sido positivo na PCR, observou-se que o mesmo foi negativo no ELISA. A presença do agente, mesmo em baixa prevalência, mostra que os bubalinos podem funcionar como um importante reservatório desse patógeno para os rebanhos bovinos da região norte do Brasil.

The aim of the study was to test the molecular and serological prevalence of Anaplasma marginale in water buffaloes of the Marajó Island, State of Pará, Brazil. For serologic research were randomly selected 800 buffaloes and for molecular research 50 of these animals were randomly chosen. To quantify the serological prevalence we used the indirect enzyme linked immunosorbent assay (iELISA) with total antigen containing proteins outer surface. To quantify the prevalence molecular was used the polymerase chain reaction (PCR) involving gene amplification fragment larger surface protein 5 (MSP5). The prevalence of positive animals in iELISA was 25% (200/800). In the PCR we detected the presence of A. marginale in 2% (1/50) of animals. Although only one animal was positive in PCR, we found that it was negative in ELISA. The presence of the agent, even in low prevalence, shows that buffaloes can act as an important reservoir for transmission of the pathogen to cattle in northern Brazil.

O objetivo do trabalho foi avaliar a eficácia de tratamentos fitoterápicos, homeopáticos e alopáticos contra ecto e endoparasitas na recria de novilhas, em dois ciclos experimentais subsequentes de 318 e 313 dias. Tratamentos: CN - controle negativo - não tratado; tratamento HH - tratado com homeopático Homeo bovis Parasitário®; tratamento CP – controle positivo – tratado com moxidectina 10%® e uma formulação acaricida contendo cypermetrina, clorpirifós e butóxido de piperonila®; tratamento HF - tratado com homeopático Fator C&MC® e tratamento FN - tratados com torta de neem® e com óleo de neem®. A contagem de ovos nas fezes (OPG), a infestação por Haematobia irritans, Rhipicephalus (Boophilus) microplus e pesagem dos animais foram realizadas em intervalos de 28 dias. No primeiro ciclo foi coletado sangue para avaliar a resposta imume. A infestação por mosca-dos-chifres não foi influenciada por nenhum dos tratamentos (P>0,05). Nos dois ciclos o número médio de carrapatos foi baixo e apenas no primeiro foi menor (P>0.05) no tratamento CP em relação aos demais. A média de OPG no tratamento CP foi mais baixa que nos demais tratamentos nos dois ciclos experimentais (P<0,05). Não houve efeito de tratamentos na resposta imune. Os animais do tratamento CP ganharam entre 22 a 30 kg de peso vivo a mais (P<0,05) que os não tratados ou tratados com medicamentos alternativos.

The aim of this study was to evaluate the efficacy of herbal, homeopathic and allopathic treatments for parasites in beef heifers during two experimental cycles of 318 and 313 days. Treatments: NC - negative control (untreated); HH - treated with homeopathic preparation Homeo bovis Parasitário®; PC - (positive control) - treated with 10% moxidectina® and an acaricide formulation of cypermethrin, chlorpyrifos and piperonyl butoxide®; HF – treated with homeopathic preparation Fator C&MC®; and FN - treated with neem cake (torta de neem®) and with neem oil (óleo de neem®). Parasite egg count (EPG), horn fly (Haematobia irritans) and tick (Rhipicephalus (Boophilus) microplus) assessment and animal weighting were performed at 28-day intervals. Blood samples were collected at the first cycle to assess the immune response. Horn fly infestation was not affected by any treatment (P>0.05). The mean number of ticks, which was low in both cycles, was lower (P<0.05) in the first cycle in animals that received PC treatment. In both experimental cycles, the mean EPG of the PC-treated animals was lower (P<0.05) than the animals receiving other treatments. Treatments had no effect on the immune response (P>0.05). The animals treated with allopathic drugs were 22 to 30 kg heavier (P<0.05) than untreated animals or animals treated with alternative drugs.

O objetivo do presente estudo foi quantificar a carga parasitária deLeishmania infantum em cães pela técnica de PCR em tempo-real (qPCR). Amostras de medula óssea, linfonodo e baço foram obtidos de 24 cães sorologicamente positivos para L. infantum que foram submetidos à eutanásia pelo serviço de vigilância oficial. Segundo os sinais clínicos, os animais foram classificados em assintomáticos ou sintomáticos. Após extração de DNA, as amostras foram submetidas à qPCR para detecção e quantificação de DNA de L. infantum. Dos 24 cães, 12,5% (3/24) foram classificados como assintomáticos e 87,5% (21/24) como sintomáticos. A PCR em tempo real detectou DNA de L. infantum em 100% dos animais, em pelo menos uma amostra biológica. Considerando cada órgão isoladamente, foi observada uma positividade de 100% em medula óssea e linfonodo, já nas amostras de baço 95,9% (23/24) foram positivas. Não foi observada diferença estatística entre a quantidade média geral de DNA entre as diferentes amostras biológicas (P = 0,518). Considerando os diferentes grupos clínicos, a carga parasitária média geral variou significantemente (P = 0,022). De acordo com os resultados obtidos não foi possível eleger a mais apropriada amostra biológica para o diagnóstico, baseado apenas na carga parasitária. Portanto, outras características como a conveniência e a facilidade de obtenção da amostra devem ser consideradas.

The aim of the present study was to quantify the parasite load ofLeishmania infantum in dogs using real-time PCR (qPCR). Bone marrow, lymph node and spleen samples were taken from 24 dogs serologically positive for L. infantum that had been put down by the official epidemiological surveillance service. According to the clinical signs the dogs were classified as asymptomatic or symptomatic. After DNA extraction, the samples were subjected to qPCR to detect and quantify L. infantum DNA. Out of the 24 dogs, 12.5% (3/24) were classified as asymptomatic and 87.5% (21/24) as symptomatic. Real-time PCR detected L. infantum DNA in all the animals, in at least one biological sample. In particular, 100% of bone marrow and lymph node scored positive, whereas in spleen, the presence of DNA was detected in 95.9% (23/24). In addition, out of 24 animals, 15 were microscopically positive to amastigote forms of L. infantum in bone marrow. No statistical significant difference was found in the overall mean quantity of DNA among the different biological samples (P = 0.518). Considering each organ separately, there was 100% positivity in bone marrow and lymph nodes, while among the spleen samples, 95.9% (23/24) were positive. Regarding the different clinical groups, the overall mean parasite load varied significantly (P = 0.022). According to the results obtained, it was not possible determine which biological sample was most suitable tissue for the diagnosis, based only on the parasite load. Therefore, other characteristics such as convenience and easily of obtaining samples should be taken into consideration.

A importância do cão como reservatório de L. infantum chagasi no meio urbano tem estimulado a realização de inúmeros trabalhos de avaliação de técnicas de diagnóstico, uma vez que este procedimento, quando realizado corretamente, torna-se um importante passo na prevenção da doença em humanos. Dentre os métodos de diagnóstico, as técnicas moleculares têm adquirido destaque. Objetivou-se neste trabalho verificar o desempenho da Reação em Cadeia da Polimerase (PCR) e da PCR em tempo real (qPCR) para diagnóstico da Leishmaniose Visceral Canina (LVC) utilizando diferentes amostras biológicas. Para tanto foram utilizados 35 cães provenientes de uma área endêmica para LVC, onde foram utilizados para o diagnóstico molecular, aspirado de medula óssea, fragmentos de linfonodo e baço. Neste estudo a qPCR foi capaz de detectar um maior número de animais positivos quando comparada com a PCR. Já entre as diferentes amostras biológicas utilizadas não foi observada diferença significativa na detecção de DNA de L. infantumchagasi por meio da PCR e qPCR. Mesmo assim, considerando a facilidade de obtenção, o linfonodo pode ser considerada como a melhor amostra para diagnóstico molecular da infecção por L. infantum chagasi.

The importance of dogs as a reservoir for Leishmania infantumchagasi in urban environments has stimulated numerous studies assessing diagnostic techniques. When performed properly, such procedures are an important step in preventing leishmaniasis in humans. Molecular methods have become prominent for this purpose. The aim of the present study was to determine the performance of the polymerase chain reaction (PCR) and real-time PCR (qPCR) for diagnosing of canine visceral leishmaniasis (CVL) using different biological samples. For this, 35 dogs from an area endemic for CVL were used. Bone marrow aspirate and lymph node and spleen fragments from these dogs were used for the molecular diagnosis. In the present study, qPCR was able to detect a greater number of positive animals than seen with PCR. Among the different biological samples used, there was no significant difference in L. infantumchagasi DNA detection between PCR and qPCR. However, considering that lymph nodes are easy to acquire, these can be considered to be the best samples for making molecular diagnoses of L. infantum chagasi infection.

This work reports a survey of Leptospira spp in pampas deer (Ozotoceros bezoarticus) in the Pantanal wetlands of the state of Mato Grosso do Sul, Brazil by serology and polymerase chain reaction (PCR). Seventy pampas deer were captured in the dry season and surveyed using PCR, microscopic agglutination test (MAT) (n = 51) and by both techniques (n = 47). PCR detected infections in two pampas deer and MAT detected infections in three. Through sequencing and phylogenetic analyses, the PCR-amplified fragment detected in deer was identified as Leptospira interrogans. Serovars Pomona and Butembo were detected using MAT and the highest titre was 200 for serovar Pomona. Epidemiological aspects of the findings are discussed.

Neste trabalho, três isolados de Plasmodium juxtanucleare foram analisados com base na morfologia, morfometria e parâmetros parasitológicos. Cada isolado foi coletado de aves (Gallus gallus) adultas infectadas naturalmente de áreas rurais de três municípios brasileiros: Seropédica (22º 48' S; 43º 41' W), no estado do Rio de Janeiro; Cruzeiro (22º 33' S; 44º 57' W), no estado de São Paulo; e Santa Bárbara do Tugúrio (21º 15' S; 43º 27' W), no estado de Minas Gerais. As amostras de sangue coletadas de cada ave infectada foram inoculadas em três grupos de dez aves (21 dias de idade). Esfregaços sangüíneos das aves infectadas experimentalmente foram realizados de dois em dois dias durante um período de 69 dias para avaliar a parasitemia. Para análises morfofisiológica e morfométrica, foram mensurados 30 indivíduos de cada estágio intraeritrocítico. Foram tomadas as medidas do diâmetro maior (DM) e diâmetro menor (dm), com os quais foi estimado o índice morfométrico (Mi=md/MD) e o tamanho (T=pab, a= md/ 2; b=MD/2). Os resultados indicaram uma parasitemia homogênea entre os três isolados, havendo diferenças apenas nas formas e tamanhos dos estádios parasitários.

In this work, three isolates of Plasmodium juxtanucleare have been analyzed based on morphological, morphometric and parasitic parameters. Each isolate was sampled from naturally infected adult chicken (Gallus gallus) from rural areas of three Brazilian municipalities: Seropédica (22º 48' S; 43º 41'W), in the state of Rio de Janeiro; Cruzeiro (22º 33' S; 44º 57'W), in the state of São Paulo; and Santa Bárbara do Tugúrio (21º 15' S; 43º 27' W), in the state of Minas Gerais. The blood samples taken from each infected chicken were inoculated in three groups of ten young chicken (21 days old). Blood smears of the experimentally infected chicken were sampled every two days until the 69th day in order to evaluate the parasitemia. For the morphological-descriptive and morphometric analyses, we measured 30 individuals from each of the intraerythocytic states, measures of the major (MD) and minor diameters (md), the estimation of morphometric index (Mi=md/MD) and size (T=pab, a= md/2; b=MD/2). The results indicated low and homogeneous parasitemia rates in the three strains, which showed differences among shape and size of the parasitic stadia displayed.

Amostras de soro obtidas de estudantes do curso de Medicina Veteriná ria da Universidade para o Desenvolvimento do Estado e da Região do Pantanal, Campo Grande, MS, Brasil, foram examinadas para a presença de anticorpos contra Toxoplasma gondii. Dos 145 soros testados, 44 (30,34%) foram positivos na hemaglutinação, com título igual ou superior a 1:16. Não foram observadas associações entre as caracterí sticas epidemiológicas examinadas, tais como hábitos alimentares (ingestão de carne bovina crua ou malpassada, vegetais crus/não lavados, produtos lácteos não pasteurizados) ou contato constante com cães e a presença de anticorpos contra T. gondii, exceto pelo percentual significativamente maior de estudantes soropositivos que relataram ter contato freqüente com gatos (P=0,03).

Serum samples obtained from students of the School of Veterinary Medicine, Universidade para o Desenvolvimento do Estado e da Região do Pantanal, Campo Grande, MS, Brazil, were examined for Toxoplasma gondii gondii antibodies. Serum samples from 44 (30.34%) of the 145 veterinary students examined were positive on the hemagglutination test, yielding a titer of 1:16 or greater. There were no relations between the epidemiological characteristics examined, such as food habits (eating of raw or rare-cooked cattle meat, raw/uncleaned vegetables, unpasteurized dairy products), frequent contact with dogs and the presence of T. gondii antibodies, except for a significantly higher percentual of seropositive students that reported frequent contact with cats (P=0.03).

De fevereiro a outubro de 1998, avaliou-se a contaminação de praças públicas de Campo Grande, MS, Brasil, por ovos de Toxocara e Ancylostoma, em amostras de fezes de cães. Das 74 praças examinadas, 42 (56,8%) estavam contaminadas por ovos de Ancylostoma, 8 (10,8%) com ovos de Toxocara e 7 (9,5%) com ambos.

From February to October 1998, the contamination of public squares of Campo Grande, MS, Brazil, by eggs of Toxocara and Ancylostoma was evaluated in dog fecal samples. Out of the 74 public parks examined, 42 (56.8%) were found to be contaminated with Ancylostoma eggs, 8 (10.8%) with Toxocara eggs and 7 (9.5%) with both.
