Um experimento foi conduzido em câmara de crescimento da Embrapa Milho e Sorgo, MG, visando avaliar características da rizosfera de genótipos de milho contrastantes na eficiência de uso de P, por meio da determinação do comprimento e densidade de pelos radiculares, do pH da rizosfera e da diversidade funcional bacteriana associada à rizosfera. Utilizou-se amostra de um Latossolo Vermelho distrófico, que, após a correção da acidez, recebeu adubação com N, K e micronutrientes. No tratamento correspondente ao nível alto de P, adicionaram-se 174 mg kg-1 de P. Como unidade experimental, utilizou-se rizobox de PVC, que recebeu 2,2 dm-3 de terra. O delineamento experimental foi o inteiramente casualizado, com três repetições, em arranjo fatorial 5 x 2, correspondendo a cinco genótipos (H1, H2 e H3 = híbridos eficientes, H4 e H5 = híbridos ineficientes) e dois níveis de P (baixo = 3 mg dm-3; alto = 29 mg dm-3). Aos 18 dias do transplantio, verificou-se que as raízes nodais dos híbridos H3 e H2 apresentaram maiores comprimentos de pelos radiculares. Em geral, o pH da rizosfera dos diferentes genótipos foi maior do que o pH não rizosférico, independentemente do nível de P. O pH da rizosfera de raízes laterais foi superior ao das nodais. Sob baixo nível de P, os híbridos H2, H5 e H4 mostraram maior variação de pH rizosférico em relação ao não rizosférico. Os híbridos H5 e H3 apresentaram maior atividade funcional microbiana na rizosfera. Sob baixo nível de P no solo, os índices de diversidade funcional microbiana, também, apresentaram maiores valores. A rizosfera dos híbridos H1, H2, H3 e H5 apresentou o mesmo perfil metabólico microbiano quando as plantas foram cultivadas sob baixo nível de P. As variações nas características da rizosfera não permitiram identificar padrões relacionados com a eficiência de uso de P nos materiais genéticos utilizados.

An experiment was conducted in a growth chamber to evaluate characteristics of the rhizosphere of maize genotypes contrasting in P-use efficiency, by determining length and density of root hairs, the rhizosphere pH and the functional diversity of rhizosphere bacteria. A sample of a Red Oxisol was limed and fertilized with N, K and micronutrients. In the treatment with the highest P level, 174 mg kg-1 P was added. Each experimental unit corresponded to a PVC rhizobox filled with 2.2 dm-3 soil. The experiment was completely randomized with three replications in a 5 x 2 factorial design, corresponding to five genotypes (H1, H2 and H3 = P-efficient hybrids, H4 and H5 = P-inefficient hybrids) and two P levels (low = 3 mg dm-3, high = 29 mg dm-3). It was found that 18 days after transplanting, the nodal roots of the hybrids H3 and H2 had the longest root hairs. In general, the pH in the rhizosphere of the different genotypes was higher than in non-rhizosphere soil, irrespective of the P level. The pH was higher in the rhizosphere of lateral than of nodal roots. At low P levels, the pH variation of the hybrids H2, H4 and H5 was greater in rhizospheric than in non-rhizospheric soil. The functional microbial activity in the rhizosphere of the hybrids H3 and H5 was highest. At low soil P levels, the indices of microbial functional diversity were also higher. The microbial metabolic profile in the rhizosphere of hybrids H1, H2, H3, and H5 remained unaltered when the plants were grown at low P. The variations in the rhizosphere properties could not be related to patterns of P-use efficiency in the tested genotypes.

O objetivo deste trabalho foi avaliar a diversidade funcional e genética de bactérias associadas à rizosfera de genótipos de milho contrastantes quanto à eficiência de uso de fósforo, por meio do teste de fontes de carbono no sistema EcoPlate e da eletroforese em gel de gradiente desnaturante (DGGE) dos fragmentos amplificados dos genes 16S ribossomais (rDNA) das bactérias. Foram coletadas amostras de solo da rizosfera de linhagens e híbridos contrastantes quanto à eficiência de uso de fósforo, cultivados em Latossolo Vermelho-Escuro fase cerrado, com baixo e alto teor de P. Bactérias da rizosfera de híbridos e linhagens eficientes, sob estresse de P, analisadas pelo sistema EcoPlate, tenderam a se agrupar conforme a análise de componentes principais, o que indica que utilizaram fontes de carbono semelhantes. Não houve diferença na diversidade bacteriana, analisada pela DGGE, entre bactérias associadas a genótipos eficientes e ineficientes no uso de P. Com base no sequenciamento do 16S rDNA, foi verificado que a rizosfera de genótipos de milho sob estresse de P parece selecionar grupos específicos de bactérias. A estrutura populacional genética e metabólica de bactérias da rizosfera foi mais influenciada pelo teor de fósforo no solo do que pela eficiência das plantas em usar o fósforo.

The objective of this work was to evaluate the functional and genetic diversity of bacteria associated to the rhizosphere of maize genotypes contrasting for phosphorus use efficiency by means of the EcoPlate carbon source test and denaturing gradient gel electrophoresis (DGGE) of amplified 16S ribosomal DNA (rDNA) fragments of bacteria. Rhizosphere soil samples of maize genotypes (hybrids and lineages) contrasting for phosphorus use efficiency cultivated in an Oxisol with high and low P content were collected. Bacteria from the rhizosphere of P-efficient maize genotypes under P stress conditions analyzed by the EcoPlate system tended to group together according to main components analysis, which indicates that these bacteria used similar carbon sources. Microbial diversity analyzed by DGGE did not differ between P-efficient and inefficient maize genotypes. Based on the 16S rDNA fragment sequencing, the rhizosphere of maize genotypes growing on low-P soils seemed to select specific bacteria species. The genetic and metabolic structure of the rhizosphere bacterial community was more strongly influenced by the level of P in the soil than by the genotypes with contrasting P-use efficiency.

Com o intuito de avaliar o efeito do estresse de P sobre atributos morfológicos do sistema radicular de genótipos de milho contrastantes quanto a eficiência na aquisição de fósforo, conduziu-se um experimento em casa-de-vegetação da Embrapa Milho e Sorgo, Sete Lagoas, MG, onde foram testados oito genótipos de milho, provenientes do programa de melhoramento da Empresa. O delineamento experimental utilizado foi o inteiramente casualizado, em esquema fatorial 8 x 2, com três repetições, correspondendo a oito genótipos (três linhagens: L1 = ineficiente, L2 e L3 = eficientes; e cinco híbridos: H1, H2 e H3 = eficientes, H4 e H5 = ineficientes) e dois níveis de P (baixo e alto). As plantas foram crescidas em solução nutritiva com duas concentrações de P equivalentes a 2,3 µM e 129 µM. A composição da solução nutritiva foi a seguinte, em mg L-1: N-NO3 152; N-NH4 18,2; Ca 141,1; K 90,1; Mg 20,8; S 18,8; Fe 4,3; Mn 0,5; B 0,27; Cu 0,04; Zn 0,15; Mo 0,08; Na 0,04 e HEDTA 20,06. Aos 18 dias do transplantio, verificou-se que, sob condições de estresse de P em solução nutritiva, os híbridos H5, H1 e H2 apresentaram os maiores valores de massa seca total e da parte aérea, em relação aos demais. De modo geral, tanto os híbridos como as linhagens, quando foram crescidos em solução com baixo nível de P, apresentaram maior relação raiz/parte aérea, do que os materiais que cresceram em solução com alto nível do nutriente. A concentração de N na parte aérea das plantas foi significativamente superior no tratamento com baixo nível de P na solução nutritiva. Os híbridos H1 e H2 e a linhagens L3 acumularam as maiores quantidades de P na parte aérea, independentemente do nível de P na solução. Esses resultados foram influenciados pelas maiores produções de produção de matéria seca apresentadas pelos referidos materiais. Independente do nível de P houve variação no conteúdo do nutriente na parte aérea dos híbridos, não sendo observado o mesmo comportamento para as linhagens.

With the objective of evaluating the effect of phosphorus stress on morphological attributes of the root system of corn genotypes contrasting in relation to phosphorus efficiency, an experiment was carried out in nutrient solution in greenhouse at Embrapa Milho e Sorgo, Sete Lagoas, MG, in the months of november and december of 2001. The experiment design was totally at random, in an 8 x 2 factorial scheme with three repetitions, corresponding to eight genotypes (three lineages: L1 = inefficient, L2 and L3 = efficient, and five hybrids: H1,H2 and H3 = efficient, H4 and H5 = inefficient) and two phosphorus levels (low = 2.3 mM and high = 129 mM). The seedlings were grown in nutrient solution with a following composition: in mg L-1: N-NO3 152; N-NH4 18,2; Ca 141,1; K 90,1; Mg 20,8; S 18,8; Fe 4,3; Mn 0,5; B 0,27; Cu 0,04; Zn 0,15; Mo 0,08; Na 0,04 e HEDTA 20,06. At 18 days after transplanting, the results showed that, under conditions of P stress in nutritive solution, the hybrids H5, H1 and H2 presented the largest values of total and shoot dry matter, in relation to the others. In general, the hybrids and inbred lines when were grown in solution with low level of P, they presented larger root-shoot relationship, than the materials grew in solution with high level of the nutrient. The concentration of N in the shoot of the plants was superior in the treatment with low level of P. The hybrids H1 and H2 and the inbred line L3 accumulated the largest amounts of P in the shoot, independently of the level of P in the solution. Independent of P level, there was variation in the nutrient content in the shoot of the hybrid, while in inbred lines were not observed the same behavior.

Objetivou-se comparar fertilizantes fosfatados em diferentes modos de aplicação, durante três cultivos sucessivos de milho, num Argissolo Vermelho já adubado anteriormente. Foi utilizado um fatorial 4 x 2+1, envolvendo quatro fontes de P (superfosfato triplo ST, termofosfato magnesiano TM, fosfato reativo de Arad FR e fosfato natural de Araxá FA), duas formas de aplicação (a lanço ou no sulco de plantio), e uma testemunha (sem P) como tratamento adicional. Foram fornecidos 180 kg ha-1 de P2O5 no primeiro cultivo, com base nos teores totais das fontes. Para os cultivos seguintes, não foi feito preparo do solo. Determinaram-se os teores de nutrientes no solo e nas folhas de milho e a produção de grãos. As respostas aos tratamentos foram mais discrepantes inicialmente e tenderam à eqüidade com os cultivos sucessivos. Nas duas primeiras safras, as fontes de maior solubilidade (ST e TM) ocasionaram as maiores produções. Os fosfatos naturais (FR e FA) apresentaram aumento de eficiência com o tempo. A aplicação localizada do FR proporcionou alta produtividade na terceira safra. O residual de antigas adubações e as condições climáticas influenciaram os efeitos dos tratamentos.

The objective of this study was to compare phosphate fertilizers in different methods of application during three successive maize cultivations in a Red Argisol (Hapludults) which had been previously fertilized. Treatments arranged in a 4x2+1 factorial design combined four P sources in the amount of 180 kg ha-1 P2O5 (triple superphosphate - TS, magnesium termophosphate - MT, Arad reactive rock phosphate - RP, and Araxá rock phosphate - AP), two application methods (broadcasted or banded in the seeding furrow), and a check plot (without P) as an additional treatment. The applied P ratio was based on the total P2O5 content of each fertilizer. After the first cultivation, the soil was no longer revolved. Soil and leaf nutrient concentrations, and the grain yield were determined. The responses to treatments were more contrasting initially and tended to level off with successive cultivations. In the first two cultivations, the most soluble sources (TS and MT) promoted greater yields. The rock phosphates (RP and AP) presented increased efficiency with time. Banded application of RP provided a higher grain yield in the third cultivation. The residual effect of previous fertilizations and the climatic conditions influenced the effects of the treatments.

O uso eficiente de fertilizantes fosfatados nos solos da região tropical ainda constitui um desafio, principalmente considerando o manejo de longo prazo. Com base nas respostas acumuladas de três cultivos sucessivos de milho, compararam-se alternativas de fornecimento de P, combinando fontes e modos de aplicação, num Argissolo Vermelho adubado em épocas passadas. O delineamento experimental foi o de blocos casualizados com quatro repetições, num esquema fatorial 4 x 3 + 1, envolvendo quatro fontes de P, na dose de 180 kg ha-1 de P2O5 (superfosfato triplo - ST, termofosfato magnesiano Yoorin - TM, fosfato reativo de Arad - FR e fosfato natural de Araxá - FA), três formas de aplicação (a lanço em área total no primeiro ano, localizada no sulco de plantio no primeiro ano e parcelada anualmente no sulco) e uma testemunha (sem aplicação de P), como tratamento adicional. O P foi fornecido, considerando os teores totais do nutriente nas fontes. Ao final dos três cultivos de milho, foram totalizados a produção de biomassa e de grãos e o acúmulo de P na parte aérea e nos grãos. Determinaram-se os teores de P residual no solo ao final do experimento. Foram calculados índices de eficiência agronômica e econômica. Foram detectadas mais diferenças em termos de absorção de P do que de produtividade de grãos. A maioria dos tratamentos proporcionou incrementos de produção semelhantes, o que, em parte, foi atribuído ao residual de adubações passadas e à eficiência genotípica do milho a P. As fontes mais solúveis, ST e TM, apresentaram desempenho similar. Para o FR, maior produção foi obtida com a aplicação parcelada no sulco, o que não se verificou no caso do FA. O parcelamento da dose total das fontes em aplicações anuais no sulco não comprometeu a produtividade do milho e propiciou maior efeito residual. Os tratamentos com maior eficiência agronômica (ST e TM) não corresponderam aos de maior eficiência econômica. Os dois fosfatos naturais (FR e FA) apresentaram relação benefício/custo mais compensadora.

The efficient use of phosphate fertilizers in soils of the tropical region is still a challenge, mainly regarding long-term soil management. Based on the cumulative response of three successive maize cultivations, alternatives of phosphorus supply combining P sources and application methods were compared in a formerly fertilized Red Argisol. Treatments arranged in a 4 x 3 + 1 factorial scheme represented by four P sources (180 kg ha-1 P2O5) as triple superphosphate - TS, magnesium termophosphate - MT, Arad reactive rock phosphate - RP, and Araxá rock phosphate - AP, and three application methods (broadcasted in the first year; along the sowing furrow in the first year; and annually split along the sowing furrow) and a check plot (without P application) as additional treatment. The applied P ratio was based on the total P2O5 content of each fertilizer. Cumulative biomass production, grain yield, and P accumulation in shoot and grain were determined. Residual available soil P was quantified at the end of the experiment. Indices of agronomic and economic efficiency were calculated. More differences between treatments were detected in P uptake than in grain yield. Most treatments provided similar yield increments, which was partly attributed to the residual effect of previous fertilizations and the genetic efficiency of P use by the corn plants. The most soluble sources, TS and MT, presented comparable performances. Unlike the AP rock phosphate, the split application of RP in the sowing furrow led to higher yields. Annual split applications along the sowing furrow did not affect the grain yield, and increased the residual effect. The treatments with greatest agronomic efficiency (TS and MT) were not the same as those with greatest economic efficiency. The two rock phosphates (RP and AP) presented the most favorable cost/benefit ratios.

O teor protéico, a expressão e a atividade de isoenzimas da peroxidase foram avaliados em ápices de raiz primária de plântulas de milho expostas a 222 µM de alumínio (Al) por 80 minutos. Duas linhagens com tolerância diferencial ao Al foram utilizadas: Cateto 237 (tolerante) e L36 (sensível). Os 20 mm apicais da raiz primária foi segmentado em intervalos de 2 mm e analisados para teor protéico e atividade da peroxidase. Os resultados demonstraram que o teor protéico ao longo do ápice da raiz da linhagem tolerante não foi afetado pelo Al, mas decresceu na linhagem sensível. O decréscimo na expressão de proteínas, verificado nos 2 mm apicais da raiz da linhagem sensível, foi devido principalmente às proteínas de baixo peso molecular, aproximadamente 43 KDa ou menor. A expressão de quase todas essas mesmas proteínas aumentou na linhagem tolerante, embora o teor protéico total não tenha sido alterado. Isto sugere que algumas destas proteínas possam ter um papel importante na tolerância das plantas aos metais, talvez como peptídeos ligantes. Enquanto a atividade da peroxidase na linhagem tolerante não alterou com a exposição ao Al, esta atividade decresceu ao longo dos 6 mm apicais da raiz na linhagem sensível. Mais isoformas aniônicas foram expressas constitutivamente na linhagem tolerante, mas sem contudo, apresentar alterações qualitativas em reposta ao Al. Os resultados demonstraram que a manutenção da expressão protéica pode ser um componente importante da tolerância das plantas ao Al, e que esta resistência está associada a expressão constitutiva de isoformas aniônicas da peroxidase.

The effects of a short-term (80 min) exposure to 222 µM aluminum (Al) on the protein content and expression and on peroxidase activity and isoenzymes in the primary root of maize were evaluated. Two inbred lines differing in their level of tolerance to Al were used: Cateto 237 (tolerant) and L36 (sensitive). The apical 20 mm of the primary root was divided into 2-mm-long segments that were analyzed for total protein content and peroxidase activity. These results demonstrate that the total protein content along the root apex was not affected by Al in the tolerant inbred line, but decreased in the sensitive line. In the apical 2 mm of the root of the sensitive line, the expression of low molecular weight proteins (43 kDa or smaller) was decreased. Expression of low molecular proteins increased in the tolerant inbred line, even though total protein content did not increase. This suggests that some of these proteins could play a role in metal tolerance, perhaps as binding peptides. While the peroxidase activity of the tolerant inbred line did not change with exposure to Al, peroxidase activity in the apical 6 mm of the root of the sensitive line decreased. The tolerant inbred line constitutively expressed more anionic peroxidase isoforms. These results demonstrate that maintenance of protein expression may be an important component of the plant's resistance to Al stress, and that resistance to Al stress is associated with the higher expression of anionic peroxidase isoforms.
