RESUMO Pitaya (Hylocereus undatus e H. costataricensis) encontra-se no grupo de frutas exóticas e seu cultivo no Brasil está em expansão, sobretudo na região semiárida do Nordeste. Nessa região os problemas de quantidade e qualidade hídrica normalmente ocasionam estresses ambientais limitando à sobrevivência das plantas, principalmente durante a germinação e estabelecimento de plântulas. Sendo assim, objetivou-se verifi car a ação mitigadora de tratamentos pré-germinativos em sementes de pitaya sob estresse salino. O experimento foi conduzido em delineamento inteiramente casualizado, em esquema fatorial 2 × 6, correspondente a duas espécies de pitayas H. undatus e H. costatariciensis e seis tratamentos pré-germinativos (T1 = 0,0 MPa (controle); T2 = estresse salino (-0,4 MPa); T3 = hidrocondicionamento + estresse salino; T4 = ácido giberélico + estresse salino; T5 = ácido salicílico + estresse salino; T6 = tiametoxan + estresse salino). Após 20 dias de semeadura, analisou-se a germinação, índice de velocidade de germinação, comprimentos de partes aérea e raiz primária, massa seca total, açúcares solúveis totais e aminoácidos livres totais. H. costaricensis mostrou-se mais tolerante à salinidade que H. undatus. O ácido salicílico estimulou a germinação e o crescimento de H. undatus, além de aumentar o teor de açúcares solúveis em H. costaricensis. O hidrocondicionamento das sementes atenuou o estresse salino durante a germinação de H. undatus e favoreceu o ganho de massa seca. O ácido giberélico estimulou a germinação de H. undatus e o crescimento de plântulas desta e de H. costaricensis, além disso aumentou os níveis de açúcares solúveis em H. undatus.

ABSTRACT Pitaya (Hylocereus undatus and H. costaricensis) is found in the group of exotic fruits and its cultivation in Brazil is expanding, especially in the semi-arid region of the Northeast. In this region, the problems of water quantity and quality usually cause environmental stresses which limit the survival of plants, especially during germination and seedling establishment. Thus, the objective was to evaluate the mitigating action of pre-germination treatments in pitaya seeds under salt stress. The experiment was conducted in a completely randomized design, in a 2 × 6 factorial scheme, corresponding to two species of pitaya (H. undatus and H. costaricensis) and six pre-germination treatments (T1 = 0.0 MPa (control); T2 = salt stress (-0.4 MPa); T3 = hydropriming + salt stress; T4 = gibberellic acid + salt stress; T5 = salicylic acid + salt stress; T6 = thiamethoxam + salt stress). After 20 days of sowing, germination, germination speed index, shoot length, primary root length, total dry mass, total soluble sugars and total free amino acids were analyzed. H. costaricensis was more tolerant to salinity than H. undatus. Salicylic acid stimulated the germination and growth of H. undatus, besides increasing the soluble sugar content in H. costaricensis. Seed hydropriming attenuated salt stress during germination of H. undatus and favored dry mass gain. Gibberellic acid stimulated the germination of H. undatus and the growth of H. undatus and H. costaricensis seedlings, in addition to increasing the levels of soluble sugars in H. undatus.

RESUMO Como os nematoides resistentes a drogas se tornaram um problema comum nas indústrias de ovinos e caprinos, métodos alternativos que utilizam produtos naturais surgiram como uma opção de tratamento anti-helmíntico viável e sustentável. Aqui, o efeito in vitro do óleo essencial extraído de Lippia gracilis Schauer foi avaliado no processo de eclosão de nematoides recuperados de caprinos naturalmente infectadas. O óleo essencial nas concentrações de 0,08% (0,008 μL/mL), 0,12% (0,012 μL/mL), e 0,16% (0,016 μL/mL)foi capaz de induzir uma inibição média de 74,7, 84 e 93%, respectivamente. A concentração efetiva necessária para inibir a eclosão de ovos em 50% dos ovos (CE50) foi de 0,03452%. Portanto, o óleo essencial de L. gracilis apresentou resultados anti-helmínticos in vitro promissores contra a eclosão de nematódeos gastrintestinais de caprinos.

ABSTRACT Since drug-resistant nematodes became a common problem in sheep and goat industries, alternative methods using natural products have emerged as a viable and sustainable anthelmintic treatment option. Here, the in vitro effect of essential oil extracted from Lippia gracilis Schauer was assessed on the hatching process of nematodes recovered from naturally infected goats. Essential oil at concentrations of 0.08% (0.008 μL/mL), 0.12% (0.012 μL/mL), and 0.16% (0.016 μL/mL) was able to induce an average inhibition of 74.7, 84 and 93%, respectively. The effective concentration required to inhibit egg hatching in 50% of eggs (EC50) was 0.03452%. Therefore, essential oil of L. gracilis showed promisor in vitro anthelmintic results against egg-hatching of goat gastrointestinal nematodes.

RESUMO: Este estudo teve como objetivo avaliar mecanismos fisiológicos e bioquímicos de adaptação de Hyptis suaveolens submetidas ao estresse osmótico decorrente da salinidade inoculadas com espécies de fungos micorrízicos. Sementes de H. suaveolens foram colocadas para germinar em vasos de polietileno contendo substrato associado, ou não, a FMA. Após o estabelecimento das plantas, foram iniciados os tratamentos com diferentes concentrações de sais (0,0; 35; 70 e 105 mM) e fungos (controle e dois tipos de FMA), perfazendo 12 tratamentos com três repetições. O delineamento experimental foi em blocos casualizados em esquema fatorial 4 x 3, perfazendo 12 tratamentos com três repetições cada. Observou-se, através de variáveis fisiológicas e bioquímicas, que a salinidade afetou todas as variáveis analisadas e o estresse promoveu redução no conteúdo de matéria seca. Verificou-se que plantas associadas com FMA apresentaram ganho de matéria seca em relação as não associadas e incremento de respostas bioquímicas e fisiológicas das plantas colonizadas no tratamento 35mM de NaCl. No entanto, o estresse salino afeta negativamente o desenvolvimento de H. suaveolens não sendo observado, portanto, atenuação dos fungos.

ABSTRACT: The objective of this study was to evaluate the physiological and biochemical mechanisms of adaptation of Hyptis suaveolens under osmotic stress due to salinity inoculations with a mycorrhizal fungal species. H. suaveolens seeds were germinated in polyethylene pots containing a substrate associated with or without arbuscular mycorrhizal fungi (AMF). After plant formation, treatments were treated with different salt concentrations (0.0, 35, 70, and 105 mM) and fungi (control and two types of AMF), totaling 12 treatments with three replicates. The experimental design used randomized blocks in a 4 x 3 factorial scheme, totaling 12 treatments with three replicates each. Salinity affected all measured physiological and biochemical variables, and the stress reduced dry matter content. Plants associated with AMF had increased dry matter compared to non-associated plants, and there were increased biochemical and physiological responses of AMF-colonized plants in the 35 mM NaCl treatment. However, saline stress negatively affected the development of H. suaveolens; and therefore, no attenuation of fungi was observed.

RESUMO O estudo teve como objetivo avaliar o enraizamento e alguns caracteres fisiológicos em estacas de Lantana camara L. (Verbenaceae) em diferentes concentrações de ácido indolbutírico (AIB) e sua associa- ção com os fungos micorrízicos arbusculares (FMA) Claroideoglomus etunicatum (W.N. Becker & Gerd.) C. Walker & A. Schüssler e Gigaspora albida N.C. Schenck & G.S. Sm. Utilizou-se o delineamento experimental inteiramente casualizado em esquema fatorial 3 × 3 com três repetições, sendo o primeiro fator composto pelos FMA e o segundo fator pelas doses de AIB (0; 1,0 e 1,5 mg.mL-1). Os resultados mostraram que a presença de FMA no substrato, assim como o AIB, isolados ou de forma interativa estimulam o enraizamento e alguns caracteres fisiológicos de estacas de L. camara.

ABSTRACT The objective of this study was to evaluate the rooting and some physiological characteristics of Lantana camara L. (Verbenaceae) stakes in different concentrations of indolebutyric acid (IBA) and its association with the arbuscular mycorrhizal fungi (AMF) Claroideoglomus etunicatum (W.N. Becker & Gerd.) C. Walker & A. Schüssler e Gigaspora albida N.C. Schenck & G.S. Sm. The experimental design was completely randomized in a 3x3 factorial scheme with three replicates, where the first factor was the AMF and the second factor by doses of IBA (0; 1.0 and 1.5 mg .mL-1). The results showed that the presence of AMF in the substrate, as well as the IBA, isolated or in association stimulates the rooting and some physiological characters of L. camara cuttings.

RESUMO: Lippia gracilis é uma planta nativa da Caatinga, rica em óleo essencial com atividade antimicrobiana. O trabalho objetivou avaliar o efeito de diferentes tipos de estacas e substratos na propagação vegetativa de L. gracilis . O experimento foi montado em delineamento inteiramente casualizado, em esquema fatorial 3 x 3, sendo 3 tipos de estacas (apical, mediana e basal) e 3 tipos de substratos (areia + húmus, argila + húmus e areia + argila + húmus), com 5 repetições cada. Aos 40 dias foram avaliadas: porcentagem de estacas brotadas e enraizadas, número médio de brotos, comprimento da maior raiz e biomassa seca dos brotos e raízes. Em todas as variáveis as estacas medianas e basais apresentaram melhores resultados do que as apicais. O substrato influenciou apenas na biomassa seca dos brotos, sendo o composto de argila + húmus superior ao areia + argila + húmus e igual, em significância, ao areia+ húmus.

ABSTRACT: Lippia gracilis is a native plant of the Caatinga, it is rich in essential oil with antimicrobial activity. The study aimed to evaluate the effect of different types of cuttings and substrates on vegetative propagation of L. gracilis. The experimental design was completely randomized, in factorial scheme 3 x 3, with 3 types of cuttings (apical, median and basal) and 3 substrates (sand + humus, clay + humus and sand + clay + humus), with 5 replications each. After 40 days were evaluated: percentage of sprouted cuttings and rooted, average number of shoots, length of roots and dry biomass of shoots and roots. In all variables the middle and basal cuttings showed better results than the apical. The substrate influenced only in the dry biomass of shoots. The substrate composed of clay + humus was better than the sand + clay + humus and equal in significance to the sand + humus.

Alecrim da Chapada (Lippia gracilis Schauer) é considerada uma planta rica em óleo essencial e possui atividade antimicrobiana comprovada, devido aos monoterpenos fenólicos carvacrol e timol. O objetivo deste trabalho foi avaliar o efeito in vitro do óleo essencial e dos extratos etanólicos de raízes e folhas de L. gracilis no controle do fungo Monosporasccus cannonballus, causador do colapso do meloeiro. O extratos foram testados nas concentrações de 2500, 5000 e 7500 ppm e o óleo essencial de L. gracilis foi avaliado nas concentrações 255, 340 e 425 ppm tendo também um controle positivo com o fungicida comercial Captan(r) e um negativo apenas com meio BDA (batata, dextrose, ágar). Discos de micélio com 3 mm de diâmetro foram inoculados no centro de placas de Petri e as medições do crescimento micelial do fungo foram realizadas 48 horas após a montagem do experimento. Verificou-se que o óleo essencial de L. gracilis nas três concentrações testadas foi eficiente para o controle do fungo, com percentuais de inibição de 100%, em comparação ao tratamento com o controle negativo (0%) que não apresentou inibição. O extrato radicular, nas concentrações de 5000 e 7500 ppm e o extrato foliar na concentração de 7500 ppm também proporcionaram um percentual de inibição de 100%.

"Alecrim da Chapada" (Lippia gracilis Schauer) is a plant which is considered to be rich in essential oil and proven to have antimicrobial activity due to the phenolic monoterpenes carvacrol and thymol. The aim of this study was to evaluate the in vitro effect of essential oil and ethanol extracts from roots and leaves of L. gracilis on the control of the fungus Monosporasccus cannonballus, the causal agent of melon collapse. The extracts were tested at the concentrations 2500, 5000 and 7500 ppm and L. gracilis essential oil was evaluated at the concentrations 255, 340 and 425 ppm; there was also a positive control with the commercial fungicide Captan(r) and a negative control only with PDA medium (potato, dextrose, agar). Mycelium discs of 3mm diameter were inoculated in the center of Petri plates and measurements of mycelial growth were performed at 48 hours after the establishment of the experiment. L. gracilis essential oil, at the three tested concentrations, was effective in controlling the fungus, showing inhibition percentages of 100%, compared to the treatment with the negative control (0%), which showed no inhibition. The root extract, at concentrations 5000 ppm and 7500 ppm, and the leaf extract, at a concentration of 7500 ppm, also provided inhibition percentage of 100%.

As flores tropicais têm contribuído para o aumento do mercado florístico no Brasil; dentre elas destacam-se as do gênero Heliconia. A propagação de helicônias que atinge maior número de mudas ocorre por via vegetativa, por meio da divisão de rizomas, aumentando a probabilidade de disseminação de doenças, podendo, num futuro próximo, comprometer a produção de flores. O cultivo in vitro surge como importante alternativa para proporcionar a produção de mudas de helicônia com elevado padrão de qualidade. Entretanto, a micropropagação de helicônias via ápices caulinares depara-se com problemas de contaminação endofítica, dificultando a multiplicação do explante. Diante disso, fez-se necessário avaliar alternativas que promovam a produção de mudas in vitro, por meio da utilização de outros explantes. Para tanto, realizou-se um experimento, utilizando embriões zigóticos, provenientes de frutos imaturos e maduros de H. bihai (L.) L. cv. Lobster Claw Two, em combinações de AIA (0; 5,70 e 11,41 µm L-1) e 2,4-D (0; 22,62; 45,24; 67,86 e 90,48 µm L-1), para induzir a formação de embriões somáticos durante 90 dias. Por meio de análise histológica, observou-se a formação de embriões somáticos apenas nos embriões zigóticos, provenientes de frutos maduros, cultivados na ausência de reguladores de crescimento.

Tropical flowers have contributed to the growth of the cut flower market in Brazil, especially plants of the genus Heliconia. Heliconias are usually propagated vegetatively, although this method can spread diseases and in the near future may severely comprise commercial flower production. In vitro cultivation represents an important potential alternative for guaranteeing the production of high-quality heliconia planting stocks. However, the micropropagation of these plants using stalk apices faces severe problems of endophytic contamination, making it difficult the supply of enough explants. Therefore, alternative techniques for the in vitro production of planting stock were investigated using other explant tissues to induce somatic embryogenesis. Zygotic embryos from immature and mature fruits of H. bihai (L.) L. cv. Lobster Claw Two were cultured in combinations of IAA (0; 5.70, e 11.41 µm L-1) and 2,4-D (0; 22.62; 45.24; 67.86 e 90.48 µm L-1) to induce somatic embryos during 90 days of culture. Histological analyses showed that somatic embryos were formed only with zygotic embryos derived from mature fruits cultivated in the absence of growth regulators.

Inicialmente os embriões procedentes de frutos imaturos e maduros de Heliconia bihai (L.) L. cv. Lobster Claw Two foram avaliados quanto à morfologia interna. Os embriões maduros foram inoculados em meio MS (1/2MS e MS completo) e GA3 (0, 2,5 e 5,0 mg L-1), utilizando sacarose ou glucose. Posteriormente, as plantas obtidas foram transferidas para meio 1/2MS acrescido de 0 ou 2.5 mg L-1 de BAP e aos 30 e 45 dias após a inoculação, avaliou-se a formação de gemas laterais. Aos 45 dias avaliou-se a variabilidade genética dessas plantas através da análise isoenzimática. Ao avaliar a morfologia interna observou-se que os embriões maduros apresentaram estádios de diferenciação superior aos embriões imaturos. Na fase de conversão do embrião em planta, observou-se que 85% dos embriões inoculados desenvolveram em plantas quando cultivados em meio ½MS com sacarose, contrastando com 41% cultivados no mesmo meio nutritivo acrescido de glucose. Na fase de multiplicação, as plantas cultivadas em meio ½MS acrescido de 2.5 mg L-1 de BAP apresentaram maior formação de gemas laterais. A análise isoenzimática apresentou diferença na intensidade de coloração e migração de algumas bandas. Esse comportamento pode estar associado com a diferença fisiológica relacionada à idade entre a planta mãe e as plantas provenientes do cultivo de embrião in vitro.

The internal morphology of embryos from immature and mature fruits of Heliconia bihai (L.) L. cv. Lobster Claw Two was examined. Embryos were inoculated into MS media (full MS and ½ MS) and GA3 (0, 2.5 and 5 mg L-1) with either sucrose or glucose. These plantlets were then replicated and transferred to MS medium (full MS or ½ MS) with 0 or 2.5 mg L-1 BAP and their multiplication was evaluated 30 and 45 days after inoculation. The genetic variability of the multiplied plants was estimated using isoenzyme analyses. The internal morphology of the mature embryos revealed their tissues to be in more advanced stages of differentiation than immature embryos. In the conversion phase, 85% of the inoculated embryos developed into plants in the ½ MS medium with sucrose, in contrast to only 41% of the embryos that were cultivated with glucose. In the multiplication phase, plants cultivated in ½ MS medium with 2.5 mg L-1 BAP demonstrated more buds. Isoenzyme analyses showed pattern changes in terms of the color intensity and the migration of some of the bands. These results may be associated with differences in the ages of the mother plants and of the plantlets obtained in vitro.

Este trabalho teve como objetivo avaliar a resposta de secções transversais de ovários e o desenvolvimento de calos embriogênicos. O experimento constou de dois ensaios. No primeiro avaliou-se combinações entre AIA (0; 5.71; 8.56; 11.42; 14.27μM) e 2,4-D (0; 13.57; 18.10; 22.62μM) e no segundo avaliou-se as concentrações de 2,4-D supracitadas, combinadas com concentrações de BA (0; 4.43; 6.65; 8.87; 11.09μM). Os calos formados foram avaliados quanto à presença de estruturas embriogênicas utilizando-se estereomicroscópio, microscópio eletrônico de varredura, além de análises citoquímicas e histológicas. Combinações entre 2,4-D e AIA induziram a formação de calos friáveis com setores pró-embriogênicos, refletindo a aquisição de competência morfogenética. Posteriormente foi observada a expressão do programa embriogênico quando massas pró-embriogências desenvolveram-se formando embriões somáticos. Esses resultados apresentam uma alternativa para a utilização de um tipo de explante que possibilita o cultivo in vitro de helicônia sem o desenvolvimento de bactérias endofíticas.

The present work evaluated the development of embryogenic callus from transversal ovary sections. The experiments were carried out under two experimental regimes using combinations of IAA (0; 5.71; 8.56; 11.42; 14.27μM) and 2,4-D (0; 13.57; 18.10; 22.62μM) or combinations of 2,4-D with BA (0; 4.43; 6.65; 8.87; 11.09μM). Assessments were made of anatomical aspects of the callus and for the presence of embryogenic structures using cytochemical and histological analyses and stereomicroscopic and scanning electronic microscopic observations. Treatments with 2,4-D and IAA produced friable calluses demonstrating cellular acquisition of morphogenetic competence as well as the formation of pro-embryogenic sectors. The expression of embryogenic program could be observed, with proembryogenic cell clusters developing into globular embryos. These results offer the possibility of using new types of explants for culturing helicons that avoid the growth of endophytic bacteria.

Objetivou-se, neste trabalho, avaliar in vitro a atividade dos extratos alcoólicos de Senna alata (L.) Roxb. sobre Monosporascus cannonballus. O delineamento experimental foi inteiramente casualizado com esquema fatorial 3 x 6 + 1, sendo constituído por três partes da planta (caule, raiz e vagem), por seis concentrações (0,25; 0,50; 50; 75; 250; 500 ppm ) mais a testemunha, com quatro repetições por tratamento. As variáveis avaliadas foram: taxa de crescimento micelial (TCM), inibição do crescimento micelial (ICM) e área abaixo da curva do crescimento micelial (AACCM). A concentração de 500 ppm foi a mais eficiente, nas três partes vegetais avaliadas. O porcentual de inibição na referida concentração foi de 28,60%, 36,70% e 27,99% para caule, raiz e vagem respectivamente. Diante dos resultados, observa-se que os extratos de S. alata indicam um forte potencial de controle para o fungo Monosporascus cannonballus.

The aim of this work was to evaluate the activity in vitro of the ethanolic extracts of Senna alata (L.) Roxb. over Monosporascus cannonballus. The experimental design was entirely randomized with factorial scheme 3 x 7 + 1, being constituted by three parts of the plant (root, stalk and green bean) for six different concentrations (0.25; 0.50, 50, 75, 250,500 ppm) plus the control, with four repeatitons for treatment. The available variables were: rate of micelial growth (TCM), inhibition of the growth micelial (ICM) and area under the curve of the growth micelial (AACCM). The concentration of 500 ppm was the most efficient ,in the three available vegetal parts. The percentual of inhibition in the concentration was of 28.60%, 36.70% e 27.99% for stalk, root and green bean respectively. Before of the results, the extracts of S. alata display high control potential for Monosporascus cannonballus fungi.

Avaliou-se a atividade antimicrobiana do óleo essencial (OE) de Lippia gracilis Schauer sobre fungos contaminantes de laboratórios de cultura de tecidos vegetais e bactérias endofíticas de helicônias. Discos de micélio dos fungos Geotrichum candidum; Trichoderma viride; Torula herbarum; Paecillomyces sp.; Aspergillus nidulans; Fusicoccum sp.; A. flavus; P. aeruginens; Curvularia lunata e A. niger, foram inoculados em meio BDA com óleo (0; 420; 440; 460; 480 e 500 µL L-1). O percentual de inibição em 420 µL L-1 foi de 100% para todos os fungos exceto paraC. lunata and A. niger, o qual foi de 95.58% and 89.40% respectivamente. Em outro experimento, suspensão das espécies Salmonela choleraceuis-diarizonae, Enterobacter asburiae, Bacillus thuringiensis, B. pumilus, Klebsiella pneumoniae, E. hormaechei, Bacillus pumilus, B. cereus, B. pumilus foi individualmente adicionada ao meio NYDA, que foi vertido em placas de Petri e, após solidificado, perfurado e preenchido com água ou óleo (420 µL L-1). Avaliou-se o diâmetro do halo de inibição. Avaliou-se também, a inibição em meio líquido. K. pneumoniae foi a espécie bacteriana menos sensível e E. hormaechei foi a mais sensível.

Antimicrobial activity of the essential oil (EO) of Lippia gracilis Schauer over fungi and heliconia endophytic bacteria was evaluated. For the fungi Geotrichum candidum; Trichoderma viride; Torula herbarum; Paecillomyces sp.; Fusicoccum sp.; P. aeruginens; Curvularia lunata; Aspergillus nidulans; A. flavus; and A. niger mycelial discs were inoculated in PDA medium with oil (0; 420; 440; 460; 480 e 500 µL L-1). The inhibition percentage in 420 µL L-1 was 100% for all fungi, except for C. lunata and A. niger, which was 95.58% and 89.40% , respectively. In other experiment, a suspension of the Salmonella choleraceuis-diarizonae, Enterobacter asburiae, Bacillus thuringiensis, B. pumilus, B. cereus, Klebsiella pneumoniae, and E. hormaechei was individually added to the solid NYDA medium dishes. In each dish, wells were drilled in medium and filled with water or EO (420 µL L-1). The diameter of the inhibition halo was assessed. K. pneumoniae was the bacterial species less sensitive and E. hormaechei was the most sensitive.
