ABSTRACT Botrytis cinerea is a ubiquitous necrotrophic filamentous fungal pathogen, causing the “grey mold” disease in a wide range of plants, including species of importance in the food industry. Recently, we reported the use of fac-[Re(I)(CO)3(2,2’-bpy)L]PF6 (C3) and fac-[Re(I)(CO)3(4,4′-dimethyl-2,2′-bpy)L]PF6 (C4) complexes as biomarkers for yeasts and bacteria. Nevertheless, the use of these complexes to stain filamentous fungi, such as Botrytis cinerea, has not been explored so far. Since Botrytis cinerea exhibits a dynamic multilayer cell wall that changes the composition during normal growth, in both their hyphae and conidia, is not possible to directly extrapolate the use of both C3 and C4 as biomarkers for Botrytis cinerea without an experimental approach. In this work, we explored new features of the luminescent compounds C3 and C4 as biomarkers for the higher filamentous fungi Botrytis cinerea, including conidia and juvenile hyphae. We found that, with the new protocol proposed, both C3 and C4 were suitable to stain conidia with a simple procedure since no cell permeabilization is required. Additionally, these results suggest that C3 and C4 can selectively stain living conidia, allowing the differentiation from non-germinating conidia. This point opens a new focus of development for rhenium (I) tricarbonyl complexes as new fluorescent biomarkers for Botrytis cinerea with potential properties as vital staining.

The secretion of enzymes involved in biocontrol from the wild T. harzianum strains Th11 and Th12 and the mutants Th11A80.1, Th11C40.1 and Th12A10.1 was studied after their cultivation in the presence of R. solani cell walls as the sole carbon source. The results showed that endoprotease activity in the supernatants from Th11A80.1 and Th11C40.1 increased 2.77 and 4.79-fold, respectively, and the P-1,3-glucanase and P-1,4-chitinase activity from Th12A10.1 increased 1.86 and 1.54-fold, respectively, compared with the corresponding parental strains. The role of the three enzymes in the biocontrol of R. solani is discussed.

La secreción de enzimas involucradas en biocontrol por las cepas silvestres de T. harzianum Th11 y Th12, y de sus mutantes Th11A80.1, Th11C40.1 y Th12A10.1, se analizó luego de cultivarlas en medio líquido usando paredes celulares de R. solani como única fuente de carbono. Los resultados mostraron que Th11A80.1 y Th11C40.1 aumentaron la secreción de endoproteasas 2,77 y 4,79 veces, respectivamente; Th12A10.1 aumentó 1,86 veces la secreción de ß-1,3-glucanasa y 1,54 veces la de ß-1,4-quitinasa al compararlas con las cepas parentales. Se discute el rol de estas tres enzimas para el biocontrol de fitopatógenos.

Biorend SC (quitosana), BC-1000 EC (extrato de toranja mais flavonóides), e ECO-100 SC (bioflavonóides mais ácidos orgânicos, fitoalexinas cítricas, ácidos graxos glicerídeos e açúcares) inibiram em 80,1%, 79,0% e 76,5%, respectivamente, a podridão causada por Botrytis cinerea quando utilizados no tratamento pós-colheita de frutos de maçã sob condições controladas.Tratamento combinado de Biorend SC, com aplicação tanto em pré como no pós-colheita, proporcionou 49,9% de inibição da podridão, enquanto BC-1000 e ECO-100 EC não foram efetivos. Nenhum desses produtos inibiu a podridão cinzenta, quando utilizados em tratamento de pré-colheita em condições controladas ou em tratamento de pós-colheita em condições comerciais. O polissacarídeo algal ulvana, utilizado nos tratamentos de pós-colheita, reduziu em 56% a podridão cinzenta das maçãs em condições controladas, mas não teve efeito inibitório nos tratamentos combinados de pré e pós-colheita. A incapacidade dos produtos em ativar mecanismos de defesa (quitinases e peroxidases) nos frutos, após o tratamento em pré-colheita, foi consistente com a falta de controle da podridão nesse tipo de ensaio. Pelos resultados, sugere-se que os produtos naturais utilizados apresentam potencial para a utilização no manejo integrado da podridão de Botrytis quando aplicados em pós-colheita.

Biorend SC (chitosan), BC-1000 EC (grapefruit extract plus bioflavonoids) and ECO-100 SC (bioflavonoids plus organic acids, citric phytoalexins, fatty acids, glycerides and sugars), respectively, suppressed grey rot of apple caused by B. cinerea by 80.1%, 79.0% and 76.5% when used as post-harvest treatments under controlled conditions. When applied as combined pre- and post-harvest treatments Biorend SC inhibited fruit rot by 49.9 %, while BC-1000 EC and ECO-100 SC were ineffective. None of the products inhibited fruit rot when applied as pre-harvest treatments under controlled conditions or as post-harvest treatments under commercial conditions. The algal polysaccharide ulvan used in post-harvest treatments suppressed grey rot by 56.0% under controlled conditions, but had no inhibitory effect on combined pre- and post-harvest treatments. The inability of products to activate defense mechanisms (chitinase and peroxidase) of fruits was consistent with the unsuccessful control of rot by pre-harvest treatment. The results suggest that the natural products used have potential for use in integrated management of Botrytis rot when applied after harvest.

In this report we describe the isolation and characterization of a gene encoding the transcription factor Acel (Activation protein of cup 1 Expression) in the white rot fungus Phanerochaete chrysosporium. Pc-acel encodes a predicted protein of 633 amino acids containing the copper-fist DNA binding domain typically found in fungal transcription factors such as Acel, Macl and Haal from Saccharomyces cerevisiae. The Pc-acel gene is localized in Scaffold 5, between coordinates 220841 and 222983. A S. cerevisiae acel null mutant strain unable to grow in high-copper medium was fully complemented by transformation with the cDNA of Pc-acel. Moreover, Northern blot hybridization studies indicated that Pc-acel cDNA restores copper inducibility of the yeast cup 1 gene, which encodes the metal-binding protein metallothionein implicated in copper resistance. To our knowledge, this is first report describing an Acel transcription factor in basidiomycetes

The development of an effective hypersensitive response (HR) in any plant system relies, not only in their gene composition and expression, but also on an effective and rapid signal transduction system. Lemon seedlings induce the phenylpropanoid pathway, which results in the de novo biosynthesis of the phytoalexin scoparone, as part of the hypersensitive response against Alternaria alternata. In order to elucidate some of the signaling elements that participate in the development of HR in lemon seedlings, we used several compounds that are known as activators or inhibitors of signal transduction elements in plants or in animal cells. Lemon seedlings treated either with cholera toxin or with phorbol 12-myristate 13-acetate (PMA), in the absence of A. alternata induced phenylalanine ammonia-lyase (PAL, E. C. 4.3.1.5) and the synthesis of scoparone, suggesting the participation of a G-protein and of a serine/threonine kinase, respectively, in signal transduction. The use of trifluoperazine (TFP), W-7, staurosporine, lavendustin A or 2,5dihydroximethyl cinnamate (DHMC) prevented PAL induction as well as scoparone biosynthesis in response to the fungal inoculation, thus allowing us to infer the participation of Calmodulin (CaM), of serine/threonine and of tyrosine protein kinases (TPK) for signal transduction in Citrus limon in response to A. alternata
