RESUMO Estabelecer os procedimentos adequados para condução do teste de germinação em espécies florestais é importante para viabilizar a comercialização de lotes de sementes, sobretudo para espécies que ainda não constam nas Instruções Normativas. Objetivou-se com este estudo determinar as condições ideais de temperatura e substrato para a realização do teste de germinação de sementes de Senna siamea, bem como: (i) caracterizar a morfo-fisiologia do estabelecimento da plântula, (ii) o período para a avaliação das plântulas, e (iii) validar a eficiência da metodologia em 10 lotes de sementes. O delineamento utilizado foi inteiramente ao acaso em arranjo fatorial 2 x 2 (temperaturas x substratos) com 12 repetições de 50 sementes para cada tratamento. Para a avaliação dos diferentes lotes, os dados foram submetidos à análise de variância e as médias comparadas pelo teste de Scott-Knott ao nível de 5% de probabilidade. Avaliou-se o percentual e a velocidade de germinação, primeira contagem, comprimento de plântulas, germinação acumulada, e morfologia de plântulas. Sementes de S. siamea possuem maior potencial germinativo em substrato rolo de papel sob temperaturas de 25 e 30 °C (86% e 87%). A primeira contagem de germinação e avaliação final de plântulas deve ser feita aos 15 e 21 dias após a semeadura, respectivamente. A espécie possui germinação do tipo epígea e suas plântulas são fanerocotiledonares. Observou-se que as condições de execução e avaliação do teste de germinação recomendadas neste estudo são adequadas na avaliação de lotes de sementes de S. siamea.

ABSTRACT Establishing the appropriate procedures for conducting germination tests in forest species is important for marketing the seeds, especially species that are not yet included in the relevant Normative Instructions. The aim of this study was to determine the ideal conditions of temperature and substrate to carry out the germination test on seeds of Senna siamea, as well as: (i) to characterise the morpho-physiology of seedling establishment, (ii) the period for seedling evaluation, and (iii) validate the efficiency of the methodology in 10 batches of seeds. The experimental design was completely randomised in a 2 x 2 factorial scheme (temperatures x substrates), with 12 replications of 50 seeds in each treatment. To evaluate the different batches, the data were submitted to analysis of variance, and the mean values compared by the Scott-Knott test at 5% probability. The percentage and speed of germination, first count, seedling length, accumulated germination, and seedling morphology were evaluated. The germination potential of S. siamea seeds is greater on paper-roll substrate at temperatures of 25 °C and 30 °C (86% and 87%). The first germination count and final seedling evaluation should be carried out 15 and 21 days after sowing, respectively. The species has epigeal germination and the seedlings are phanerocotyledonous. It was found that the method for carrying out and evaluating the germination test recommended in this study is suitable for evaluating batches of S. siamea seeds.

ABSTRACT Among the main problems that compromise the development of forest species are water and salt stresses, especially in the early stages of development, when seedlings are more sensitive. Thus, the objective of this study was to evaluate the morphophysiological and biochemical changes in Enterolobium contortisiliquum seedlings subjected to abiotic stresses. The experimental design was randomized complete blocks (RCB), composed of five treatments, with four replicates, and the experimental plot consisted of twenty plants. From the 31st day after sowing, when the seedlings reached approximately 15 cm in height, they began to receive the treatments, which consisted of different levels of electrical conductivity of irrigation water (0.3; 2; 4; 6; 8 dS m-1) for salt stress and periods of water restriction (0; 4; 8; 12 and 16 days without irrigation) for water stress. The traits evaluated were: plant height, root length, stem diameter, number of leaves, leaf area, total dry matter, Dickson quality index, height/shoot dry matter ratio, total soluble sugars, starch, total amino acids and proline. An increase in the electrical conductivity of irrigation water and days without irrigation compromised the development of E. contortisiliquum seedlings, which was intensified from 2 dS m-1 and four days without irrigation for salt and water stresses, respectively. The mechanism of survival of E. contortisiliquum to abiotic stresses involves reduction in the growth and quality of seedlings, with activation of biochemical defense mechanisms of the species.

ABSTRACT The fungi presence during seed maturation and dispersion is related to biotic and abiotic factors. These microorganisms can cause loss seeds physiological quality in unfavorable climatic conditions. We hypothesize that sanitary quality of seeds produced is affected by tree and origin climatic condition. The aim was evaluated fungi incidence and seeds quality of Pityrocarpa moniliformis from different trees and two climatic conditions (Aw: rainy tropical; and BSh: warm semiarid). We collected seeds from 28 parent trees, evaluating separately the physiological and sanitary quality of the seeds produced in each tree. Different trees originated seeds with differences in physiological quality and fungi incidence. The climatic condition of the trees' origin affected the amount and fungi species present in the seeds produced, with a predominant incidence of the genera Alternaria spp., Aspergillus spp., Botrytis spp., Fusarium spp., Penicillium spp. and Rhyzopus sp. The Alternaria spp. incidence is correlated with seeds of low physiological quality. Therefore, we conclude that Pityrocarpa moniliformis trees under a tropical rainy climate produce seeds with superior physiological and sanitary quality compared to those located in a warm semiarid climate.

RESUMO A presença de fungos potencialmente prejudiciais durante a maturação e dispersão das sementes tem relação com fatores bióticos e abióticos. Esses microrganismos podem causar perda da qualidade fisiológica de sementes em condições climáticas desfavoráveis. Nesta pesquisa, levantou-se a hipótese de que a qualidade sanitária das sementes produzidas é afetada pela árvore e condição climática de origem. Assim, objetivou-se avaliar a incidência de fungos e a qualidade de sementes de Pityrocarpa moniliformis provenientes de diferentes árvores e duas condições climáticas (Aw: tropical chuvoso; e BSh: semiárido quente). Coletaram-se sementes de 28 árvores, avaliando-se separadamente a qualidade fisiológica e sanitária das sementes produzidas em cada árvore. Diferentes árvores originaram sementes com diferenças na qualidade fisiológica e incidência de fungos. A condição climática de origem das árvores afetou a quantidade e a espécie fúngica presente nas sementes produzidas, com incidência predominante dos gêneros Alternaria spp., Aspergillus spp., Botrytis spp., Fusarium spp., Penicillium spp. e Rhyzopus sp, sendo a incidência de Alternaria spp. correlacionada com sementes de baixa qualidade fisiológica. Portanto, conclui-se que árvores de Pityrocarpa moniliformis sob clima tropical chuvoso produzem sementes de qualidade fisiológica e sanitária superior em relação àquelas localizadas em clima semiárido quente.

Resumo: A tecnologia de aplicação do plasma atmosférico consiste em um método rápido e prático, que apresenta potencial de modificação da superfície tegumentar de sementes com dureza tegumentar. O objetivo desta pesquisa foi avaliar o efeito da aplicação do plasma frio atmosférico em sementes de Pityrocarpa moniliformis, verificando suas implicações sobre a impermeabilidade do tegumento, germinação e vigor. As sementes foram submetidas ao plasma por 1,5; 2,0; 3,0; 4,0 e 5,0 minutos, utilizando-se como controle, sementes sem nenhum tratamento. Após a aplicação, determinou-se a molhabilidade do tegumento, curva de embebição e condutividade elétrica da solução de embebição. As sementes também foram submetidas ao teste de germinação, tendo-se, a partir dos seus resultados determinado a viabilidade, mediana, uniformidade e assimetria da germinação. As sementes submetidas ao plasma por 4,0 e 5,0 minutos apresentaram os menores ângulos de contato aparente, 64 e 61°, respectivamente, caracterizando maior molhabilidade do tegumento, entre os tratamentos testados. Maior germinação foi observada nos tratamentos com aplicação do plasma quando comparado com o controle. A tecnologia de aplicação do plasma atmosférico tem potencial para ser utilizada como acelerador da germinação de sementes de P. moniliformis.

Abstract: Atmospheric plasma application technology consists of a fast and practical method, which has the potential to modify the surface of seeds with integumentary hardness. The aim of this research was to evaluate the effect of applying atmospheric cold plasma on Pityrocarpa moniliformis seeds, assessing its implications on the seed coat impermeability, germination and vigor. Cells were subjected to plasma for 1.5, 2.0, 3.0, 4.0 and 5.0 minutes, using seeds without any treatment as a control. After application, the seed coat wettability, imbibition curve and electrical conductivity of the soaking solution were determined. Seeds were also subjected to the germination test, and its results were used to determine viability, median, uniformity and asymmetry of germination. Seeds subjected to plasma for 5.0 and 4.0 minutes showed the lowest apparent contact angles, 64 and 61°, respectively, characterizing greater wettability of the seed coat among the treatments tested. Higher germination was observed in the treatments with plasma application when compared to the control. Atmospheric plasma application technology has the potential to be used as an accelerator of P. moniliformis seed germination.

Resumo: Recentemente está crescendo a necessidade de tecnologias não invasivas, rápidas e precisas que possam estimar a qualidade das sementes. Entre essas tecnologias, destaca-se a análise de imagens de raios X para avaliação da morfologia interna das sementes. Assim, o objetivo deste estudo foi avaliar a eficiência do software SARS na análise de radiografias digitais de sementes de braquiária, denominado SARS (Sistema de Análise de Radiografias de Sementes) na avaliação da qualidade física das sementes. Cinco lotes comerciais de sementes de Urochloa decumbens cv. Basilisk foram utilizados. As sementes foram produzidas na safra de 2018/2019. As imagens radiográficas das sementes foram analisadas por meio do SARS, por meio das quais foram obtidas características físicas. As sementes foram submetidas ao teste de germinação, no qual foram mensuradas variáveis relacionadas à qualidade fisiológica. Foi possível observar que as sementes com maior potencial de germinação e vigor apresentaram correlações fortes e significativas com algumas das variáveis físicas obtidas pelo SARS. Assim, a alta correlação do comprimento de plântulas e a densidade relativa das sementes é importante para validar o método de análise de imagens radiográficas de sementes. O SARS mostrou-se uma ferramenta eficiente para análise de radiografias digitais de sementes de braquiária, gerando descritores que dão suporte a análise morfométrica e interna das sementes, que podem ser associados à sua qualidade fisiológica. Os parâmetros físicos obtidos pela técnica têm estreita relação com a germinação e o vigor das sementes.

Abstract: Recently is growing the need for non-invasive, fast, and accurate technologies that can predict seed quality. Between these technologies, X-ray image analysis stand out for evaluation of the internal morphology of the seeds. Thus, the aim of the present study was to evaluate the efficiency of a specialized software for analyzing digital radiographs of Urochloa decumbens seeds called SARS (Sistema de Análise de Radiografias de Sementes - Seed Radiograph Analysis System). Five comercial seed lots of U. decumbens cv. Basilisk were used. The seed lots were produced in the 2018/2019 crop season. Radiographic images of the seeds were analyzed in SARS, through which physical characteristics were obtained. The seeds were then subjected to germination test, in which variables related to the physiological quality were evaluated. It was possible to observe that the seeds with greater germination and vigor showed strong and significant correlations with some of the physical variables obtained using SARS. Thus, high correlation of seedling length and relative seed density is important for validating the seed radiographic image analysis method. SARS proved to be an efficient tool for analyzing digital radiographs of U. decumbens seeds. It can generate descriptors which support morphometric and internal analysis of the seeds. Physical parameters obtained by using the technique have close relationship with the germination and vigor of the seeds.

Resumo A utilização de técnicas que envolvam a análise computadorizada de plântulas na avaliação da qualidade de lotes de sementes tem se mostrado muito eficiente. Assim, objetivou-se, neste trabalho, verificar a eficiência do Sistema de Análise de Plântulas (SAPL) na detecção de diferenças de vigor entre lotes de sementes de moringa, em comparação às informações fornecidas por testes de vigor tradicionalmente utilizados. Para tanto, determinou-se o teor de água das sementes e a sua qualidade fisiológica pelos testes de germinação, primeira contagem de germinação, índice de velocidade de germinação, massa seca de plântulas, emergência, índice de velocidade de emergência, envelhecimento acelerado tradicional, envelhecimento acelerado com solução salina e utilizando-se o SAPL. A caracterização fisiológica das sementes determinou que os lotes 1 e 3 apresentaram qualidade superior aos demais, sendo o lote 4 o menos vigoroso. Os parâmetros obtidos na análise computadorizada (comprimento da parte aérea, comprimento da raiz primária, comprimento total de plântula, índice de crescimento, índice de uniformidade e índice de vigor) com o software SAPL foram eficientes para diferenciar os lotes de sementes em comparação aos testes tradicionalmente utilizados, não somente entre lotes de baixo e alto vigor, mas também para aqueles de qualidade intermediária, corroborando com os resultados obtidos nos testes tradicionalmente empregados na avaliação da qualidade de sementes.

Abstract The use of techniques involving a computerized analysis of seedlings in evaluating the seed lot quality has shown to be of great efficiency. Thus, the aim of this work was to verify the efficiency of the Seedling Analysis System (SAPL) in detecting vigor differences between moringa seed lots in comparison to the information provided by usual vigor tests. Therefore, the water content and the physiological quality of seeds by germination test, first count test, germination speed index, emergency speed index, traditional accelerated aging test, and accelerated aging test using saline solution were determined by SAPL. The experimental design was completely randomized, using four replicates. The parameters obtained in the computerized analysis (shoot and primary root lengths, total seedling length, and growth, uniformity, and vigor index) by the SAPL software were efficient in distinguishing seed lots in comparison to the usual tests, and not only among lots showing low and high vigor, but also for those showing intermediate quality.

RESUMO A utilização de ciclos de hidratação descontínua pode atuar na melhoria da germinação de sementes e crescimento inicial de plântulas sob estresse hídrico. Nesse sentido, objetivou-se avaliar o efeito da hidratação descontínua na germinação de sementes de Mimosa caesalpiniifolia e Pityrocarpa moniliformis sob estresse hídrico. Para isso, sementes dessas espécies foram submetidas a 0 (sem hidratação), 2 e 4 ciclos de hidratação (18 h) e desidratação (12 e 18 h, respectivamente), com posterior germinação sob estresse hídrico simulado para os potenciais osmóticos 0,0 (água destilada); -0,2; -0,4; -0,6 e -0,8 MPa. As plântulas foram avaliadas por meio dos testes de germinação, índice de velocidade de germinação, tempo médio de germinação e comprimento de plântulas. A hidratação descontínua em sementes de M. caesalpiniifolia e P. moniliformis prejudica a germinação e o vigor de plântulas sob estresse hídrico até -0,6 MPa. No entanto, a utilização de dois ciclos em P. moniliformis favorece a germinação sob estresse hídrico mais severo (-0,8 MPa).

ABSTRACT The use of discontinuous hydration cycles can act in improving seed germination and seedling initial growth under water stress. In this context, the objective of this study was to evaluate the effect of discontinuous hydration on the germination of Mimosa caesalpiniifolia and Pityrocarpa moniliformis seeds under water stress. For this, seeds of these species were subjected to 0 (without hydration), 2 and 4 hydration cycles (18 h) and dehydration (12 and 18 h, respectively), with subsequent germination under water stress simulated for osmotic potentials of 0.0 (distilled water), -0.2, -0.4, -0.6 and -0.8 MPa. Seedlings were evaluated using tests of germination, germination speed index, mean germination time and seedling length. Discontinuous hydration in M. caesalpiniifolia and P. moniliformis seeds hampers germination and vigor of seedlings under water stress up to -0.6 MPa. However, the use of two cycles in P. moniliformis favors germination under more severe water stress (-0.8 MPa).

Resumo: A inclusão de técnicas de análise de imagens para a avaliação precisa e rápida da qualidade de sementes florestais é altamente desejável. Entre as técnicas de imagem disponíveis, destacam-se o uso de imagens radiográficas digitais de sementes e a análise de imagens de plântulas, ainda pouco utilizadas na determinação da qualidade de sementes florestais. Objetivou-se com este trabalho, avaliar a viabilidade do uso da análise de imagens de sementes e plântulas na avaliação da qualidade física e fisiológica de sementes de Senna siamea. Imagens radiográficas de sementes de cinco lotes foram obtidas e, a partir delas, foram gerados descritores morfológicos e de integridade tecidual. Após a obtenção das imagens das sementes, estas foram submetidas ao teste de germinação e crescimento de plântulas, dos quais se extraíram variáveis relacionadas à qualidade fisiológica. As plântulas geradas foram escaneadas e analisadas por meio do software ImageJ. Os dados foram analisados por meio da análise de variância, correlação e análise de componentes principais. Os resultados mostraram diferenças entre os lotes de sementes quanto a sua qualidade fisiológica e integridade física de tecidos internos. Foram observadas correlações significativas entre as variáveis obtidas com a análise radiográfica e pelos testes de caracterização fisiológica das sementes. O uso das técnicas de análise de radiografias de sementes e de imagens de plântulas permitem acessar informações de integridade física e fisiológica de sementes de S. siamea.

Abstract: The inclusion of image analysis techniques for the accurate and rapid assessment of the quality of forest seeds is highly desirable. The use of digital radiographic images of seeds and the analysis of seedling images, still little used in determining the quality of forest seeds, are among the available imaging techniques. This study aimed to assess the feasibility of using the analysis of images of seeds and seedlings in the assessment of the physical and physiological quality of Senna siamea seeds. Radiographic images of seeds were obtained from five seed lots, allowing generating morphological and tissue integrity descriptors. These seeds were then subjected to germination and seedling growth tests, which allowed obtaining variables related to physiological quality. The generated seedlings were scanned and analyzed using the software ImageJ. The data were analyzed using analysis of variance, correlation, and principal component analysis. The results showed differences between seed lots in terms of physiological quality and physical integrity of internal tissues. Significant correlations were observed between the variables obtained with the radiographic analysis and seed physiological characterization tests. The use of techniques to analyze seed radiographs and seedling images allows access to information on the physical and physiological integrity of S. siamea seeds.

Resumo: Métodos não-destrutivos e de alto desempenho têm sido desenvolvidos para avaliação da qualidade de sementes. O objetivo deste estudo foi relacionar parâmetros obtidos a partir da análise gratuita e automatizada de radiografias digitais de sementes de melão híbrido com o seu potencial fisiológico. Foram utilizadas amostras de sementes comerciais de três cultivares híbridas de melão (Cucumis melo L.), cada uma representada por três lotes. Foram obtidas imagens radiográficas das sementes, das quais foram geradas determinações de área, perímetro, circularidade, densidade relativa, densidade integrada e preenchimento da cavidade interna de sementes, por meio de uma macro (PhenoxXray) desenvolvida para o software ImageJ®. Após o teste de raios-X, as sementes foram submetidas ao teste de germinação, a partir do qual foram obtidas variáveis relacionadas à qualidade fisiológica. Observou-se variabilidade entre lotes para as características físicas e fisiológicas. Os resultados demonstraram que o uso da macro PhenoXray permite a fenotipagem em larga escala das radiografias de sementes de maneira simples, rápida, consistente e totalmente gratuita. A metodologia é eficiente na obtenção de dados morfométricos e de integridade tecidual em sementes de melão, e os parâmetros gerados apresentam estreita relação com atributos fisiológicos da qualidade das sementes.

Abstract: Non-destructive and high throughput methods have been developed for seed quality evaluation. The aim of this study was to relate parameters obtained from the free and automated analysis of digital radiographs of hybrid melons’ seeds to their seeds’ physiological potential. Seeds of three hybrid melon (Cucumis melo L.) cultivars from commercial lot samples were used. Radiographic images of the seeds were obtained, from which area, perimeter, circularity, relative density, integrated density and seed filling measurements were generated by means of a macro (PhenoXray) developed for ImageJ® software. After the X-ray test, seed samples were submitted to the germination test, from which variables related to the physiological quality of the seeds were obtained. Variability between lots was observed for both physical and physiological characteristics. Results showed that the use of the PhenoXray macro allows large-scale phenotyping of seed radiographs in a simple, fast, consistent and completely free way. The methodology is efficient in obtaining morphometric and tissue integrity data of melon seeds and the generated parameters are closely related to physiological attributes of seed quality.

RESUMO Novas e refinadas tecnologias, baseadas em procedimentos computadorizados, criam potencial para automatização de avaliações laboratoriais, alternativa para a redução de custos e aumento da capacidade de tomada de decisão quanto ao destino de lotes de sementes. O presente trabalho teve como objetivo avaliar a eficiência do software de processamento digital de imagens de plântulas, o SAPL®, na caraterização do potencial fisiológico de sementes de milho, em comparação com informações fornecidas por testes de vigor recomendados para essa espécie. Dez lotes de sementes de milho foram submetidos às avaliações de germinação, primeira contagem de germinação, índice de velocidade de germinação, emergência de plântulas, teste de frio e condutividade elétrica. Os resultados desses testes foram comparados com os dados obtidos com a análise de imagem, que avaliou o desenvolvimento de plântulas aos três e cinco dias, contados a partir do início do teste de germinação, mediante a obtenção de valores de comprimento de plântulas, índice de crescimento, uniformidade de desenvolvimento, índice de vigor e índice de vigor corrigido. O SAPL® é uma alternativa consistente e promissora para avaliação do potencial fisiológico de sementes de milho, tendo apresentado alta correlação de suas variáveis com os testes tradicionais utilizados. A avaliação pode ser realizada no terceiro e quinto dia após a instalação do teste de germinação.

ABSTRACT New and refined technologies based on computerized procedures create the potential for automating laboratory evaluations, constituting an alternative to reduce costs and increase the decision-making capacity regarding the destination of seed lots. The objective of this study was to evaluate the efficiency of the Sistema de Análise de Plântulas (SAPL ®) free digital image processing software of seedlings in characterizing the physiological potential of maize seeds in comparison with information provided by vigor tests recommended for this species. Ten lots of maize seeds were submitted to germination, first germination count, germination speed index, seedling emergence, cold test and electrical conductivity. The results of these tests were compared with the data obtained from the image analysis, which evaluated the development of seedlings at three and five days counted from the beginning of the germination test, by obtaining values of seedling length, growth, development uniformity, vigor index and corrected strength index. SAPL ® is a consistent and promising alternative for evaluating the physiological potential of maize seeds, presenting a high correlation of its variables with the traditional tests used. The evaluation can be performed on the third and fifth day after the germination test is installed.

Resumo Os métodos que possibilitam a rápida avaliação do vigor são de grande interesse no controle de qualidade em empresas produtoras de sementes, visto que o teste de lixiviação de potássio tem se mostrado uma ferramenta eficiente na avaliação de lote de sementes, como da espécie Moringa oleifera, a qual apresenta alto potencial nos diferentes subprodutos da planta, podendo ser empregado na agricultura e indústria farmacêutica. O objetivo do trabalho foi adequar a metodologia do teste de lixiviação de potássio em sementes de moringa. Os experimentos foram realizados em laboratório e em casa de vegetação, utilizando-se quatro lotes de sementes, submetidos aos testes de germinação, emergência de plântula, comprimento total e massa seca de plântulas além do teste de frio, para a caracterização fisiológica, e do teste de lixiviação de potássio. Este último foi realizado com amostras de 25 e 50 sementes, colocadas em copos plásticos contendo 70 e 100 mL de água destilada a 25 °C por períodos de 1, 2, 3, 4, 5 e 6 horas. O delineamento empregado foi inteiramente casualizado com quatro repetições por tratamento. Pode-se afirmar que, em todas as avaliações realizadas, apenas um lote se destacou em relação aos demais, pois, apresentou menor quantidade de lixiviados de potássio e maior potencial fisiológico em condições de laboratório e em campo. Os testes de emergência e de frio e as combinações de amostras de 50 sementes e os volumes 70 mL de água destilada por 1, 5 e 6 horas, 50 sementes em volumes de 100 de água destilada por 1, 2, 3, 4, 5 e 6 horas de embebição, foi a metodologia do teste de lixiviação de potássio mais adequado para classificar lotes de sementes de Moringa oleifera em função de sua qualidade fisiológica.

Abstract Methods that allow rapid evaluation of the vigor are of great interest in the quality control seeds used in the companies, since the potassium leaching test has been shown as being an efficient tool in the evaluation of many species such as Moringa oleifera. This species presents high potential in the different byproducts of the plant, being able to be used in agriculture and pharmaceutical industry. The objective of this work was to adapt the methodology of potassium leaching test in moringa seeds. The experiments were carried out in a seed laboratory and in a greenhouse, using four lots of seed, submitted to germination tests, seedling emergence, total length and dry mass of seedlings and the cold test, for the physiological characterization, besides the potassium leaching test. The latter was performed with samples of 25 and 50 seeds, placed in plastic cups containing 70 and 100 mL of distilled water at 25 °C for periods of 1, 2, 3, 4, 5 and 6 h. The design was in DIC with four replicates per treatment, and the results submitted to ANOVA, when significant by the F test, the means compared by the Tukey test at 5 %. It can be affirmed that in all the evaluations carried out, only one lot stood out in relation to others, because it presented less amount of potassium leachate and greater physiological potential under laboratory and field conditions. The emergency and cold tests and combinations of samples of 50 seeds and volumes 70 mL of distilled water for 1, 5 and 6 hours, 50 seeds in volumes of 100 distilled water per 1, 2, 3, 4, 5 and 6 hours of imbibition, was the methodology of the potassium leaching test more adequate to classify lots of seeds of Moringa oleifera due to their physiological qualities.

Resumo A morfologia das sementes e as modificações físicas sofridas por elas durante a germinação podem estar associadas ao vigor. Por isso, objetivou-se com este trabalho monitorar o incremento de área em sementes de moringa durante a embebição, por meio da análise de imagens, com o intuito de se determinar a sua qualidade fisiológica a partir deste incremento. Utilizaram-se sementes de quatro lotes de moringa com e sem tegumento, fotografadas antes e durante a embebição, nos períodos de 8, 16 e 24 horas, nas temperaturas de 15 e 20°C. Calculou-se o incremento de área (%) através da análise das imagens, utilizando-se o programa ImageJ. Os resultados de cada período avaliado foram comparados com os testes usualmente utilizados na avaliação do vigor de sementes. O delineamento foi inteiramente casualizado, com quatro repetições de 50 sementes por tratamento. Os dados foram submetidos a ANOVA e comparados pelo teste de Tukey. Os lotes 1, 2 e 4 foram considerados de melhor qualidade, enquanto o lote 3 apresentou qualidade inferior, quando comparado aos demais lotes testados. As sementes de moringa seguiram o padrão trifásico de embebição, com a protrusão da raiz primária ocorrendo em 72 horas. Foi possível avaliar o percentual de incremento de área por meio do programa ImageJ, sendo o período de 8 horas de embebição a 15°C o que demonstrou maior eficiência para classificação dos lotes quanto a sua qualidade fisiológica.

Abstract The seed morphology and the physical modifications undergone by them during germination may be associated with vigor. Therefore, the aim of this work was to monitor the increase of area in Moringa seeds during imbibition, by means of image analysis, in order to determine their physiological quality. Seeds of four Moringa lots with and without tegument, photographed before and during soaking, were used in the periods of 8, 16 and 24 hours, at temperatures of 15 and 20°C. The area increase (%) was calculated by image analysis using the ImageJ program. The results of each evaluated period were compared with the traditional tests of germination and vigor. The design was entirely randomized, with four replicates of 50 seeds per treatment. Data were submitted to ANOVA and compared by the Tukey test. Lots 1, 2 and 4 were considered better quality, while lot 3 presented lower quality when compared to the other lots tested. Moringa seeds followed the three-phase pattern of imbibition, with protrusion of the radicle occurring in 72 hours. It was possible to evaluate the percentage of increment of area through the program ImageJ, being the period of 8 hours of imbibition at 15°C which demonstrated greater efficiency to classify the lots as to their quality.

RESUMO O uso de novas abordagens metodológicas e a inclusão de avaliações não destrutíveis na determinação de qualidade das sementes florestais tem se mostrado de grande importância para o desenvolvimento da produção florestal. O objetivo deste trabalho foi avaliar a eficácia do uso de ferramenta semi-automatizada de medição de imagens digitais obtidas no teste de raios X em lotes de sementes de Leucaena leucocephala e sua relação com a qualidade fisiológica das sementes. Para tanto, foram utilizadas sementes de cinco lotes colhidos de árvores-matrizes localizadas na região de Viçosa, MG. O estudo foi realizado por meio da análise de imagens radiográficas das sementes, para as quais foram obtidos o percentual de sementes com danos (predação e fungos), e medidas de área, perímetro, circularidade, densidade relativa e densidade integrada das sementes. Após o teste de raios X, as sementes foram submetidas ao teste de germinação, em que se extraíram dez variáveis relacionadas à qualidade fisiológica das sementes, dentre elas um índice de vigor. O uso de ferramentas de medição semi-automatizada associado ao teste de raios X é eficaz para avaliar a morfologia interna das sementes de Leucaena leucocephala e suas variáveis apresentam relação com a qualidade fisiológica das sementes.

ABSTRACT The use of new methodological approaches and inclusion of non-destructive evaluations in determining the quality of forest seeds has proven to be of great importance for the development of forest production. The aim of this study was to evaluate the effectiveness of the use of a semi-automated tool for measurement of digital images obtained by the X-ray test in seed lots of Leucaena leucocephala and their relationship to seed physiological quality. Seeds were collected from five mother trees in Viçosa, MG. The study was performed through analysis of radiographic images of the seeds. It was determined the percentage of damaged seeds (predation and fungi) and measured: the area, perimeter, circularity, relative density, and integrated density of the seeds. After the X-ray test, a germination test was performed on the seeds, from which ten variables related to seed physiological quality were extracted, including a vigor index. The use of semi-automated measurement tools associated with the X-ray test was effective for evaluating the internal morphology of Leucaena leucocephala seeds, and the variables determined are related to seed physiological quality.

RESUMO A obtenção de informações baseadas em imagens é uma abordagem poderosa para capturar e quantificar dados sobre o vigor de sementes. No entanto, sistemas comerciais que facilitam o processamento e a aquisição de imagens são, muitas vezes, de custo proibitivo. Objetivou-se avaliar a eficiência do Sistema de Análise de Plântulas (SAPL®), para analisar o potencial fisiológico de sementes de soja, em comparação com informações fornecidas por testes de vigor tradicionalmente recomendados para a espécie. Nove lotes de sementes de soja foram submetidos a avaliações de germinação, velocidade de germinação, emergência de plântulas, teste de frio e condutividade elétrica. Nas análises com o SAPL®, foram avaliadas plântulas com quatro e seis dias, contados a partir do início do teste de germinação, que resultaram em valores de comprimento de plântulas, crescimento, uniformidade de desenvolvimento, índice de vigor e índice de vigor corrigido. Os lotes avaliados com emergência superior a 90 % apresentaram índice de vigor superior a 600 e 800, no quarto e sexto dias, respectivamente. Os índices gerados pelo SAPL®, exceto o índice de uniformidade, apresentaram correlações positivas e altas com os testes tradicionais (> 0,80). O SAPL® é eficiente para identificar diferenças no vigor de lotes de sementes de soja.

ABSTRACT Obtaining image-based information is a powerful approach to capture and quantify seed vigor data. However, commercial systems that facilitate the processing and acquisition of images are often cost prohibitive. This study aimed to evaluate the efficiency of the Seedling Analysis System (Sistema de Análise de Plântulas - SAPL®), in order to analyze the physiological potential of soybean seeds, in comparison with the information provided by vigor tests which are traditionally recommended for this species. Nine lots of soybean seeds were submitted to germination, germination speed, seedling emergence, cold test and electrical conductivity tests. In the SAPL® analyzes, seedlings of four and six days, counted from the beginning of the germination test, were evaluated, resulting in values for seedling length, growth, development uniformity, vigor index and corrected vigor index. The evaluated lots with emergence greater than 90 % showed a vigor index higher than 600 and 800, respectively in the fourth and sixth days. The indexes generated by SAPL®, except for the uniformity index, presented positive and high correlations with the traditional tests (> 0.80). SAPL® is efficient in identifying differences in the vigor of soybean seed lots.

RESUMO A germinação de sementes de mamão é lenta e desuniforme, normalmente em função de inibidores presentes na sarcotesta; no entanto, alguns procedimentos de retirada podem acarretar danos físicos, interferindo, de forma negativa, na qualidade fisiológica e no desempenho em campo. Objetivou-se avaliar a qualidade física e fisiológica de sementes de mamão, submetidas a métodos de remoção da sarcotesta, por meio de análise de imagens. Sementes de mamão Formosa ‘Tainung 01' foram submetidas aos seguintes métodos de remoção da sarcotesta: fermentação + peneira, areia + peneira, peneira e liquidificador + peneira. Após a retirada da sarcotesta, as sementes foram submetidas a testes de geminação, primeira contagem de germinação, velocidade de germinação e comprimento de plântula. Para a análise de imagem, as sementes sem sarcotesta foram submetidas ao teste de raios X e à análise computadorizada de imagens de plântulas pelo software SVIS®. Os métodos utilizados mostraram-se promissores somente para a retirada da sarcotesta, enquanto o método peneira + liquidificador, além da melhor retirada, obteve alta qualidade física e fisiológica das sementes. Entretanto, a utilização do software SVIS® permitiu identificar a alta qualidade física e fisiológica das sementes com a utilização do método liquidificador + peneira, que promoveu maior eficiência na remoção da sarcotesta.

ABSTRACT The germination of papaya seeds is slow and uneven, usually due to inhibitors present in the sarcotesta; however, some removal procedures may cause physical damage, negatively interfering in the physiological quality and field performance. This study aimed to evaluate the physical and physiological quality of papaya seeds submitted to methods of sarcotesta removal by image analysis. Papaya Formosa ‘Tainung 01' seeds were submitted to the following methods of sarcotesta removal: fermentation + sieve, sand + sieve, sieve and blender + sieve. After the sarcotesta removal, the seeds were submitted to tests of germination, first germination count, germination speed and seedling length. For the image analysis, the seeds without sarcotesta were submitted to the X-ray test and to a computerized analysis of seedling images by the SVIS® software. The methods used were only promising for the sarcotesta removal, while the blender + sieve method obtained the highest physical and physiological seed quality, in addition to performing the best sarcotesta removal. However, the image analysis using the SVIS® software allowed to identify the high physical and physiological seed quality using the blender + sieve method, which promoted the highest efficiency in removing the sarcotesta.
