A fragment of 466 base pairs from a highly variable peripheral region of the mitochondrial D-loop of horses was amplified and analyzed by single stranded conformational polymorphism (SSCP). Fourteen distinct SSCP variants were detected in 100 horses belonging to four breeds (Arabian, ARB; Thoroughbred, TB; Argentinian Creole, ARC; and Peruvian Paso from Argentina, PPA). Each breed showed four to eight SSCP variants, many of which were shared between two or three of the studied breeds. Arabian horses were the most variable (eigth variants), with three variants unique to the breed. PPA and ARC showed two and one characteristic SSCP variants, respectively, while TB shared all its variants with at least one of the other breeds. An analysis based on the presence/absence of the variants revealed a closer relationship between PPA and TB, which was not completely unexpected considering the mixed ancestry of the PPA mares. The results also confirm the efficiency of SSCP to detect variability in horse mitochondrial DNA.

A indução de aneuploidia por agentes físicos e químicos usando diferentes sistemas de teste foi avaliada. O efeito de raios-X, cafeína, acetaldeído, etanol, dietilestilbestrol, propionaldeído e hidrato de cloral foi estudado por contagem cromossômica em células diplóides embriônicas de hamster chinês. A habilidade aneugênica de cloreto de cádmio, sulfato de cádmio, dicromato de potássio, cloreto de crômio, cloreto de níquel e sulfato de níquel foi avaliada por meio de análise de anáfase-telófase em células de ovário de hamster chinês. A contagem cromossômica em fibroblastos humanos (linhagem celular MRC-5) foi empregada para avaliar o efeito de ácido cacodílico, cloreto de cádmio, sulfato de cádmio e dicromato de potássio. Finalmente, a indução de micronúcleos positivos e negativos para cinetocoro por cloreto de cádmio, sulfato de cádmio, dicromato de potássio, cloreto de crômio e cloreto de níquel foi estudada usando anticorpos CREST. Quando o efeito de agentes diferentes foi determinado por contagem cromossômica, observou-se um aumento de células hipoplóides mas não de hiperplóides. A análise anáfase-telófase mostrou que sais metálicos aumentaram a freqüência de cromossomos "lagging". Este achado foi confirmado pelo aumento de micronúcleos positivos para cinetocoro usando anticorpos CREST. Portanto, a perda cromossômica poderia ser considerada a principal causa de aneuploidia induzida.

The induction of aneuploidy by physical and chemical agents using different test systems was evaluated. The effect of X-rays, caffeine, acetaldehyde, ethanol, diethylstilbestrol, propionaldehyde, and chloral hydrate was studied by chromosome counting in Chinese hamster embryonic diploid cells. Aneugenic ability of cadmium chloride, cadmium sulfate, potassium dichromate, chromium chloride, nickel chloride, and nickel sulfate was assessed by means of anaphase-telophase analysis in Chinese hamster ovary cells. Chromosome counting in human fibroblasts (MRC-5 cell line) was employed to evaluate the effect of cacodilic acid, cadmium chloride, cadmium sulfate, and potassium dichromate. Finally, the induction of kinetochore-positive and kinetochore negative micronuclei by cadmium chloride, cadmium sulfate, potassium dichromate, chromium chloride, and nickel chloride was studied using CREST antibodies. When the effect of different agents was determined by chromosome counting, an increase of hypoploid but not of hyperploid cells was observed. Anaphase-telophase analysis showed that metal salts increased the frequency of lagging chromosomes. This finding has been confirmed by the increment of kinetochore-positive micronuclei using CREST antibodies. Therefore, chromosome loss could be considered as the main cause of induced aneuploidy.

Tripulantes de vôos internacionais estão expostos a radiações ionizantes originadas da colisão de partículas cósmicas com átomos da atmosfera. O grau de exposição varia de acordo com a altitude, latitude e a atividade solar. A análise citogenética mostrou um aumento significante no número de cromossomos dicêntricos (14,5 ± 0,76 por 1000 células) em membros ativos da tripulação, assim como em tripulantes aposentados (7,5 ± 0,59 por 1000 células), em relação a controles (0,5 ± 0,05 por 1000 células). Em tripulantes em atividade a freqüência de cromossomos em anel também aumentou (2,0 ± 0,31 por 1000 células), mas isso não ocorreu em tripulantes aposentados. Estas observações sugerem um alto grau de exposição do pessoal de bordo a radiações ionizantes. Conseqüentemente, o acompanhamento citogenético de tripulantes de aviões cronicamente expostos a baixas doses de radiação é um campo aberto para futuras investigações.

Air crew members of international flights are exposed to ionizing radiations originated from the collision of cosmic particles with atoms of the atmosphere. The degree of exposure varies according to the altitude, latitude, and solar activity. The cytogenetical analysis showed a significant increase of dicentric chromosomes (14.5 ± 0.76 per 1,000 cells) in active air crew members as well as in retired air crew members (7.5 ± 0.59 per 1,000 cells) in relation to controls (0.5 ± 0.05 per 1,000 cells). In active members the frequency of ring chromosomes was also increased (2.0 ± 0.31 per 1,000 cells) but not found in retired crew members. These observations suggest a high degree of exposure to ionizing radiations of air crew personnel. Consequently, the cytogenetic follow-up of air crews chronically exposed to low radiation doses is an open field for further investigations.

O grau de similaridade entre agentes químicos e físicos quanto a sua capacidade de induzir translocações recíprocas foi analisado por meio de técnicas de análise multivariada. Analisou-se o efeito de três diferentes doses de raios gama, quatro doses de raios-X e diferentes doses de adriamicina, mitomicina C, tio-tepa e bleomicina. Os dados foram arranjados em uma matriz básica por 2 métodos: análise de cluster e ordenação. Dois grupos principais foram encontrados, um incluindo doses de 9 e 10 Gy e o outro incluindo as outras doses de radiação ionizante e as substâncias químicas. Vários subgrupos foram encontrados dentro do segundo grupo. Usando dados de presença/ausência, não se encontrou um padrão específico de indução de danos cromossômicos para os agentes físicos e químicos. O aumento na freqüência de translocação recíproca observado com 9 e 10 Gy deveu-se a um aumento no tipo de configurações multivalentes. Esta variabilidade pode ser dependente da dose. Igualmente, a similaridade observada no segundo grupo entre as substâncias químicas e as doses menores de radiação ionizante também pode ser dependente da dose.

The degree of similarity between chemical and physical agents in their capacity to induce reciprocal translocations was analyzed by means of multivariate analysis techniques. The effect of three different doses of gamma rays, four doses of X-rays and different doses of adriamycin, mitomycin C, thio-tepa and bleomycin was analyzed. Data were arranged in a basic matrix by two methods: cluster analysis and ordination. Two main groups were found, one including doses of 9 and 10 Gy and the other including the remaining lower doses of ionizing radiation and the other chemicals. Various subgroups were found within the second group. Accordingly, using presence/absence data there was not a specific pattern of chromosomal damage induction for physical and chemical agents. The increase in the frequencies of reciprocal translocation observed with 9 and 10 Gy was due to an increase in the kind of multivalent configurations. This variability could be dose dependent. Likewise, the similarity observed in the second group between the chemicals and the lower doses of ionizing radiation could also be dose dependent.

As evidências de que a aneuploidia durante a mitose pode ser um fator na etiologia de malignidades somáticas estão cada vez mais fortes. A análise de alterações em anáfase-telófase da mitose é um teste útil para a avaliação da capacidade aneuploidogênica e clastogênica de substâncias químicas. Vários metais têm sido identificados como carcinogênicos para o homem e para animais. Contudo, os mecanismos de ação permanecem obscuros. No presente estudo, a capacidade aneugênica e clastogênica do sulfato de cádmio, do dicromato de potássio e do cloreto de níquel foi analisada usando o teste anáfase-telófase. Células do ovário do hamster chinês cultivadas por dois ciclos foram tratadas com o composto desejado por 8 horas antes da colheita das células. Foram quantificadas as freqüências de células com pontes de cromatina, lagging cromossomos e lagging fragmentos cromossômicos. O índice mitótico foi determinado pela contagem do número de células em mitose por 1000 células em cada lamínula e foi expresso como uma porcentagem do número de placas mitóticas. A análise estatística foi feita usando o método "G". Análises de correlação e de regressão foram realizadas para avaliar as variações do índice mitótico. O crômio e o cádmio foram clastogênicos e aneugênicos e aumentaram as freqüências dos três tipos de aberrações avaliadas; o níquel teve apenas atividade aneugênica porque ele aumentou a freqüência de lagging cromossomos. Estes resultados indicam que o teste anáfase-telófase é sensível o suficiente para detectar as relações dependentes da dose que podem distinguir as atividades clastogênicas e/ou aneugênicas e que os resultados obtidos usando o teste anáfase-telófase foram semelhantes aos obtidos pela contagem cromossômica.

There is increasing evidence that aneuploidy during mitosis may be a factor in the etiology of somatic malignancy. The analysis of alterations in anaphase-telophase of mitosis is a useful test for evaluating the aneuploidogenic and clastogenic ability of chemicals. Several metals have been found to be carcinogenic to humans and animals. However, the underlying mechanisms remain unclear. In the present study the aneugenic and clastogenic abilities of cadmium sulfate, potassium dichromate and nickel chloride were analyzed using the anaphase-telophase test. Chinese hamster ovary (CHO) cells cultured for two cycles were treated with the desired compound for 8 h before cell harvesting. The frequency of cells with chromatin bridges, lagging chromosomes and lagging chromosomal fragments was scored. The mitotic index was determined by counting the number of mitotic cells per 1,000 cells on each coverslip and was expressed as a percentage of the number of mitotic plates. Statistical comparisons were done using the "G" method. Correlation and regression analyses were performed to evaluate variations of the mitotic index. Chromium and cadmium were clastogenic and aneugenic and increased the frequencies of the three types of aberrations scored; nickel had only aneugenic activity because it increased the frequency of lagging chromosomes. These results indicate that the anaphase-telophase test is sufficiently sensitive to detect dose-response relationships that can distinguish clastogenic and/or aneugenic activities and that the results obtained using the anaphase-telophase test were similar to those obtained by chromosome counting.

Muitas raças de gado foram submetidas a alta pressão de seleção para caracteres de produção. Conseqüentemente, a estrutura genética e a distribuição alélica da população poderiam diferir em raças sob alta pressão de seleção, quando comparadas a raças não selecionadas. Foi feita a análise das freqüências dos genes de k-caseína, aS1-caseína e prolactina em rebanhos de gado Creole argentino (AC) e Hosltein argentino (AH). Os valores de FST calculados mediram o grau de diferenciação genética de subpopulações, dependendo de variações na freqüência dos genes. A raça AC apresentou variação consideravelmente maior entre os rebanhos nos loci de aS1-caseína e k-caseína. Estratégias de conservação devem considerar a população inteira de AC de forma a manter a variabilidade genética encontrada nesta raça nativa.

Many cattle breeds have been subjected to high selection pressure for production traits. Consequently, population genetic structure and allelic distribution could differ in breeds under high selection pressure compared to unselected breeds. Analysis of <FONT FACE="Symbol">k</font>-casein, <FONT FACE="Symbol">a</font>S1-casein and prolactin gene frequencies was made for Argentine Creole (AC) and Argentine Holstein (AH) cattle herds. The calculated FST values measured the degree of genetic differentiation of subpopulations, depending on the variances of gene frequencies.The AC breed had considerably more variation among herds at the <FONT FACE="Symbol">a</font>S1-casein and <FONT FACE="Symbol">k</font>-casein loci. Conservation strategies should consider the entire AC population in order to maintain the genetic variability found in this native breed.

Gene frequencies at the k-casein locus were estimated in six different herds (N = 180) of Argentinean Creole cattle. The results showed a strong influence of subdivision and independent evolution on the divergence of the observed gene frequencies. These results suggest that the population structure of these herds favor the maintenance of polymorphism, which is of crucial importance for the long-time survival of populations.
