Resumo O burro doméstico possui um papel fundamental como animal de tração na economia rural do Paquistão, onde a população desse animal está aumentando a cada ano. A região de controle de mtDNA completa de 40 burros amostrados aleatoriamente foi ampliada por PCR e sequenciada bidirecionalmente por intermédio de primers específicos. Haplótipos distintos de mtDNA obtidos no estudo atual (KY446001 − KY446011) foram submetidos a medidas de haplótipo (h) e diversidade de nucleotídeos (π) por meio de DnaS, bem como análises filogenéticas, de rede e AMOVA. Havia um total de 27 sítios polimórficos presentes em 11 haplótipos de mtDNA exclusivos dos 40 animais estudados de diferentes regiões. A rede de união de vizinhos e a rede de união mediana ilustram a divisão de todos esses haplótipos em duas linhagens núbias e somalis bem definidas, confirmando a origem materna africana do burro doméstico do Paquistão. Os parâmetros de diversidade h (0,967 ± 0,037) e π (0,02917 ± 0,00307) revelaram altos níveis de diversidade genética em burros paquistaneses. AMOVA demonstrou apenas 1% de diferenças genéticas entre as duas linhagens maternas de mtDNA, apontando a falta de subestrutura populacional em burros paquistaneses, como é o caso da população mundial de burros domésticos. Os burros paquistaneses têm origem materna africana e altos níveis de diversidade de mtDNA. A alta diversidade genética pode ser por causa da reprodução não seletiva e de heteroplasmia. Aqui, fornecemos o primeiro relatório sobre a diversidade do mtDNA da região de controle em burros domésticos do Paquistão.

Abstract Domestic donkey plays a key role as a draft animal in rural economy of Pakistan where its population is increasing every year. The complete mtDNA control region of forty randomly sampled donkeys was PCR- amplified and sequenced bi-directionally using specific primers. Distinct mtDNA haplotypes obtained in the current study (KY446001−KY446011) were subjected to haplotype (h) and nucleotide diversity (π) measures using DnaS as well as to phylogenetic, Network, and AMOVA analyses. There were a total 27 polymorphic sites present within 11 unique mtDNA haplotypes from the studied 40 animals from different regions. Neighbor-joining network and median-joining network both illustrated the splitting of all these haplotypes into two well-defined Nubian and Somali lineages, confirming African maternal origin of Pakistani domestic donkey. Diversity parameters h (0.967± 0.037) and π (0.02917± 0.00307) were found to reveal high levels of genetic diversity in Pakistani donkeys. AMOVA demonstrated only 1% of genetic differences between two mtDNA maternal lineages, pointing to lack of population substructure in Pakistani donkeys as is the case with worldwide domestic donkey population. Pakistani donkeys have African maternal origin and high levels of mtDNA diversity. High genetic diversity may be due to non-selective breeding and heteroplasmy. We herein provide the first report on mtDNA diversity of control region in Pakistani domestic donkey.

Resumo O estudo foi desenhado para investigar o efeito do óleo de coco nos níveis de alguns parâmetros hepáticos e hematológicos em coelhos intoxicados com tetracloreto de carbono. Também a capacidade antioxidante do óleo de coco para várias concentrações foi avaliada com base na porcentagem de eliminação de radicais livres (DPPH). Os animais experimentais foram divididos em cinco grupos, oito coelhos em cada grupo. Estes foram: grupo A (controle normal), grupo B (controle tóxico), grupo C (controle padrão), grupo D (tratado com óleo de coco 50 mL/kg de peso corporal após intoxicação por CCl4), grupo E (tratado com óleo de coco 200 mL/kg de peso corporal após intoxicação por CCl4). Os efeitos observados foram comparados com um fármaco hepatoprotetor padrão silimarina (50 mL/kg de peso corporal). O óleo de coco (200 mL/kg de peso corporal) reduziu significativamente (P<0,05) os níveis séricos elevados de alanina transaminase (ALT), aspartato transaminase (AST) e fosfatase alcalina (ALP), quando comparado a um coelho controle tóxico. Os resultados dos coelhos tratados com extrato foram semelhantes aos do grupo de coelhos administrados com silimarina. O tratamento com raiz de óleo de coco e silimarina não causou alterações significativas nos valores de RBC, Plaquetas, (Hb), (MCH) e (HCT). No entanto, observou-se aumento significativo (P<0,05) na contagem total de leucócitos. O presente estudo sugeriu que o óleo de coco pode ser usado como uma alternativa fitoterápica (precisa de mais exploração, ou seja, para detectar seu composto bioativo e sua eficácia) para atividade hepatoprotetora.

Abstract The study was designed to investigate the effect of Coconut Oil on the levels of some liver and hematological parameters in carbon tetrachloride intoxicated rabbits. Also the antioxidant capacity of Coconut Oil for various concentrations was assessed on the basis of percent scavenging of (DPPH) free radical. Experimental animals were divided into five groups, eight rabbits in each group. These were: group A (Normal control), group B (Toxic control), group C (Standard control), group D (Treated with Coconut Oil 50 mL/kg body weight after CCl4 intoxication), group E (Treated with Coconut Oil 200 mL/kg body weight after CCl4 intoxication). The effects observed were compared with a standard hepatoprotective drug silymarine (50 mL/kg body weight). The Coconut Oil (200 mL/kg body weight) significantly (P<0.05) reduced the elevated serum levels of alanine transaminase (ALT), aspartate transaminase (AST) and alkaline phosphatase (ALP) when compared to a toxic control rabbits. The results of extract treated rabbits were similar to silymarine administered rabbits group. Treatment with Coconut Oil root and silymarine caused no significant changes in RBC, Platelets, (Hb), (MCH) concentration and (HCT) values. However, significant (P<0.05) increase was observed in the total WBC count. The present study suggested that Coconut Oil can be used as an herbal alternative (need further exploration i.e to detect its bioactive compound and its efficacy) for hepatoprotective activity.

Resumo A bioacumulação de metais pesados tóxicos no corpo humano pode causar efeitos adversos à saúde, cuja gravidade depende de sua dosagem e duração da exposição. Neste estudo, filhotes de duas espécies diferentes de peixes comestíveis, ou seja, Tor putitora (Mahseer) e Ctenopharyngodon idella (carpa-capim), foram expostos a diferentes concentrações de nitrato de chumbo em um ambiente controlado de aquário por três diferentes períodos de duração (14, 28 e 60 dias). A bioacumulação de chumbo em diferentes órgãos, incluindo brânquias, pele, músculos, fígado, intestino e bexiga natatória dos peixes, foi avaliada por espectrometria de absorção atômica. Geralmente, a maior concentração de chumbo foi observada nas brânquias e a menor nos músculos para ambas as espécies em cada dosagem e duração experimental. Na exposição de 14 dias, o padrão relativo de bioacumulação em diferentes órgãos foi observado como brânquia > fígado > pele > intestino > bexiga natatória > músculo para ambas as espécies de peixes. Da mesma forma, o padrão de bioacumulação observado em 28 dias de exposição foi: brânquia > fígado > intestino > pele > bexiga natatória > músculo em ambas as espécies. Já o padrão de exposição de 60 dias foi observado como brânquia > fígado > intestino > bexiga natatória > músculo > pele. Os dados mostram que a carpa-capim armazenou maiores concentrações de chumbo do que a Mahseer, o que pode ser atribuído ao fato de serem onívoras. Além disso, a menor bioacumulação foi registrada nos músculos até o 56º dia de exposição, após o que a concentração aumentou de forma constante nos músculos. O padrão observado destaca a importância da duração da exposição ao chumbo; a exposição crônica pode resultar em sua bioacumulação em concentrações tóxicas nos músculos, o que é particularmente preocupante porque os músculos dos peixes são muito consumidos como alimento em todo o mundo.

Abstract Bioaccumulation of toxic heavy metals in the human body can give rise to adverse health effects, the severity of which depends upon their dosage and duration of exposure. In this study, yearlings of two different species of edible fish, i.e., Tor putitora (Mahseer) and Ctenopharyngodon Idella (grass carp), were exposed to different concentrations of lead nitrate in a controlled environment of aquarium for three different lengths of duration (14, 28, and 60 days). The bioaccumulation of lead in different organs, including gills, skin, muscles, liver, intestine, and swim bladder of the fish, was assessed using atomic absorption spectrometry. Generally, the highest lead concentration was observed in the gills and lowest in the muscles for both species at each experimental dosage and duration. In 14-days exposure, the relative pattern of bioaccumulation in different organs was observed as gill > liver > skin > intestine > swim bladder > muscle for both fish species. Similarly, the pattern of bioaccumulation observed in 28-days exposure was as: gill > liver > intestine > skin > swim bladder > muscle in both species. Whereas, pattern in 60-days exposure was observed as gill > liver > intestine > swim bladder > muscle > skin. The data shows that grass carp had stored higher concentrations of lead than Mahseer, which may be attributed to the fact that they are omnivorous. Furthermore, the lowest bioaccumulation was recorded in the muscles until the 56th day of the exposure, after which the concentration steadily increased in the muscles. The observed pattern highlights the importance of exposure’s duration to lead; chronic exposure could result in its bioaccumulation at toxic concentrations in the muscles, which is particularly of concern because the fish muscles are heavily consumed as food worldwide.

Resumo O presente estudo teve como objetivo investigar a relação entre polimorfismos em genes desintoxicantes (GSTM1, GSTT1 e GSTP1) e sua associação com câncer colorretal (CCR) em tabagistas da etnia pashtun. Os polimorfismos nos genes selecionados foram genotipados em um estudo de caso-controle composto por 100 pacientes do sexo masculino com CCR, confirmados histologicamente, e 100 controles saudáveis, pareados por ano de nascimento e sexo usando o método PCR-RFLP. Os genótipos GSTM1 nulo e GSTT1 nulo contribuíram significativamente para o risco de CCR nos casos (OR = 3,131, IC 95%: 1,451-6,758, P = 0,004; OR = 3,541, IC 95%: 1,716-7,306, P = 0,001, respectivamente), enquanto a associação observada para GSTP1 Val/Val (1,139, IC 95%: 0,356-3,644, P = 0,826) não apresentou significância estatística. O GSTM1 nulo e o GSTT1 nulo combinados mostraram um risco 41 vezes maior (IC 95%: 4,945-351,950, P = 0,001) para CCR, enquanto os genótipos GSTM1 nulo e GSTP1 Ile/Val ou Val/Val combinados apresentaram risco cerca de 3 vezes maior (IC 95%: 1,196-7,414, P = 0,019) para CCR. Da mesma forma, os genótipos combinados GSTT1 nulo e GSTP1 Ile/Val ou Val/Val tiveram um risco para CRC cerca de 3 vezes maior (95% CI: 1,285-8,101, P = 0,013). Na combinação de três genótipos GST, os genótipos GSTM1 nulo, GSTT1 nulo e GSTP1 Ile/Val ou Val/Val apresentaram um risco 22 vezes maior (IC 95%: 2,441-212,106, P = 0,006) para CRC. Nossos achados sugerem que o polimorfismo GSTM1 e GSTT1 e sua combinação com GSTP1 podem estar associados à suscetibilidade ao CRC da população pashtun de Khyber Pakhtunkhwa, Paquistão, viciada em Naswar.

Abstract The current study aimed to investigate the relationship between polymorphisms in detoxifying (GSTM1, GSTT1, and GSTP1) genes and their association with colorectal cancer (CRC) in tobacco addicts of Pashtun ethnicity. Polymorphisms in the selected genes were genotyped in a case-control study consisting of 100 histologically confirmed male CRC patients and 100 birth-year and gender-matched healthy controls using the PCR−RFLP method. The GSTM1 null, and GSTT1 null genotypes were significantly contributed to the risk of CRC in the cases (OR= 3.131, 95% CI: 1.451−6.758, P = 0.004, and OR= 3.541, 95% CI: 1.716−7.306, P = 0.001, respectively), whereas the association observed for GSTP1 Val/Val (1.139, 95% CI: 0.356−3.644, P = 0.826) did not show statistical significance. The combined GSTM1 null and GSTT1 null showed a 41-fold increased risk (95% CI: 4.945−351.950, P = 0.001), while, the combined GSTM1 null and GSTP1 Ile/Val or Val/Val variant genotypes exhibited about 3-fold (95% CI: 1.196−7.414, P = 0.019) increased risk to CRC. Similarly, the combined GSTT1 null and GSTP1 Ile/Val or Val/Val variant genotypes showed about a 3-fold (95% CI: 1.285−8.101, P = 0.013) increased risk of CRC. In the combination of three GST genotypes, the GSTM1 null, GSTT1 null, and GSTP1 Ile/Val or Val/Val variant genotypes demonstrated a more than a 22-fold (95% CI: 2.441−212.106, P = 0.006) increased risk of CRC. Our findings suggest that GSTM1 and GSTT1 polymorphism and its combination with GSTP1 may be associated with CRC susceptibility in the Naswar addicted Pashtun population of Khyber Pakhtunkhwa, Pakistan.

Resumo Marcadores bioquímicos, como proteínas, são muito importantes para determinar a diversidade genética entre espécies de plantas em determinada população, o que, por sua vez, é muito importante para criadores e agricultores, pois eles podem selecionar facilmente a variedade mais adequada para crescer em certa localidade. Nesse sentido, o presente estudo tem como objetivo avaliar a diversidade genética em germoplasma de Acacia modesta por meio da técnica de Eletroforese em Gel de Poliacrilamida com Dodecil Sulfato de Sódio (SDS-PAGE). Cerca de 40 genótipos foram submetidos à análise SDS-PAGE, em que foi observado um total de 12 bandas polipeptídicas no eletroforetograma. Destes, 16,67% eram monomórficos, enquanto os 83,33% restantes eram polimórficos. As variações encontradas em B-2, 4, 5, 6, 7, 8, 9, 10, 11 e 12 foram de 20, 22,50, 32,50, 10, 2,50, 22,50, 15, 5, 2,50 e 75%, respectivamente. A contribuição do lócus para a discordância genética foi de 83,33%. A análise de agrupamento classificou todos os genótipos em 9 agrupamentos. Os genótipos em um cluster foram idênticos em relação ao perfil de proteínas e apresentaram menor variação genética intraespecífica, enquanto diferenças foram encontradas em outros genótipos.

Abstract Biochemical markers such as protein are very important to determine genetic diversity among plant species in a given population which in turn is very important for breeders and farmers as they can then easily select the most appropriate variety to grow in a given locality. In this connection, the present study is aimed to evaluate genetic diversity in Acacia modesta germplasm through Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis (SDS-PAGE) technique. About 40 genotypes were subjected to SDS-PAGE analysis where a total of 12 polypeptide bands were observed in electrophoretogram. Out of which 16.67% were monomorphic while the remaining 83.33% were polymorphic. Variation found in B-2, 4, 5, 6, 7, 8, 9, 10, 11 and 12, were 20, 22.50, 32.50, 10, 2.50, 22.50, 15, 5, 2.50 and 75% respectively. Locus contribution toward genetic disagreement was 83.33%. Cluster analysis sorted all the genotypes into 9 clusters. The genotypes in one cluster were identical regarding protein profiling and showed less intra-specific genetic variation whereas differences were find from other genotypes.

Resumo A contaminação dos recursos naturais com metais pesados liberados pelas siderúrgicas é a principal causa da poluição do solo e da água no Dargai Malakand, localizado no lado norte do Paquistão. Portanto, o presente estudo teve como objetivo determinar o teor de metais pesados em amostras de solo e água desta área. O cultivo de plantas silvestres (nove plantas nativas: Pteris vittata, Populus nigra, Eucalyptus camaldulensis, Persicaria maculosa, Arundo donax, Xanthium strumarium, Verbascum thapsus, Ricinus communis e Parthenium hysterophorus) foi testado quanto às suas capacidades de fitorremediação, que é um meio ambiente, geralmente utilizado, e abordagem de baixo custo para eliminar metais pesados de solos e águas poluídas. Amostras de solo, água e efluentes foram retiradas dos locais contaminados de sete siderúrgicas no distrito de Dargai, Malakand, e submetidas à análise de metais pesados. Com base no fator de bioconcentração (BCF) e fator de translocação (TF) calculados, o maior BCF para zinco foi registrado para as raízes de Pteris vittata (3,93), enquanto o menor valor foi observado para as folhas de Verbascum thapsus (0,306). A raiz de Pteris vittata apresentou o maior BCF para ferro (1,618), enquanto as folhas de Ricinus communis apresentaram o menor (0,023). O maior valor de BCF para cromo foi maior para raízes de Populus nigra (0,717), enquanto o menor valor foi registrado para folhas de Persicaria maculosa (0,031). Para os metais selecionados, Fe, Zn e Cr, os maiores TF foram registrados para Pteris vittata (0,988), Verbascum thapsus (0,944) e Xanthium strumairum (0,968), respectivamente. Portanto, recomenda-se que essas plantas sejam cultivadas próximas a siderúrgicas para recuperar metais pesados de efluentes industriais, solo poluído e água poluída.

Abstract The contamination of natural resources with heavy metals released from steel mills is the primary cause of soil and water pollution in the Dargai Malakand, located on the northern side of Pakistan. Therefore, the present study was aimed to determine the level of heavy metals in soil and water samples of this area. The wild plant growing (nine native plants: Pteris vittata, Populus nigra, Eucalyptus camaldulensis, Persicaria maculosa, Arundo donax, Xanthium strumarium, Verbascum thapsus, Ricinus communis and Parthenium hysterophorus) there were then tested for their phytoremediation capabilities which is an environmentally friendly, generally utilized, and low-cost approach to eliminate heavy metals from polluted soils and water. Soil, water, and effluent samples were taken from the contaminated sites of seven steel mills in Dargai District Malakand and subjected to heavy metals analysis. Based on bioconcentration factor (BCF) and translocation factor (TF) calculated, The highest BCF for zinc was recorded for Pteris vittata roots (3.93), while the lowest value was observed for Verbascum thapsus leaves (0.306). Pteris vittata root showed the highest BCF for iron (1.618), while Ricinus communis leaves showed the lowest (0.023). The highest BCF value for chromium was highest for Populus nigra roots (0.717), while the lowest value was recorded for Persicaria maculosa leaves (0.031). For the selected metals; Fe, Zn and Cr the highest TF were recorded for Pteris vittata (0.988), Verbascum thapsus (0.944) and Xanthium strumairum (0.968) respectively. Therefore, it is recommended that these plants should be grown near to steel mills to reclaim heavy metals from industrial effluent, polluted soil as well as from polluted water.

Abstract Emergence of plasmid mediated colistin and extended spectrum β-lactamases (ESBL) resistant genes has been impacted the efficacy of colistin and β-lactams drugs like 3rd, 4th generation cephalosporin. Current study was aimed to investigate antimicrobial resistance genes (ARGs) among Escherichia coli isolates from meat producing commercial broilers in Pakistan. Two hundred (n=200) fecal samples were collected during January-2018 to August-2019. For isolation of E. coli, pink colonies on MacConkey agar were transferred to EMB agar. Metallic sheen color colonies were tested biochemically using API-20E kit. The molecular identification of E. coli (n=153) was targeted by amplification of uid gene through polymerase chain reaction (PCR) and different ARGs i.e. gentamicin, streptomycin, tetracycline, colistin, β-lactams drugs, quinolone and ampicillin followed by sequence analysis. Genotypically, followed by phenotypically of resistant ARGs of isolated PCR-confirmed E. coli (153) shoed resistant against gentamicin (aac(3)-IV), streptomycin (aadA1), tetracycline (tetA), colistine (mcr-1), ampicillin (bla-TEM) and bla-CTX-M were 86%, 88%, 86%, 88%, 83% & 77% respectively. 33/38 (86%) of the isolate was positive for quinolone resistance. Colistine (mcr-1), ESBLs (bla-TEM) and (bla-CTX-M) resistance genes were 88%, 83% and 77% respectively. About 33 isolated E. coli harbored the both mcr-1 and ESBLs genes. All of E. coli isolates were found sensitive to ceftriaxone (CTX-30) and imipenem (IMP-10). The Isolated E. coli showed single or multi clade decadency. The E. coli and ARGs sequences showed single or multi clade decadency. This is first comprehensive study from Pakistan that described the molecular evidences of ARGs and their co-existence in single isolates originated from commercial poultry. Commercial chicken (Broilers) can act as melting pot of antibiotic resistance genes for human being. It is alarming situation for surveillance of antibiotic resistance program because of more regulated prescription of antimicrobial agents in Pakistan

Resumo O surgimento de colistina mediada por plasmídeo e genes de resistência a β-lactamases de espectro estendido (ESBL) afetou a eficácia de medicamentos colistina e β-lactâmicos, como as cefalosporinas de 3ª e 4ª geração. O presente estudo teve como objetivo investigar genes de resistência antimicrobiana (ARGs) entre isolados de Escherichia coli em frangos de corte comerciais no Paquistão. Duzentas (n = 200) amostras fecais foram coletadas durante janeiro de 2018 a agosto de 2019. Para o isolamento de E. coli, colônias rosas em ágar MacConkey foram transferidas para ágar EMB. As colônias de cores de brilho metálico foram testadas bioquimicamente usando o kit API-20E. A identificação molecular de E. coli (n = 153) foi direcionada pela amplificação do gene uid através da reação em cadeia da polimerase (PCR) e diferentes ARGs, ou seja, gentamicina, estreptomicina, tetraciclina, colistina, medicamentos β-lactâmicos, quinolona e ampicilina, seguido de análise de sequência. Genotipicamente, seguido por fenotipicamente de ARGs resistentes de E. coli isoladas foram confirmadas por PCR (153) como resistente contra gentamicina (aac(3)-IV), estreptomicina (aadA1), tetraciclina (tetA), colistina (mcr-1), ampicilina (bla-TEM) e bla-CTX-M, demonstrando resultados de 86%, 88%, 86%, 88%, 83% e 77%, respectivamente. Cerca de 33/38 (86%) do isolado foi positivo para resistência às quinolonas. Os genes de resistência à colistina (mcr-1), ESBLs (bla-TEM) e (bla-CTX-M) foram 88%, 83% e 77%, respectivamente. Cerca de 33 E. coli isoladas continham os genes mcr-1 e ESBLs. Todos os isolados de E. coli foram considerados sensíveis à ceftriaxona (CTX-30) e imipenem (IMP-10). A E. coli isolada apresentou decadência de um ou vários clados. As sequências de E. coli e ARGs apresentaram decadência de um ou vários clados. Este é o primeiro estudo abrangente do Paquistão que descreveu as evidências moleculares de ARGs e sua coexistência em isolados únicos originados de aves comerciais. Dessa forma, é possível concluir que o Frango comercial (Broilers) pode atuar como caldeirão de genes de resistência a antibióticos para o ser humano. É uma situação alarmante para a vigilância do programa de resistência a antibióticos devido à prescrição mais regulamentada de agentes antimicrobianos no Paquistão

ABSTRACT The current study was conducted to elucidate the clinical effect of ochratoxin A in Gallus gallusdomesticus and to ameliorate its toxic effects by the development and characterization of highly porous carbon-based adsorbent derived from coconut shell. A series of experiments were performed on one day-old chicks (Group A to F). Clinical signs of the positive control (group B fed on 400 ppb ochratoxin A contaminated feed) included depression, diarrhea, increased water intake, low body weight, high degree of genotoxicity, swollen and hemorrhagic kidneys and liver. Serum alanine amino transferase (ALT), alkaline phosphatase (ALP), creatinine and urea concentration significantly increased while albumin, globulin and total proteins were found significantly low. All these lead to increased mortality. Among adsorbents treated groups, chickens in group C (0.3% of the prepared adsorbent mixed in OTA contaminated feed) and D (0.6% of the prepared adsorbent mixed in Ochratoxin A (OTA) contaminated feed) showed very low pathological effects while group E (0.9% of the prepared adsorbent mixed in OTA contaminated feed) and F (1.2% of the prepared adsorbent mixed in OTA contaminated feed) showed normal behavior, blood biochemistry and well maintained histological structure like that in group A. As a whole, the adsorbents treated groups fed with ochratoxin A contaminated feed, supplemented with different levels of the prepared adsorbent helped to ameliorate the toxic effects of OTA and nearly showed normal clinical signs.
