Tobacco farmers are routinely exposed to complex mixtures of inorganic and organic chemicals present in tobacco leaves. In this study, we examined the genotoxicity of tobacco leaves in the snail Helix aspersa as a measure of the risk to human health. DNA damage was evaluated using the micronucleus test and the Comet assay and the concentration of cytochrome P450 enzymes was estimated. Two groups of snails were studied: one fed on tobacco leaves and one fed on lettuce (Lactuca sativa L) leaves (control group). All of the snails received leaves (tobacco and lettuce leaves were the only food provided) and water ad libitum. Hemolymph cells were collected after 0, 24, 48 and 72 h. The Comet assay and micronucleus test showed that exposure to tobacco leaves for different periods of time caused significant DNA damage. Inhibition of cytochrome P450 enzymes occurred only in the tobacco group. Chemical analysis indicated the presence of the alkaloid nicotine, coumarins, saponins, flavonoids and various metals. These results show that tobacco leaves are genotoxic in H. aspersa and inhibit cytochrome P450 activity, probably through the action of the complex chemical mixture present in the plant.

Cashew (Anacardium occidentale) is a medicinal plant native to Brazil and also yields a nutritious fruit juice. Its large pulpy pseudo-fruit, referred to as the cashew apple, contains high concentrations of vitamin C, carotenoids, phenolic compounds and minerals. Natural and processed cashew apple juice (CAJ/cajuina) are amongst the most popular juices in Brazil, especially in the north-east. Both juices have antioxidant potential and suppress mutagenicity of hydrogen peroxide. In the present study we evaluated the inhibitory effects of CAJ/cajuina on Aflatoxin B1(AFB1)-induced mutation, using the Salmonella/microsome assay with the experimental approaches of pre-, co- and post-treatments. Both CAJ/cajuina suppress AFB1-induced mutagenesis in strain TA102 when applied in co- and in post-treatment. Possible mechanisms for anti-mutagenicity in co-treatment are (a) interaction with S9 enzymes, (b) metabolization to non-mutagenic compounds of AFB1 or (c) inactivation of S9 potential. Total suppression of AFB1 mutagenicity was observed in co-treatment with both CAJ and cajuina. Post-treatment anti-mutagenicity of both juices suggests a modulation of activity of error-prone DNA repair. CAJ/cajuina may be considered promising candidates for control of genotoxicity of AFB1 and may thus be considered as health foods with anti-carcinogenic potential. This promising characteristic warrants further evaluation with in vivo studies.

Vários procedimentos terapêuticos, diagnósticos e profiláticos usados em hospitais apresentam um risco genético. Para avaliar o risco genotóxico ocupacional, 42 trabalhadores do Hospital de Clínicas de Porto Alegre, RS, Brasil, ocupacionalmente expostos a chumbo (uso de soldas), óxido de etileno (área de esterilização), drogas antineoplásicas (enfermeiros e farmacêuticos) e radiação ionizante foram comparados com 42 indivíduos não expostos. A análise de linfócitos binucleados apresentou um aumento estatisticamente não significativo (P = 0.058) na freqüência de micronúcleos. Quando analisados separadamente, os grupos expostos a drogas antineoplásicas e radiação ionizante apresentaram um aumento estatisticamente significativo (P = 0.038 e P = 0.0217, respectivamente) na freqüência de micronúcleos. As freqüências de pontes dicêntricas e anomalias de fuso sugerem a ação de clastogênicos nestes dois grupos. Fatores como idade, sexo e hábitos de fumo e álcool não apresentaram correlação com as alterações genéticas observadas. Estes resultados sugerem que os procedimentos de segurança adotados foram muito importantes para proteger os trabalhadores da exposição a agentes mutagênicos e que estas medidas devem ser melhoradas nas áreas de radiologia e quimioterapia.

Many therapeutic, diagnostic and prophylactic procedures used in hospitals are of potential genetic risk. An evaluation was made of genotoxic occupational risk in 42 workers from the Hospital de Clínicas de Porto Alegre, RS, Brazil, who had been occupationally exposed to lead (solder), ethylene oxide (sterilization area), antineoplastic drugs (nurses and pharmacists) or ionizing radiation. They were compared with 42 unexposed individuals. There was an increase in the frequency of binucleated cytochalasin-blocked lymphocytes with micronuclei, though it was not significant (P = 0.058). The groups exposed to antineoplastic drugs and radiation had a significant increase in micronuclei frequency (P = 0.038 and P = 0.022, respectively). The high frequencies of dicentric bridges suggest the action of clastogenics in these two groups. These results suggest that the safety procedures adopted were very important to protect workers from exposure to mutagenic agents and should be improved in the radiological and chemotherapeutical areas.

A mais importante vantagem do Ensaio Cometa é a possibilidade do seu uso em qualquer organismo e tipo celular. Além de ser barato e rápido, se obtem resultados em poucas horas. Devido a isto é que ele vem sendo recomendado como teste inicial para monitoramento genotóxico ambiental. Neste trabalho investigaram-se adaptações na técnica para trabalhos de campo, com o uso de roedores endêmicos de região de mineração de carvão. O organismo utilizado foi o Ctenomys torquatus, capturado em duas diferentes áreas: Pelotas (região sem mineração de carvão-controle) e Candiota (zona de mineração de carvão). Foi coletado sangue periférico de 18 roedores, que após marcação foram liberados. O sangue foi protegido da luz e mantido sob refrigeração, e processado in loco. As concentrações das agaroses e as condições alcalinas de lise e eletroforese foram modificadas a partir das metodologias sugeridas pelas revisões existentes. As amostras restantes foram mantidas em RPMI 1640 (1:10) (4ºC) e pela nossa experiência podem ser utilizadas por até 4 dias. Foram analisadas 50 células por animal. As células foram classificadas de 0-4, segundo seu grau de lesão, além de serem medidas em unidades arbitrárias. Foi demonstrado aumento significante no nível de dano do DNA no grupo de Candiota (P < 0,05 - teste t), quando comparado com Pelotas. Ainda se demonstrou correlação entre o índice de danos e as medidas celulares. Estes resultados, neste estudo piloto, indicam que o carvão induz efeitos no DNA que são detectáveis através do Ensaio Cometa, o que indica a viabilidade deste teste para o biomonitoramento ambiental.

The main advantages of single-cell gel electrophoresis (SCG) are its applicability to any eukaryotic organism and cell type, its low cost and the short time required to obtain results. These properties make the SCG assay particularly useful in screening for environmental genotoxicity. The present study describes a modified version of this technique for use in field work with native rodents and examines some factors which influence the outcome of the assay. Wild rodents (Ctenomys torquatus, "tuco-tuco") from a region close to a strip coal mine and from a region with no coal mines were used. Animals from the coal mining region had significantly more DNA damage than those from the control area. The use of this SCG technique for direct sampling in the field should facilitate environmental genotoxicity studies with natural populations, without the need to remove the animals from their habitat or to sacrifice them.

A ação clastogênica de duas concentrações (18,75 e 37,5 mg/kg) do alcalóide pirrolizidínico integerrimina, obtido do Senecio brasiliensis, foi testada em camundongos após diferentes tempos do tratamento agudo. Após este tratamento, o alcalóide foi capaz de induzir aumento na freqüência de eritrócitos policromados micronucleados presentes tanto na medula óssea quanto no sangue periférico

The pyrrolizidine alkaloid integerrimine, obtained from Senecio brasiliensis, was tested by acute dosing at two concentrations (18.75 and 37.5 mg/kg), and at different times, to establish its ability to induce micronuclei in mouse erythrocytes. This alkaloid was able to increase the frequency of micronucleated polychromatic erythrocytes in both, bone marrow and peripheral blood erythrocytes

Resumo Foi determinada a prevalência de microfilaremia em uma amostra representativa de adolescentes e adultos, em 13 aldeias, de índios amazônicos brasileiros em julho-agosto de 1976. Através de esfregaço de sangue periférico corados com Giemsa e de preparações de culturas de linfócitos de sangue periférico, ou ambas, foram testadas 533 pessoas com idade acima de 10 anos e 68 crianças com menos de 10 anos. A Mansonella ozzardi foi a única espécie de microfilária encontrada. A prevalência foi altamente aldeia-específica. Em quatro (4) das 13 aldeias, houve casos de não detecção de microfilaremia pelos métodos usados. Em quatro (4) outras aldeias, as prevalências encontradas para os residentes de 10 anos e mais velhos foram em excesso de 60% para cada método utilizado. Em aldeias onde a microfilaremia foi documentada, apenas cinco (13%) do total de 38 crianças testadas foram positivas. Houve uma tendência geral da prevalência aumentar com o aumento da idade, A razão de prevalência de microfilaremia entre homens e mulheres foi de aproximadamente 1,4:1 para o teste de esfregaço de sangue periférico e exatamente 1:1 para o outro. Em cada 5 aldeões com microfilaremia-positiva testados com ambos os métodos, detectamos uma mais alta prevalência da microfilaremia com as preparações feitas de cultura de linfócitos.

Summary The prevalence of microfilaremia among a representative sample of the adolescents and adults in 13 Brazilian Amazon Indian villages was determined for July-August, 1976. With Giemsa-stained peripheral blood smears, preparations from peripheral blood lymphocyte cultures, or both, 533 persons aged 10 years and older and 68 children below age 10 were tested. Mansonella ozzardi was the only blood-borne microfilarial species found. Prevalence was highly village-specific. In four of 13 villages persons tested had no detectable microfilaremia by the methods used. In four other villages the overall prevalences for residents aged 10 years and older were in excess of 60% by one of the methods used. In villages where microfilaremia was documented only five (13%) of the total 38 children tested were positive. There was a general trend for prevalence to rise with increasing age. The ratio of the prevalence of microfilaremia among males to that among females was approximately 1.4:1 with peripheral blood smear test results but exactly 1:1 with the other method. In each of the five microfilaremia-positive villages tested using both methods, we detected a higher prevalence of microfilaremia with the preparations made from the lymphocyte culture studies.
