Resumo A procura por insumos que substituam produtos de alto custo, como óleo e farinha de peixe, na fabricação de rações para uso na aquicultura é crescente, sendo necessário garantir a qualidade nutricional das dietas. A silagem produzida a partir de resíduos de pescado e de vegetais apresenta-se como uma alternativa de baixo custo e eficiente fonte proteica. O objetivo do presente estudo foi avaliar as respostas fisiológicas e bioquímicas de alevinos de tambaqui alimentados com quatro níveis de inclusão de silagem em ração comercial com 28% de proteína bruta, em dois períodos: 45 e 90 dias. Cada tratamento foi realizado em três repetições, com 10 tambaquis em cada caixa experimental de 100 L. Ao término de cada período estabelecido, amostras sanguíneas foram coletadas de cinco animais de cada repetição para determinação das variáveis hematológicas e bioquímicas. Peso, comprimento total e índices hepatossomático e lipossomático de espécimes alimentados com silagem não mostraram alterações significativas em ambos os períodos de avaliação, bem como os valores de hematócrito. A concentração de hemoglobina de tambaqui após 45 dias de alimentação aumentou quando foi fornecida dieta com inclusão de 20% de silagem. A contagem de eritrócitos, volume corpuscular médio e hemoglobina corpuscular média não apresentaram alterações entre os tratamentos, em ambos os períodos. As proteínas totais aumentaram significativamente no plasma de tambaquis que receberam dietas com inclusão de 5 e 10% de silagem, avaliados após 45 dias de alimentação. Evidenciou-se que os grupos com inclusão de silagem na dieta não apresentaram alterações significativas nos parâmetros avaliados, assim não sendo prejudiciais à higidez do tambaqui. Portanto, o uso da silagem como suplemento alimentar durante o cultivo do tambaqui é uma alternativa sustentável para produtores, por promover a redução do descarte de resíduos de pescado e da agricultura.

Abstract The demand for products to replace high-cost raw materials, such oil and fish meal, in the manufacture of feed for use in aquaculture, while also guaranteeing the nutritional quality of the diets, is increasing. Silage produced with fish and vegetables residues is a low-cost and efficient protein source. The objective of the present study was to evaluate the physiological and biochemical responses of tambaqui fingerlings fed four different levels of silage included in commercial feed with 28% crude protein, over two periods: 45 and 90 days. Each treatment was carried out over three replications, with 10 tambaqui in each 100 L experimental tank. At the end of each established period, blood samples were collected from five animals from each repetition to determine the hematological and biochemical variables. Body weight and total length, hepatosomatic and liposomal indices and hematocrit of specimens fed with diets supplemented with silage did not exhibit significant changes in both assessment period. After 45 days of feeding, the hemoglobin concentration increased when tambaqui were fed a diet including 20% silage. The red blood cell count, mean corpuscular volume and mean corpuscular hemoglobin did not change between treatments in either period. The total protein concentrations increased significantly in the plasma of tambaqui fed with diets with the inclusion of 5 and 10% of silage, evaluated after feeding for 45 days. It was found that the groups which had silage included in their diet did not exhibit significant alterations in the evaluated parameters, and the diet was therefore not consider harmful to the health of tambaqui. Therefore, the use of silage as a feed supplement during tambaqui farming is a sustainable alternative for producers, as it leads to a reduction of impacts of fish and vegetables waste disposal.

Resumo As armadilhas extracelulares de neutrófilos (NETs) foram relatadas pela primeira vez como uma estratégia microbicida para neutrófilos ativados. Por meio de uma resposta imunológica contra vários estímulos, os neutrófilos liberam seu DNA ligado a proteínas de grânulos, núcleo e citoplasma (por exemplo, elastase e mieloperoxidase). Até o momento, os NETs têm sido implicadas em danos aos tecidos durante processos inflamatórios intensos, principalmente quando sua liberação depende da geração de radicais de oxigênio. Os flavonóides são agentes antioxidantes e anti-inflamatórios, e destes, a quercetina é comumente encontrada em nossa dieta diária. Portanto, a quercetina pode exercer alguma atividade protetora contra o dano tecidual induzido por NETs. Em nossos ensaios in vitro, a quercetina reduziu a liberação das NETs, mieloperoxidase (MPO) e elastase a partir de neutrófilos estimulados com forbol 12-miristato 13-acetato (PMA). A atividade dessas enzimas também foi diminuída na presença de quercetina. A quercetina também reduziu o efeito citotóxico dos NETs sobre células alveolares (linha celular A549). Além disso, os ensaios in silico indicaram interações favoráveis entre a quercetina e as proteínas da NET (MPO e elastase). No geral, nossos resultados demonstram que a quercetina diminui os efeitos deletérios das NETs, reduzindo sua liberação à partir de neutrófilos ativados e diminuindo a atividade enzimática de MPO e elastase, possivelmente por meio de interação direta.

Abstract Neutrophil extracellular traps (NETs) were first reported as a microbicidal strategy for activated neutrophils. Through an immunologic response against several stimuli, neutrophils release their DNA together with proteins from granules, nucleus, and cytoplasm (e.g., elastase and myeloperoxidase). To date, NETs have been implicated in tissue damage during intense inflammatory processes, mainly when their release is dependent on oxygen radical generation. Flavonoids are antioxidant and anti-inflammatory agents; of these, quercetin is commonly found in our daily diet. Therefore, quercetin could exert some protective activity against tissue damage induced by NETs. In our in vitro assays, quercetin reduced NETs, myeloperoxidase (MPO), and elastase release from neutrophils stimulated with phorbol 12-myristate 13-acetate (PMA). The activity of these enzymes also decreased in the presence of quercetin. Quercetin also reduced the cytotoxic effect of NETs on alveolar cells (A549 cell line). Further, in silico assays indicated favorable interactions between quercetin and NET proteins (MPO and elastase). Overall, our results demonstrate that quercetin decreases deleterious cellular effects of NETs by reducing their release from activated neutrophils, and diminishing the enzymatic activity of MPO and elastase, possibly through direct interaction.

Resumo O conteúdo de dopamina nas estruturas cerebrais tem sido relacionado à excitabilidade neuronal e várias abordagens têm sido utilizadas para estudar este fenômeno durante o período de vulnerabilidade às crises. No presente trabalho, descrevemos os efeitos da depleção de dopamina após a administração de 6-hidroxidopamina (6-OHDA) na região pars compacta da substância negra de ratos submetidos ao modelo de epilepsia com pilocarpina. A susceptibilidade ao estado de mal epiléptico induzido pela pilocarpina, bem como a frequência de crises espontâneas e recorrentes durante o período crônico do modelo foi determinada. Sendo o hipocampo uma das principais estruturas afetadas no desenvolvimento desse modelo experimental de epilepsia, os níveis de dopamina nessa região foram determinados após a administração da droga. No primeiro experimento, 62% (15/24) dos ratos pré-tratados com 6-OHDA e 45% (11/24) daqueles que receberam ácido ascórbico como solução controle evoluíram para crises límbicas motoras e para o estado de mal epiléptico, após a administração de pilocarpina. A gravidade das crises durante o período agudo do modelo foi significativamente maior nos ratos epilépticos experimentais (56,52%) do que nos ratos controle (4,16%). No segundo experimento, não houve diferença significante entre os grupos quanto à frequência de crises na fase crônica do modelo. Nossos dados mostraram que a dopamina pode desempenhar um papel importante na gravidade das crises na fase aguda da pilo, o que parece ser exercido por sua ação inibitória da dopamina sobre a expressão motora das crises.

Abstract The dopamine content in cerebral structures has been related to neuronal excitability and several approaches have been used to study this phenomenon during seizure vulnerability period. In the present work, we describe the effects of dopamine depletion after the administration of 6-hidroxidopamine (6-OHDA) into the substantia nigra pars compacta of male rats submitted to the pilocarpine model of epilepsy. Susceptibility to pilocarpine-induced status epilepticus (SE), as well as spontaneous and recurrent seizures (SRSs) frequency during the chronic period of the model were determined. Since the hippocampus is one of main structures in the development of this experimental model of epilepsy, the dopamine levels in this region were also determined after drug administration. In the first experiment, 62% (15/24) of 6-OHDA pre-treated rats and 45% (11/24) of those receiving ascorbic acid as control solution progressed to motor limbic seizures evolving to SE, after the administration of pilocarpine. Severeness of seizures during the model´s the acute period, was significantly higher in epileptic experimental rats (56.52%), than in controls (4.16%). In the second experiment, the frequency of seizures in the model’s chronic phase did not significantly change between groups. Our data show that dopamine may play an important role on seizure severity in the pilo’s model acute period, which seems to be due to dopamine inhibitory action on motor expression of seizure.

Abstract It is described for the first time the synthesis and characterization of NiMn2O4 employing the Mn ores from the Amazon Region as starting material, instead of using commercial reagents as normally occurs in its synthesis process. Initially, an Mn ore was transformed into a single product, the octahedral layer (OL) manganese oxide with Ni2+ ions interlayer (Ni-OL-2), which was used as a matrix to obtain NiMn2O4 by thermal decomposition. The following techniques were used for characterization: X-ray diffraction (XRD), X-ray fluorescence spectroscopy (XRF), infrared spectroscopy (FTIR), thermal analysis (TG-DTA), scanning (SEM) and transmission (TEM) electron microscopies. The results revealed that Mn oxide ore composed of nsutite mineral transformed into a well-ordered crystalline Ni-OL-2 phase, after the hydrothermal and cation exchange process. The thermal decomposition of Ni-OL-2 produced the nickel manganite phase at 850 °C with spherical ball morphology and crystal sizes in the range of 200-300 nm.

Resumo A herança da característica de fruto sem sementes de Annona squamosa ainda não foi esclarecida. Técnicas moleculares podem auxiliar em programas de melhoramento, principalmente na seleção assistida do gene de interesse. O gene INO pode estar relacionado ao desenvolvimento da semente dessas frutas. O objetivo foi investigar a herança da ausência de sementes em Annona squamosa e a conservação do gene INO nos genótipos Annona squamosa e Annona cherimola x Annona squamosa avaliando sua homologia com banco de dados de genes INO. A geração F1 foi obtida pelo cruzamento do mutante 'Brazilian seedless' (genitor masculino) (P1) com o tipo selvagem com sementes (A. squamosa) (M1 e M2, genitores femininos). O gene INO foi estudado em A. squamosa, mutante e selvagem (P1, M1, M2 e M3) e na cultivar Gefner atemoya (A. cherimola x A. squamosa) (M4). O DNA foi extraído de folhas jovens, e quatro conjuntos de primers específicos flanqueando o gene INO foram amplificados. A característica sem sementes foi identificada como estenospermática nos frutos do parental P1, sugerindo herança monogênica com dominância completa. A alta similaridade de sequência das amplificações do gene INO nos acessos de pinha (M1, M2, M3) e na cultivar de atemóia Gefner (M4) reforça a hipótese de sua conservação.

Abstract The inheritance of the seedless fruit characteristic of Annona squamosa has not yet been explained. Molecular techniques may aid breeding programs, mainly in the assisted selection of the target gene. The INO gene may be related to seed development in these fruits. The objective of the present paper was to investigate the inheritance of seedlessness in the 'Brazilian seedless' sugar apple and INO gene conservation in Annona squamosa and Annona cherimola x Annona squamosa genotypes by assessing their homology with the INO database genes. The F1 generation was obtained by crossing the mutant 'Brazilian seedless' (male genitor) (P1) with the wild-type A. squamosa with seeds (M1 and M2, female genitors). The INO gene was studied in mutant and wild-type A. squamosa (P1, M1, M2 and M3) and in the Gefner atemoya (A. cherimola x A. squamosa) (M4) cultivar. The DNA was extracted from young leaves, and four sets of specific primers flanking the INO gene were amplified. The seedless characteristic was identified as stenospermatic in the fruits of parental P1, suggesting monogenic inheritance with complete dominance. High sequence similarity of the INO gene amplifications in the sugar apple accessions (M1, M2, M3) and the atemoya cultivar Gefner (M4) reinforces the hypothesis of their conservation.

Abstract Human Respiratory Syncytial Virus (hRSV) infection results in death and hospitalization of thousands of people worldwide each year. Unfortunately, there are no vaccines or specific treatments for hRSV infections. Screening hundreds or even thousands of promising molecules is a challenge for science. We integrated biological, structural, and physicochemical properties to train and to apply the concept of artificial intelligence (AI) able to predict flavonoids with potential anti-hRSV activity. During the training and simulation steps, the AI produced results with hit rates of more than 83%. The better AIs were able to predict active or inactive flavonoids against hRSV. In the future, in vitro and/or in vivo evaluations of these flavonoids may accelerate trials for new anti-RSV drugs, reduce hospitalizations, deaths, and morbidity caused by this infection worldwide, and be used as input in these networks to determine which parameter is more important for their decision.

Resumo A infecção pelo Vírus Sincicial Respiratório Humano (hRSV) resulta na morte e hospitalização de milhares de pessoas em todo o mundo a cada ano. Infelizmente, não existem vacinas ou tratamentos específicos para tais infecções. A testagem de centenas, ou mesmo milhares, de moléculas promissoras é um desafio para a ciência. Neste trabalho, nós integramos propriedades biológicas, estruturais e físico-químicas para treinar e aplicar o conceito de inteligência artificial (IA) capaz de prever flavonoides com potencial atividade anti-hRSV. Durante as etapas de treinamento e simulação, a IA produziu resultados com taxas de acerto superiores a 83%, sendo capaz de prever flavonoides ativos ou inativos contra o hRSV. No futuro, avaliações in vitro e/ou in vivo desses flavonoides poderão acelerar os testes de novas drogas anti-RSV, reduzir hospitalizações, mortes e morbidade causadas por essa infecção. Além disso, a validação futura destes dados poderá determinar qual parâmetro tem maior peso na decisão da inteligência.

Resumo Para a indústria calçadista, Sobral, no Ceará, se destaca como o mais expressivo território produtivo, fato que se justifica pela localização de unidades produtivas da Grendene. Este artigo buscou analisar os aspectos gerais das interações territoriais da produção de calçados da Grendene-Sobral com base nos circuitos espaciais da produção. Para a realização desta demanda, os procedimentos metodológicos consistiram em: a) revisão de literatura arrimada em autores especialistas no tema; b) levantamento documental e estatístico e c) pesquisa de campo. Por intermédio da pesquisa, constatou-se, entre outros resultados, que Sobral recebe todas as ordens e determinações da produção, caracterizando-se como um território usado para a produção propriamente dita. Assim sendo, o produto é idealizado no Sul, produzido em Sobral e distribuído para o mercado consumidor nacional e global, o que explica a multiescalaridade dos insumos, a organização do processo produtivo e a multiescalaridade da distribuição e consumo da produção calçadista da empresa. Resumen Para la industria del calzado, Sobral, en Ceará, se destaca como el territorio productivo más expresivo, hecho que se justifica por la ubicación de las unidades productivas de Grendene. Este artículo buscó analizar los aspectos generales de las interacciones territoriales en la producción de calzado de Grendene-Sobral a partir de los circuitos espaciales de producción. Para llevar a cabo esta demanda, los procedimientos metodológicos consistieron en: a) revisión bibliográfica a partir de autores especializados en el tema; b) levantamiento documental y estadístico yc) investigación de campo. Abstract For the footwear industry, Sobral, in Ceará, stands out as the most expressive productive territory, a fact that is justified by the location of Grendene's production units. This article sought to analyze the general aspects of territorial interactions in the production of footwear by Grendene-Sobral based on the spatial circuits of production. Abstract Antibiotics have shown less efficiency against resistance of pathogenic microorganisms. As a result, research centers have sought therapeutic alternatives against multidrug resistance of bacteria to antibiotics, one of which is using plant extracts. Bamboo extracts are used for several medicinal purposes. This study aimed to evaluate the antibacterial potential of hydroalcoholic extracts of culms and leaves of the species Guadua aff. lynnclarkiae on strains of Staphylococcus aureus, Streptococcus pneumoniae, and Klebsiella pneumoniae. We evaluated the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC). Only the leaves of G. aff. lynnclarkiae showed a bactericidal effect for all tested strains with MBC ranging from 1.55 mg ml-1 to 25 mg ml-1. The culms had bacteriostatic action with MIC ranging from 1.55 mg ml-1 to 6.25 mg ml-1, and bactericidal action at the concentration of 6.25 mg ml-1 only for S. aureus. This study provides bases for the use of this Amazonian native bamboo in bioprospecting.

Resumo Os antibióticos têm mostrado menor eficiência contra a resistência de microrganismos patogênicos. Assim sendo, centros de pesquisa têm buscado alternativas terapêuticas contra a multirresistência das bactérias aos antibióticos, sendo uma delas o uso de extratos vegetais. Extratos de bambu têm sido usados para diversos fins medicinais. Este trabalho teve como objetivo avaliar o potencial antibacteriano de extratos hidroalcoólicos de colmos e folhas da espécie Guadua aff. lynnclarkiae em cepas de Staphylococcus aureus, Streptococcus pneumoniae e Klebsiella pneumoniae. Avaliamos a concentração inibitória mínima (CIM) e a concentração bactericida mínima (CBM). Apenas as folhas de G. aff. lynnclarkiae mostraram efeito bactericida para todas as cepas testadas com CBM variando de 1,55 mg ml-1 a 25 mg ml-1. Os colmos apresentaram ação bacteriostática com CIM variando de 1,55 mg ml-1 a 6,25 mg ml-1, e ação bactericida na concentração de 6,25 mg ml-1 apenas para S. aureus. Este estudo fornece bases para o uso deste bambu nativo da Amazônia na bioprospecção.

ABSTRACT Introduction Nasosinusal Adenocarcinomas are rare neoplasms that account for less than 5% of head and neck cancers. Occupational risk factors that have been linked to a higher incidence of Nasosinusal Adenocarcinomas include chronic exposure to wood dust and/or leather, nickel and chromium compounds, formaldehyde and other solvents, and polycyclic aromatic hydrocarbons. Occupations with exposure to dense wood dust are particularly associated with the intestinal subtype of adenocarcinoma, the most frequent location of which occurs in the ethmoid sinus and upper nasal cavity. The report of this clinical case aims to raise awareness among health professionals for the surveillance and early diagnosis of these occupational cancers. Description of the Clinical Case The authors present the case of an 81-year-old man who worked for 56 years as a joiner in the furniture industry. In his occupational history he denied the use of Personal Protective Equipment, namely respiratory protection. In a surveillance consultation with his family physician, he reported epistaxis and occasional nasal obstruction of the right nostril with an evolution of about three months. During the evaluation, a neoformative lesion was diagnosed in the right nasal fossa and the patient was immediately referred to Otorhinolaryngology consultation. A biopsy of the lesion revealed "morphological aspects of intestinal adenocarcinoma". After evaluation in group consultation, he was proposed for endoscopic sinus surgery and adjuvant radiotherapy. Discussion/Conclusion Occupational exposure to wood dust should lower the physician's threshold of suspicion for symptoms suggestive of occupational cancer. The long latency period for the development of adenocarcinoma and the higher survival of people treated in the early stages of the disease make periodic medical surveillance in the early detection of nasal adenocarcinoma necessary.

RESUMO Introdução Os Adenocarcinomas Nasossinusais são neoplasias raras que correspondem a menos de 5% dos cancros da cabeça e do pescoço. Os fatores de risco ocupacional que estão relacionados com uma maior incidência de Adenocarcinomas Nasossinusais incluem a exposição crónica a pó de madeira e/ou de couro, níquel e compostos de crómio, formaldeído e outros solventes, além de hidrocarbonetos aromáticos policíclicos. As profissões com exposição a pó de madeiras densas estão particularmente associadas ao subtipo de adenocarcinoma do tipo intestinal, cuja localização mais frequente ocorre no seio etmoidal e na parte superior da cavidade nasal. O relato deste caso clínico tem como objetivo sensibilizar os profissionais de saúde para a vigilância e diagnóstico precoce destes cancros ocupacionais. Descrição do Caso Clínico Os autores apresentam o caso de um homem de 81 anos, que trabalhou durante 56 anos como marceneiro na indústria do mobiliário. No seu histórico ocupacional nega utilização de Equipamento de Proteção Individual, nomeadamente a nível respiratório. Numa consulta de vigilância com o seu médico de família referiu epistaxes e obstrução nasal ocasional da narina direita com cerca de três meses de evolução. Na avaliação foi diagnosticada uma lesão neoformativa da fossa nasal direita e foi encaminhado com urgência para consulta de Otorrinolaringologia. Realizou biópsia da lesão que revelou “aspetos morfológicos de adenocarcinoma do tipo intestinal”. Depois de avaliado em consulta de grupo, foi proposto para cirurgia endoscópica nasossinusal e radioterapia adjuvante. Discussão/Conclusão A exposição ocupacional ao pó de madeira deve sensibilizar o médico a suspeitar de sintomas sugestivos de cancro ocupacional. O longo período de latência para o desenvolvimento do adenocarcinoma e a maior sobrevivência das pessoas tratadas nas fases iniciais da doença tornam necessária uma vigilância médica periódica para deteção precoce do adenocarcinoma nasal.

The present work aims to evaluate the manufacturing processes of free-standing graphene oxide (GO)/reduced graphene oxide(rGO)/carbon nanotube (MWCNT) papers-like and the effect on electrical conductivities after annealing treatment. The pure GO and the hybrid papers were obtained by vacuum filtration and submitted to mild thermal annealing at 400 °C under an argon atmosphere. Analyses comparing the carbon paper-like materials before and after thermal treatment were done by thermogravimetric analysis (TGA), Raman spectroscopy, scanning electron microscopy (SEM), and electrical conductivity measurements. The characterization results show the annealing process effect on the morphology of paper materials. The GO papers and the hybrid (GO/rGO/MWCNT) exhibited good free-standing structure properties. The SEM results suggest the formation of gas void due to the thermal treatment. TGA and Raman results showed the thermal reduction and restoration of the carbon sp2 network of the graphene oxide induced by thermal treatment. The thermal treatment was responsible for changing the almost insulator characteristic of free-standing pure GO paper, achieving an electrical conductivity of 752 S/m, and improving 45% of the free-standing hybrid carbon nanomaterials papers (GO/rGO/MWCNT) electrical conductivity, achieving the value of 1020 S/m.

BACKGROUND: South Africa (SA) has one of the highest gun-related mortality rates in the world - 20 people per day. The available data, however, do not reflect the substantial number of patients suffering non-lethal firearm injuries. Gunshot-related injury has been recognised as a highly costly healthcare problem by individual treating centres in SA and other countries; however, no 'national picture' has been examined in detailOBJECTIVES: To explore the burden of gunshot-related orthopaedic injuries across SAMETHODS: A multicentre research network was established in SA, and 37 orthopaedic units across 9 provinces participated. A prospective, observational cohort study was performed during a 2-week period in 2019. Patients were screened, enrolled and reported by local orthopaedic teams. Patients were included if they had at least one acute gunshot-related orthopaedic fracture referred to the orthopaedic service. Patients were asked additional questions around baseline health-related quality-of-life (HRQOL) and personal circumstances. Follow-up was at 8 weeks after injuryRESULTS: Thirty-seven centres enrolled 135 patients over the 2-week study period. Western Cape Province had the highest number of reported cases (n=52; 39%), followed by Gauteng (n=35; 26%) and KwaZulu-Natal (n=29; 21%). The median age of patients was 30.5 years and the majority were male (89%). Forty-three percent of patients had been either shot or stabbed prior to this injury. Fifty-two percent of all patients required fracture fixation surgery and 11% required wound debridement without fracture fixation. HRQOL data were collected successfully at baseline, but follow-up data were available for <25% of casesCONCLUSIONS: Gunshot-related orthopaedic injuries represent a significant burden of disease in the SA healthcare environment. This study highlights several areas for further research in the management of the injuries and associated outcomes

RESUMO Marcadores sorológicos são rotineiramente utilizados na prática clínica para o estadiamento de linfomas e para a determinação de seu prognóstico em humanos. No entanto, pouco se sabe sobre sua utilização em cães, mesmo os linfomas sendo neoplasias com alta prevalência nessa espécie. No presente estudo, as concentrações séricas do receptor solúvel de interleucina-2 (sIL-2R) e do antígeno do câncer 125 (CA 125) foram mensurados em 10 cães saudáveis e em 15 cães com linfoma cutâneo, utilizando-se o kit ELISA canino e a leitura em um Stat Fax modelo 2100 (sIL-2R), bem como o kit ELISA humano e a leitura pelo ELISYS UNO humano (CA 125). Os resultados mostraram que não houve diferença significativa (P<0,05) nas concentrações dos marcadores entre os grupos. Além disso, os resultados não apontaram significância clínica no estadiamento tumoral e estabelecimento do prognóstico em cães diagnosticados com linfoma cutâneo.
